Developing protocols for recombinant adeno-associated virus-mediated gene therapy in space.

With the advent of the era of International Space Station (ISS) and Mars exploration, it is important more than ever to develop means to cure genetic and acquired diseases, which include cancer and AIDS, for these diseases hamper human activities. Thus, our ultimate goal is to develop protocols for gene therapy, which are suitable to humans on the earth as well as in space. Specifically, we are trying to cure the hemoglobinopathies, beta-thalassemia (Cooley's anemia) and sickle cell anemia, by gene therapy. These well-characterized molecular diseases serve as models for developing ex vivo gene

Sympathetic outflow to muscle in humans during short periods of microgravity produced by parabolic flight.

We have investigated the changes in muscle sympathetic nerve activity (MSNA) from the tibial nerve during brief periods of microgravity (microG) for approximately 20 s produced by parabolic flight. MSNA was recorded microneurographically from 13 quietly seated human subjects with their knee joints extended in a jet aircraft simultaneously with the electrocardiogram, the blood pressure wave (measured with a Finapres), the respiration curve, and the thoracic fluid volume (measured by impedance plethysmography). During quiet and seated parabolic flight, MSNA was activated in hypergravity and was

Microgravity tissue engineering.

Tissue engineering studies were done using isolated cells, three-dimensional polymer scaffolds, and rotating bioreactors operated under conditions of simulated microgravity. In particular, vessel rotation speed was adjusted such that 10 mm diameter x 2 mm thick cell-polymer constructs were cultivated in a state of continuous free-fall. Feasibility was demonstrated for two different cell types: cartilage and heart. Conditions of simulated microgravity promoted the formation of cartilaginous constructs consisting of round cells, collagen and glycosaminoglycan (GAG), and cardiac tissue constructs

Eccentric contraction-induced injury in normal and hindlimb-suspended mouse soleus and EDL muscles.

The primary objective of this study was to compare the magnitude of injury in mouse extensor digitorum longus (EDL) and soleus muscles induced by high-force eccentric contractions. A second objective was to study the effect of altering the daily loading of the muscles through hindlimb suspension (HS) on the injury. One of two protocols was performed in vitro: 1) 15 eccentric contractions (n = 20: 10 EDL and 10 soleus muscles) or 2) 15 isometric contractions (n = 20: 10 EDL and 10 soleus muscles). After the protocol, the decrements in contractile performance and lactate dehydrogenase (LDH) rele

The effect of space flight on human cellular immunity.

T-lymphocyte responsiveness to mitogens is depressed by an average of 56% in humans (129 subjects) tested during and after space flight. Although there is not yet conclusive evidence of a clinical significance of the test, it is clear that factors of space flight like stress, closed environment and cosmic radiation may affect immune responsiveness. The data obtained from space crews may be compared to the depression seen in subjects undergoing heavy physical stress of head down tilt bedrest. Recently, delayed hypersensitivity [correction of hypersensivity] response was tested on crews of the U

[Clinical and experimental use of Cogimir].

This paper introduces 3 applications of a computer-based methodology for the assessment of cognitive abilities which has been developed during the Cogimir space project: an experimental study comparing gender-related lexical decision abilities, an investigation of hemisphere-specific reading in a subject with a partial split-brain syndrome, and a follow-up study measuring the effect of rehabilitation in a patient with frontal brain damage. Computerized tasks have several advantages over conventional neurobehavioral measurement procedures, among them exact stimulus and response control, the abi

Effects of head-down tilt for 10 days on the compliance of the leg.

The purpose of this investigation was to measure lower leg compliance before, during and after a 10-day period of bedrest at head-down tilt to test the hypothesis that leg compliance and the capacity for venous pooling is increased by the adaptation to stimulated microgravity. Venous occlusion plethysmography with multiple proximal occlusion pressures was used to obtain compliance measurements in six male subjects. Calf circumference decreased significantly during the tilt (corresponding to a decrease in cross sectional area of 7%) and had not returned to baseline seven days after the end of t

[Activation of respiratory sinus arrhythmia in monkeys during space flight].

During the first day in space the rhesus monkey, called Vernyi, flown on Cosmos-1667 showed higher values of the daytime, night, and daily respiratory sinus arrhythmia (RSA) when compared to those observed before launch and before or the first day of a simulation study. During orbital flight, the mean daily value of RSA was in the range 13.08-16.00 beats/min. In the simulation study, this parameter increased from 4.68 beats/min during the first day to 15.16 beats/min on the 6th day. The mean RSA values during days 1 through 3 were significantly higher in real than in simulation flight. Analysi

Neurosciences research in space: future directions.

Future research in the neurosciences can best be understood in the context of NASA's life sciences goals in the near term (1990-95), mid term (1995-2000), and long term (2000 and beyond). Since NASA is planning short-duration Spacelab and International Microgravity Laboratory (IML) flights for many years to come, the acute effects of exposure to microgravity will continue to be of experimental and operational interest in the near term. To this end, major new areas of research will be devoted to ground-based studies of preflight adaptation trainers and their efficacy in preventing or reducing t

[The C-cell system of the thyroid in rats following a flight on the Kosmos 1667 biosatellite].

Histological, electron-microscopic and morphometric investigations of the thyroid gland of Wistar SPF male rats (aged 3 months) flown for 7 days on Cosmos-1667 showed that its parenchyma was functionally active and changed but little as compared to the controls. However, at an acute stage of adaptation to microgravity C-cells showed morphological signs of their functional decline: the number of low activity cells and cells whose cytoplasm contained secretory granules increased, the volume of nuclei decreased significantly (by 16.2% as compared to the control), and dystrophic changes seen ultra

Some results of the effect of space flight factors on Drosophila melanogaster.

Two experiments with Drosophila melanogaster males were performed aboard the Salyut 6 orbital station. Mutagenic effects of a 8 day space flight on sex chromosome nondisjunction and intergene recombination in chromosome II were studied. The space flight factors (SFF) increased the frequency of chromosome nondisjunction and recombination. The model experiments showed that the combined effects of vibration and acceleration do not cover the whole spectrum of space flight mutagenic factors. These data suggest that heavy space ions are mainly responsible for the observed effect.

The prevention of motion sickness in orbital flight.

A question has arisen whether zero gravity qualifies as just another "motion environment" in which motion sickness may be elicited, or if one or more undetermined etiological factors are present which render such terms as "motion environment" and "motion sickness" inappropriate. The question is of more than academic interest for the reason that in the forthcoming Shuttle Program we have either one problem or two: one if we are concerned solely with prevention, but two if we must tackle the problem of covert etiological factors as well. This problem points up the need to define motion sickness

Neuroprotective and neuromodulatory role of agmatine in mitigating simulated microgravity-induced cognitive and behavioral deficits in rats.

Extended periods of microgravity during orbital flights can impair astronauts' cognitive abilities, including learning and memory, posing a persistent health concern in the field of aerospace medicine. The study examined the pharmacological effects of agmatine and its influence on simulated neurobehavioral changes in rats under microgravity conditions. Rats were exposed to simulated microgravity (SMG) conditions using the hindlimb unloading (HU) model for 28 days and evaluated for behavioural alterations using the open field test, elevated plus maze, and forced swim test, and cognitive deficit

Evaluation of Acquired Antibiotic Resistance in Escherichia coli Exposed to Long-Term Low-Shear Modeled Microgravity and Background Antibiotic Exposure.

The long-term response of microbial communities to the microgravity environment of space is not yet fully understood. Of special interest is the possibility that members of these communities may acquire antibiotic resistance. In this study, Escherichia coli cells were grown under low-shear modeled microgravity (LSMMG) conditions for over 1,000 generations (1000G) using chloramphenicol treatment between cycles to prevent contamination. The results were compared with data from an earlier control study done under identical conditions using steam sterilization between cycles rather than chloramphe

Changes in multifidus and abdominal muscle size in response to microgravity: possible implications for low back pain research.

PURPOSE: In microgravity, muscle atrophy occurs in the intrinsic muscles of the spine, with changes also observed in the abdominal muscles. Exercises are undertaken on the International Space Station and on Earth following space flight to remediate these effects. Similar effects have been seen on Earth in prolonged bed rest studies and in people with low back pain (LBP). The aim of this case report was to examine the effects of microgravity, exercise in microgravity and post-flight rehabilitation on the size of the multifidus and antero-lateral abdominal muscles. METHODS: Ultrasound imaging w
