Astrocytes expressing hyperphosphorylated tau protein without glial fibrillary tangles in argyrophilic grain disease.

Argyrophilic grain disease (AgD), a frequent type of late onset dementia, is characterized by the occurrence of Gallyas-stained neuropil gra … s in stellate cells. The high expression of CD44 indicate that they probably correspond to reactive astrocytes. Unlike astrocytic plaques in corticobasal degeneration (CBD), where AT8 reactivity is accumulating in distal astrocytic processes, tau reactivity in AgD was found in all astrocytic cell compartments. The absence of glial fibrillary tangles further …

Influence of nitric oxide on cellular and mitochondrial integrity in oxidatively stressed astrocytes.

Astrocytes provide protection and trophic support to neurons, but like neurons are vulnerable to oxidative stress. Decreased function of astrocytes resulting from oxidative stress could contribute to neurodegeneration. Our goal is to understand the intracellular events associated with oxidative stress in astrocytes. Because nitric oxide (NO) has been implicated as a contributor to oxidative stress in the brain, we examined in this study whether NO contributed to oxidative stress in astrocytes. Stimulation of NO decreases superoxide levels, preserves mitochondrial membrane potential, and decrea…

Taurine release is enhanced in cell-damaging conditions in cultured cerebral cortical astrocytes.

The release of preloaded [3H]taurine from cultured cerebral cortical astrocytes was studied under various cell-damaging conditions, including hypoxia, ischemia, aglycemia and oxidative stress, and in the presence of free radicals. Astrocytic taurine release was enhanced by K+ (50 mM), veratridine (0.1 mM) and the ionotropic glutamate receptor agonist kainate (1.0 mM). Metabotropic glutamate receptor agonists had only weak effects on taurine release. Similarly to the swelling-induced taurine release the efflux in normoxia seems to be mediated mainly by DIDS-(diisothiocyanostilbene-2,2'-disulpho…

Measles virus antigen in macrophage/microglial cells and astrocytes of subacute sclerosing panencephalitis.

In two patients with subacute sclerosing panencephalitis (SSPE) of 10 and 25 months duration we demonstrated by immunohistochemistry the presence of measles-virus nucleocapsid antigen (MVNA) in CD68+ cells and astrocytes of brain tissues. In both cases, CD68+ hematogenous monocyte/ macrophages and perivascular microglial cells (Mphi) were found infiltrating the brain parenchyma, and often partially or completely invested by perivascular reactive astrocytes expressing glial fibrillary acidic protein (GFAP). Mphi with cytoplasmic MVNA were often seen in the Virchow-Robin spaces and in close asso…

A monoclonal antibody that specifically recognizes a novel mitochondrial protein of human astrocytes.

We established a monoclonal antibody to human astrocytes using a human glial cell-rich fraction as the immunogen. The antibody, named PRAS-4, specifically labeled populations of astrocytes in a fine granular manner immunohistochemically. In formalin-fixed, paraffin-embedded tissue sections, PRAS-4-positive astrocytes were extensively distributed in the gray matter of the central nervous system, namely the cerebral cortex, basal ganglia, diencephalon, midbrain, various nuclei of the brain stem, cerebellar cortex and nuclei, and spinal cord. In the white matter, a few positive astrocytes were lo…

Polarity of processes with Golgi apparatus in a subpopulation of type I astrocytes.

The Golgi apparatus-complex (GA), is a key organelle involved in several posttranslational modifications of polypeptides destined for lysosomes, plasma membranes and secretion. As reported from this laboratory, certain astrocytes in rat brain contain cisternae of the GA not only in perikarya, but also in processes. In order to further investigate which type of astrocytes contain GA in processes we conducted the present study using primary cultures of rat astrocytes and organelle specific antibodies against the GA and the rough endoplasmic reticulum (RER). While the perikarya of all cells conta…

Immunohistochemical localization of glial fibrillary acidic protein (GFAP) in rat pineal stalk astrocytes.

In the present work, the presence and distribution of astrocytes in the rat pineal stalk is investigated applying an immunohistochemical technique for the demonstration of glial fibrillary acidic protein (GFAP) on Epon-embedded semithin sections (0.5 micron thick). GFAP-immunoreactive cells are evenly and regularly distributed along the entire pineal stalk. The GFAP-immunoreactive cells display a stellate shape showing variable numbers of cell processes that are mainly oriented parallel to the longitudinal stalk axis. Astrocytic processes show a clear tendency to encircle the remaining element…

Early loss of astrocytes in herpes simplex virus-induced central nervous system demyelination.

Immunohistochemistry was used to study herpes simplex virus type 1-induced central nervous system demyelination in the trigeminal root entry zone of mice inoculated with herpes simplex virus type 1 by the corneal route. There was no change in peripheral nervous system myelin as shown by immunostaining for P0 glycoprotein. Double immunoperoxidase staining for herpes simplex virus type 1 antigens and glial fibrillary acidic protein showed that most of the infected cells were astrocytes. Glial fibrillary acidic protein immunostaining was completely lost in the inferior medial portion of the trige…

Nicotine protects astrocytes expressing alpha-synuclein against aminochrome cytotoxicity: Implications for Parkinson's disease.

Astrocytes containing alpha-synuclein (α-Syn) are a cytopathological finding in post-mortem samples of patients with Parkinson's disease (PD). Aminochrome, a subproduct of dopamine oxidation, can induce formation of neurotoxic α-Syn oligomers, astrocyte reactivity, and astrocyte cell death. Nicotine, on the other hand, has been shown to have a protective effect against aminochrome cytotoxicity in substantia nigra dopaminergic cells. However, whether nicotine can also protect against aminochrome toxicity in α-Syn-expressing astrocytes is not known. To address this question, we used the human gl…

APOE4 and INPP5D converge on membrane mechanics to regulate endocytosis in human astrocytes.

Disrupted endocytosis is an early feature of Alzheimer's disease (AD), but how genetic risk factors functionally impact this pathway remains unclear. Using isogenic human iPSC-derived astrocytes, we show that the AD risk variant, APOE4, impairs clathrin-mediated membrane curvature, clathrin-mediated endocytosis, and alters plasma membrane lipid saturation and tension. Compared to APOE3 astrocytes, APOE4 cells display an accumulation of flat clathrin structures, reduced maturation of clathrin-coated pits, decreased early endosomes, and reduced endocytic uptake. We then identify the AD risk gene…

Characterizing the metabolomes of microglia, astrocytes and neurons in ageing and Alzheimer's brains.

Neurons and glia are distinct in their morphology, development and function, possessing unique transcriptomes and proteomes, but little is known about their metabolomes. The challenge of brain cell metabolic profiling is to obtain a large number of cells for reliable analysis. Here we purified microglia, astrocytes and neurons from mouse brains, identifying >70 metabolites through targeted metabolomics and 9,854 metabolite features via untargeted metabolomics. We systematically characterized cell type-enriched metabolites and metabolic pathways, revealing an enrichment of glutathione (GSH) and…

Mycotoxin Alternariol Induces Ca2+ Dysregulation and Cell Death in Astrocytes: Mechanistic Toxicological Insights.

Alternariol (AOH) is a low-molecular-weight mycotoxin produced by Alternaria species, frequently found in cereal grains, tomatoes, and processed foods. While its genotoxic and endocrine-disrupting effects are well established, emerging studies suggest a neurotoxic potential that remains poorly understood. Astrocytes-key regulators of neuronal support and calcium (Ca2+) balance in the central nervous system (CNS)-are an underexplored target of AOH toxicity. Here, we investigated the effects of AOH on Ca2+ signaling and viability in human astrocytes (Gibco Human Astrocyte, GHA cells). AOH exposu…

The α7-nicotinic acetylcholine receptor (α7nAChR) has driven extensive research over the past three decades for its pro-cognitive potential. … re most sensitive to schizophrenia-related cognitive impairments. By contrast, loss of α7nAChR from astrocytes produces profound behavioral alterations that are cognitive domain-specific, are time-of-day dependent, coincide with reduced levels of the N-methyl D-aspartate receptor (NMDAR) co-agonist D-serine, and are fully restored by D-serine supplementation …

Inhibition of endocannabinoid degradation in astrocytes reprograms glial reactivity and prevents seizure sequelae.

BACKGROUND: Temporal lobe epilepsy (TLE) is the most common form of focal epilepsy and is characterized by a pathological cascade of excitotoxicity that leads to neuroinflammation, progressive neuronal loss, and subsequent cognitive decline. Despite its prevalence, effective therapies remain lacking. Previous studies have demonstrated that the dysregulation of the endocannabinoid system contributes to epileptic activity. In particular, inactivation of monoacylglycerol lipase (MAGL), the key rate-limiting enzyme responsible for the degradation of the endocannabinoid 2-arachidonoylglycerol (2-AG

APOE3 astrocytes can rescue lipid abnormalities and dystrophic neurites of APOE4 human neurons.

Lipid abnormalities are emerging as key pathogenic mechanisms in neurodegenerative diseases, such as Alzheimer's disease, Parkinson's disease, and Lewy body dementia. Astrocytes in the brain provide apolipoprotein E (APOE) proteins and influence neuronal metabolism and health. Using live-cell imaging and objective neurite imaging techniques, we induced cellular lipid load (cholesterol and triglycerides) by inhibiting the lysosomal cholesterol transport protein NPC1 in human neuron-astrocyte cocultures and examined the effects of CRISPR-edited APOE3 and APOE4 human astrocytes on the rescue of d…

Regulatory mechanisms and research progress of astrocytes in the neuro-endocrine-immune network.

The neuro-endocrine-immune (NEI) network is a key regulatory network that maintains organismal homeostasis and participates in stress responses. Its function depends on the dynamic coupling among the nervous, endocrine, and immune systems. Astrocytes, as abundant and functionally diverse glial cells in the central nervous system, are increasingly recognized as important participants in the NEI network. Growing evidence indicates that astrocytes do not merely provide structural support in the traditional sense, but also exhibit marked regional heterogeneity and functional diversity. Through bro…
