X-ray crystallographic structure of BshB, the zinc-dependent deacetylase involved in bacillithiol biosynthesis.

Many gram-positive bacteria produce bacillithiol to aid in the maintenance of redox homeostasis and degradation of toxic compounds, including the antibiotic fosfomycin. Bacillithiol is produced via a three-enzyme pathway that includes the action of the zinc-dependent deacetylase BshB. Previous studies identified conserved aspartate and histidine residues within the active site that are involved in metal binding and catalysis, but the enzymatic mechanism is not fully understood. Here we report two X-ray crystallographic structures of BshB from Bacillus subtilis that provide insight into the Bsh

Bedforms as Biocatalytic Filters: A Pumping and Streamline Segregation Model for Nitrate Removal in Permeable Sediments.

Bedforms are a focal point of carbon and nitrogen cycling in streams and coastal marine ecosystems. In this paper, we develop and test a mechanistic model, the "pumping and streamline segregation" or PASS model, for nitrate removal in bedforms. The PASS model dramatically reduces computational overhead associated with modeling nitrogen transformations in bedforms and reproduces (within a factor of 2 or better) previously published measurements and models of biogeochemical reaction rates, benthic fluxes, and in-sediment nutrient and oxygen concentrations. Application of the PASS model to a dive

Realtime (31)P NMR Investigation on the Catalytic Behavior of the Enzyme Adenylate kinase in the Matrix of a Switchable Ionic Liquid.

The integration of highly efficient enzymatic catalysis with the solvation properties of ionic liquids for an environmentally friendly and efficient use of raw materials such as wood requires fundamental knowledge about the influence of relevant ionic liquids on enzymes. Switchable ionic liquids (SIL) are promising candidates for implementation of enzymatic treatments of raw materials. One industrially interesting SIL is constituted by monoethanol amine (MEA) and 1,8-diazabicyclo-[5.4.0]-undec-7-ene (DBU) formed with sulfur dioxide (SO2) as the coupling media (DBU-SO2-MEASIL). It has the abili

Strategies for regeneration of nicotinamide coenzymes emphasizing self-sufficient closed-loop recycling systems.

Biocatalytic reduction reactions depending on nicotinamide coenzymes require an additional reaction to regenerate the consumed cofactor. For preparative application the preferred method is the simultaneous coupling of an in situ regeneration reaction. There are different strategically advantageous routes to achieve this goal. The standard method uses a second enzyme and a second co-substrate, for example formate and formate dehydrogenase or glucose and glucose dehydrogenase. Alternatively, a second substrate is employed which is converted by the same enzyme used for the primary reaction. For e

Cytochrome P450 OxyBtei catalyzes the first phenolic coupling step in teicoplanin biosynthesis.

Bacterial cytochrome P450s form a remarkable clade of the P450 superfamily of oxidative hemoproteins, and are often involved in the biosynthesis of complex natural products. Those in a subgroup known as "Oxy enzymes" play a crucial role in the biosynthesis of glycopeptide antibiotics, including vancomycin and teicoplanin. The Oxy enzymes catalyze crosslinking of aromatic residues in the non-ribosomal antibiotic precursor peptide while it remains bound to the non-ribosomal peptide synthetase (NRPS); this crosslinking secures the three-dimensional structure of the glycopeptide, crucial for antib

CARD-FISH analysis of a TCE-dechlorinating biocathode operated at different set potentials.

Bioelectrochemical systems (BES) are increasingly being considered for bioremediation applications, such as the reductive transformation of chlorinated hydrocarbons in subsurface environments. These systems typically rely on a polarized solid-state electrode (i.e. a cathode) serving as electron donor for the microbially catalyzed reductive dechlorination of chlorinated contaminants. The microorganisms involved in dechlorinating biocathodes are not still identified. Particularly, it is not clear whether the same microorganisms responsible for the reductive dechlorination in 'conventional' biore

(In)organic anions as carbonic anhydrase inhibitors.

Carbonic anhydrases (CAs, EC 4.2.1.1) are widespread enzymes in all life kingdoms with five distinct genetic families known to date, the α-, β-, γ-, δ- and ζ-CAs. With the exception of the δ-class, which is less investigated to date, enzymes from the remaining classes found in vertebrates, corals, fungi, bacteria and archaea have been studied for their inhibition with simple inorganic anions as well as more complex inorganic and organic ones. In this paper we review the available data for the inhibition of these enzymes with all anions except sulfonamides and their bioisosteres (sulfamates, su

Impact of organic solvents on the catalytic performance of halohydrin dehalogenase.

Biocatalytic transformations in organic synthesis often require the use of organic solvents to improve substrate solubility and promote the … tivity in the ring-closure reaction and logP of the solvent. Knowledge of such a relationship makes biocatalysis with organic solvents more predictable, which may reduce the need to experiment with a variety of solvents in the future. The results revealed a high enzyme compatibility with hydrophobic solvents (e.g., n-heptane) in terms of activity and stability …

Computational Redesign of an ω-Transaminase from Pseudomonas jessenii for Asymmetric Synthesis of Enantiopure Bulky Amines.

ω-Transaminases (ω-TA) are attractive biocatalysts for the production of chiral amines from prochiral ketones via asymmetric synthesis. However, the substrate scope of ω-TAs is usually limited due to steric hindrance at the active site pockets. We explored a protein engineering strategy using computational design to expand the substrate scope of an (S)-selective ω-TA from Pseudomonas jessenii (PjTA-R6) toward the production of bulky amines. PjTA-R6 is attractive for use in applied biocatalysis due to its thermostability, tolerance to organic solvents, and acceptance of high concentrations of i…

Engineering a Paraburkholderia pallida Tannase for Improved Catalytic Efficiency, Thermal Stability, and Green Tea Clarification.

Tannases can reduce haze and astringency in tea beverages, but their application is often limited by poor thermal stability and catalytic efficiency. A tannase from Paraburkholderia pallida (Tanpp) was engineered using active-site mutagenesis and computational hotspot prediction. The double mutant F384T/N307A showed the best performance, with a 2.7-fold increase in catalytic efficiency and an 11-fold longer half-life at 50 °C than wild-type Tanpp. Structural modeling and molecular dynamics simulations suggested that N307A widened the substrate-entry region, whereas F384T compacted the catalyti

Spatial localization immobilization of metals and enzymes at Pickering emulsion droplet interfaces for chemo-enzymatic cascade catalysis.

Developing chemo-enzymatic cascade catalysis in catalytic asymmetric synthesis is of utmost significance, but such processes often suffer from low reaction efficiency because of incompatible reaction environments and suppressed intermediate transportation. High-Throughput Screening of Isomeric Reaction Products by Droplet Microfluidics Coupled to Cyclic Ion Mobility-Mass Spectrometry.

Engineered biocatalysts enable highly selective chemical transformations with low environmental impact. Development of biocatalysts by directed evolution requires screening many enzyme variants for improved catalytic properties. The low throughput of commonly used label-free screening methods, e.g., liquid chromatography-mass spectrometry (LC-MS), becomes the rate-limiting step in biocatalyst development, limiting the coverage of protein sequence space explored. Direct MS methods have been applied to biocatalyst screening; however, these methods cannot be used to evaluate isomer selectivity. I

Construction of Engineered Escherichia coli and Optimization of Conditions for Carcinine Synthesis via Multi-Enzyme Cascade Catalysis.

Carcinine is an imidazole dipeptide with potent antioxidant and antiglycation properties, although its chemical synthesis currently relies on severely environmentally harmful processes. In this work, a multi-enzyme cascade biotransformation system comprising 4'-phosphopantetheinyl transferase and non-ribosomal peptide synthetase was constructed. To overcome the limitations arising from stochastic spatial distribution and suboptimal mass transfer associated with independent enzymes, a fusion protein strategy was adopted. The two enzymes were fused via a flexible genetic linker within plasmid pE

Combinatorial Lid-Domain Engineering and Computational Design Enable Simultaneous Enhancement of Activity and Thermostability of D-Allulose 3-Epimerase.

d-Allulose is a promising low-calorie rare sweetener; however, its industrial production is limited by the insufficient catalytic activity and thermostability of d-allulose 3-epimerases (DAEases). In this study, a combinatorial engineering strategy integrating lid-domain modulation and computational design was developed to improve enzyme performance. Lid engineering identified a key mutation (N120D) associated with enhanced catalytic turnover, while computational screening identified beneficial mutations (T69S, D71E, and S128A). The best mutant, T69S/D71E/N120D/S128A, exhibited a 1.9-fold incr

An Engineered Fusion Enzyme-MOF Platform for Enhanced Biotransformation of Ginsenoside Rd to Rare Ginsenoside Rh2.

Rare ginsenoside Rh2 has attracted increasing interest for its antitumor, anti-inflammatory, and metabolic regulatory activities. Enzymatic biotransformation provides a green and selective route for Rh2 production, but current systems suffer from inefficient cascade conversion and limited enzyme stability. Here, two β-glucosidases enabling the stepwise and selective conversion of 20(S)-ginsenoside Rd to 20(S)-ginsenoside Rh2 were identified and assembled into a fusion biocatalyst through linker engineering. Compared with the parental two-enzyme cascade under equimolar enzyme loading, the optim

Immobilized horseradish peroxidase hybrid biocatalysts for degradation of pharmaceuticals and other emerging micropollutants.

With the increasing detection of emerging pollutants (EPs), such as pharmaceuticals, personal care products, and pesticides, in water bodies, there is a growing research priority to develop efficient methods for their degradation. In the present study, the degradation of 21 EPs from different classes was investigated using horseradish peroxidase (HRP) covalently immobilized onto two photocatalytic supports, TiO2 and ZnO. The hybrid biocatalysts (TiO2-HRP and ZnO-HRP) were used to degrade EPs with and without the presence of a redox mediator, 1-hydroxybenzotriazole (HOBT). The results revealed
