Decreased plasma tryptophan concentration in major depression: relationship to melancholia and weight loss.

Plasma total tryptophan (TRP) concentration was significantly lower in 31 patients with major depression compared to a healthy control group. The ratio of plasma TRP concentration to that of other branch chain amino acids (the TRP:BCAA ratio) was also decreased. Further analysis revealed that the decrease in plasma TRP and TRP:BCAA ratio was most apparent in patients with major depression and melancholia. Overall, women but not men had significantly decreased plasma tryptophan concentrations, perhaps because of a contributory effect of weight loss; this latter effect, however, could not be dis…

Enzymatic studies on tryptophan metabolism disorder in rats chronically exposed to carbon disulfide.

Eight-week-old female Wistar rats were exposed to carbon disulfide for 6 hr a day, 5 days a week, for 12 weeks in inhalation chambers. Then the activities of the main enzymes of tryptophan metabolism (i.e., L-tryptophan 2,3-dioxygenase, indoleamine 2,3-dioxygenase, kynurenine 3-hydroxylase, kynureninase, and kynurenine aminotransferases) in their tissues were determined. The results showed that exposure to carbon disulfide caused a significant increase in the activities of kynureninase and kynurenine-2-oxoglutarate aminotransferase in the kidneys, but only a slight increase in their activities…

Tryptophan loading may reverse tolerance to opiate analgesics in humans: a preliminary report.

Five patients on chronic opiate medication to treat low-back and leg pain were determined to have developed opiate tolerance on the basis of their failure to obtain significant relief (rated on a subjective pain scale and by the degree of straight leg-raising they were able to endure) after receiving 30 mg of morphine administered i.v. in divided doses over 35 min. After these patients' diets had been supplemented with 4 g/day of L-tryptophan for 2-9 weeks, they achieved significant relief from pain when the opiate tolerance test was re-administered, and were able to lead more active lives whi…

Effect of diet restriction on some key enzymes tryptophan-NAD pathway in rats.

Dietary intake of rats was restricted by feeding varying amounts of a 20% protein diet. After 6 weeks of feeding, some key enzymes of the tryptophan and nicotinic acid-NAD pathway, liver nicotinamide nucleotide concentration, and urinary metabolites of tryptophan and nicotinic acid were studied. With an increase in diet restriction, liver tryptophan oxygenase (EC 1.13.1.12) activity increased. Quinolinate phosphoribosyltransferase (EC 2.4.2.a) activity, on the other hand, was found to decrease with moderate diet restriction up to 50% restriction, but increased again with more severe diet restr…

C-mannosyl tryptophan dynamics in a mouse model of the peritoneal dissemination of ovarian cancer.

C-Mannosyl tryptophan (C-Man-Trp), a unique monomeric glycosyl amino acid, is up-regulated in the blood of ovarian cancer patients; however, the underlying mechanisms remain unclear. In the present study, C-Man-Trp production and its dynamics were investigated in female B6C3F1 mice transplanted with mouse ovarian cancer OV2944-HM-1 (HM-1) cells. After transplantation, C-Man-Trp levels increased in the plasma, urine, ascites, peritoneal exudate cells (PECs), and tumor masses of mice. Furthermore, changes in the transcriptional expression of C-Man-Trp metabolism-related genes, C-mannosyltransfer…

Gut microbiota and the tryptophan-kynurenine pathway in anxiety: new insights and treatment strategies.

Anxiety disorders are mental health disorders characterized by long-lasting fear, worry, nervousness, and alterations in gut microbiota (GM). The GM is a vital modulator of brain function through the gut-brain axis, which acts as the neural pathway between the central and peripheral nervous systems. Dysbiosis of GM plays an essential role in anxiety development because of alterations in the vagus nerve, increased intestinal permeability, and altered breakdown of tryptophan (TRP). The Kynurenine (KYN) pathway plays a crucial role in the pathogenesis of anxiety disorders, primarily through its n…

Tryptamine accumulates in cheese mainly via the decarboxylation of tryptophan by lactic acid bacteria.

Few microorganisms are known to decarboxylate L-tryptophan, thereby producing tryptamine, a neuromodulator biogenic amine (BA) that can accumulate in cheese. Since lactic acid bacteria (LAB) are largely responsible for the production of other BA in this product, it was hypothesised that they would also be the main agents of tryptamine production. Using a rapid test based on the natural fluorescence of tryptamine, thousands of bacterial isolates from several cheese samples were screened. Only 1.4% of all isolates (43 out of 2982) were able to synthesise tryptamine and secrete it into the cultur…

Toxicology and safety study of L-tryptophan and its impurities for use in broiler feed.

L-tryptophan has been utilized as a feed additive in animal nutrition to improve growth performance, as well as a dietary supplement to alleviate various emotional symptoms in humans. Despite its benefits, concerns regarding its safety arose following the outbreak of eosinophilia-myalgia syndrome (EMS) among individuals who consumed L-tryptophan. The causative material of EMS was determined to be not L-tryptophan itself, but rather L-tryptophan impurities resulting from a specific manufacturing process. To investigate the effect of L-tryptophan and its impurities on humans who consume meat pro…
