Neuronal cell cultures as a model for assessing neurotoxicity induced by encephalitic viruses.

Primary dispersed and organotypic cultures were prepared from selected brain areas and spinal cords of rat (Sprague-Dawley) and mouse (SJL/OLA(F) Ness-Ziona) fetuses and neonates. Following fiber regeneration, synapse formation and myelination, cultures were infected with one of the following viruses: Rabies CVS-21 strain, Sindbis Alphavirus, West-Nile Flavivirus and Theiler Murine Encephalomyelitis virus. Light and electron microscopical studies showed clear differences in the target cells for virus infection; time of viral replication and in the intensity and specificity of the cytopathic ef…

[Reproduction and interferogenic activity of togaviruses].

A comparative study of the correlation between reproduction and the interferon-inducing activity of viruses in chick embryo fibroblast cultures was carried out with members of different groups of togaviruses: alphavirus (Venezuelan equin encephalomyelitis viru, VEE) and flavivirus (Saint Louis encephalitis virus, SLE). The correspondence between cycles of accumulation of intracellular and extracellular viruses and the dynamics of interferon production the synthesis of which began early in the stage of exponential virus growth and correlated with the dynamics of their reproduction, was determin…

Detection of persistent viremia by a live attenuated vaccine strain of the chikungunya virus in a blood donor.

We report here the case of a blood donor who tested positive for chikungunya RNA by nucleic acid testing introduced in Guadeloupe (a French island in the Caribbean) to prevent the transmission of arboviral infections by transfusion. The strain was isolated, and further investigations revealed the presence of the IXCHIQ live attenuated vaccine strain in blood 1 month after vaccination.

A high throughput antiviral screening platform for alphaviruses based on Semliki Forest virus expressing eGFP reporter gene.

Alphaviruses, which contain a variety of mosquito-borne pathogens, are important pathogens of emerging/re-emerging infectious diseases and potential biological weapons. Currently, no specific antiviral drugs are available for the treatment of alphaviruses infection. For most highly pathogenic alphaviruses are classified as risk group-3 agents, the requirement of biosafety level 3 (BSL-3) facilities limits the live virus-based antiviral study. To facilitate the antiviral development of alphaviruses, we developed a high throughput screening (HTS) platform based on a recombinant Semliki Forest vi…

Virus-specific thermostability and heat inactivation profiles of alphaviruses.

Serological diagnosis is a critical component for disease surveillance and is important to address the increase in incidence and disease burden of alphaviruses, such as the chikungunya (CHIKV) and Ross River (RRV) viruses. The gold standard for serological diagnosis is the plaque reduction neutralization test (PRNT), which demonstrates the neutralizing capacity of serum samples after the removal of complement activity and adventitious viruses. This procedure is normally performed following inactivation of the virus at 56°C for 30min. Although this protocol has been widely accepted for the inac…

Differential proteome analysis of chikungunya virus infection on host cells.

BACKGROUND: Chikungunya virus (CHIKV) is an emerging mosquito-borne alphavirus that has caused multiple unprecedented and re-emerging outbreaks in both tropical and temperate countries. Despite ongoing research efforts, the underlying factors involved in facilitating CHIKV replication during early infection remains ill-characterized. The present study serves to identify host proteins modulated in response to early CHIKV infection using a proteomics approach. METHODOLOGY AND PRINCIPAL FINDINGS: The whole cell proteome profiles of CHIKV-infected and mock control WRL-68 cells were compared and a…

Chikungunya: a potentially emerging epidemic?

Chikungunya virus is a mosquito-borne emerging pathogen that has a major health impact in humans and causes fever disease, headache, rash, nausea, vomiting, myalgia, and arthralgia. Indigenous to tropical Africa, recent large outbreaks have been reported in parts of South East Asia and several of its neighboring islands in 2005-07 and in Europe in 2007. Furthermore, positive cases have been confirmed in the United States in travelers returning from known outbreak areas. Currently, there is no vaccine or antiviral treatment. With the threat of an emerging global pandemic, the peculiar problems

Prophylaxis and therapy for Chikungunya virus infection.

BACKGROUND: Chikungunya virus (CHIKV) is a recently reemerged arbovirus responsible for a massive outbreak of infection in the Indian Ocean region and India that has a very significant potential to spread globally because of the worldwide distribution of its mosquito vectors. CHIKV induces a usually self-limited disease in humans that is characterized by fever, arthralgia, myalgia, and rash; however, cases of severe CHIKV infection have recently been described, particularly in adults with underlying condition and neonates born to viremic mothers. METHODS: Human polyvalent immunoglobulins were

[Chikungunya in paediatrics: epidemic of 2005-2006 in Saint-Denis, Reunion Island].

UNLABELLED: During the southern-hemispheric 2005-2006 summer, Reunion Island was struck by an epidemic of Chikungunya (Chik), which affected more than a third of the overall population. OBJECTIVES: Our objective was to describe pediatric cases of Chick. MATERIAL AND METHODS: We conducted a retrospective descriptive monocentric study of confirmed pediatric cases of Chik recruited at Saint-Denis' departmental hospital during the peak of the epidemic (January 1st to April 30th 2006). RESULTS: Eighty-six children aged 10 days to 18 years were included. In addition to the typical clinical presen

Rapid infection of salivary glands in Culiseta melanura with eastern equine encephalitis virus: an electron microscopic study.

Transmission electron microscopy was used to determine if eastern equine encephalitis (EEE) virus infects and replicates in the salivary glands of Culiseta melanura after 3 days of extrinsic incubation (EI). The Cs. melanura studied were from a colony strain, were orally infected, and had EI periods of 55-69 hours. Both naked nucleocapsids and enveloped virions were present in aggregates, suggestive of viral replication, within salivary gland acinar cells. Nucleocapsids were present in the cytoplasm below the plasma membrane that lined apical cavities. Enveloped virions occurred in the salivar

Venezuelan and western equine encephalitis virus E1 liposome antigen nucleic acid complexes protect mice from lethal challenge with multiple alphaviruses.

Eastern, Venezuelan and western equine encephalitis viruses (EEEV, VEEV, and WEEV) are mosquito-borne viruses that cause substantial disease in humans and other vertebrates. Vaccines are limited and current treatment options have not proven successful. In this report, we vaccinated outbred mice with lipid-antigen-nucleic acid-complexes (LANACs) containing VEEV E1+WEEV E1 antigen and characterized protective efficacy against lethal EEEV, VEEV, and WEEV challenge. Vaccination resulted in complete protection against EEEV, VEEV, and WEEV in CD-1 mice. Measurements of bioluminescence and plaque red

Engineering drug-responsive replication machinery for precise control of self-amplifying RNA.

Precise, reversible control of gene expression from self‑amplifying RNA (saRNA) remains difficult, limiting the therapeutic flexibility of this otherwise potent platform. Although alphavirus‑derived saRNAs encode non‑structural proteins that drive RNA replication and offer an intrinsic regulatory point, no existing approach has enabled direct, drug‑dependent control of this machinery for high‑fidelity modulation of expression. Here we engineer saRNA constructs whose replication is activated by the approved small‑molecule drug trimethoprim, using drug‑responsive degradation domains fused to ind…

A WEEV 6K viroporin activates MLKL-dependent necroptosis to drive inflammatory injury.

Neurotropic alphavirus infection is characterized by neuronal injury and sustained neuroinflammation, yet the viral determinants linking membrane perturbation to host inflammatory damage remain poorly defined. Here, we show that the Western equine encephalitis virus (WEEV) 6K protein functions as an ER-localized viroporin, displaying stable single-channel activity with broad cation permeability. Consistent with its ion channel activity, 6K expression perturbs intracellular Ca²⁺ store homeostasis and is associated with cellular injury phenotypes. Mechanistically, 6K predominantly engages MLKL-m…

Chlorinated biscoumarins inhibit chikungunya virus replication in cell-based and animal models.

Biscoumarin derivatives were evaluated for antiviral activity against chikungunya virus (CHIKV), a re-emerging mosquito-borne alphavirus with no approved treatment. Compounds 3 and 4 demonstrated potent in vitro antiviral efficacy, with EC₅₀ values of 2.85 ± 0.42 µM and 3.08 ± 0.45 µM (SI > 20) for compound 3 in Vero and HEK293 cells, respectively. Compound 4 showed comparable potency in Vero cells but was less effective in HEK293 cells. Time-of-addition and replicon assays suggested that both compounds act at a post-entry step, likely inhibiting viral RNA replication. In vivo, a single oral d…
