Constitutive phenolics of Harpephyllum caffrum (Anacardiaceae) and their biological effects on human keratinocytes.

Assessment of the UV protecting potential of an aqueous methanol leaf extract of Harpephyllum caffrum proved that it possesses a distinct radical scavenging effect and inhibits the production of the proinflammatory cytokine IL-6 by human keratinocytes (HaCaT cells) following UV radiation. Phytochemical investigation of this extract led to isolation and structural determination of the hitherto unknown phenolics, kaempferol 3-O-(2″-sulphatogalactopyranoside), its quercetin analogue and 3-methoxyellagic acid 4-O-galactopyranoside in addition to 18 known phenolic compounds. The structures were det…

Modeling of protein and phenolic compound removal from aqueous solutions by electrocoagulation.

Electrocoagulation is a technique basically applied in water and wastewater treatment, but which has a number of potential applications in polymer, protein, drug, and vaccine delivery. In this work, we correlate the current applied between the electrodes to the removal of phenolic compounds or protein from aqueous solutions, but the principle can also be applied to other biological compounds such as plant pigments and sugars. Simple and time-dependent models were developed based on the complex formation between these biological substances and the aluminium hydroxide gel phase. The models devel…

Inhibition of platelet aggregation by olive oil phenols via cAMP-phosphodiesterase.

The aim of the present study was to confirm that olive oil phenols reduce human platelet aggregability and to verify the hypothesis that cAMP- and cGMP- phosphodiesterases (PDE) could be one of the targets of the biological effect. Four extracts from oils characterized by a high phenol content (HPE), and low phenol levels (LPE) were prepared and analyzed qualitatively and quantitatively by HPLC-UV and electrospray ionization-MS/MS. Human washed platelets stimulated with thrombin were used for the aggregation assay. Human platelet cAMP-PDE and recombinant PDE5A1 were used as enzyme source. Plat…

Potentials of phenolic molecules of natural origin and their derivatives as anti-HIV agents.

Identification of phenolic compounds and their derivatives interfering the several steps of the viral life cycle of the human immunodeficiency virus type 1 (HIV-1) is focused for the development of novel molecules for the treatment of AIDS. Several phenolic compounds isolated and characterized from natural sources have been studied in detail and found to exhibit inhibitory effects against different steps of the HIV-1 life cycle, including virus-cell fusion and virus absorption, reverse transcription, integration (IN) and proteolytic cleavage. In the review, we are summarizing some strong evide…

Inhibitory effect of dietary phenolic compounds on Chlamydia pneumoniae in cell cultures.

Chlamydial infections are very common worldwide. All chlamydial species have a tendency to cause persistent infections, which have been asso … t cells at studied concentrations. Our study revealed direct antichlamydial effect for selected polyphenolic compounds against C. pneumoniae, in vitro. We also demonstrated the ability of some of the investigated compounds to accumulate inside cells or into cell membranes and cause inhibition, even when present only prior to infection.

CuCl2-catalyzed PCDD/F formation and congener patterns from phenols.

Homologue and isomer patterns of polychlorinated dibenzo-p-dioxin (PCDD) and polychlorinated dibenzofuran (PCDF) in CuCl2-catalyzed formation were studied in an isothermal flow reactor using a distribution of 20 phenols as measured in municipal waste incinerator (MWI) exhaust gases. A mixture of 20 phenols was synthesized and used as reactants for this study because phenols are known to be key precursors in the formation of PCDD/F. Experiments were conducted at 400 degrees C. The 92% of nitrogen (N2) and 8% of oxygen (O2) were used as a carrier gas. PCDD/F homologue and isomer patterns with di…

Extraction of anthocyanins and other phenolics from black currants with sulfured water.

Health benefits of fruits, vegetables, and red wine are attributed to anthocyanins and other phytochemicals. In this research, the extraction of phenolics from black currants was optimized using different SO(2) concentrations (28, 300, 700, 1100, and 1372 ppm), temperatures (6, 20, 40, 60, and 74 degrees C), and solvent to solid ratios (S/S) (6, 20, 40, 60, and 74 mL/g). Surface response methodology was used to optimize yields of anthocyanins and total phenolics, as well as their antiradical and antioxidant activities. The extraction of phenolics varied with the SO(2) concentration, S/S, and t…

Molecular mechanisms of chemopreventive effects of selected dietary and medicinal phenolic substances.

Recently, considerable attention has been focused on identifying naturally occurring chemopreventive substances capable of inhibiting, retarding, or reversing the multi-stage carcinogenesis. A wide array of phenolic substances, particularly those present in dietary and medicinal plants, have been reported to possess substantial anticarcinogenic and antimutagenic activities. The majority of these naturally occurring phenolics retain antioxidative and anti-inflammatory properties which appear to contribute to their chemopreventive or chemoprotective activity. Capsaicin (trans-8-methyl-N-vanillyl…

[Analysis of contact dermatitis by cytokines. III.--Sensitization and crossreaction of phenolic compounds].

We examined sensitization and crossreaction by the guinea pig maximization test (GPMT) with phenolic compounds. Skin specimens were collected from earlobes of BALB/c mice on 24 h after elicitation phase of contact hypersensitivity reaction (CHR) with phenolic compounds. The expression of cytokines of skin specimens was examined by the reverse transcriptase/polymerase chain reaction (RT-PCR) method. Consequently, phenolic compounds which showed positive reaction in GPMT expressed IL-2 and IFN-gamma on 24 h after elicitation, and some phenolic compounds showed marked crossreaction. Therefore, it…

The role of oestradiol on the dermal penetration of phenol, alone or in a mixture, in ovariectomized rats.

The human body, as well as that of animals, is almost entirely covered by skin. As a result, skin is considered the first line of defence against various types of chemicals. Many factors can affect the rate of dermal penetration of these agents either by enhancing or decreasing penetration. The aim of this study was to demonstrate the effect of oestradiol on the dermal penetration of phenol, a major chemical found in industry and hazardous waste sites. The effect of oestradiol on the dermal penetration of phenol was assessed by applying (14)C-phenol alone or in combination with trichloroethyle…

Quantitation of the phenolic glycolipid of Mycobacterium leprae and relevance to glycolipid antigenemia in leprosy.

Chemical and immunologic procedures have been developed for quantitation, in the body fluids of patients with leprosy, of phenolic glycolipid I, the major specific antigen of the leprosy bacillus. Serum samples were extracted with CHCl3/CH3OH and fractionated on columns of silicic acid. Thin-layer chromatography with a sensitivity of about 500 ng allowed detection of the glycolipid in untreated lepromatous and borderline patients, and high-pressure liquid chromatography gave a quantitation of 0.8-3.7 micrograms/ml of serum from four patients. An ELISA-inhibition assay with polyclonal antibodie…

Determination of amphetamine, norephedrine, and their phenolic metabolites in rat brain by gas chromatography.

A specific analytical procedure for the quantitation of amphetamine (I), norephedrine (III), and their amphoteric metabolites, p-hydroxy-amphetamine (II) and p-hydroxynorephedrine (IV), in biological samples using electron-capture gas chromatography (GC-EC) is described. The procedure utilizes the ion-pairing reagent, bis(2-ethylhexyl)phosphoric acid, which frees the amines from most contaminants and permits the efficient extraction of the amphoteric compounds (as acetates) from the aqueous solution. Amines I and III and acetylated amines II and IV were perfluoroacetylated prior to GC-EC analy

Antimicrobial phenolic secondary metabolites from the flowers of Narcissus tazetta L.

Bioassay-guided fractionation of the ethyl acetate antimicrobial fraction from the hydroalcoholic extract of the fresh Narcissus tazetta L. flowers afforded a new antimicrobial compound, tazettin (1), along with 3-caffeoylquinic acid methyl ester (2), methyl-3,4-dihydroxybenzoate (3), and syringic acid (4). The compounds were assigned by NMR and (+)-HRESIMS analyses. Antimicrobial assays revealed that 1 potently inhibited Staphylococcus aureus (18 mm, MIC = 14.9 µM) and Candida albicans (16 mm, MIC = 29.8 µM. Compound 2 exhibited similar potency against S. aureus (16 mm, MIC = 10.8 µM) and C.

Phenolic contaminants generate persistent phenoxyl radicals to accelerate antibiotic degradation.

Water pollution by coexisting multiple contaminants presents escalating challenges to environmental remediation and public health protection. In advanced oxidation processes, contaminant interactions are invariably regarded as detrimental, introducing competitive reactions and matrix interferences that diminish treatment efficiency. However, phenolic compounds-a prevalent class of recalcitrant water pollutants-possess latent oxidative capabilities that remain strategically unexploited. Whether their reactivity can be harnessed to accelerate, rather than impede, the removal of priority contamin…

Effects of apple phenolics on the human metabolome: modulation of key metabolic pathways.

Apples are widely recognized for their potential health benefits, partly attributed to their phenolic compounds. However, their impact on human metabolism remains incompletely understood. This study investigated metabolic effects of apple-derived phenolic compounds using untargeted metabolomics approach across multiple biofluids. In a crossover intervention study, 30 healthy men consumed a phenolic-rich apple juice or a placebo for two weeks. Blood, urine and saliva samples were collected before and after each intervention and analyzed by direct infusion ultra-high resolution mass spectrometry…
