A novel role for the mineralocorticoid receptor in glucocorticoid driven vascular calcification.

Vascular calcification, which is common in the elderly and in patients with atherosclerosis, diabetes and chronic renal disease, increases the risk of cardiovascular morbidity and mortality. It is a complex, active and highly regulated cellular process that resembles physiological bone formation. It has previously been established that pharmacological doses of glucocorticoids facilitate arterial calcification. However, the consequences for vascular calcification of endogenous glucocorticoid elevation have yet to be established. Glucocorticoids (cortisol, corticosterone) are released from the a…

Converging, Synergistic Actions of Multiple Stress Hormones Mediate Enduring Memory Impairments after Acute Simultaneous Stresses.

UNLABELLED: Stress influences memory, an adaptive process crucial for survival. During stress, hippocampal synapses are bathed in a mixture of stress-released molecules, yet it is unknown whether or how these interact to mediate the effects of stress on memory. Here, we demonstrate novel synergistic actions of corticosterone and corticotropin-releasing hormone (CRH) on synaptic physiology and dendritic spine structure that mediate the profound effects of acute concurrent stresses on memory. Spatial memory in mice was impaired enduringly after acute concurrent stresses resulting from loss of sy…

Differential neuroendocrine responsiveness to morphine in Lewis, Fischer 344, and ACI inbred rats.

Preclinical evidence suggests there is a link between the responsiveness to stress and the propensity to self-administer drugs of abuse. Our previous findings, for example, have shown a significant positive correlation between the locomotor response to novelty and the acquisition of morphine self-administration in Lewis (LEW), Fischer 344 (F344) and ACI inbred rat strains. As an extension of this work, we now report on the neuroendocrine responses (i.e., corticosterone and prolactin secretion) evoked by morphine administration in these same inbred strains. Male LEW, F344, and ACI rats were sur…

A second enzyme protecting mineralocorticoid receptors from glucocorticoid occupancy.

We have confirmed that A6 cells (derived from kidney of Xenopus laevis), which contain both mineralocorticoid and glucocorticoid receptors, do not normally possess 11 beta-hydroxysteroid dehydroxgenase (11 beta-HSD1 or 11 beta-HSD2) enzymatic activity and so are without apparent "protective" enzymes. A6 cells do not convert the glucocorticoid corticosterone to 11-dehydrocorticosterone but do, however, possess steroid 6 beta-hydroxylase that transforms corticosterone to 6 beta-hydroxycorticosterone. This hydroxylase is cytochrome P-450 3A (CYP3A). We have now determined the effects of 3 alpha,5…

Chronic Stress Segregates Mice into Distinct Behavioral Phenotypes Based on Glucocorticoid Sensitivity.

Chronic stress alters hypothalamic-pituitary-adrenal (HPA) axis function, affecting corticosterone regulation and adaptive responses. Understanding individual variability in stress adaptation requires identifying distinct HPA axis response patterns. Here, we assessed HPA axis sensitivity in male C57BL6 mice exposed to 30 days of chronic social defeat stress (CSDS). Negative feedback integrity was evaluated using the dexamethasone suppression test (DST), with corticosterone measured after saline or low-dose dexamethasone administration at days 10 and 30. Behavioral testing (open field, elevated…

Acute stress induces a rapid increase of testosterone in a songbird: implications for plasma testosterone sampling.

Testosterone (T) and glucocorticoids, such as corticosterone, have both been shown to be important for the way vertebrates in general, and birds specifically, react to their immediate environment. For both corticosterone and T, many sources of variation in plasma levels have been demonstrated. Interestingly, a small number of studies on bird species have indicated that acute stress can have a positive effect on plasma T levels, analogous to what has been observed for plasma corticosterone levels. Using captive male European starlings (Sturnus vulgaris), we provide the first evidence in songbir…

The role of corticosteroids in the acquisition of sensitization to locomotor stimulant effects of MK-801.

In the present study, we investigated the role of corticosterone and glucocorticoid receptors in the acquisition of sensitization to locomotor stimulant effects of MK-801 in rats. MK-801 (two doses, 0.4 mg/kg i.p. each, given twice, 48 h apart) evoked sensitization, observed as enhancement of the locomotor activity to a challenging dose of MK-801 (0.4 mg/kg) but not of a stereotypy-like activity. Pharmacological manipulations which deplete endogenous corticosterone, i.e., administration of the corticosterone synthesis inhibitor metyrapone (two injections, 150 and 50 mg/kg, given at 24 and 2 h …

St. John's Wort Extract Ze 117 and Escitalopram Alter Plasma and Hippocampal Lipidome in a Rat Model of Chronic-Stress-Induced Depression.

Chronic stress is a key factor in the development of depression. It leads to hyperactivation of the hypothalamic-pituitary-adrenal (HPA) axis, which in turn increases the formation of glucocorticoids (GCs). Chronically elevated GC levels disrupt neuroplasticity and affect brain lipid metabolism, which may, ultimately, contribute to the development of depression. This study aimed to investigate the effects of the antidepressants St. John's Wort extract and escitalopram on lipid metabolism in vivo. Therefore, repeated corticosterone injections were used to induce depression-like behavior in rats…

Systolic blood pressure was measured, using an indirect tail method, in conscious male rats at several time intervals after the intracerebro … ctive glucocorticoid agonist RU 28362 (10 ng) increased and decreased blood pressure, respectively. Corticosterone given intracerebroventricularly (10-100 ng) did not affect blood pressure unless the dose was increased to 1 microgram. Tyrosine hydroxylase-expressing neurons in the paraventricular nucleus of the hypothalamus regulate peripheral leukocytes during inflammatory pain.

BACKGROUND: People suffering from pain are more susceptible to infection due to the pain-induced alterations of immune function. The release of glucocorticoids (GCs) via the activation of the hypothalamus-pituitary-adrenal (HPA) axis has been considered as a major explanation for regulating glucose and energy mobilization, and immune function in response to stress. However, whether the abnormal immune function induced by inflammatory pain is associated with the excess release of corticosterone remains elusive. Central and peripheral neuronal circuits and immune organ innervation have been show…

Pineal and adrenal effects on calcium homeostasis in the rat.

In human infants and newborn rats, white light at the intensity used to treat hyperbilirubinemia lowers serum calcium concentration. Occipital shielding or (in newborn rats) exogenous melatonin prevents this effect. Propranolol, by inhibiting melatonin synthesis, also causes hypocalcemia, which is preventable by melatonin. Metyrapone or adrenalectomy prevents hypocalcemia after light exposure or propranolol. Exogenous corticosterone lowers serum calcium; this is prevented by supplementary melatonin. In adult rats, the change in calcium after light, propranolol, or corticosterone is minimal. Af…

[Mineralocorticoids in cervical mucous and female blood serum: levels in the preovulatory phase of the menstrual cycle].

Preovulatory cervical mucous of normal cycles was checked in 8 women for the presence of deoxycorticosterone (DOC), corticosterone and aldosterone. Cervical mucous DOC varied from lower to higher levels, and corticosterone concentration was several fold lower than in the blood serum. No aldosternone was detected in the cervical mucous. High cervical mucous DOC levels may indicate its synthesis by glandular cells of the cervix. Low corticosterone cervical mucous levels may indicate its transfer from the blood.

Influence of melatonin on rat thyroid, adrenals and testis secretion during the day.

The effect of pinealectomy and exogenous melatonin on circadian rhythm of triiodothyronine (T3), thyroxin (T4), corticosterone and testosterone in sham-operated and pinealectomized rats were investigated. The hormones concentration were RIA-measured and the circadian rhythm secretion were analysed by cosinor method. The findings suggest that pinealectomy abolishes the rhythmical character of corticosterone secretion and disturbs the circadian rhythm of T3, T4 and testosterone. Exogenous melatonin has the suppressive effect of diurnal secretion of T3, T4 and testosterone in pinealectomized rats…

Dopamine D2-like receptors modulate freezing response, but not the activation of HPA axis, during the expression of conditioned fear.

Considering the complexity of aversive information processing and defensive response expression, a combined action of stress modulators may be required for an optimal performance during threatening situations. Dopamine is now recognized as one of the most active modulators underlying states of fear and anxiety. On the other hand, activation of hypothalamic-pituitary-adrenocortical (HPA) axis, which leads to the release of corticosterone in rodents, has been considered a key part of the stress response. The current study is an extension of prior work investigating modulatory effects of dopamine…

Glucocorticoid metabolism and Na+ transport in chicken intestine.

The role of aldosterone in regulation of electrogenic Na+ transport is well established, though mineralocorticoid receptors bind glucocorticoids with similar binding affinity as aldosterone and plasma concentration of aldosterone is much lower than glucocorticoids. In mammals, the aldosterone specificity is conferred on the low-selective mineralocorticoid receptors by glucocorticoid inactivating enzyme 11beta-hydroxysteroid dehydrogenase (11HSD) that converts cortisol or corticosterone into metabolites (cortisone, 11-dehydrocorticosterone) with lower affinity for these receptors. The present s…

Sexual Differences in Behavior Patterns and Hormonal Balance under Normal and Mild Stress Conditions in Inbred Wistar Rats.

Sex differences were studied under normal conditions and after a single mild stress in inbred Wistar rats. It was found that intact females had higher blood corticosterone and leptin levels than males. In females, locomotor activity and the number of active defense reactions in elevated plus maze were higher, while the number of passive defense reactions was lower than in males. After stress, the blood corticosterone and leptin levels increased in both females and males, while the level of the main sex hormones decreased. We found for the first time that leptin and corticosterone levels positi…

Plasma steroid-binding globulin mediation of differences in stress reactivity in alternative male phenotypes in tree lizards, Urosaurus ornatus.

Plasma steroid-binding globulins, for example, corticosteroid-binding globulin and sex hormone-binding globulin (SHBG), have been identified … e and employ a satellite strategy. The two types of males have similar basal levels of total plasma corticosterone and testosterone. However, testosterone levels of nonterritorial males are more sensitive than those of territorial males to negative regulation by stress-induced increases in corticosterone. Effect of the renin-angiotensin system on the exacerbation of adrenal glucocorticoid steroidogenesis in diabetic mice: Role of angiotensin-II type 2 receptor.

Prior investigation shows an increase in the activity of both hypothalamus-pituitary-adrenal (HPA) axis and the renin-angiotensin system (RAS) in diabetic patients. Moreover, activation of angiotensin-II type 1 receptor (AT1) has been associated with adrenal steroidogenesis. This study investigates the role of RAS on the overproduction of corticosterone in diabetic mice. Diabetes was induced by intravenous injection of alloxan into fasted Swiss-webster mice. Captopril (angiotensin-converting enzyme inhibitor), Olmesartan (AT1 receptor antagonist), CGP42112A (AT2 receptor agonist) or PD123319 (…
