Three-Dimensional Organoid-like Co-Culture of Human Endometrial Endothelial and Stromal Cells to Study Endometriosis-Associated Responses.

Three-dimensional (3D) endothelium-stromal co-cultures were established using human endometrial cells from biopsy of healthy women (n = 13) and serum samples from both healthy and endometriotic women (n = 5). For 3D construction, stromal cells were mixed with extracellular matrix components, followed by endothelial cell seeding. Morphological analysis confirmed the organization of tissue-like structures. Immunofluorescence and flow cytometry verified the expression of specific stromal and endothelial markers (Cytokeratin, Vimentin, Insulin-like growth factor-binding protein 1, and von Willebra

T cells co-expressing high levels of CD29 and CD99 show increased cytotoxic potential and are upregulated in Sjögren's disease.

This study aimed to investigate the role of CD29 and CD99 in human T lymphocytes and elucidate its significance in Sjögren's disease (SjD). ScRNA-Seq data were utilized to analyze CD29 and CD99 expression in PBMCs. CD29 and CD99-expressing T lymphocyte proportions in SjD patients and healthy controls were determined by flow cytometry. Bulk RNA-Seq was performed to identify differential gene expression between the CD29hiCD99hi and CD29intCD99int subsets. The roles of CD29 and CD99 were elucidated through the stimulation of T lymphocytes with anti-CD29 and anti-CD99. CD29 and CD99-related subset

Co-Exposure to Aflatoxin B1 and Patulin Induces Hepatic Injury in Mice and HepG2 Cells by Activating Oxidative Stress and Apoptosis.

Aflatoxin B1 (AFB1) and patulin (PAT) are prevalent foodborne mycotoxins with hepatotoxic potential, but the hepatic effects of combined exposure remain largely unclear. This study investigated the hepatotoxic consequences of co-exposure to AFB1 and PAT using no-observed adverse effect levels (NOAELs) in C57BL/6 mice and low-cytotoxic concentrations in HepG2 cells selected by viability screening. Mice and cells were assigned to four groups: control, AFB1, PAT and AFB1 + PAT. Exposure to either toxin individually did not cause evident liver injury, whereas co-exposure significantly elevated ala

A machine-learning capillary electrophoresis-based pattern to identify suspected sickle cell trait and α-thalassemia co-inheritance.

OBJECTIVES: Sickle cell trait (HbAS) and α-thalassemia are common conditions. Although their hematological laboratory findings have been extensively characterized individually, there is limited evidence on the hematological pattern when they are co-inherited. Early and simple diagnosis of HbAS/α-thalassemia allows for optimization of the clinical approach and provision of appropriate genetic counselling. The objective is to identify, using machine learning algorithms, the most relevant hematological quantities and to establish a useful cut-off value for screening patients with suspected HbAS/α

Mycobacterium tuberculosis and SARS-CoV-2 Co-Infection: An Efficient Cell Model to Study Lung Host-Pathogen Interactions.

In less than a year, SARS-CoV-2 managed to displace Mycobacterium tuberculosis (Mtb) as the leading cause of death worldwide due to a single infectious agent. Both pathogens affect the respiratory tract, mainly the lungs. However, the impact that a possible Mtb + SARS-CoV-2 co-infection can have on the host response is still unknown. Herein we depict a rigorous system to evaluate the complex interaction between two simultaneous infections in vitro in a lung epithelial cell line (A549). Overall, the process includes eight steps: (1) Mtb culture, (2) cell maintenance, (3) preparation of viral st

A pancreatic cancer organoid-macrophage co-culture using starPEG-heparin hydrogel deciphers tumor-immune cell interactions.

Macrophages are among the most abundant immune cells in the pancreatic ductal adenocarcinoma (PDAC) tumor microenvironment (TME) and play a key role in regulating the immunosuppressive niche that facilitates tumor growth. Although recent three-dimensional (3D) culture systems using patient-derived materials have advanced our understanding of tumor biology, most models lack key cellular TME components and thus fail to capture tumor-immune cell interactions. To address this gap, we developed an in-vitro 3D co-culture model incorporating PDAC patient-derived organoids (PDOs) and macrophages withi

Immune Modulation in Peripheral Blood of Cystic Fibrosis Patients Following Ex Vivo Co-Culture with Mesenchymal Stem Cells.

BACKGROUND: Cystic fibrosis (CF) is an inherited multisystemic disease. Despite advances in treatment, many patients still experience progressive lung dysfunction. Dental follicle-derived mesenchymal stem cells (DF-MSCs) possess significant immunomodulatory potential in inflammatory airway diseases. We evaluated the effects of DF-MSCs on lymphocyte proliferation, CD4+CD25+FoxP3+ regulatory T cell (Treg) frequency, and cytokine responses in peripheral blood mononuclear cells (PBMCs) from CF patients. METHODS: PBMCs from 20 CF patients and 20 matched healthy controls were isolated by density gr

Poly(d,l-lactide-co-glycolide) Nanoparticles Encapsulating Doxorubicin for Improved Treatment in Cholangiocarcinoma and Drug-Resistant Cells.

Cholangiocarcinoma (CCA), a malignancy of the bile duct epithelium, represents a significant public health issue in the Greater Mekong Subregion, including Thailand. Its aggressive characteristics and late-stage diagnosis lead to poor prognosis and elevated mortality rates. Chemotherapy faces limitations, including the requirement for high and frequent dosages, low cellular uptake, and side effects. To address these challenges, poly(d,l-lactide-co-glycolide) (PLGA) nanoparticles (NPs) encapsulating doxorubicin (DOX), a chemotherapeutic drug, were developed via a modified nanoprecipitation tech

A co-culture system to study the effects of Poly I:C-activated microglia on the differentiation of murine primary neural stem cells.

Studies in rodents have shown that systemic inflammation induced by prenatal exposure to the viral mimetic polyinosinic:polycytidylic acid (Poly I:C) triggers maternal immune activation. Cytokines released by the maternal immune system can cross the placenta and enter fetal circulation. In the fetal brain, embryonic microglia may produce additional cytokines and other inflammatory mediators in response to maternally derived cytokines. This resulting cytokine imbalance is suggested to impair neurogenesis and brain development, potentially contributing to the onset of neuropsychiatric disorders

GSNCASCR: An R Package to Identify Differentially Co-Expressed Curated Gene Sets with Single-Cell RNA-Seq Data.

(1) Differential co-expression analysis between two phenotypes with a known gene set helps to uncover gene regulation alterations. (2) GSNCASCR uses CSCORE to estimate the gene pair correlations for network reconstruction and GSNCA to quantify the structure changes of co-expression networks of the predefined gene sets. It also ranks genes based on their "importance" in the weighted network. The method is implemented with free R software (version 0.1.0, available on GitHub), allowing users to analyze their data with the help of demo vignettes included in the package. (3) With analysis of both s

Co-modification of amylase by acrolein with formaldehyde or methylglyoxal: Impact on enzyme function, structure, and cellular stress in two cell lines.

Reactive carbonyl species (RCS), such as acrolein (Acr), formaldehyde (FA), and methylglyoxal (MGO) are highly reactive aldehydes co-existing in processed foods and food materials. RCS may modify the digestive enzymes individually while whether they act together on the enzymes remains unknown. In this study, the co-action of RCS on α-amylase was studied. The results show that when Acr is present together with MGO or FA, they simultaneously modify amylase to form carbonylated proteins and inhibit the activity of amylase. The co-action of RCS increases the modification sites of amylase, enhances

Co-expression of PD1+ and HLA-DR+ in CD8+ T cells is increased in tonsils of children with EBV primary and persistent infection.

INTRODUCTION: Epstein-Barr virus (EBV) infects B lymphocytes and establishes lifelong persistence in the B cells. While systemic T-cell responses have been well characterized, the local immune response at the site of viral entry in children from undeveloped countries remains poorly understood. METHODS: Tonsillar CD4 and CD8 T cells in 32 pediatric patients undergoing tonsillectomy were classified as primary infected (PI), EBV carriers (EC), and non-infected children by serology. T-cell subsets were assessed by flow cytometry, whereas LMP1 and EBNA2 viral proteins were evaluated by immunohisto

Immune cells in dorsal root ganglia are associated with pruritus in a mouse model of allergic contact dermatitis and co-culture study.

The interaction between the neuroimmune system plays a crucial role in itch sensation, yet most research has focused on immune cells within the skin. Our study seeks to explore the presence and functions of immune cells within the dorsal root ganglia (DRG) in the context of allergic contact dermatitis (ACD). Immunofluorescence and histological staining techniques were employed to identify immune cells, including T-cells, basophils, mast cells, and dendritic cells (DCs), within the DRG of BALB/c mice sensitized and challenged with toluene diisocyanate (TDI). Our findings revealed an increase in
