Refugia-based strategies associated with a combination of anthelmintic drugs belonging to different drug classes are becoming more common management practices to mitigate anthelmintic resistance (AR) in gastrointestinal nematodes (GIN) in small ruminants. Though refugia-based strategies have been largely demonstrated in small ruminants, cattle veterinarians and producers are considering such management strategies in grazing cattle production systems. Implementing refugia-based strategies lowers the amount of anthelmintics used in the herd and therefore slows the progression of AR by allowing a…

The safety and efficacy of benzimidazole anthelmintics for the treatment of rat lungworm disease (neuroangiostrongyliasis) have been questioned regardless of numerous experimental animal studies and clinical reports. In this review, 40 of these experimental animal studies and 104 clinical reports are compiled with a focus on albendazole. Among the 144 articles involving an estimated 1034 patients and 2561 animals, 4.1% were inconclusive or vague regarding the use of benzimidazoles. Of the remaining 138 articles, 90.5% found benzimidazoles to be safe and effective (885 patients, 2530 animals), …

This guideline is aimed at those who are involved in the assessment of anthelmintic efficacy in ruminant livestock species (bovine, ovine and caprine). The intent is to provide a framework that can be adopted worldwide for the testing of anthelmintics in ruminants, such that studies carried out in different countries can be compared and thereby unnecessary duplication can be reduced. Recommendations are made for the selection, housing and feeding of study animals, the type of studies required, the method used to conduct those studies, the assessment of results and the standards for defining an…

In the last decade there have been numerous reports of anthelmintic resistant cyathostomins in many parts of the world. The objective of the present study was to evaluate the efficacy of the commercially available anthelmintics against cyathostomin egg shedding in the Mid-Atlantic region of the United States. A total of 989 horses from 67 different farms located in southeastern Pennsylvania, northern Delaware, and northeastern Maryland were treated with fenbendazole, oxibendazole, pyrantel pamoate, ivermectin, or moxidectin at their recommended dosages. Fecal egg count reduction testing was us…

Seven dairy-goat farms, located in central, western France, were studied in order to assess the variability in susceptibility of the lungworm Muellerius capillaris first-stage larvae (L1) to 3 different anthelmintics in relation to farm origin, by means of motility tests. The motility tests were performed by mixing larval suspensions with pyrantel (PYR), thiabendazole (TBZ) or ivermectin (IVE) solutions (3 concentrations x 3 incubation durations). The same anthelmintic tests were repeated 7 and 21 d after febantel (probenzimidazole) treatment of the goats. Before the treatment of goats, the av…

About 1.5 billion people are infected with at least one of three soil-transmitted helminths (STHs) - roundworm, hookworm, and whipworm- which have devastating outcomes for growth, nutrition, cognition, and school attendance, trapping them in poverty. The WHO currently approves only two anthelmintics with only one mechanism of action for mass drug administration (MDA), and resistance has been documented. This calls for new drugs and new strategies to reduce worm burden. Previous research, beginning with larval stage 4 (L4) C. elegans as a model, used a health-rating system to score individual w…

BACKGROUND: The soil-transmitted nematodes (STNs) or helminths (hookworms, whipworms, large roundworms) infect the intestines of ~1.5 billion of the poorest peoples and are leading causes of morbidity worldwide. Only one class of anthelmintic or anti-nematode drugs, the benzimidazoles, is currently used in mass drug administrations, which is a dangerous situation. New anti-nematode drugs are urgently needed. Bacillus thuringiensis crystal protein Cry5B is a powerful, promising new candidate. Drug combinations, when properly made, are ideal for treating infectious diseases. Although there are s

We describe a case of anthelmintic resistance on one of the largest organic small ruminant farms in Denmark. The flock was established in 2007 by purchase of animals from other Danish farms and had history of clinical parasitism, high mortality of young stock and anthelmintic treatment failure. In October 2011, 40 lambs and 40 kids were selected for a faecal egg count reduction test (FECRT) with fenbendazole (FBZ), ivermectin (IVM), moxidectin (MOX) and levamisole (LEV). Lambs were treated with the recommended sheep dose of each product while kids received the sheep dose of IVM, 1.5× sheep dos

Helminth infections remain among the most prevalent neglected tropical diseases worldwide, with angiostrongyliasis representing an emerging … compound-specific effects on parasite larvae, supporting biological differentiation from reference anthelmintics. This activity was accompanied by a favorable selectivity profile, as no cytotoxic effects were observed in human and animal cell lines at concentrations well above those required for antiparasitic activity (CC50 >200 μM). Moreover, spathulenol did …

Gastrointestinal nematode (GIN) infections represent one of the most significant constraints to the health and productivity of small ruminants worldwide, leading to substantial economic losses and animal welfare concerns. Control of these parasites has historically relied on the extensive use of anthelmintic drugs; however, the rapid emergence and global spread of anthelmintic resistance (AR) have compromised their effectiveness. Resistance is now reported across all major classes of anthelmintics and is particularly prevalent in key species such as Haemonchus contortus. The development of AR …

This study evaluated the anthelmintic activity of a hydroalcoholic extract (HA-E) from Dalbergia palo-escrito aerial parts against Haemonchus contortus using a bioguided approach. The egg hatching inhibition (EHI) assay was conducted to determine activity. The HA-E was partitioned into an aqueous fraction (Aq-F) and an ethyl acetate fraction (EtOAc-F). The EtOAc-F was fractioned by column chromatography into seven subfractions (PeF1 to PeF7). HA-E and EtOAc-F achieved 100% EHI at 50 and 1.0 mg/mL, respectively. Subfractions PeF1, PeF3, and PeF4a exhibited strong activity, with EC90 values of 0

BACKGROUND: Mass deworming programs using anthelmintic drugs are a common public health strategy for controlling soil-transmitted helminth (STH) infections in high-risk, low-income countries. However, whether routine deworming induces long-term changes in gut microbiome composition remains unclear. We used longitudinal data to examine how anthelmintics alter the gut microbiome profiles of Ethiopian children.
METHODS: Fecal samples were collected from school-aged children enrolled in a mass deworming program in Jimma, Ethiopia, before and after receiving a 400 mg dose of albendazole. Baseline …

BACKGROUND: Senna alata is widely recognized for its wide range of therapeutic benefits, including its antioxidant activity. Our previous research demonstrated its non-toxic nature in mice and its modulatory effects on the antioxidant system of the zoonotic parasite Hymenolepis diminuta. However, understanding the mode of action of S. alata during active parasitic infection is essential for evaluating its antiparasitic potential. This study aims to explore the in vivo efficacy of alcoholic leaf extract of S. alata against H. diminuta infection in a rodent model, focusing on both antiparasitic

Parasitic helminths are a major constraint on livestock production worldwide, with significant implications for food security and animal and human health. As reliance on conventional anthelmintic drugs becomes increasingly unsustainable due to widespread anthelmintic resistance, chemical residues, and environmental concerns, there is an urgent need for innovative, biologically derived alternatives such as polyunsaturated fatty acids (PUFAs). This review evaluates PUFAs as biologically derived agents that could complement or partially replace conventional dewormers. We integrate molecular, cell
