Activation of mTOR coincides with autophagy during ligation-induced atrophy in the rat submandibular gland.

Salivary gland atrophy is a common consequence of pathology, including Sjögren's syndrome, irradiation therapy and obstructive sialadenitis. During severe atrophy of the rat submandibular gland caused by excretory duct ligation, the majority of acinar cells disappear through apoptosis, whereas ductal cells proliferate and dedifferentiate; yet, the gland can survive in the atrophic state almost indefinitely, with an ability to fully recover if deligated. The control mechanisms governing these observations are not well understood. We report that ~10% of acinar cells survive in ligation-induced a

Schwann cells in the inner ear: development, disease, and regeneration.

Schwann cells are classically known as the constituent supporting cells of the peripheral nervous system. Beyond the scope of merely myelinating axons of the more saliently known neurons, Schwann cells comprise the majority of peripheral nervous system tissue. Through the lens of the inner ear, additional properties of Schwann cells are becoming elucidated. Therein, the process of myelin formation in development is more aptly understood as a homeostatic oscillation of differentiation status. Perpetual interaction between neural and non-neural cells of the inner ear maintains an intricate balan

Sinusoidal cell-derived biomarker scores predict diagnosis and prognosis in chronic liver disease.

BACKGROUND: Sinusoidal cells are central drivers of chronic liver disease (CLD) progression, yet current biomarkers fail to capture their phenotypic states. We aimed to develop sinusoidal cell-specific biomarkers for the diagnosis and prognosis of CLD. METHODS: Single-cell RNA sequencing data were analyzed to identify cell-type-specific signatures for liver sinusoidal endothelial cell capillarization, hepatic stellate cell activation, and macrophage polarization. These signatures were integrated into three sinusoidal scores (endothelial, mesenchymal, and macrophage) reflecting dedifferentiati

Distinct coexisting pathologies of oral malignant peripheral nerve sheath tumor with melanin pigmentation and indolent small lymphocytic lymphoma.

Spindle cell malignancies of the oral cavity are uncommon and pose considerable diagnostic challenges due to overlapping histomorphology and immunohistochemical features. Malignant peripheral nerve sheath tumor (MPNST) with heterologous differentiation is particularly rare. Moreover, its coexistence with a synchronous hematological malignancy such as small cell lymphoma (SLL) is exceedingly unusual. A male 62 year, presented with a progressively enlarging, pigmented, ulcero-proliferative painless mass over the right upper alveolus extending to the hard palate. Imaging revealed a heterogeneousl

MCSP+ metastasis founder cells activate immunosuppression early in human melanoma metastatic colonization.

To investigate the early, poorly understood events driving metastatic progression, we searched for the earliest detectable disseminated cancer cells (DCCs), also often referred to as disseminated tumor cells (DTCs), in sentinel lymph node (SLN) biopsies of 492 patients with stage I-III melanoma. Using micromanipulator-assisted isolation of rare DCCs, single-cell mRNA and DNA sequencing, codetection by indexing immunofluorescence imaging and survival analysis, we identified melanoma-associated chondroitin sulfate proteoglycan (MCSP)+ melanoma cells as metastasis founder cells (MFCs). We found t

Identification, structural, and functional characterization of a new early gene (6A3-5, 7 kb): implication in the proliferation and differentiation of smooth muscle cells.

Arterial smooth muscle cells (SMCs) play a major role in atherosclerosis and restenosis. Differential display was used to compare transcription profiles of synthetic SMCs to proliferating rat cultured SMC line. An isolated cDNA band (6A3-5) was shown by northern (7 kb) to be upregulated in the proliferating cell line. A rat tissue northern showed differential expression of this gene in different tissues. Using 5' RACE and screening of a rat brain library, part of the cDNA was cloned and sequenced (5.4 kb). Sequence searches showed important similarities with a new family of transcription facto

Mixed low grade and high grade endometrial stromal sarcoma of uterus: differences on immunohistochemistry and chromosome in situ hybridisation.

A case of a 64 year old woman with a tumour of the uterus is reported. The patient presented with postmenopausal bleeding and subsequently underwent total hysterectomy and bilateral salpingo-oophorectomy. Sections of the tumour showed a low grade endometrial stromal sarcoma coexisting with areas consistent with high grade sarcoma. The sarcoma cells, in both the low and high grade areas, were positive for vimentin and negative for desmin and cytokeratin on immunohistochemistry. While the sarcoma cells in the low grade region showed immunoreactivity for oestrogen and progestogen receptors, those

Diffusion-weighted MR imaging: diagnosing atypical or malignant meningiomas and detecting tumor dedifferentiation.

BACKGROUND AND PURPOSE: Atypical and malignant meningiomas are uncommon tumors with aggressive behavior and higher mortality, morbidity, and recurrence compared with benign tumors. We investigated the utility of diffusion-weighted (DW) MR imaging to differentiate atypical/malignant from benign meningiomas and to detect histologic dedifferentiation to higher tumor grade. MATERIALS AND METHODS: We retrospectively compared conventional and DW MR images (b-value 1000 s/mm(2)) acquired on a 1.5T clinical scanner between 25 atypical/malignant and 23 benign meningiomas. The optimal cutoff for the ab

Mixed neuroendocrine-non-neuroendocrine neoplasm with mucinous adenocarcinoma and amphicrine carcinoma components in the bile duct: an autopsy case.

We report the first case of bile duct mixed neuroendocrine-non-neuroendocrine neoplasm (MiNEN) that had a mucinous carcinoma component. An 88-year-old man with biliary obstruction was diagnosed as having distal bile duct cancer using imaging examinations and endoscopic biopsy. The patient received the best supportive care without surgical resection for 13 months until death. An autopsy revealed a bulky mass involving the distal bile duct and multiple metastases in intra-abdominal lymph nodes, the liver, and the lungs. The primary cancer was microscopically diagnosed as a MiNEN, which consisted

Richter's syndrome in chronic lymphocytic leukemia.

Over an 18-year period a distinctive large cell lymphoreticular neoplasm (Richter's transformation) developed in 9 patients with chronic lymphocytic leukemia. Clinical findings at the onset of Richter's transformation were remarkably uniform and consisted of the abrupt onset of fever, marked asymmetric lymphadenopathy with the formation of masses, splenomegaly, and hepatomegaly. All patients underwent rapid clinical deterioration followed by death within six and a half months. Earliest infiltrates of large lymphoreticular cells were identified in the lymph nodes in 3 of 4 patients and the bone

Targeting syndecan-2 inhibits papillary thyroid cancer invasiveness and de-differentiation.

BACKGROUND: The high incidence of recurrence and metastatic disease remain the major issues for papillary thyroid cancer (PTC) patients. Previous studies have demonstrated that Syndecan-2 (SDC2) plays a key role in multiple cancers progression. However, the potential role of SDC2 in PTC progression and recurrence remains unclear. METHODS: First, we performed bioinformatics analysis and western-blotting analysis to explore the potential prognostic value of SDC2 in PTC. Then we applied transient siRNA knockdown and plasmid overexpression to alter SDC2 expression level in PTC cell line B-CPAP an

Study of proliferative processes and nuclear estradiol and progesterone receptors in myocytes in pregnant and postpartum mouse uterus.

Numerical densities of the nuclei were morhometrically evaluated in all myocytes and myocytes expressing nuclear estrogen- and progesterone-receptor complexes, which were revealed immunohistochemically with monoclonal antibodies in C57Bl/6 mice. It was shown that the above quantitative parameters of myometrial cells after the first pregnancy were similar to those in nonpregnant mice by day 10 after delivery. In the third pregnancy, especially developed after the second interrupted pregnancy, proliferation processes in the myometrium were not completed by postpartum day 10, but dramatically pro

Sarcomatoid salivary duct carcinoma of the oral cavity.

This report recounts a new case of sarcomatoid variant of salivary duct carcinoma in a 45-year-old woman. The 1.5-cm polypoid mass protruded from the retromolar area and focally extended into the pharyngeal wall. The patient was free of disease for 11 years. The tumor showed a biphasic malignancy; nondescript sarcomatous proliferation predominated over ductal carcinoma population, with perceptible blending of the two components. The sarcomatoid areas had no heterologous properties. Discohesive pleomorphic cells were immunopositive for vimentin, epithelial membrane antigen and AE1/AE3 at a sign

Activation of P2X7R Inhibits Proliferation and Promotes the Migration and Differentiation of Schwann Cells.

In the vertebrate nervous system, myelination of nerve fibers is crucial for the rapid propagation of action potentials through saltatory conduction. Schwann cells-the main glial cells and myelinating cells of the peripheral nervous system-play a crucial role in myelination. Following injury during the repair of peripheral nerve injuries, a significant amount of ATP is secreted. This ATP release acts to trigger the dedifferentiation of myelinating Schwann cells into repair cells, an essential step for axon regeneration. Subsequently, to restore nerve function, these repair cells undergo rediff

Articular Chondrocyte Phenotype Regulation through the Cytoskeleton and the Signaling Processes That Originate from or Converge on the Cytoskeleton: Towards a Novel Understanding of the Intersection between Actin Dynamics and Chondrogenic Function.

Numerous studies have assembled a complex picture, in which extracellular stimuli and intracellular signaling pathways modulate the chondrocyte phenotype. Because many diseases are mechanobiology-related, this review asked to what extent phenotype regulators control chondrocyte function through the cytoskeleton and cytoskeleton-regulating signaling processes. Such information would generate leverage for advanced articular cartilage repair. Serial passaging, pro-inflammatory cytokine signaling (TNF-α, IL-1α, IL-1β, IL-6, and IL-8), growth factors (TGF-α), and osteoarthritis not only induce dedi

Metastatic adenoid cystic carcinoma with high-grade transformation ("dedifferentiation") in pleural effusion and neck lymph node: A diagnostic challenge on cytology?

High-grade transformation (HGT) or "dedifferentiation" is an uncommon phenomenon among salivary gland carcinomas including adenoid cystic carcinoma (ACC), which is important to recognize because it is associated with increased tumor aggressiveness, with a high propensity for lymph node and distant metastases. ACC with HGT is histologically characterized by a distinct population of poorly differentiated cells with loss of the typical biphasic ductal and myoepithelial differentiation seen in conventional ACC, associated with pleomorphism, necrosis and increased mitotic activity. We report the cy

SOX9: dosage- and context-dependent functions in pancreatic development and β-cell biology.

SOX9 is a highly conserved transcription factor (TF) belonging to the SRY-related HMG-box (SOX) family and the high-mobility group (HMG) class of DNA-binding proteins. SOX9 integrates structural flexibility, DNA-dependent dimerization, and context-specific cofactor interactions to orchestrate organogenesis. In the pancreas, SOX9 acts as a dosage-sensitive gatekeeper: high expression in multipotent progenitors maintains proliferation and prevents premature endocrine differentiation, while its timely downregulation is a prerequisite for NEUROG3 (NGN3) induction and β-cell maturation. Genetic and

[Sexuality in the second half of the life span].

The central dysregulated pathway of clear cell (cc) renal cell carcinoma (RCC), the von Hippel Lindau/hypoxia inducible factor-α axis, is a key regulator of intracellular iron levels, however the role of iron uptake in human RCC tumorigenesis and progression remains unknown. We conducted a thorough, large-scale investigation of the expression and prognostic significance of the primary iron uptake protein, transferrin receptor 1 (TfR1/CD71/TFRC), in RCC patients. TfR1 immunohistochemistry was performed in over 1500 cores from 574 renal cell tumor patient tissues (primary tumors, matched benign

Thyroid Hormone Receptor β Suppresses Cancer Cell Activity by Differential Regulation of Glycogen Metabolism.

Anaplastic thyroid cancer (ATC) is one of the most lethal endocrine cancers with no enduring therapies. Thyroid hormone receptor β (TRβ), a recognized tumor suppressor, modulates the transcriptome altering gene expression in numerous intracellular signaling pathways. Our recent studies revealed that TRβ agonism inhibits glycogen metabolism in ATC cells. Our goal in the present study was to delineate the molecular mechanisms by which TRβ regulates glycogen synthesis and breakdown. In ATC cells, activation of TRβ induced changes in expression of genes and proteins in glycogen signaling concordan
