Furazolidone has normally been administered as a non-absorbable antimicrobial agent for use in gastrointestinal infections. However, in India and Mexico it has been used successfully for the treatment of typhoid fever. We measured concentrations of furazolidone by high performance liquid chromatography (HPLC) in several biological fluids, after a single oral dose (5 mg/kg). A sensitive, reliable, rapid quantitative method was developed for the N,N'-dimethyl derivatives of the 5,5'-disubstituted barbiturates (NNDM-barbiturates) after liquid-liquid extraction of 0.5-mL urine volumes. Each barbiturate was identified by GC/MS through the retention time for the total ion current and selected ion monitoring of four ion currents for each analyte. Chromatographic selectivity and position specificity were examined by using deletion and substitution analogues of calcitonin (CT), a 32-amino acid polypeptide. The biological activity of CT has been shown to be related to structural features, including a regular spacing of hydrophobic and hydrophilic residues in positions 8-22. Detection of antibody to Coccidioides immitis F antigen is of proved value in the diagnosis of coccidioidomycosis. This antibody is demonstrable by use of an immunodiffusion assay with reference coccidioidin antigen and antiserum to C. immitis. Using a combination of lectin affinity and immunoaffinity chromatography, we isolated the F antigen from coccidioidin and prepared monospecific antibody to the purified antigen. The availability of these reagents will enable the development of a sensitive and specific assay for detecting serologic reactivity to this antigen.

High pressure liquid chromatography (HPLC) was demonstrated to be a good tool for the separation, identification and quantitation of corticosteroids (CS) extracted from homogenized tissue of adrenal glands and adrenal tumors in patients with hypercorticism. A chromatographic system consisting of Sorbax-SIL and Sorbax-CN columns, organic solvent extraction and a UV detector was used to analyze both more polar and less polar corticosteroids in the adrenal tissue.

When extracts of Escherichia coli are filtered through a Sepharose column containing covalently bound rifamycin a protein is bound which can be eluted either with a high concentration of urea or more specifically with low concentrations of rifamycins. Its Mr is 18,000 +/- 1,000 in the presence of dodecylsulfate, in its absence 36,000 +/- 3,000. The association constant of the protein for rifampicin is 2.4 +/- 0.5 x 10(-4) M with two binding sites per dimer as determined by equilibrium dialysis. Large amounts of this protein are released from the cells by an osmotic shock.

A method is described for the determination of unconjugated terbutaline in plasma. The assay is based on gas chromatography chemical ionization mass spectrometry with ammonia as the reagent gas. 24 h-urine excretion of aldosterone-18-glucuronide was determined with and without preceding paper chromatography in patients with essential hypertension (n = 50). Radioimmunoassay was performed with a highly specific antibody against aldosterone. Urine specimens determined without chromatography gave significantly higher values than aliquots which were estimated at the same time with chromatography. The pyrolysis gas chromatography mass spectrometry of Ibuprofen, Fenoprofen, Naproxen and Ketoprofen, a series of anti-inflammatory propionic acid derivatives, is shown to proceed via decarboxylation and elimination to yield characteristic ethyl and vinyl fragments. The pyrogram enables the identification of the drug to be achieved as the pure compound, in a formulated dosage form or excreted in urine. The presence of metabolites derived from Ibuprofen causes four new fragments to be observed in the urine pyrogram. The identification of 16 components of the control urine pyrogram is presented.
