Perception of Radiation Risk by Japanese Radiation Specialists Evaluated as a Safe Dose Before the Fukushima Nuclear Accident.

From October to December 2010, just before the radiological accident at the Fukushima Daiichi nuclear power plant, 71 radiation professionals from radiation facilities in Japan were asked what they considered as a "safe dose" of radiation for themselves, their partners, parents, children, siblings, and friends. Although the 'safe dose' they noted varied widely, from less than 1 mSv y to more than 100 mSv y, the average dose was 35.6 mSv y, which is around the middle point between the legal exposure dose limits for the annual average and for any single year. Similar results were obtained from o

Cerebral radioprotection by pentobarbital: dose-response characteristics and association with GABA agonist activity.

Pentobarbital reduces cerebral radiation toxicity; however, the mechanism of this phenomenon remains unknown. As an anesthetic and depressant of cerebral metabolism, pentobarbital induces its effects on the central nervous system by stimulating the binding of gamma-aminobutyric acid (GABA) to its receptor and by inhibiting postsynaptic excitatory amino acid activity. The purpose of this study is to investigate the role of these actions as well as other aspects of the radioprotective activity of pentobarbital. Fischer 344 rats were separated into multiple groups and underwent two dose-response

Deletional self-tolerance to a melanocyte/melanoma antigen derived from tyrosinase is mediated by a radio-resistant cell in peripheral and mesenteric lymph nodes.

Self-tolerance to melanocyte differentiation Ags limits the ability to generate therapeutic antimelanoma responses. However, the mechanisms responsible for CD8 T cell tolerance to these Ags are unknown. We have used a newly generated TCR-transgenic mouse to establish the basis of tolerance to one such Ag from tyrosinase. Despite expression of tyrosinase transcripts in the thymus, central deletion does not shape the tyrosinase-specific CD8 T cell repertoire. We demonstrate that this endogenously expressed melanocyte Ag is constitutively presented in both peripheral and mesenteric lymph nodes, l

[Effects and mechanism of a locally combined treatment with radiation and heat therapy (author's transl)].

An animal experimental study on solid Walker-tumors was undertaken to determine the therapeutic effects utilizing hyperthermy in the form of combined decimeter wave megavolt treatment. Noted was the superior effect of combination therapy as compared to exclusive use of radiation therapy. Hyperthermy therapy alone does inhibit tumor growth but not involution. This inhibiting characteristic of hyperthermy probably changes cellular mitotic rhythm in the form of cellular synchronisation. The indications for combined hyperthermy megavolt treatment is hypoxic malignant tumors of low radiation sensit

Radiation Resilient Synthetic Antiferromagnets-Based Neuromorphic Device for Sea Surface Temperature Reconstruction.

Reconstruction of sea surface temperature is critical for marine monitoring, yet conventional edge devices based on complementary metal-oxid … evices retain over 92% of their performance after 1 Mrad (Si) γ-irradiation, demonstrating inherent radiation tolerance arising from strong antiferromagnetic exchange coupling. By mapping the nonlinear conductance response of SAF onto the cross-attention mechanism of a Perceiver IO architecture, we achieve accurate reconstruction of sea surface temperature fields fro …

In vivo dosimetry for testicular and scalp shielding in total skin electron therapy using a radiophotoluminescence glass dosimeter.

Mycosis fungoides (MF) is a common, low-grade non-Hodgkin's lymphoma of skin-homing T lymphocytes that can be treated via skin-directed radiotherapy. Our institution has implemented total skin electron therapy (TSET) with a 4.3 m source-to-surface distance (SSD) and 6 MeV electron beams with a beam spoiler. A 35-year-old male undergoing TSET desired to avoid radiotherapy-induced hair loss and temporary infertility; therefore, leakage dose to scalp and testicles was reduced with a special radiation shield composed of stacked lead sheets. The shields for the scalp and scrotal were of 3 mm and 6

Down-regulation of the human CDC16 gene after exposure to ionizing radiation: a possible role in the radioadaptive response.

We have used the method of differential display of mRNAs to search for changes in gene expression associated with radioadaptation triggered by low doses of ionizing radiation in human lymphoblasts. We isolated a cDNA designated PB13 that was down-regulated as early as 1 h after irradiation with 4 Gy in cells adapted by a pre-exposure to a dose of 2 cGy, compared to 3 h in nonadapted cells (4 Gy alone). Northern analysis confirmed the differential expression of a 2.4-kb transcript that was repressed for at least 10 h after irradiation. The major part of the PB13 cDNA was identical to the human

[Stereotactic radiotherapy of brain metastases in complex situations].

Stereotactic radiation therapy of brain metastases is a treatment recognized as effective, well tolerated, applicable for therapeutic indications codified and validated by national and international guidelines. However, the effectiveness of this irradiation, the evolution of patient care and the technical improvements enabling its implementation make it possible to consider it in more complex situations: proximity of brain metastases to organs at risk; large, cystic, haemorrhagic or multiple brain metastases, combination with targeted therapies and immunotherapy, stereotactic radiotherapy in p

Phloroglucinol inhibits ultraviolet B radiation-induced oxidative stress in the mouse skin.

PURPOSE: Previously we demonstrated that phloroglucinol (1,3,5-trihydroxybenzene) protected human HaCaT keratinocytes against ultraviolet B (UVB, 280-320 nm)-induced oxidative stress in vitro by scavenging intracellular reactive oxygen species (ROS). The current study investigated whether phloroglucinol could similarly protect the mouse skin against UVB-induced oxidative tissue damage in vivo. MATERIALS AND METHODS: Male 7-week-old Balb/c mice were divided into the following untreated normal control, phloroglucinol only-treated, vehicle plus UVB (30 or 60 mJ/cm(2))-exposed, and phloroglucinol

Evaluation of a single-isocenter technique for axillary radiotherapy in breast cancer.

The aim of this study was to develop a technique for axillary radiotherapy that minimizes the risk of radiation-induced damage to the surrounding normal tissue (i.e., arm, shoulder, lung, esophagus, and spinal cord) while keeping the risk of a nodal recurrence to a minimum. A planning study was performed in 20 breast cancer patients. The target volume of the axillary treatment encompassed the periclavicular and axillary lymph node areas. The 3-dimensional (3D) computed tomography (CT) information in this study was used to outline the lymph node areas and the organs at risk (i.e., the esophagus

IFN signaling at the nexus of the radiotherapy response in malignant peripheral nerve sheath tumors.

Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive sarcomas that constitute a major cause of mortality in individuals with neurofibromatosis type 1 (NF-1) and exhibit highly variable responses to radiotherapy. In this issue of the JCI, Zhu and colleagues integrated functional genomics, single-cell transcriptomics, and analysis of human tumors to show that type I IFN signaling shapes both tumor-intrinsic radiation sensitivity of MPNSTs and local recruitment and activation of T cells. Their findings establish IFN signaling as a central coordinator of the radiotherapy response in MP

Exploring treatment complexity in maxillary ameloblastoma: A case study on the efficacy of radiotherapy with transcriptomic and literature insights.

In this report, we describe the case of a patient initially experiencing symptoms including nasal obstruction and rhinorrhoea, leading to surgeries and a diagnosis of ameloblastoma. Despite complete surgical resections, MRI follow-ups indicated tumour regrowth, prompting a shift to radiotherapy. The treatment plan involved a high-dose (60Gy in 30 fractions of 2Gy) volumetric modulated arc therapy. During treatment, the patient experienced minimal side effects and showed significant clinical improvement, including tumour size reduction and resolution of facial paralysis. Follow-up MRI confirmed

The radiomimetic enediyne, 20'-deschloro-C-1027 induces inter-strand DNA crosslinks in hypoxic cells and overcomes cytotoxic radioresistance.

The ability of the radiomimetic anti-tumor enediyne C-1027 to induce DNA inter-strand crosslinks (ICLs), in addition to the expected DNA strand breaks, is unique among traditional DNA targeted cancer therapies. Importantly, radiation therapy and most radiomimetic drugs have diminished effect in hypoxic environments due to decreased induction of DNA strand breaks, which is an oxygen requiring process. However, C-1027's induction of ICLs is enhanced under hypoxia and it is actually more potent against hypoxic cells, overcoming this common tumor resistance mechanism. In this study, an analog of C

Effect of relative humidity on Deinococcus radiodurans' resistance to prolonged desiccation, heat, ionizing, germicidal, and environmentally relevant UV radiation.

To test the effect of humidity on the radiation resistance of Deinococcus radiodurans, air-dried cells were irradiated with germicidal 254 nm UV, and simulated environmental UV or γ-radiation and survival was compared to cells in suspension. It was observed that desiccated cells exhibited higher levels of resistance than cells in suspension toward UV or γ-radiation as well as after 85°C heat shock. It was also shown that low relative humidity improves survival during long-term storage of desiccated D. radiodurans cells. It can be concluded that periods or environments in which cells exist in a

Telomere length modulates human radiation sensitivity in vitro.

The molecular basis of the interindividual differences of normal individuals to ionizing radiation is poorly understood. Several studies in telomerase KO mice with short telomeres have uncovered an inverse relationship between telomere length and radiation sensitivity. The present work aims to determine if chromosome radiosensitivity is correlated with telomere length in healthy individuals. With this purpose, individual radiosensitivity was determined by the micronucleus assay in peripheral blood lymphocytes from two groups of individuals of the same age but with highly heterogeneous telomere

[Features of the radiosensitivity of cells from ataxia-telangiectasia patients].

Cell survival, single and double DNA strand breaks formation and removal, spontaneous and induced chromosome aberrations and sister chromatid exchange (SCE) levels in gamma-irradiated cells of patients with ataxia-telangiectasia (AT) were studied. Except SCE all of the above indexes of AT cells sensitivity, were higher, than in normal human cells, but lower, than it is commonly characteristic of AT cells in literature. A conclusion is that the analysed AT cells belong to the AT-variant form. Possible mechanisms of high radiosensitivity of AT cells, accompanied by radioresistance of DNA replica
