Vitamin D attenuates myofibroblast differentiation and extracellular matrix accumulation in nasal polyp-derived fibroblasts through smad2/3 signaling pathway.

To investigate the potential role of vitamin D (1,25(OH)2D3) in preventing the development of nasal polyps, we examined the effect of vitamin D on myofibroblast differentiation and extracellular matrix (ECM) production in TGF-β1-induced nasal polyp-derived fibroblasts (NPDFs) and elucidated the mechanisms underlying its inhibitory effect. 1,25(OH)2D3 significantly reduced expression levels of α-SMA, a myofibroblast marker, and fibronectin, a representative ECM component, in a dose-dependent manner in TGF-β1-induced NPDFs. 1,25(OH)2D3 suppressed activated Smad2/3 in time-course. Up-regulation o

TGFβ/Smad2/3-Mediated Crosstalk Between Vocal Fold Fibroblasts and Myoblasts In Vitro.

OBJECTIVES: Traditionally, disorders of the vocal fold (VF) mucosa and underlying musculature have been regarded as mutually exclusive entities. However, emerging evidence from other organ systems suggests mucosal and muscle compartments engage in reciprocal interactions with functional consequences. We hypothesized that similar crosstalk exists in the VF, whereby fibrotic mucosa influences adjacent muscle. To model this process, we stimulated human VF fibroblasts (HVOX) with TGF-β1, a central mediator of fibrosis, and examined the effects on rat VF myoblasts (rVF-Mbs), as well as reciprocal i

Construction of an Esophageal Cancer Prognostic Model Based on Mitochondrial Energy Metabolism-Related Genes and Mechanism of INHBA in Promoting Malignant Progression via the Smad2/3 Pathway.

OBJECTIVE: Esophageal cancer (ESCA) has poor prognosis and lacks reliable biomarkers. This study aimed to construct a mitochondrial energy metabolism-related prognostic model and identify key regulatory genes. METHODS: TCGA-ESCA and GSE53625 data were used as training and validation cohorts, respectively. Mitochondrial energy metabolism scores were calculated by ssGSEA. Differential expression analysis and WGCNA identified key genes, followed by consensus clustering, Cox/LASSO regression, enrichment analysis, immune infiltration, TIDE evaluation, and nomogram development. A mutation update on the LDS-associated genes TGFB2/3 and SMAD2/3.

The Loeys-Dietz syndrome (LDS) is a connective tissue disorder affecting the cardiovascular, skeletal, and ocular system. Most typically, LDS patients present with aortic aneurysms and arterial tortuosity, hypertelorism, and bifid/broad uvula or cleft palate. Initially, mutations in transforming growth factor-β (TGF-β) receptors (TGFBR1 and TGFBR2) were described to cause LDS, hereby leading to impaired TGF-β signaling. More recently, TGF-β ligands, TGFB2 and TGFB3, as well as intracellular downstream effectors of the TGF-β pathway, SMAD2 and SMAD3, were shown to be involved in LDS. This empha

Smad2 protects against TGF-β1/Smad3-mediated collagen synthesis in human hepatic stellate cells during hepatic fibrosis.

With structural similarity but functional diversity, Smad2 and Smad3 interact with each other to mediate transforming growth factor-β (TGF-β)-triggered signaling transduction. However, in the hepatic fibrosis, the detailed roles of R-Smads, and interaction between Smad2 and Smad3 are still undefined. In this setting, we established a rat model of CCl4-induced hepatic fibrosis in vivo and TGF-β1-treated hepatic stellate cell model in vitro to detect whether Smad2 and Smad3 play distinct roles in mediating liver fibrogenesis. Results indicated that both phosphorylation of Smad2 and Smad3 were de

Erythropoietin decreases carbon tetrachloride-induced hepatic fibrosis by inhibiting transforming growth factor-beta.

BACKGROUND: In addition to hematopoietic effect, the erythropoietin is known as a multifunctional cytokine with anti-fibrosis and organ-protective activities. The purpose of this study was to evaluate the effect of recombinant human erythropoietin (rhEPO) on hepatic fibrosis and hepatic stellate cells (HSCs). METHODS: Carbon tetrachloride (CCl(4)) induced hepatic fibrosis mice models were used for in vivo study and HSCs line for in vitro study. CCl(4) and rhEPO (0, 200 or 1000 U/kg) was injected intraperitoneally in BALB/c mice three times a week for 4 weeks. Immunohistochemistry and immunobl

LASS2 alleviates liver fibrosis by interacting with THBS1, TFRC, and APOE to inhibit TGF-β1 signaling and ferroptosis, and by reshaping the hepatocyte lipid secretome to suppress paracrine HSC activation.

BACKGROUND: Liver fibrosis (LF) lacks effective therapies. Longevity assurance homolog 2 (LASS2), a key ceramide synthase, contributes to liver homeostasis, but its role in LF remain unclear. METHODS: Bioinformatics revealed LASS2 correlation with classic fibrotic factors, and its expression was assessed in fibrotic mouse livers and activated LX-2 cells. Gain- and loss-of-function studies were performed in vivo (liver-specific AAV) and in vitro (recombinant adenovirus), followed by phenotypic and mechanistic analyses. Protein interactions were mapped by proteomics and co-immunoprecipitation (

Nebivolol ameliorates sepsis-evoked kidney dysfunction by targeting oxidative stress and TGF-β/Smad/p53 pathway.

Sepsis is a potential fetal organ destruction brought on through an overzealous immunologic reaction to infection, causing severe inflammation, septic shock, and damage to different organs. Although there has been progress in the identification and controlling of clinical sepsis, the fatality rates are still significant. This study, for the first time, intended to examine the possible ameliorative impact of Nebivolol, a β1-adrenergic antagonist antihypertensive drug, against nephrotoxicity resulted from cecal ligation and puncture (CLP)-induced sepsis in rats, on molecular basis. The Smad transcriptional corepressor TGIF recruits mSin3.

The homeodomain protein TG-interacting factor (TGIF) represses transcription by histone deacetylase-dependent and -independent means. Heterozygous mutations in human TGIF result in holoprosencephaly, a severe genetic disorder affecting craniofacial development, suggesting that TGIF is critical for normal development. After transforming growth factorbeta (TGFbeta) stimulation, Smad proteins enter the nucleus and form transcriptional activation complexes or interact with TGIF, which functions as a corepressor. The relative levels of Smad corepressors and coactivators present within the cell may

Morin attenuates diethylnitrosamine-induced rat liver fibrosis and hepatic stellate cell activation by co-ordinated regulation of Hippo/Yap and TGF-β1/Smad signaling.

Despite great progress in understanding the activation of hepatic stellate cells (HSCs) during liver fibrosis, therapeutic approaches to inhibit HSC activation remain very limited. Recent reports highlight Yes-associated protein (Yap) and transforming growth factor-β1 (TGF-β1) as critical regulators of HSC activation and henceforth a compound targeting Hippo/Yap and TGF-β1/Smad pathways would be a potential anti-fibrotic candidate. Morin, a dietary flavonoid, was earlier reported to inhibit HSC proliferation and induction of apoptosis of cultured HSCs, mainly by suppressing Wnt/β-catenin and N

Cardiac myocyte-specific ablation of follistatin-like 3 attenuates stress-induced myocardial hypertrophy.

Transforming growth factor-β family cytokines have diverse actions in the maintenance of cardiac homeostasis. Follistatin-like 3 (Fstl3) is an extracellular regulator of certain TGF-β family members, including activin A. The aim of this study was to examine the role of Fstl3 in cardiac hypertrophy. Cardiac myocyte-specific Fstl3 knock-out (KO) mice and control mice were subjected to pressure overload induced by transverse aortic constriction (TAC). Cardiac hypertrophy was assessed by echocardiography and histological and biochemical methods. KO mice showed reduced cardiac hypertrophy, pulmonar

Rupestonic Acid of Artemisia Rupestris L. Extract Treats Pulmonary Fibrosis in COPD by Targeting TGF-β1.

Pulmonary fibrosis (PF) is the final stage of lung damage, such as chronic obstructive pulmonary disease (COPD), with no effective treatment. Transforming growth factor beta 1 (TGF-β1) is a key protein involved in fibrosis and regulating inflammation. Therefore, targeting components of TGF-β1 is an effective strategy for controlling PF. Artemisia rupestris L, a perennial herb of rupestris belonging to Artemisia, has been prescribed as a treatment for pulmonary inflammation. We investigated the effects and mechanisms of Artemisia rupestris L ethanol extract (EEAR) on PF induced by cigarette smo

Anti-fibrotic effects of evogliptin in fibroblasts derived from post-burn hypertrophic scars.

Hypertrophic scar (HTS) frequently occurs after burn injury, characterized by persistent fibroblast activation, overexpression of α-smooth muscle actin (αSMA) and extracellular matrix (ECM) components. Complete scarless regeneration remains incompletely understood. Dipeptidyl peptidase-4 (DPP4) is a cell-surface serine protease, its inhibitors (DPP4i) have been associated with fibrosis. However, the functional role of evogliptin in HTS formation remains unclear. This investigated its effects on fibrosis markers in hypertrophic scar fibroblasts (HTSFs). Human normal fibroblasts (HNFs) and HTSFs

Calycosin ameliorates inflammation and M1 macrophage polarization via Spon2 in LPS-triggered MH-S alveolar macrophages.

Acute lung injury (ALI) remains a critical inflammatory condition with limited therapeutic interventions. This study explores the anti-inflammatory potential of calycosin (CAL), a bioactive flavonoid, in lipopolysaccharide (LPS)-induced MH-S alveolar macrophages cell line, with particular focus on macrophage polarization mechanisms. Through CCK-8 cytotoxicity assessment and subsequent experimental grouping (control, LPS, and LPS + CAL), we demonstrated CAL's ability to significantly suppress LPS-triggered inflammatory mediators including IL-1β, TNF-α, and IL-6 at both transcriptional and prote

Molecular characterization of suburothelial fibrosis in murine acute recurrent bladder inflammation.

Chronic fibrosis replaces functional organ tissue with scar tissue by overproduction of a thick and stiff extracellular matrix. Bladder fibrosis decreases bladder compliance, ultimately resulting in overactive bladder. The phenoconversion of fibroblasts into myofibroblasts is the defining feature of fibrosis. Recently, regionally distinct populations of bladder platelet-derived growth factor receptor alpha positive (PDGFRα+) cells were identified as fibroblasts. Because of this heterogeneity, the identity of the bladder fibroblast cells that undergo phenotypic conversion into myofibroblasts is

Molybdenum interferes with MMPs/TIMPs expression to reduce the receptivity of porcine endometrial epithelial cells.

To investigate the effect of trace element molybdenum (Mo) on the receptivity of porcine endometrial epithelial cells (PEECs) and evaluate Mo toxicity and its potential molecular mechanisms, Mo-treated PEECs models were established by incubating the cells with various concentrations of medium containing Mo (0, 0.005, 0.020, 0.200, and 5 mmol/L MoNa2O4·2H2O). The results showed that Mo disrupted the morphology and ultrastructure of PEECs, triggered blurred cell edges, cell swelling, cell cycle arrest, and increased apoptosis. At the molecular level, Mo treatment activated the TGF-β1/SMAD2 and P

Downregulation of Elovl5 promotes breast cancer metastasis through a lipid-droplet accumulation-mediated induction of TGF-β receptors.

Metastatic breast cancer cannot be cured, and alteration of fatty acid metabolism contributes to tumor progression and metastasis. Here, we were interested in the elongation of very long-chain fatty acids protein 5 (Elovl5) in breast cancer. We observed that breast cancer tumors had a lower expression of Elovl5 than normal breast tissues. Furthermore, low expression of Elovl5 is associated with a worse prognosis in ER+ breast cancer patients. In accordance with this finding, decrease of Elovl5 expression was more pronounced in ER+ breast tumors from patients with metastases in lymph nodes. Alt
