The accessory molecule Lgp55 plays a role early in murine fetal thymocyte differentiation.

A rat IgM monoclonal antibody, PA3-795, inhibits the antigen-specific responses of mouse T-cell hybridomas. It recognizes a heavily glycosylated cell-surface protein, designated Lgp55, that is detectable after activation on mature T cells. During fetal life, Lgp55 is found at high levels on newly immigrant thymic T-cell precursors prior to surface expression of other T-lineage molecules. High levels of expression are also found on thymocytes in the outer cortex of adult mice. Thymocytes at later stages of differentiation bear decreasing amounts of surface Lgp55, and none is detectable on "sing

Bradykinin suppresses alcohol intake and plays a role in the suppression produced by an ACE inhibitor.

The possible role of the endogenous kinins in the control of alcohol intake was assessed in two experiments. In Experiment 1, naive rats, maintained on ad lib food and water, were given daily 40-min access to a 6% (w/v) alcohol solution and water. Daily intraperitoneal (IP) injections of captopril (20 mg/kg) significantly reduced alcohol intake, while pretreatment with subcutaneous (SC) injections of the bradykinin antagonist [D-Phe7]-bradykinin (100-300 micrograms/kg) attenuated the suppressive effect of captopril on alcohol intake. The saline vehicle or the bradykinin antagonist alone did no

Soy of dietary source plays a preventive role against the pathogenesis of prostatitis in rats.

This study examined the effects of diet on the development of prostatitis in male rats. Adult male rats were placed on either of two specially formulated diets which differed from one another by the presence or absence of soy as the protein source. A third group of rats (control) was fed standard laboratory rat chow which also includes soy as a source of protein. After 11 weeks, it was found that rats maintained on soy-free diet developed prostatitis mainly in the lateral lobe of the prostate. Increased severity and incidence of prostatitis in rats maintained on the soy-free diet coincided wit

Tumor necrosis factor plays a protective role in experimental murine cutaneous leishmaniasis.

The ability of mice to resist infection with L. major correlated directly with the capacity of their LNC to produce TNF in response to in vitro parasite challenge. Blocking TNF in vivo by passively administering anti-TNF antibodies exacerbated the course of L. major infection, resulting in substantially larger cutaneous lesions and elevated numbers of parasites within those lesions. In addition, treatment of infected mice with exogenous rHuTNF afforded host protection as evidenced by smaller lesion size and decreased parasite counts. Taken together, these results suggest a central role for TNF

[The "HELLP syndrome" without fragmentocytes: do drug-induced antibodies play a role?].

This is a case report on a nulliparous women, who developed the HELLP syndrome following intravenous tocolysis. Contrary to our expectations, neither schistocytes nor signs of intravasal coagulation were apparent; however, we found Fenoterol-associated anti-erythrocyte antibodies. However, the low titre of these antibodies does not explain the massive haemolysis. It is suggested, that these antibodies represent a causal factor in developing the HELLP syndrome.

Do circulating factors play a role in the pathogenesis of minimal change nephrotic syndrome?

This review examines the studies which have been undertaken to test the hypothesis that minimal change nephrotic syndrome of childhood (MCNS) is a primary immune disorder and that there is altered T-cell function which results in release of a circulating factor. This factor alters glomerular permeability, perhaps by modifying charge sites in the glomerular capillary bed, and results in selective proteinuria. The abnormalities in immune function observed in MCNS are summarized, as are the studies of circulating factors which have been identified. Although some agents have been shown to alter ca

T-cell suppressor factors play an integral role in preventing fetal rejection.

In previous studies we showed that the blocking of T-cell suppressor factors (TsF) with monoclonal antibody could completely ablate pregnancy, and demonstrated the presence of TsF in fetal and maternal tissues. In our current study we used a monoclonal antibody specific for TsF to determine the time during gestation when TsF is most integral in the maintenance of pregnancy. Significant decreases in the number of viable pregnancies when monoclonal antibody was administered on days 3, 4 and 5 were demonstrated. In addition, ELISAs were used to measure the levels of TsF in tissues from pregnant a

Evaluation of the role hemoglobin in cerebrospinal fluid plays in producing contractions of cerebral arteries.

Many investigators have concluded that hemoglobin is the spasmogen responsible for cerebral vasospasm. The present study was designed to ascertain whether the contractile responses of isolated canine basilar arteries to xanthochromic cerebrospinal fluid from subarachnoid hemorrhage patients was associated with hemoglobin concentration as measured spectrophotometrically. The results clearly showed that spasmogenicity and hemoglobin content were not correlated. The magnitude and duration of the arterial responses varied greatly, further indicating that more than a single factor was responsible.

Do food antigens play a role in the pathogenesis of some cases of human glomerulonephritis?

Circulating immune complexes after a test meal were measured with three methods (PEG precipitation, Clq-ELISA and the indirect granulocyte phagocytosis test) in 10 controls, two symptomless persons with selective IgA deficiency and 14 patients with various types of glomerulonephritis, of which two patients (with idiopathic membranous glomerulopathy and local focal glomerulonephritis) also had selective IgA deficiency. The PEG and Clq-ELISA test did not show significant differences between the groups. In the two symptomless persons with selective IgA deficiency and in the patient with local foc

Overweight/obesity and gait abnormality: what role does the brain play in the relationship?

BACKGROUND: Gait is related to weight factors and regulated by the nervous system. Emerging evidence indicates that adiposity impacts brain health. This study explores the relationship between anthropometric indices and gait performance, and investigates the role of brain parameters in the relationship. METHODS: This study included 710 community-dwelling older adults from the Taizhou Imaging Study. Nine anthropometric indices were calculated to assess body weight using standard formulas based on measured height, weight, waist circumference (WC), and hip circumference. Gait assessment includes

TGFα plays essential roles in angiogenesis and cognitive recovery after ischemic stroke.

Stroke is a major global health issue and imposes a financial burden on both survivors and caregivers. Current treatments for ischemic stroke, such as intravenous thrombolysis and endovascular thrombectomy, have strict time constraints, leaving a substantial need for therapies that improve tissue recovery. Enhancing revascularization is essential for creating a supportive environment for brain repair. Our previous study shows that transforming growth factor alpha (TGFα) is critical in promoting white matter integrity and long-term recovery by protecting oligodendrocytes after stroke. We detect

Adipose tissue plays a crucial role in obesity-induced postoperative neurocognitive disorders.

BACKGROUND: Postoperative neurocognitive disorder (PND) is characterized by cognitive deficits that emerge after surgery. Although often reversible within weeks or months, PND can persist and lead to significant central dysfunction and an increased risk of dementia. Given that obesity is a chronic low-grade inflammatory condition previously associated with structural and functional brain alterations, we hypothesized and demonstrated that it could be a significant risk factor for PND. METHODS: We assessed the incidence of PND and cognitive performance in obese individuals undergoing cardiac or

TgDDP is a novel DIX domain protein that plays an essential role in Toxoplasma endodyogeny.

UNLABELLED: Apicomplexan parasites are protozoan pathogens responsible for major human diseases, including toxoplasmosis, malaria, and cryptosporidiosis. Toxoplasma gondii has emerged as a model for studying cell division in tissue coccidians. Unlike higher eukaryotes that divide by binary fission, Toxoplasma replicates via internal budding (endodyogeny), yet many factors governing daughter cell biogenesis remain poorly understood. Here, we characterize TgDDP (T. gondii DIX-DnaJ domain protein), a previously unannotated protein containing both DIX and DnaJ domains-a unique combination restrict

Accumulation of newly synthesized docosahexaenoic acid plays an essential role in heart regeneration.

Adult zebrafish and neonatal mice can fully regenerate their hearts after partial amputation through the proliferation of preexisting cardiomyocytes (CMs). However, the adult mammalian heart has limited regenerative capability following cardiac damage. The reason for this phenomenon remains elusive. Here, we find that docosahexaenoic acid (DHA) is accumulated only in the injured hearts of zebrafish and neonatal mice, but not of adult mice, which coincides with the upregulation of DHA synthesis genes in CMs, fibroblasts, and macrophages near the injury areas. Inhibition of Fads2, a DHA synthesi

Guided and unguided neural organoids play complementary roles in studying neurodevelopment and neuroinflammation.

OBJECTIVE: This study aimed to compare guided dorsal forebrain neural organoids with unguided neural organoids, focusing on differences in structural organization, cellular composition, and functional properties. METHODS: Using the same human induced pluripotent stem cell line, we applied two established differentiation protocols in parallel to generate guided and unguided neural organoids. RESULTS: Guided neural organoids exhibited reproducible cytoarchitecture, relatively homogeneous morphology, and robust network activity, making them particularly well-suited for modeling specific aspects

Pyroptosis Plays a Key Role Through Macrophages in Primary Biliary Cholangitis of Mice.

BACKGROUND AND AIMS: Primary biliary cholangitis (PBC) is an autoimmune intrahepatic cholestatic disease with both environmental and genetic participation. In this study, we aim to investigate the involvement of pyroptosis in PBC mice based on prior bioinformatic analysis of PBC patients and explore the immune cell populations potentially involved. METHODS: KEGG pathway enrichment analysis was performed using the GSE119600 dataset from the Gene Expression Omnibus (GEO) database, which includes whole-blood samples from PBC patients (n = 90) and non-liver disease CTRs (n = 47).
