Monoclonal antibody to a proximal nephrogenic renal antigen: immunohistochemical analysis of formalin-fixed, paraffin-embedded human renal cell carcinomas.

An IgG1 murine monoclonal antibody (PN-15) to an epitope preserved in formalin-fixed, paraffin-embedded tissue sections of renal cell carcinomas was developed following immunization of mice with the microsomal fraction of human renal cortical tissue homogenates. An immunohistochemical screening procedure was utilized against fixed, embedded tissue sections of renal cell carcinomas. This antibody identified a Mr 200,000 determinant on immuno-Western blot analysis, and the recognized epitope was shown to reside in the carbohydrate domain of a Mr 200,000 glycoprotein (gp200). Analysis of its tiss…

Demonstration of human papillomavirus types in paraffin processed tissue from human ano-genital lesions by in-situ DNA hybridisation.

A sensitive in situ hybridization technique for the demonstration of human papillomavirus (HPV) employing a biotin-streptavidin polyalkaline phosphatase complex has been successfully applied to formalin-fixed, paraffin processed tissue obtained from a selected series of patients with ano-genital lesions. Benign condylomata from males and females showed the presence of HPV 6 and 11. Two cases of vulval intraepithelial neoplasia showed HPV 16. Four cases of squamous carcinoma of the anal canal also showed HPV 16 in the tumour or in the adjacent pre-invasive neoplastic epithelium. A case of malig…

[Mineral oil pneumonia caused by throat gargles containing lipoid paraffin. Diagnosed by biochemical examination of the sputum (author's transl)].

A 20-year-old man developed a pneumonia after using throat gargles containing lipoid paraffin over 5 years. The lung biopsy showed a lipoid pneumonia and the bodyplethysmographic examination a ventilation disorder indicating a starting lung fibrosis. By the means of gaschromatography the identity of the lipoid in the sputum and in the gargle sample was established and the diagnosis continued. The disease, its pathophysiology and iatrogenic etiology are discussed and the consequencies concerning the ENT-specialist are stressed.

Integrated assessment of dermal and inhalation exposure to chlorinated paraffins using personal silicone wristbands and measurement of skin contamination.

Chlorinated paraffins (CPs), specifically short-chain (SCCPs) and medium-chain (MCCPs), are widely used industrial chemicals of concern due to persistence, bioaccumulation, and toxicity. This study assessed integrated personal exposure (dermal and inhalation) to SCCPs and MCCPs using silicone wristbands (SWBs) and hand wipes (HWs) in 20 volunteers over five days. Participants wore two SWBs simultaneously: a non-removable wristband (N) worn continuously and a removable wristband (R) taken off during sleep and hygiene. SCCPs and MCCPs were detected in all SWBs. Median concentrations in SWBs(N) w…

Gestational exposure to short-chain chlorinated paraffins induces sex-specific hepatic steatosis in adult mouse offspring.

Short-chain chlorinated paraffins (SCCPs), ubiquitous persistent organic pollutants, have been increasingly implicated in metabolic disorders; however, their developmental effects on hepatic lipid metabolism in offspring remain poorly understood. This study aimed to investigate whether gestational SCCPs exposure induces hepatic lipid accumulation in adult offspring and elucidate the underlying mechanisms. Pregnant C57BL/6J mice were orally administered SCCPs (0, 1, 10, or 100 mg/kg) from gestational day 0 (GD0) to GD18. At 12 weeks of age, male offspring exposed to the medium dose of SCCPs dev…

Application of an in situ hybridization method for fungal diagnosis in formalin-fixed, paraffin-embedded allergic fungal rhinosinusitis tissue.

Allergic fungal rhinosinusitis (AFRS) is a chronic inflammatory disease of the sinuses that can involve serious late complications; thus, prompt diagnosis is essential to determine the appropriate treatment. The most important diagnostic element of AFRS is the detection of noninvasive fungi within eosinophilic mucin. However, the rarity of fungal hyphae in ARFS makes it difficult to specifically identify them using histochemical staining alone. In this study, we designed a new in situ fungal detection probe for the diagnosis of fungi in formalin-fixed, paraffin-embedded AFRS tissues. Tissue se…

Modification of the Hi-C Technology for Molecular Genetic Analysis of Formalin-Fixed Paraffin-Embedded Sections of Tumor Tissues.

Molecular genetic analysis of tumor tissues is the most important step towards understanding the mechanisms of cancer development; it is also necessary for the choice of targeted therapy. The Hi-C (high-throughput chromatin conformation capture) technology can be used to detect various types of genomic variants, including balanced chromosomal rearrangements, such as inversions and translocations. We propose a modification of the Hi-C method for the analysis of chromatin contacts in formalin-fixed paraffin-embedded (FFPE) sections of tumor tissues. The developed protocol allows to generate high…

Combined RNAscope and immunohistochemistry staining on duodenal paraffin sections as a new tool to reveal cytolytic potential of intraepithelial lymphocytes.

Immunohistochemistry (IHC) is a consolidated technique for the identification of surface and cytoplasmic antigens in cells or tissue sections using specific antibodies, yet simultaneous detection of two markers on the same cell may be difficult to achieve. Here we develop a protocol to perform a double staining using RNAscope, a new in-situ hybridization (ISH) technology, to visualize perforin transcripts, and classical IHC to visualize either CD8 or TcRγδ positive intraepithelial lymphocytes (IELs) in small intestinal paraffin sections of celiac disease (CD) patients. This double assay will a…

Multiplexed Barcoding Image Analysis for Immunoprofiling and Spatial Mapping Characterization in the Single-Cell Analysis of Paraffin Tissue Samples.

Multiplexed imaging technology using antibody barcoding with oligonucleotides, which sequentially detects multiple epitopes in the same tissue section, is an effective methodology for tumor evaluation that improves the understanding of the tumor microenvironment. The visualization of protein expression in formalin-fixed, paraffin-embedded tissues is achieved when a specific fluorophore is annealed to an antibody-bound barcode via complementary oligonucleotides and then sample imaging is performed; indeed, this method allows for the use of customizable panels of more than 40 antibodies in a sin…

Development of a sample preparation method for micro-proteomics analysis of the formaldehyde-fixed paraffin-embedded liver tissue samples.

Liver micro-proteomics based on the routinely used formaldehyde-fixed paraffin-embedded (FFPE) samples is valuable for innovative research, but the technical approach for sample preparation is often challenging. In this study, we aimed to develop a method for sample preparation for micro-proteomics on using the FFPE liver samples. We collected 2000 individual cells per batch from FFPE liver slices with laser capture microdissection and used them as test samples. We used the microscale fresh-frozen liver samples or HepG2 cells as control samples. For the FFPE samples, we first established a pro…

Formalin-Fixed Paraffin-Embedded (FFPE) samples are not a beneficial replacement for frozen tissues in fetal membrane microbiota research.

Formalin-Fixed Paraffin-Embedded (FFPE) tissues are routinely collected, archived, and used for clinical diagnosis, including maternal and neonatal health. Applying FFPE samples to microbiota research would be beneficial to reduce preparation, storage and costs associated with limited available frozen samples. This research aims to understand if FFPE fetal membrane samples are comparable to frozen tissues, which are the current gold standard for DNA microbiota analysis. Extracted DNA from nine matched paired patients were sequenced by Illumina sequencing of the V4 16S rRNA gene region. This in…

A sheet pocket to prevent cross-contamination of formalin-fixed paraffin-embedded block for application in next generation sequencing.

Formalin-fixed paraffin-embedded (FFPE) blocks are used as biomaterials for next-generation sequencing of cancer panels. Cross-contamination is detected in approximately 5% of the DNA extracted from FFPE samples, which reduces the detection rate of genetic abnormalities. There are no effective methods available for processing FFPE blocks that prevent cells from mixing with other specimens. The present study evaluated 897 sheets that could potentially prevent cell transmission but allow for the movement of various solvents used in FFPE blocks. According to the International Organization for Sta…

Histological paraffin-embedded block: a good alternative specimen to detect the use of opiates at least 20 years ago.

PURPOSE: Since the solely certain remnants of a performed autopsy are formalin-fixed paraffin-embedded (FFPE) samples, stored in the archives of every institute of legal medicine, we managed to extract molecules of toxicological interest from these specimens. METHODS: We assessed the analysis of ten fresh liver samples collected from heroin-related deaths and then histologically processed the same samples. The embedded blocks were then extracted by means of a new extracting method and the eluates were measured. We also selected five toxicological cases of heroin-related fatalities that were e…

A Simplified Method for the Histochemical Detection of Iron in Paraffin Sections: Intracellular Iron Deposits in Central Nervous System Tissue.

Although all cells contain iron, most histochemical methods fail to reveal the presence of iron within many cells of the central nervous system (CNS), particularly neurons. Previously, a sensitive method was developed that limited the extraction of iron in paraffin sections, and this method revealed staining within neurons. However, the staining was often too robust making it difficult to discern discrete intracellular structures. In 1970, a study incorporated acetone in an iron histochemical procedure to facilitate the demarcation of staining features. In the present study, both acetone and l…

Performance of the MeltPro MTB Assays in the Diagnosis of Drug-Resistant Tuberculosis Using Formalin-Fixed, Paraffin-Embedded Tissues.

OBJECTIVES: The MeltPro MTB assays for detection of resistance to antituberculosis (TB) drugs perform well in genotypic drug susceptibility testing (DST) of clinical samples, but their effectiveness with formalin-fixed, paraffin-embedded (FFPE) tissues is unknown. METHODS: FFPE tissues were obtained from 334 patients with TB. Susceptibility to rifampicin (RIF), isoniazid (INH), and fluoroquinolones was examined using the MeltPro MTB assays, with Xpert MTB/RIF (Xpert) and/or phenotypic DST (pDST) results as references. Samples with discordant results were analyzed by multiplex polymerase chain…

Nonmalignant Formalin-Fixed Paraffin-Embedded Tissues as a Source to Study Germline Variants and Cancer Predisposition: A Systematic Review.

Background: Archived formalin-fixed paraffin-embedded (FFPE) specimens from nonmalignant tissues derived from cancer patients are a vast and potentially valuable resource for high-quality genotyping analyses and could have a role in establishing inherited cancer risk. Methods: We systematically searched PubMed, Ovid MEDLINE, and Scopus databases for all articles that compared genotyping performance of DNA from nonmalignant FFPE tissue with blood DNA derived from cancer patients irrespective of tumor type. Two independent researchers screened the retrieved studies, removed duplicates, excluded …
