Designing Highly Potent Side-Chain Lactam-Bridged Cyclic Competence-Stimulating Peptide-Based Quorum-Sensing Modulators in Streptococcus oligofermentans.

Streptococcus oligofermentans, a Gram-positive bacterium found in the oral microbiome, shows promise as an oral probiotic for preventing dental caries. It exhibits a reverse correlation with Streptococcus mutans, a key caries-causing pathogen, likely due to its production of hydrogen peroxide, a process mediated by quorum sensing (QS). In this work, we set out to develop novel lactam-based cyclic analogues of the competence stimulating peptide (CSP) signal utilized by S. oligofermentans for QS activation. To this end, we first conducted a ring position scan, where we determined the best positi…

Effects of caffeic acid phenethyl ester against multi-species cariogenic biofilms.

Dental caries is a biofilm-related disease, widely perceived to be caused by oral ecological imbalance when cariogenic/aciduric bacteria obtain an ecological advantage. Compared with planktonic bacteria, dental plaques are difficult to remove under extracellular polymeric substance protection. In this study, the effect of caffeic acid phenethyl ester (CAPE) on a preformed cariogenic multi-species biofilm was evaluated, which was comprised of cariogenic bacteria (Streptococcus mutans), commensal bacteria (Streptococcus gordonii), and a pioneer colonizer (Actinomyces naeslundii). Our result reve…

An electrochemical sensing platform for early prediction of dental caries.

Dental caries, with a global prevalence of 29 %, represents a major public health challenge. Traditional diagnostic methods often delay intervention, leading to irreversible tooth damage. In this study, we present a saliva-based electrochemical sensing platform for the early prediction of dental caries through the simultaneous detection of three key biomarkers: Streptococcus mutans, Ca2+, and pH. A nanocomposite electrode, constructed from Prussian blue, metal-organic framework, polypyrrole, and carboxylated multi-walled carbon nanotubes, provided a highly active electrochemical surface with r…

SloR-SRE binding to the S. mutans mntH promoter is cooperative.

UNLABELLED: Streptococcus mutans is a commensal member of the plaque microbiome. It is especially prevalent when dietary sugars are available for S. mutans fermentation, generating acid byproducts that lower plaque pH and foster tooth decay. S. mutans can survive in the transient conditions of the mouth, in part because it can regulate the uptake of manganese and iron during periods of feast when metal ions are available, and famine when they are limited. S. mutans depends on a 25kDa metalloregulatory protein, called SloR, to modulate the uptake of these cations across the bacterial cell surfa…

Synergistic Antibacterial Strategy of AIEgens Photosensitizer CPTQ for Photodynamic Therapy and Ferroptosis-like Bacterial Death to Prevent Dental Caries.

Dental caries, primarily caused by Streptococcus mutans (S. mutans) biofilms, remains a global health concern. Antimicrobial photodynamic therapy (aPDT) has shown promise in controlling cariogenic bacteria by generating reactive oxygen species (ROS), but the aggregation-caused quenching (ACQ) effect of traditional photosensitizers limits its efficacy. In this study, we developed a novel aggregation-induced emission (AIE) photosensitizer, CPTQ, with an enhanced photodynamic antibacterial efficacy and intrinsic antibacterial properties. CPTQ exhibited superior ROS generation, effective bacterial…

Biofilm ecology associated with dental caries: understanding of microbial interactions in oral communities leads to development of therapeutic strategies targeting cariogenic biofilms.

A biofilm is a sessile community characterized by cells attached to the surface and organized into a complex structural arrangement. Dental caries is a biofilm-dependent oral disease caused by infection with cariogenic pathogens, such as Streptococcus mutans, and associated with frequent exposure to a sugar-rich diet and poor oral hygiene. The virulence of cariogenic biofilms is often associated with the spatial organization of S. mutans enmeshed with exopolysaccharides on tooth surfaces. However, in the oral cavity, S. mutans does not act alone, and several other microbes contribute to cariog…
