The PI3K/Akt/FOXO1 signaling axis mediates acetaminophen hepatotoxicity by disrupting metabolic homeostasis and oxidative stress.

Acetaminophen (APAP) is a widely used antipyretic-analgesic drug. At high or cumulative doses, it can induce serious liver injury, though the full spectrum of its hepatotoxic mechanisms remains unclear. This study comprehensively explored its pathogenic mechanisms by integrating network toxicology, molecular docking, in vivo and in vitro validations. Multi-source database screening identified 27 cross-targets associated with APAP hepatotoxicity. Protein-protein interaction (PPI) network and topological analyses revealed core targets central to APAP-induced liver injury. Gene Ontology (GO) and

AI-derived constrained conditional model for screening marker genes through integrated high-throughput transcriptome big data.

Currently, a large volume of high-throughput transcriptome data are accessible through various public databases. This brings urgent need to integrate transcriptome data sourced from numerous independent studies to extract common characteristics to precisely address specific research questions. Referring to this, we innovatively designed a model framework that leverages integrated transcriptome data with declarative constraints. Here the constraint is used to incorporate the AI-derived prior knowledge (genes in certain biological pathways, functions or user-specified terms) into the model and d

PregBase: A comprehensive knowledge base for clinically relevant knowledge representation and biomarker prediction in pregnancy.

Pregnancy complications are a leading cause of maternal and neonatal mortality worldwide. Understanding their underlying mechanisms is hindered by dispersed evidence across thousands of studies and complex biological interactions. We present PregBase, a comprehensive knowledge base for pregnancy research comprising automated extraction, validation, and exploration. PregBase was constructed by utilising large language models (LLMs) to extract relationships from literature; 13 LLMs were benchmarked across seven prompting strategies, with ensemble shuffle prompting outperforming single-strategy a

Tetranectin is significantly elevated in the synovial fluid of patients with full-thickness rotator cuff tears.

Rotator cuff tears are one of the most common shoulder disorders and can lead to pain, weakness, and limited mobility. Although the clinical condition has been widely studied, its molecular mechanisms are still not fully understood. In this exploratory study, we analyzed the proteomic profile of synovial fluid in patients with full-thickness rotator cuff tears and compared the results to those from a control group. Proteomic analysis was performed on synovial fluid samples from seven patients with full-thickness tears and three patients with partial-thickness tears, who served as the control g

Transcriptomic Analysis of Plac1 Ablation Reveals Broad Alterations in Signaling Pathways Essential for Prenatal Development and Overlap with a Preeclampsia-Associated Signature.

Plac1 is an X-linked gene essential for placental and embryonic development. A knockout (KO) mouse model was used to define placental gene expression changes associated with Plac1 loss at E16.5 and E18.5 using gene expression microarray. Genes exhibiting at least a 1.5-fold change and FDR < 0.05 were considered significant. At E16.5, 717 genes were downregulated and 796 upregulated in KO placentas relative to wild type (WT), whereas at E18.5, 1121 genes were downregulated and 1151 upregulated. Subsets of highly and uniquely dysregulated genes were examined by gene-level curation alongside syst

E3 Ubiquitin Ligases in Neurodevelopmental Disorders.

Neurodevelopmental disorders (NDDs) comprise a heterogeneous group of conditions that may present with developmental delay, intellectual disability, language impairment, epilepsy, autism-like features, motor abnormalities, or syndromic manifestations. Ubiquitination is an essential post-translational mechanism that regulates protein stability, localization, trafficking, and signaling. Because E3 ubiquitin ligases determine substrate specificity in this process, abnormalities in E3 ligases or E3-associated complexes can selectively affect proteins required for brain development and neuronal fun

Demonstration of extracellular traps in tuberculous pleural effusion from adult patients.

In pleural tuberculosis (TB), pleural effusion is typically characterized by a lymphocytic-rich exudate; however, neutrophils and macrophages predominate during the initial phase of infection. These immune cells are known to combat pathogens via phagocytosis, degranulation, and formation of extracellular traps (ETs)-a recently recognized defence mechanism. Although in-vitro and in-vivo studies have demonstrated ETs formation during Mycobacterium tuberculosis infection, their presence in clinical samples of patients with pleural TB has not been previously demonstrated. In this study, we employe

A Pilot Proteomics Analysis of Mycobacterium tuberculosis Lacking a Putative Short-Chain Dehydrogenase Rv0148.

Mycobacterium tuberculosis (Mtb) is an intracellular pathogen that survives in host cells by resisting hostile antimicrobial defenses. However, the molecules and mechanisms that contribute to Mtb's intracellular survival are not fully understood. We have previously reported that Rv0148, a putative short-chain dehydrogenase/reductase, plays a significant role in Mtb stress response and virulence in in vitro and in vivo models. To further understand the role of Rv0148 in regulating global functions of Mtb, we performed comparative pilot proteomic analysis between Δrv0148 mutant, and wild-type (W

Transcriptome analysis reveals no obvious unintended effects in the spleen of CRISPR/Cas9-mediated CD163 and pAPN double-knockout pigs.

Gene editing provides powerful tools for farm animal breeding. Our group previously obtained CD163/pAPN double-knockout (DKO) pigs via CRISPR/Cas9 and somatic cell nuclear transfer. These pigs are not only resistant to three infectious viruses but also maintain normal production performance. However, unintended effects of CRISPR/Cas9 tools may pose potential risks to animal well-being or safety. This study aimed to characterize the differences in splenic gene expression between wild-type (WT) and DKO pigs, providing a basis for safety evaluation of gene-edited animals. A comprehensive transcri

Transcriptomic analysis of immune and metabolic responses in largemouth bass (Micropterus salmoides) challenged with Citrobacter freundii.

Citrobacter freundii, a common zoonotic pathogen affecting humans, livestock and fish, is recognized for its substantial impact on largemouth bass (Micropterus salmoides) mortality. However, the mechanisms of C. freundii infection in largemouth bass remain poorly understood. Based on the results of extracellular enzyme activity detection, it was speculated that the production of extracellular enzymes may be related to bacterial pathogenicity. To elucidate the immune response mechanism, transcriptomic analysis was conducted on spleen tissues from largemouth bass infected with C. freundii. Follo

Vacuole and mitochondria patch protein Mcp1 of Saccharomyces boulardii impairs the oxidative stress response of Candida albicans by regulating 2-phenylethanol.

BACKGROUND: Vacuole and mitochondria patch (vCLAMP) protein Mcp1 is crucial in eukaryotic cells response to environmental stress, but the mechanism of Mcp1 in Saccharomyces boulardii (S. boulardii) against pathogenic fungi is unclear. RESULTS: This work first explored the role of Mcp1 in S. boulardii against Candida albicans (C. albicans). The results showed that Mcp1 located on the vacuolar and mitochondrial membrane of S. boulardii. Overexpression of Mcp1 inhibited the adhesion and hyphal formation of C. albicans in vitro. The mice model of intestinal infection revealed that WT-pGK1-MCP1 mu

Comprehensive high-throughput sequencing analysis and verification of miRNAs isolated from bronchial epithelial cells undergoing radiation-induced epithelial-mesenchymal transition.

Radiation-induced pulmonary fibrosis (RIPF) constitutes a significant complication in radiotherapy for various cancers, often severely limiting its efficacy. Recent studies suggest that epithelial-mesenchymal transition (EMT) plays a vital role in the pathogenesis of RIPF. Elucidating the involvement of microRNAs (miRNAs) in EMT could provide valuable insights into the mechanisms underlying RIPF and potentially reveal therapeutic targets. In this study, twelve dishes of BEAS-2B cells were irradiated with 6 Gy 60Co γ-rays, and RNA was extracted at 0, 6, and 48 h after irradiation. High-throughpu

Efficient analysis of toxicity and mechanisms of isoflurane on postoperative delirium by network toxicology and molecular docking.

Postoperative delirium (POD), a prevalent neurological complication in older surgical patients, adversely affects recovery. Isoflurane, a common inhalational anesthetic, exhibits neurotoxic potential, but its role in POD pathogenesis remains unclear. Network toxicology and molecular docking strategies identified 38 shared targets between isoflurane (PubChem/ChEMBL) and POD (GeneCards). Protein-protein interaction networks (STRING/Cytoscape) screened eight core genes: catechol-O-methyltransferase (COMT), angiotensin l-converting enzyme (ACE), solute carrier family 6, member 3 (SLC6A3), cathepsi

Direct connexin-26 interactions with membrane proteins functionally relevant to the cochlea.

Connexin 26, the protein encoded by the GJB2 (Gap junction protein beta 2) gene, is expressed in different tissues, including the cochlea and skin. Pathogenic DNA alterations in GJB2 cause autosomal recessive nonsyndromic hearing loss, whereas some GJB2 variants may lead to deafness-associated skin disorders. Genes encoding proteins of the Connexin26 molecular complex may fit as candidates to explain genetic hearing loss of yet unknown etiology. In search for Connexin26 direct protein partners, 120 million clones of a human fetal brain cDNA library were screened for interaction with full-lengt
