Genome-wide analysis of the nucleus accumbens identifies DNA methylation signals differentiating low/binge from heavy alcohol drinking.

Alcohol-use disorders encompass a range of drinking levels and behaviors, including low, binge, and heavy drinking. In this regard, investigating the neural state of individuals who chronically self-administer lower doses of alcohol may provide insight into mechanisms that prevent the escalation of alcohol use. DNA methylation is one of the epigenetic mechanisms that stabilizes adaptations in gene expression and has been associated with alcohol use. Thus, we investigated DNA methylation, gene expression, and the predicted neural effects in the nucleus accumbens core (NAcc) of male rhesus macaq

Alcohol-induced impairment in adolescents admitted to inpatient treatment after heavy episodic drinking: effects of age and gender.

OBJECTIVE: In Germany and many other countries, the number of adolescent heavy episodic drinking-induced hospital admissions (HEDHA) in pediatric units markedly increased during the past decade. A low level of response to alcohol in young adults is associated with high risk for later development of alcohol use disorders (AUDs). METHOD: We performed a retrospective chart review of all 1,123 HEDHA cases in adolescents aged 11-17 years who were admitted to one of the pediatric inpatient units covering the cities of Dresden, Pirna, and Rostock, Germany, between 2000 and 2008. Blood alcohol concen

Attempted Training of Alcohol Approach and Drinking Identity Associations in US Undergraduate Drinkers: Null Results from Two Studies.

There is preliminary evidence that approach avoid training can shift implicit alcohol associations and improve treatment outcomes. We sought to replicate and extend those findings in US undergraduate social drinkers (Study 1) and at-risk drinkers (Study 2). Three adaptations of the approach avoid task (AAT) were tested. The first adaptation - the approach avoid training - was a replication and targeted implicit alcohol approach associations. The remaining two adaptations - the general identity and personalized identity trainings - targeted implicit drinking identity associations, which are rob

Stress moderates the effect of childhood trauma and adversity on recent drinking in treatment-seeking alcohol-dependent men.

OBJECTIVE: This study sought to clarify the relationship between childhood trauma and adversity with later alcohol consumption and the moderating effects of adult psychosocial stress. METHOD: Seventy-seven recently abstinent alcohol-dependent men attending residential treatment programs were assessed. Childhood trauma/adversity was assessed with the Childhood Trauma Questionnaire (CTQ), drinks per drinking day (DDD) with the Timeline Follow Back, and chronic psychosocial stress with the UCLA Stress Interview. Drinking and stress were retrospectively assessed for 6 months prior to the present

Treating individuals with social anxiety disorder and at-risk drinking: phasing in a brief alcohol intervention following paroxetine.

Paroxetine alone is not sufficient to decrease alcohol use in socially anxious alcoholics seeking anxiety treatment. We tested the hypothesis that adding a brief-alcohol-intervention (BI) to paroxetine would decrease alcohol use. All subjects (N=83) had a diagnosis of social anxiety disorder, endorsed drinking to cope with anxiety, were NIAAA-defined at-risk drinkers, and were randomized to either paroxetine alone, or paroxetine plus BI. Both groups showed significant improvement in both social anxiety severity (F(5,83)=61.5, p<0.0001) and drinking to cope (e.g. F(4,79)=23, p<0.0001) and these

Where the individual meets the ecological: a study of parent drinking patterns, alcohol outlets, and child physical abuse.

BACKGROUND: Despite well-known associations between heavy drinking and child physical abuse, little is known about specific risks related to drinking different amounts of alcohol in different drinking venues. This study uses a context-specific dose-response model to examine how drinking in various venues (e.g., at bars or parties) is related to physically abusive parenting practices while controlling for individual and psychosocial characteristics. METHODS: Data were collected via a telephone survey of parents in 50 cities in California, resulting in 2,163 respondents who reported drinking in

Does family history of alcohol problems influence college and university drinking or substance use? A meta-analytical review.

AIMS: Family history of alcohol use problems is a reliable determinant of alcohol use and problems in the population at large, but findings are inconsistent when this issue is examined in college and university students. No quantitative summary of this literature has been reported to date. The purpose of this study was to conduct a meta-analysis on the effects of family history on substance use and abuse in college and university students. METHODS: A two-group contrast meta-analysis was conducted to evaluate the differences in substance use and abuse between family history-positive and -negat

Permanent impairment of birth and survival of cortical and hippocampal proliferating cells following excessive drinking during alcohol dependence.

Experimenter-delivered alcohol decreases adult hippocampal neurogenesis and hippocampal-dependent learning and memory. The present study used clinically relevant rodent models of nondependent limited access alcohol self-administration and excessive drinking during alcohol dependence (alcohol self-administration followed by intermittent exposure to alcohol vapors over several weeks) to compare alcohol-induced effects on cortical gliogenesis and hippocampal neurogenesis. Alcohol dependence, but not nondependent drinking, reduced proliferation and survival in the medial prefrontal cortex (mPFC).

Markers of oxidative stress and systemic vasoconstriction in pregnant women drinking > or =48 g of alcohol per day.

BACKGROUND: The precise pathway by which alcohol causes the characteristic features of fetal alcohol spectrum disorders is unknown. Proposed mechanisms for fetal injury from maternal alcohol use include cellular damage from oxidative stress and impaired fetal oxygenation related to maternal systemic vasoconstriction. Our objective was to compare the levels of urinary markers of oxidative stress and systemic vasoconstriction between women consuming large amounts of alcohol during pregnancy and women who did not drink alcohol during pregnancy. METHODS: Pregnant women consuming > or =48 g alcoho

Calcium channel agonist (-)-BAY k 8644 suppresses free and limited access intake of alcohol in genetic drinking rats.

Drugs which possess selective actions on a given voltage operated calcium (Ca2+) channel (VOCC) are reportedly involved in the pharmacological actions of alcohol. Recently it was shown that the 1,4-dihydropyridine (-)-BAY k 8644, an L-type VOCC agonist, reduces alcohol intake relatively selectively in the genetic drinking AA rat. This study determined whether (-)-BAY k 8644 would alter volitional alcohol drinking in two other genetic models of alcoholism, male P rats and a new strain of male and female high ethanol preferring (HEP) rats. By use of a standard 10-day preference test for water ve…

Prenatal Stress and Prenatal Alcohol Alter the Adult Dopamine System and Alcohol Consumption: Dopamine Drives Drinking Behavior in a Prospective 20-Year Longitudinal Experiment with Rhesus Macaques.

Given the prevalence of alcohol use and stress during pregnancy, we examined the effects in offspring of prenatal alcohol and stress on the dopamine system and drinking behavior in a primate model. In a 20-year prospective longitudinal experiment, we studied alcohol-naive adult rhesus monkeys of both sexes bred from mothers randomly assigned during pregnancy to consume moderate alcohol, be exposed to mild stress, both, or neither. Positron emission tomography (PET) was used to measure dopamine D2-type receptor (D2) and transporter (DAT) availability in substantia nigra/ventral tegmental area (

Optimizing the Measurement of Information on the Context of Alcohol Consumption Within the Drink Less App Among People Drinking at Increasing and Higher Risk Levels: Mixed-Methods Usability Study.

BACKGROUND: There is a growing public health evidence base focused on understanding the links between drinking contexts and alcohol consumption. However, the potential value of developing context-based interventions to help people drinking at increasing and higher risk levels to cut down remains underexplored. Digital interventions, such as apps, offer significant potential for delivering context-based interventions as they can collect contextual information and flexibly deliver personalized interventions while addressing barriers associated with face-to-face interventions, such as time constr

Youth Involvement in Family Drinking and Parental Permission to Sip/Taste Alcohol: A Test of an Early Risk Pathway to Perceived Parental Approval of Underage Alcohol Use.

BACKGROUND: Several alcohol-specific parenting behaviors are associated with adolescent alcohol use. An important next step in the literature is integrating these behaviors into unified risk pathways, which have the potential to inform preventive interventions and public health messaging. Youth involvement in family drinking (i.e., asking children to fetch, pour, and open drinks) reflects parental-approved contact with alcohol, which may present the opportunity for sipping/tasting alcohol with parental permission. In turn, sipping/tasting alcohol with parental permission may increase adolescen

Circadian clock genes Bmal1 and Per2 in the nucleus accumbens are negative regulators of alcohol-drinking behavior in mice.

Alcohol use disorder (AUD) is a leading cause of morbidity and mortality worldwide, characterized by cycles of heightened craving and excessive, uncontrolled consumption of alcohol despite progressive decline in physical and mental health. Voluntary alcohol consumption is influenced by a variety of environmental and genetic factors, including circadian clock genes, whose effects are modulated in a sex-specific manner. The sex-specific role of clock genes in alcohol drinking was identified through selective ablation of Bmal1 and Per2 from neurons of the mouse striatum; however, the contribution…

Evaluating effects of feedback on college students' self-reports of alcohol consumption and standard drink free-pours.

Excessive alcohol consumption is a concern on college campuses, yet most data are gathered from self-report surveys, which require college students to identify standard drink sizes. Unfortunately, they typically cannot define or pour standard drinks, rendering the reliability and validity of these data questionable. Researchers have suggested the provision of feedback to college students might improve these skills. However, evidence for this is limited by lack of repeated measures, little evaluation of the effects of feedback on observable behavior (e.g., free-pour), and aggregate data. The cu
