Pulmonary toxicity of trichloroethylene: induction of changes in surfactant phospholipids and phospholipase A2 activity in the mouse lung.

Trichloroethylene (TCE) is a common organic solvent in use as a dry cleaning agent as well as an inhalant anesthetic. Nevertheless the effects of this material on the pulmonary surfactant which prevents alveolar collapse at maximal expiration is not known. Therefore, we have examined the effect of TCE on the intra- and extracellular surfactant pools and the activity of phospholipase A2, an enzyme which controls the remodeling of phosphatidylcholine to dipalmitoylphosphatidylcholine, the primary constituent of the pulmonary surfactant. Male CD-1 mice were treated ip with 2500 or 3000 mg/kg TCE.

Effect of phospholipids on thyroid oligosaccharyltransferase activity and orientation. Evaluation of structural determinants for stimulation of N-glycosylation.

Oligosaccharyltransferase solubilized by Nonidet P-40 was found to have a highly specific lipid requirement which is consistent with the lability of the enzyme when removed from its membrane association. Enzyme activity as measured by the N-glycosylation of a hexapeptide acceptor was greatly stimulated and stabilized by phosphatidylcholine (PC) while other naturally occurring phosphoglycerides had minimal effect. The quaternary ammonium group of PC was observed to be involved in the interaction with the enzyme as modification of the choline moiety by removal of methyl groups resulted in a prog

The decline of arachidonic acid incorporation into phospholipids is particularly evident, indicating that aging affects mainly the utilization of polyunsaturated fatty acids.

Relative effects of different surfactants on intestinal absorption and the release of proteins and phospholipids from the tissue.

The actions of anionic, cationic and non-ionic surfactants on the absorptive capability of rat jejunal tissue in vivo were compared with their effects on the amounts of protein and phospholipid released from the mucosal surface under the same conditions. Release of a comparatively small amount of protein was accompanied by large increases in the absorption rates of both L-valine and salicylate, whereas much larger quantities of phospholipid were released before any increase in absorption were observed. Much of the released material appeared to be derived from mucus which was partly degraded af

Phospholipid analogs and beyond: pleiotropic roles of betaine lipids in phycology, ecology, and biomedicine.

Betaine lipids (BLs) are a class of non-phosphorus glycerolipids widely found in microalgae, fungi, bacteria, mosses, and ferns. For decades, the central paradigm regarding the biological function of BLs has been their role in substituting for phospholipids under phosphate starvation-a crucial adaptive mechanism for survival in oligotrophic environments. However, accumulating evidence indicates that the functional diversity of BLs is far more extensive. Recent studies have demonstrated their distinct roles in fatty acid metabolism, adaptation to various environmental stresses, and ecological i…

Phospholipid-induced bijel formation for fabricating plant-based cheese with meltability and stretchability.

Developing plant-based cheeses that fully replicate the authentic melting and stretching behavior of Mozzarella cheese remains a significant hurdle in food science. In this work, we propose a phospholipid-induced strategy to engineer plant-based cheese with excellent meltability and stretchability based on soy glycinin coacervates. We demonstrate that phospholipid critically governs the structure transition from a phase-separated emulsion gel to a bicontinuous interfacially jammed emulsion gel (bijel). Phase behavior mapping identified a narrow compositional window in which both meltability an

A Bioactive Phospholipid Promotes Rapid Lung Progenitor Activation via AP-1.

UNLABELLED: Upon injury to the distal lung, alveolar type 2 cells (AT2s) must make a discrete switch from surfactant factories to stem cells capable of regeneration, which involves both proliferation and differentiation into oxygen-exchanging alveolar type 1 (AT1) cells. However, the discrete signals and molecular pathways facilitating this fundamental switch in AT2 functionality are uncertain. Here we demonstrate that the bioactive lipid lysophosphatidic acid (LPA), typically associated with driving fibrosis, is an extremely efficient inducer of this state change in comparison to previously i

The effect of adding human Milk phospholipid analogs on the digestive properties of infant formula.

Phospholipids are important structural components of the milk fat globule membrane (MFGM) and contribute to fat globule integrity and lipid digestion. In this study, human milk phospholipid analogs (HMPAs) were prepared by blending yak ghee sphingomyelin-rich phospholipids (SMPL), soybean lecithin (SL), egg yolk lecithin (EYL), and sunflower lecithin (SSL). The optimized HMPAs blend contained 39% SL, 11% EYL, 18% SSL, and 32% SMPL, giving the highest calculated similarity score (88.315) to the selected human milk phospholipid and fatty acid targets. HMPAs were then combined with commercial MFG…

Iteratively Designed Phospholipid-Targeted Supramolecular Sonosensitizers for Precision Treatment of Pulmonary Infections.

Fungal infections represent a significant global health threat, exacerbated by the scarcity of new therapeutic options, increasing drug resistance, and host toxicity. Sonodynamic therapy (SDT) offers a promising approach to overcoming antimicrobial resistance by enabling the localized generation of cytotoxic reactive oxygen species (ROS) within deep-seated lesions. Nevertheless, reports of SDT for in vivo antifungal treatment remain scarce, largely due to suboptimal ROS generation efficiency, poor biofilm penetration, and a lack of fungal specificity. Herein, we report the iterative design of

Driving a Stimuli-Responsive Wedge in the Packing of Phospholipid Membranes Using Bolaamphiphile Intercalants.

Bolaamphiphiles─amphiphilic molecules with polar groups at each of the two ends of a hydrophobic tail with pH-sensitive spontaneous molecular curvatures, endow membranes of extremophiles with an exquisite balance between stability (or robustness) and adaptability (or plasticity). But how the presence (or real-time insertion) of bolaamphiphiles influences lamellar lipid membranes is poorly understood. Using a combination of time-resolved confocal fluorescence microscopy, in situ small-angle X-ray and neutron scattering (SAXS and SANS), and neutron spin echo (NSE) measurements, we monitor here t

Secreted phospholipase A2 regulates intercellular communications by coordinating extracellular phospholipid metabolism.

Lipids play fundamental roles in life. In essence, "phospholipase A2" (PLA2) indicates a group of enzymes that release fatty acids and lysophospholipids by hydrolyzing the sn-2 position of glycerophospholipids. To date, more than 50 enzymes that possess PLA2 or related lipid-metabolizing activities have been identified in mammals and are subdivided into several families in terms of their structures, catalytic mechanisms, tissue/cellular localizations, and evolutionary relationships. Among the PLA2 superfamily, the secreted PLA2 (sPLA2) family contains 11 isoforms in mammals, each of which has …

Rational design and translational advancement of phospholipid-based nanocarriers for targeted cancer therapy.

Источник пометил публикацию как отозванную

Phospholipid-derived nanocarriers represent a versatile and chemically customizable class of drug delivery systems that self-assemble into bilayered vesicles due to their intrinsic amphiphilicity. These systems can encapsulate both hydrophilic and hydrophobic drugs through non-covalent interactions and manipulation of lipid phase behavior. This review examines the molecular and supramolecular principles underlying the formation, stability, and functional performance of key phospholipid-based nanocarriers-including liposomes, transferosomes, ethosomes, invasomes, phytosomes, pharmacosomes, and
