Staphylococcal superantigen-like protein 10 induces necroptosis through TNFR1 activation of RIPK3-dependent signal pathways.

Staphylococcal aureus (S. aureus) infection can lead to a wide range of diseases such as sepsis and pneumonia. Staphylococcal superantigen-like (SSL) proteins, expressed by all known S. aureus strains, are shown to be involved in immune evasion during S. aureus infection. Here, we show that SSL10, an SSL family protein, exhibits potent cytotoxicity against human cells (HEK293T and HUVEC) by inducing necroptosis upon binding to its receptor TNFR1 on the cell membrane. After binding, two distinct signaling pathways are activated downstream of TNFR1 in a RIPK3-dependent manner, i.e., the RIPK1-RI

Administration of Vaccines in Dairy Sheep and Goat Farms: Patterns of Vaccination, Associations with Health and Production Parameters, Predictors.

This paper reports findings regarding patterns of vaccine usage in sheep and goat farms, in 325 sheep flocks and 119 goat herds throughout Greece. The objectives of the study were (a) to describe the patterns of vaccine administration in small ruminant farms and (b) to highlight factors that were associated with vaccinations in the farms. Vaccination against brucellosis was performed in all farms into the study. Among optional vaccinations, anti-clostridial vaccination was most frequently performed (in 97.8% of farms), followed by vaccination against contagious agalactia, (56.5% of farms), pne

Clonal expansion of T lymphocytes causes arthritis and mortality in mice infected with toxic shock syndrome toxin-1-producing staphylococci.

Erosive arthritis is a common and feared complication of staphylococcal infection. The reason(s) for the progressive course of the arthritis is unknown. It has been recently established that enterotoxins produced by Staphylococcus aureus display superantigen properties leading to stimulation of T cells carrying distinct T cell receptor V beta elements. This finding provides a potential connection between staphylococcal exoproteins and endogenous immune mechanisms participating in the infectious process. We have recently describe successful induction of infections arthritis in mice after intrav

[Special emergencies in dermatology].

Emergency cases in dermatology are rare but potentially life-threatening conditions. They comprise a broad spectrum of diseases which require immediate hospitalization or even intensive care. A rapid diagnosis and appropriate therapy is necessary since some of these emergency cases are associated with a high mortality and with severe disabling complications. Typical examples are: the bacterial infections Waterhouse-Friderichsen syndrome, necrotizing erysipelas and necrotizing fasciitis, as well as staphylococcal and streptococcal toxic shock syndromes. Another special emergency situation is ve

[Use of fluoroquinolone chemotherapy in orthopedics and traumatology].

In the submitted review the authors analyze hitherto published results with the use of new antimicrobial chemotherapeutic drugs--fluoroquinolones in the treatment of infections of the bones, joints, soft tissues and also in the peroperative prophylaxis in orthopaedics and traumatology. Quinolones are one of the drugs of choice which may be successful in the treatment of staphylococcal and gram-negative chronic osteomyelitis which are resistant to classical treatment and also the drug of choice in the treatment of septic arthritis and ostitis.

[Investigation of food poisoning of staphylococcal etiology using a comparison of the extracellular proteins of the isolated causative agents].

Materials on the study of four outbreaks of alimentary toxinfections of staphylococcal etiology, carried out by the method of the identification of staphylococci by the spectra of their extracellular proteins, are presented. As the result of this study, the sources and transfer routes of infections have been established. The method of the identification of staphylococci by the exoprotein spectra has made it possible to find out the epidemiological chain with greater accuracy. Such identification may serve as an auxiliary method in epidemiological surveillance.

[Long-term antibiotic suppressive therapy for an infected thoracic aorta graft].

BACKGROUND: Infections of arterial vascular grafts are among the most dreaded complications in vascular surgery. Infection in a thoracic aorta graft poses particular challenges. Depending on the local anatomy, extraanatomic bypass, otherwise the cornerstone in the management of infected vascular grafts, is usually impossible. MATERIAL AND METHODS: We present a case report on a patient with an infected thoracic aorta graft and discuss the choice of antibiotics for long-term suppressive therapy. RESULTS: In the course of the 52 months since the insertion of the aortic graft, the patient experi

Graft infection and bacteremia with Listeria monocytogenes in a patient receiving hemodialysis.

Patients receiving hemodialysis are particularly susceptible to infection. We report a case of Listeria monocytogenes bacteremia and graft infection developing in a patient receiving hemodialysis. Vancomycin hydrochloride therapy was initiated in anticipation of a staphylococcal infection, and continued as the patient's clinical course improved. Ultimately the arteriovenous graft required excision. Identification of the organism and drug susceptibilities are described. To our knowledge, this is the first case report of both an L monocytogenes arteriovenous graft infection and the use of vancom

Purpura fulminans secondary to rickettsial infections: A case series.

Purpura fulminans (PF) is a descriptive term used to describe a heterogeneous group of disorders characterized by rapidly progressive purpuric lesions that may develop into extensive areas of skin necrosis, and peripheral gangrene. This rare disorder is associated with laboratory evidence of consumptive coagulopathy and is often fatal. PF is usually associated with many infections, most notably with meningococcal, staphylococcal, and streptococcal infections. However, there are very few reports of this entity with spotted fever and scrub typhus from India. Rickettsial infections are an underdi

[Opportunistic microorganisms in urinary tract diseases].

The microflora of clinical material (urine, urethral discharge and smear, vaginal discharge and smear, feces in colonic dysbacteriosis) from patients with urinary tract infections (UTI) in the Moscow Region was studied. Escherichia coli and Staphylococcus epidermidis have been found to be the most common agents isolated from the urine of UTI patients. Pseudomonas aeruginosa and other staphylococcal species and enterococci account for a smaller proportion of infections. High- (E. coli and S. epidermidis) and moderate-priority pathogens are translocated into the urinary tract from the colonic bi

Concept of empiric therapy with antibiotic combinations. Indications and limits.

It appears that the use of antibiotic combinations, especially synergistic ones, is indicated for the management of gram-negative bacillary … s care when using them. Besides gram-negative bacillary sepsis in granulocytopenic patients, severe staphylococcal infections might represent an indication for the use of combination therapy, especially in patients with compromised mechanisms of defense against infection.

Submasseteric Infection: A Rare, Deep Space Cheek Infection Causing Trismus.

Submasseteric space infections are rare at any age but particularly so in primary school children. The origin of the infection is usually odontogenic, from pericoronitis in a third molar. Submasseteric inflammation is a deep facial space inflammation, often progressing to mature abscess, and usually misdiagnosed as staphylococcal or streptococcal lymphadenitis or pyogenic parotitis. The hallmark of a masticatory space infection is trismus. The cardinal signs of this infection include a firm mass in the body of the masseter muscle with overlying cellulitis with trismus.

Review of mupirocin ointment in the treatment of impetigo.

The introduction of mupirocin ointment gives the pediatrician a reliable topical alternative to oral antibiotic therapy for group A streptococcal and staphylococcal impetigo. It is as effective as oral antibiotics and is associated with fewer adverse effects. In superficial skin infections that are not widespread, mupirocin ointment offers several advantages. It is highly active against the most frequent skin pathogens, even those resistant to other antibiotics, and the topical route of administration allows delivery of high drug concentrations to the site of infection.

Conversion of chronic staphylococcal mastitis to acute gangrenous mastitis after neutropenia in blood and bone marrow produced by an equine anti-bovine leukocyte serum.

A stock strain of Staphylococcus aureus of mastitis origin, characterized by alpha-, beta-, and delta-toxins, was used to produce chronic mastitis of 20 to 300 days' duration in 6 lactating mammary quarters of 4 cows. Early acute Streptococcus agalactiae mastitis was produced in 1 additional mammary quarter of 1 cow. Equine anti-bovine leukocyte serum (EABLS) was administered to all cows by continuous intravascular drip for 12 to 32 hours. Neutropenia in blood and partial depletion of neutrophil reserve in bone marrow were produced. Chronic subclinical staphylococcal mastitis in 2 quarters of

[Right endocarditis in disseminated aspergillosis].

A 53 year old man with an anaplastic bronchial carcinoma was hospitalised for septic shock and acute respiratory distress after a cutaneous, probably staphylococcal infection, and died in spite of anti-staphylococcal antibiotherapy. The autopsy showed pulmonary, cardiac, cerebral and renal aspergillosis. A right heart aspergillous endocarditis, very rare in this pathology, was also discovered but there were no cardiac valves lesions. The patient was in an "immunodepressed" state as usually observed in pulmonary aspergillosis. The endocardial localisation of aspergillosis and the "pseudo-miliar

Allergy-A New Role for T Cell Superantigens of Staphylococcus aureus?

Staphylococcus aureus superantigens (SAgs) are among the most potent T cell mitogens known. They stimulate large fractions of T cells by cross-linking their T cell receptor with major histocompatibility complex class-II molecules on antigen presenting cells, resulting in T cell proliferation and massive cytokine release. To date, 26 different SAgs have been described in the species S. aureus; they comprise the toxic shock syndrome toxin (TSST-1), as well as 25 staphylococcal enterotoxins (SEs) or enterotoxin-like proteins (SEls). SAgs can cause staphylococcal food poisoning and toxic shock syn

Staphylococcal scalded skin syndrome: potentiation by immunosuppression in mice; toxin-mediated exfoliation in a healthy adult.

Staphylococcal scalded skin syndrome, associated with exfoliative toxin produced by phage group II Staphylococcus aureus, has recently been reported in an adult receiving immunosuppressive therapy. To determine the effect of immunosuppression on the development of the staphylococcal scalded skin syndrome, experimental animals were treated with prednisolone, azathioprine, or a combination of both drugs utilizing the clinical isolate from the adult with scalded skin syndrome. The mean lethal dose and mean exfoliating dose were identical and were 6,000-fold lower in animals receiving both drugs o
