PEGylated murine Granulocyte-macrophage colony-stimulating factor: production, purification, and characterization.

Granulocyte-macrophage colony-stimulating factor (GM-CSF) regulates proliferation, differentiation, and function of hematopoietic progenitor cells. Aside from expansion of hematopoietic cells, GM-CSF has shown efficacy in other diseases, including Crohn's disease. While GM-CSF being clinically used in humans, the ability to perform mechanistic studies in murine models is difficult due to the limited availability and rapid clearance of murine GM-CSF in the peripheral blood. To address these issues, we efficiently expressed murine GM-CSF under the control of the AOX1 gene promoter in Pichia past

Mobilization of human lymphoid progenitors after treatment with granulocyte colony-stimulating factor.

Hemopoietic stem and progenitor cells ordinarily residing within bone marrow are released into the circulation following G-CSF administration. Such mobilization has a great clinical impact on hemopoietic stem cell transplantation. Underlying mechanisms are incompletely understood, but may involve G-CSF-induced modulation of chemokines, adhesion molecules, and proteolytic enzymes. We studied G-CSF-induced mobilization of CD34+ CD10+ CD19- Lin- and CD34+ CD10+ CD19+ Lin- cells (early B and pro-B cells, respectively). These mobilized lymphoid populations could differentiate only into B/NK cells o

In vitro and in vivo characterization of smooth small colony variants of Brucella abortus S19.

Brucella abortus is known to produce chronic infections in both humans and a variety of animal species. However, the mechanisms underlying the persistence of the bacteria in the presence of an ongoing immune response are still unknown. In this respect we made use of the observation that in vitro grown B. abortus S19 exhibits heterogenicity in colony size when plated onto TS agar, while experimental infection of mice uniformly results in the in vivo selection of the small colony variant. We demonstrate that the spontaneous smooth small colony variant is characterized not only by a slower growth

A role for G-CSF (granulocyte-colony stimulating factor) in the central nervous system.

Antiepileptic drugs (AEDs) are used extensively to treat multiple non-epilepsy disorders, both in neurology and psychiatry. This article provides a review of the clinical efficacy of AEDs in non-epilepsy disorders based on recently published preclinical and clinical studies, and attempts to relate this efficacy to the mechanism of action of AEDs and pathophysiological processes associated with the disorders. Some newer indications for AEDs have been established, while others are under investigation. The disorders where AEDs have been demonstrated to be of clinical importance include neurologic

Comparative morphologic and immunohistochemical investigation of spontaneously occurring thymomas in a colony of European hamsters.

This study documents the characteristics of a large series of spontaneously occurring thymomas in a laboratory colony of European hamsters (Cricetus cricetus). Thymomas are rare organotypic neoplasms originating from the thymic epithelial compartment. Because the hamster thymomas largely resembled their human counterparts, the recent World Health Organization (WHO) classification of human thymic epithelial tumors was used. Forty hamsters of both sexes aged 3-29 months were examined macroscopically and histologically. In 22 (55%) of the 40 animals, necropsy revealed enormous whitish masses in t

Change in colony morphology of Candida lusitaniae in association with development of amphotericin B resistance.

It is not uncommon to see amphotericin B treatment failure in patients with systemic infection caused by Candida lusitaniae. We report a patient with stage IV ovarian carcinoma and C. lusitaniae sepsis whose treatment with amphotericin B failed. The initial blood isolate was susceptible to amphotericin B in vitro; however, the MIC for a blood isolate recovered 7 weeks after treatment began showed a fourfold increase. Direct subculture of two positive blood samples obtained within a week of the patient's death showed the coexistence of two distinct colony color variants on CHROMagar Candida (CA

Risk and benefit of treatment of severe chronic neutropenia with granulocyte colony-stimulating factor.

The Severe Chronic Neutropenia International Registry (SCNIR) was established in 1994 following four phase I/II and one phase III clinical trial on the use of filgrastim (recombinant human granulocyte colony-stimulating factor [r-metHuG-CSF]) as a treatment for severe chronic neutropenia (SCN). A primary purpose of the SCNIR is to monitor SCN patients treated with filgrastim for adverse events that might occur over time. As of December 31, 2000, 832 patients with SCN (384 congenital, 160 cyclic, 288 idiopathic) were enrolled. Clinical trial and Registry data show that filgrastim is an effectiv

Mutation of macrophage colony stimulating factor (Csf1) causes osteopetrosis in the tl rat.

Osteopetrosis results from a heterogeneous group of congenital bone diseases that display inadequate osteoclastic bone resorption. We recently mapped tl (toothless), a mutation that causes osteopetrosis in rats, to a genetic region predicted to include the rat Csf1 gene. In this study, we sequenced the coding sequence of the rat Csf1 gene to determine if a mutation in Csf1 could be responsible for the tl phenotype. Sequencing revealed a 10-base insertion in the coding sequence of mutant animals that produces a frameshift and generates a stop codon early in the mutant Csf1 coding sequence. The

Mandibular gland chemistry of grass-cutting ants: species, caste, and colony variation.

The compositions of the alarm pheromones of two species of grass-cutting ant, Atta bisphaerica and A. capiguara, were examined, and caste and colony variations quantified. The pheromones of A. bisphaerica and A. capiguara were remarkably similar and were composed of a complex mixture of volatiles in which 4-methyl-3-heptanone and 2-heptanone were the most abundant compounds. Small but consistent intraspecific differences were found between the worker castes and between individual colonies. The results support the view that alarm pheromones are rarely species specific. The possible importance o

The effect of granulocyte colony stimulating factor on the immune parameters in experimental obstructive jaundice.

BACKGROUND/AIMS: Obstructive jaundice is an important clinical problem. It may cause complications such as renal insufficiency, cardiovascular sequels, coagulation defects, gastrointestinal bleeding, delayed wound healing, secondary biliary cirrhosis and sepsis. We investigated the effect of GM-CSF on immunological parameters in the experimental obstructive jaundice. METHODOLOGY: In our experimental study we studied four groups that consisted of 28 rats. The 1st group consisted of sham rats, the 2nd group consisted of sham and GM-CSF applied rats and the 3rd and 4th groups consisted of rats t

A colony blot immunoassay to detect enteroinvasive Escherichia coli and Shigella in water samples.

AIMS: The aim of the study was to develop a colony blot immunoassay to detect Shigella and enteroinvasive Escherichia coli (EIEC) in water. METHODS AND RESULTS: Spiked samples were filtered through nitrocellulose membranes. Colony prints on the filters were tested with a monoclonal antibody specific to IpaC, an antigen coded by the invasion plasmid of Shigella and EIEC. Invasive pathogens could be successfully detected with the technique, even in the presence of a large number of non-pathogenic bacterial cells. The method was significantly more sensitive in identifying pathogen-containing sam

Reproduction and mortality in a colony of captive greater cane rats, Thryonomys swinderianus, Temminck.

The reproductive performance and mortalities in a colony of captive greater cane rats, Thryonomys swinderianus, were monitored from 1992 to 1998 at the Grasscutter Domestication Centre, Achimota, Ghana. The animals were kept in cages and exposed to constant lighting from a 100 W electric light bulb during the night. The diet consisted mainly of freshly cut Panicum maximum (guinea grass) fed ad libitum, with occasional supplements of cassava and cane sugar. The results indicate that the mean litter size and litter weight were 2.9 +/- 0.51 and 439.4 +/- 81.23 g, respectively. These figures are l

Chlamydia pneumoniae infection in a breeding colony of African clawed frogs (Xenopus tropicalis).

More than 90% of a breeding colony of clawed frogs (Xenopus tropicalis) imported to the United States from western Africa died in an epizootic of chlamydiosis. Chlamydial inclusions were observed by light and electron microscopy in the liver of an infected frog. Chlamydia pneumoniae was isolated in cell cultures from four frogs. A cutaneous infection by a chytridiomycete fungus observed in two frogs could have been a cofactor in the die-off.ous Diseases

Effect of granulocyte/colony-stimulating factor on the onset of the adult respiratory distress syndrome.

To evaluate the effect of granulocyte/colony-stimulating factor (G-CSF) on the onset of the adult respiratory distress syndrome (ARDS), we investigated whether the incidence of ARDS due to pulmonary infection differed between the G-CSF group which received chemotherapy with G-CSF and historical controls without G-CSF. We evaluated 132 patients with hematological malignancy in complete remission without any main organ dysfunction who had been treated between April 1983 and December 1997. We compared the incidence of ARDS due to pulmonary infection between those who received G-CSF and those who

Prevalence of enterotropic and polytropic mouse hepatitis virus in enzootically infected mouse colonies.

Mouse hepatitis virus (MHV) causes the most prevalent viral infection in contemporary laboratory mouse colonies. According to their primary replication site, different strains of MHV segregate into two overlapping biotypes, enterotropic and polytropic. These two groups vary greatly in disease pattern, pathogenicity, immune response, and duration of infection. Historically, the polytropic MHV strains represent the extensively studied prototype strains that have minimal enterotropism, whereas enterotropic MHV strains have been less characterized. Anecdotal reports suggest that most MHV strains e

High host specificity of Crataerina melbae (Diptera: Hippoboscidae) in a mixed colony of birds.

We looked for louse flies (Diptera, Hippoboscidae) in a mixed colony of 9 species of birds from 1991 to 1997. Alpine swifts (Apus melba) exhibited an unusually high prevalence (85.9%) by Crataerina melbae (Rondani, 1879). No birds from the other 8 breeding bird species were parasitized by this louse fly. We suggest that the number of potential swiftlike hosts as well as the size of hosts may account for the differential prevalence of the louse flies within the colony.

Elevated serum granulocyte colony-stimulating factor levels in patients with Graves' disease.

OBJECTIVE: Agranulocytosis is a serious side effect of anti-thyroid drugs (ATD). Granulocyte colony-stimulating factor (G-CSF) is one of the cytokines that increase granulocyte number. The aim of this study was to examine the sequential variation of serum G-CSF levels in patients with Graves' disease before and after ATD therapy. PATIENTS: Sixty-three patients with Graves' disease were studied before, during and after treatment with methimazole (MMI). Serum samples from 71 healthy subjects were used as controls. DESIGN AND MEASUREMENTS: Serum levels of G-CSF were measured by a novel chemilum

Carbimazole induced agranulocytosis: rescue with human recombinant granulocyte colony stimulating factor.

We report on a 25 yr old woman with primary autoimmune hyperthyroidism who was treated with carbimazole. Forty-three days later she developed agranulocytosis (64 x 10(6)/l). With 4 days treatment with recombinant human Granulocyte Colony Stimulating factor the granulocyte count rose to normal--4,350 cells x 10(6)/l. This is considerably faster than the rate of spontaneous recovery which usually takes one to 2 weeks.
