[Outbreaks Due to Parasites: Examples from the World and Türkiye].

Outbreaks due to parasites can occur in various parts of the world and in different periods. These outbreaks can be caused by water and food, as well as by human-to-human or vector-borne transmission. Cryptosporidium spp. and Giardia intestinalis were among the pathogens that affected most people in water-borne outbreaks occurred in the world between 2010-2014. The chlorine resistance of both Cryptosporidium spp. and Giardia spp. leads to the widespread detection of these parasites in waterborne outbreaks. These two protozoans cause self-limiting watery diarrhea in immunocompetent individuals,…

Critical role for isoprenoids in apicoplast biogenesis by malaria parasites.

Isopentenyl pyrophosphate (IPP) is an essential metabolic output of the apicoplast organelle in Plasmodium falciparum malaria parasites and is required for prenylation-dependent vesicular trafficking and other cellular processes. We have elucidated a critical and previously uncharacterized role for IPP in apicoplast biogenesis. Inhibiting IPP synthesis blocks apicoplast elongation and inheritance by daughter merozoites, and apicoplast biogenesis is rescued by exogenous IPP and polyprenols. Knockout of the only known isoprenoid-dependent apicoplast pathway, tRNA prenylation by MiaA, has no effe…

Malaria parasites release vesicle subpopulations with signatures of different destinations.

Malaria is the most serious mosquito-borne parasitic disease, caused mainly by the intracellular parasite Plasmodium falciparum. The parasit … en the two subpopulations. These results shed light on the sophisticated mechanism by which malaria parasites utilize EV subpopulations as a communication tool to target different cellular destinations or host systems.

Assessing PfGARP-Mediated Apoptosis of Blood-Stage Plasmodium falciparum Parasites.

Apoptosis is conventionally regarded as an evolutionarily conserved and genetically controlled process of programmed cell death confined to metazoan organisms. However, recently, conserved features of apoptosis have also been demonstrated in unicellular eukaryotes (Holzmuller et al. Parasitology 132:S19-S32, 2006; Le Chat et al. Mol Biochem Parasitol 153:41-47, 2007; Madeo et al. Curr Opin Microbiol 7:655-660, 2004; Welburn et al. Parasitology 132:S7-S18, 2006; Jensen et al. Science 216:1230-1233, 1982) including malaria parasites (Al-Olayan et al. Int J Parasitol 32:1133-1143, 2002; Ch'ng et …

New insights into apicoplast metabolism in blood-stage malaria parasites.

The apicoplast of Plasmodium falciparum is the only source of essential isoprenoid precursors and Coenzyme A (CoA) in the parasite. Isoprenoid precursor synthesis relies on the iron-sulfur cluster (FeS) cofactors produced within the apicoplast, rendering FeS synthesis an essential function of this organelle. Recent reports provide important insights into the roles of FeS cofactors and the use of isoprenoid precursors and CoA both inside and outside the apicoplast. Here, we review the recent insights into the roles of these metabolites in blood-stage malaria parasites and discuss new questions …

Gastrointestinal parasites in immunocompromised patients; A comparative cross-sectional study.

Gastrointestinal parasites (GIPs), including helminths and protozoa species, are a major health problem in many parts of the world. About 3.5 billion people are affected by the parasites worldwide. GIPs are one of the leading causes of death among immunocompromised individuals and can cause serious clinical complications, especially in people with Human Immunodeficiency Virus (HIV)/AIDS, hemodialysis patients, and transplant recipients. This study aimed to compare the prevalence of GIPs among immunocompromised patients and immunocompetent individuals in Lorestan province, West Iran. In the cur…

Malaria Parasites Hijack Host Receptors From Exosomes to Capture Lipoproteins.

Malaria parasites cannot multiply in host erythrocytes without cholesterol because they lack complete sterol biosynthesis systems. This suggests parasitized red blood cells (pRBCs) need to capture host sterols, but its mechanism remains unknown. Here we identified a novel high-density lipoprotein (HDL)-delivery pathway operating in blood-stage Plasmodium. In parasitized mouse plasma, exosomes positive for scavenger receptor CD36 and platelet-specific CD41 increased. These CDs were detected in pRBCs and internal parasites. A low molecular antagonist for scavenger receptors, BLT-1, blocked HDL u…

Dormant Plasmodium falciparum Parasites in Human Infections Following Artesunate Therapy.

BACKGROUND: Artemisinin monotherapy of Plasmodium falciparum infection is frequently ineffective due to recrudescence. Artemisinin-induced d … eding in vitro cultures, and genetic mutations in the K13 gene were investigated. RESULTS: Dormant parasites were observed in post-AS blood samples of 3D7- and K13-infected participants. The molecular signature of dormancy, an up-regulation of acetyl CoA carboxylase, was detected in 3D7 and K13 samples post-AS, but not in pre-AS samples. Posttreatment samp …

A case of a child with four parasites in Bulgaria.

Children are more susceptible to intestinal parasitic diseases than adults due to their lack of hygiene habits and close social interactions with their peers. We present a case of a 5-year-old boy with diarrhea and abdominal pain. The stool sample and perianal swab were examined in LINA laboratory, Burgas, Bulgaria. We detected four parasites Giardia (Lamblia) intestinalis, Blastocystis hominis, Hymenolepis nana and Entamoeba coli. Microbiological stool examination shows negative results for Candida spp., Salmonella spp. and Shigella spp. Patient's peripheral blood tests were normal. The calpr…

Pathogenic role of mitogen activated protein kinases in protozoan parasites.

Protozoan parasites with complex life cycles have high mortality rates affecting billions of human lives. Available anti-parasitic drugs are inadequate due to variable efficacy, toxicity, poor patient compliance and drug-resistance. Hence, there is an urgent need for the development of safer and better chemotherapeutics. Mitogen Activated Protein Kinases (MAPKs) have drawn much attention as potential drug targets. This review summarizes unique structural and functional features of MAP kinases and their possible role in pathogenesis of obligate intracellular protozoan parasites namely, Leishman…

Occurrence of intestinal parasites in Mayan children from Yucatán, Mexico.

Parasitic intestinal infections occur worldwide and affect the poorest and deprived populations. In Mexico, indigenous peoples live with the highest levels of poverty and marginalization. This study aimed to analyze the occurrence of intestinal parasites among Mayan children from the villages of Xkalakdzonot and Paraíso in Yucatán State. Stool samples from 83 children were examined using two coproparasitological tests (formalin-ethyl acetate concentration and FLOTAC Pellet techniques). The overall prevalence of intestinal parasites in children was 65.1%. Six protozoa and four helminths were id…

Quantification of Leishmania Parasites in Murine Models of Visceral Infection.

Visceral leishmaniasis (VL) is mainly caused by Leishmania donovani (India and East Africa), and Leishmania infantum (Mediterranean Basin and South America) infections. Although murine models of visceral infection lack the clinicopathological aspects of VL in humans, they have been proven useful at advancing our knowledge in the Leishmania field. Indeed, these models have been used not only to better understand the pathophysiology of the infection but also in drug and vaccine development. This chapter focuses on the protocols used to experimentally infect mice and to quantify parasite burdens
