Systemic mastocytosis associated with t(8;21)(q22;q22) acute myeloid leukemia.

Although KIT mutations are present in 20-25% of cases of t(8;21)(q22;q22) acute myeloid leukemia (AML), concurrent development of systemic mastocytosis (SM) is exceedingly rare. We examined the clinicopathologic features of SM associated with t(8;21)(q22;q22) AML in ten patients (six from our institutions and four from published literature) with t(8;21) AML and SM. In the majority of these cases, a definitive diagnosis of SM was made after chemotherapy, when the mast cell infiltrates were prominent. Deletion 9q was an additional cytogenetic abnormality in four cases. Four of the ten patients f

Targeting the NF-kappaB signaling pathway in Notch1-induced T-cell leukemia.

T-cell acute lymphoblastic leukemia (T-ALL), unlike other ALL types, is only infrequently associated with chromosomal aberrations, but it was recently shown that most individuals with T-ALL carry activating mutations in the NOTCH1 gene. However, the signaling pathways and target genes responsible for Notch1-induced neoplastic transformation remain undefined. We report here that constitutively active Notch1 activates the NF-kappaB pathway transcriptionally and via the IkappaB kinase (IKK) complex, thereby causing increased expression of several well characterized target genes of NF-kappaB in bo

[Autologous bone marrow transplantation as a treatment for autoimmune disease: mechanisms and results].

Autoimmune diseases are characterized by immune response against self antigens. One of the current research interests in this field is oriented toward development of tolerance. One of the newest options in the search for tolerance is autologous bone marrow transplantation: a variant of bone marrow transplant in which the patient's own hematopoietic stem cells are reinfused after myeloablative therapy. The idea of using bone marrow transplant in treatment of autoimmune diseases derived from observing remission in autoimmune diseases in patients transplanted due to coexisting neoplastic disease.

Hodgkin's lymphoma in systemic onset juvenile rheumatoid arthritis after treatment with low dose methotrexate.

We describe the occurrence of malignant lymphoma as a possible complication of immunosuppression associated with low dose methotrexate (MTX) therapy for juvenile rheumatoid arthritis (JRA). A 6-year-old girl with systemic onset JRA who had received low dose MTX therapy for 16 months developed diffuse peripheral lymphadenopathy and enlargement of the lymph nodes in the mediastinum, hilum of the lungs, and liver. Lymph node histology disclosed mixed cellularity Hodgkin's lymphoma; the neoplastic cells were positive for CD30 and CD15, but negative for Epstein-Barr virus RNA or EBV latent membrane

A case of malignant mastocytosis with circulating mast cell precursors: biologic and phenotypic characterization of the malignant clone.

The phenotypic and biologic properties of malignant cells in a case of aggressive mastocytosis with multi-organ involvement, circulating mast cell precursors and absence of skin infiltrates were analyzed. Circulating mast cell precursors were detected by immunostaining using antibodies against mast cell tryptase as well as by electron microscopy. These progenitors were tryptase+/chymase- (MCT) and accounted for 10 to 20% of nucleated mononuclear blood cells (MNC). A subset of them contained metachromatic granules. As assessed by combined toluidine blue/immunofluorescence staining, the granulat

Roles of TIF1β in Leukemic Stem Cell Through SETDB1-Dependent and Independent Mechanisms.

TIF1β/TRIM28/KAP1 has been recognized as a scaffold protein that partners with KRAB-ZFPs and heterochromatin complexes to enforce gene silencing. In embryonic and pluripotent stem cells, it maintains self-renewal by silencing endogenous retroelements through the establishment of heterochromatin. While these canonical functions have been extensively examined in embryonic stem (ES) cells, accumulating evidence also highlights its diverse contributions to cancer biology. We herein focused on the oncogenic role of TIF1β in leukemic progression, contrasting this with its physiological roles in hema

Infection, stem cells and cancer signals.

The association of cancer with preceding parasitic infections has been observed for over 200 years. Some such cancers arise from infection of tissue stem cells by viruses with insertion of viral oncogenes into the host DNA (mouse polyoma virus, mouse mammary tumor virus). In other cases the virus does not insert its DNA into the host cells, but rather commandeers the metabolism of the infected cells, so that the cells continue to proliferate and do not differentiate (human papilloma virus and cervical cancer). Cytoplasmic Epstein Barr virus infection is associated with a specific gene transloc

Induction of eosinophilic colonies by interleukin-5 on acute myeloblastic leukaemic cells.

The effect of interleukin-5 (IL-5) on blast colony formation of acute myeloblastic leukaemia (AML) cells was studied. IL-5 showed a weak but significant colony stimulatory effect in one out of six cases studied. IL-5 induced eosinophilic colony growth in this case, and chromosomal and histochemical analyses strongly suggested that these eosinophilic colonies were derived from leukaemic blast progenitor cells. To our knowledge, this is the first report that IL-5 induces eosinophilic colonies on fresh AML cells. A possible induction of differentiation of AML blast cells by IL-5 is suggested.

Cellular distribution of a human I-A-like (DS/DC) antigen on normal and neoplastic lymphoid cells.

Previous biochemical studies have shown that B cell specific, monoclonal antibody, McAb 33.1, reacts with a class II antigen that represents a human analogue of murine I-A (DS/DC) antigens (J. Exp. Med. 158:1924, 1983). McAb 33.1 recognizes a polymorphic human B lymphocyte specific antigen present on mu+, B1+ peripheral B cells, B lymphoid cell lines, activated B cells, and neoplastic B lymphoid cells. Of 100 HLA-D/DR typed EBV-transformed lymphoblastoid cell lines tested, only those from DR3,3 and DR7,7 individuals failed to react with McAb 33.1. The 33.1 antigen is present at lower concentra

piR-823 and piR-20326 Differentially Regulate Stemness Programs in Ovarian Cancer Stem-like Cells.

Ovarian cancer stem-like cells (OCSCs) are major drivers of tumor recurrence and chemoresistance. Although PIWI-interacting RNAs (piRNAs) have emerged as important regulators of cancer stem cell (CSC) biology, their roles in OCSCs remain poorly understood. We performed a systematic analysis of public datasets to identify ovarian cancer-associated piRNAs and selected piR-823 and piR-20326 for functional validation. Their expression was evaluated by quantitative reverse transcription polymerase chain reaction (qRT-PCR)following piRNA mimic and inhibitor transfections in three-dimensional (3D) ov

Expression profiling of inhibitory immune checkpoints in colorectal cancer stem cells and their association with tumor immunity and immunotherapy biomarkers.

BACKGROUND: Colorectal cancer stem cells (CSCs) represent a rare subpopulation within tumors endowed with self-renewal capabilities and are recognized as key drivers of tumor initiation, metastasis, and therapeutic resistance. These cells also display immunomodulatory properties that enable them to evade immune surveillance. However, the mechanisms underlying their immune evasion, including the role of immune checkpoints (ICPs), remain poorly understood. Therefore, this study aimed to characterize the inhibitory ICP expression landscape in colorectal CSC-enriched models and to evaluate its ass

SPINK1-COL18A1 crosstalk shapes epigenome and drives cancer stemness in pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is a devastating cancer with increasing incidence and a dismal prognosis. Here, we uncover serine protease inhibitor Kazal type 1 (SPINK1) as a putative determinant of PDAC progression with a previously unrecognized role in epigenomic regulation. We show that SPINK1 expression, which is highly dynamic across PDAC progression, is associated with key aggressive cancer phenotypic states and regulates cancer cell stemness and plasticity in both in vitro and patient samples. Mechanistically, our results suggest a new signaling axis where SPINK1 interacts with

Spheroid cultures reveal cancer stem cell enrichment and distinct chemotherapeutic response in triple-negative breast cancer.

BACKGROUND: Three-dimensional (3D) cancer spheroids mimic key morphological and biological features of solid tumors, providing a valuable model for evaluating drug responses. Here, we investigated whether intrinsic cancer stem cell composition and tumor subtype determine spheroid formation capacity, transcriptional profiling, and therapeutic response in breast cancer cell lines. METHODS: Using the liquid overlay technique, we optimized conditions for reproducible spheroid generation using 10 human cancer cell lines derived from breast cancer, glioblastoma, and malignant melanoma. Barrett's oesophageal adenocarcinoma encompasses tumour-initiating cells that do not express common cancer stem cell markers.

Accumulating evidence has suggested that tumours have a hierarchical organization in which only the cancer stem cells (CSCs) have tumour-initiating properties. Several surface antigens have been employed to isolate CSCs from various malignancies, although not from oesophageal adenocarcinoma (EA). We tested whether Barrett's oesophagus (BE) and EA might serve as a model for the CSC concept. In vivo assays were performed by transplantation of serially diluted bulk EA cells into NOD-SCID mice to establish the presence and frequency of tumour-initiating cells. These were found to be present as ca.

ZBTB20 promotes malignancy and stemness in lung squamous cell carcinoma through the transcriptional repression of KCTD1 and activation of Hedgehog signaling.

Prostate Potassium channel tetramerization domain containing 1 (KCTD1) is a tumor suppressor that inhibits the Hedgehog (Hh) signaling pathway. The aim of this study was to investigate the role of KCTD1 in lung squamous cell carcinoma (LUSC) and its impact on the Hedgehog (Hh) pathway. Jervine, a known Hh signaling inhibitor, was used to validate the function KCDT1. After ascertaining the expression pattern of KCTD1 in LUSC tissues, gain- and loss-of-function experiments were conducted to analyze the impact of KCTD1 on the malignant behavior and stemness of LUSC cells malignancy, and the activ

Exosomal lncRNA Mir100hg derived from cancer stem cells enhance glycolysis and promote metastasis of melanoma through miR-16-5p and miR-23a-3p.

Increasing evidence demonstrate that the significant role of long non-coding RNA (lncRNA) in metastasis and the remodeling of the tumor microenvironment. However, the precise mechanisms of lncRNAs in cancer metastasis are still poorly understood. The function of lncRNA-Mir100hg in melanoma and its involvement in mediating communication between tumor stem cells and non-stemness tumor cells remains unknown. We found that Mir100hg is upregulated in melanoma stem cells (CSCs) known as OLSD. Furthermore, Mir100hg can be transferred from OLSD to non-stem cancer cells (OL) through exosomes. Once Mir1

Dissecting inherent intratumor heterogeneity in patient-derived glioblastoma culture models.

BACKGROUND: Molecular profile of glioblastoma multiforme (GBM) revealed 4 subtypes, 2 of which, proneural and mesenchymal, have been predominantly observed, with the latter displaying a more aggressive phenotype and increased therapeutic resistance. Single-cell RNA sequencing revealed that multiple subtypes actually reside within the same tumor, suggesting cellular heterogeneity in GBM. Further, plasticity between these 2 subtypes is observed during tumor recurrence and in response to radiation therapy. METHODS: Patient-derived GBM stemlike cells were cultured as neurospheres. These cells wer

GSC State Plasticity Drives Therapeutic Resistance and Recurrence in Glioblastoma.

Glioblastoma (GBM) is the most aggressive primary brain tumor in adults, and treatment failure is closely linked to the presence of GBM stem cells (GSCs). Rather than representing a fixed marker-defined state, GSC identity is better understood as a dynamic and reversible stem-like state. GBM cells exhibit transcriptional plasticity, most clearly reflected by the proneural-to-mesenchymal transition, although direct interconversion among neural progenitor cell-like, oligodendrocyte precursor cell-like, astrocyte-like, and mesenchymal-like states remains unproven. GBM cells can also transition bi
