A Network Pharmacology Approach for Uncovering the Mechanism of 'Kouchuangling' in Radiation-induced Oral Mucositis Treatment.

BACKGROUND: Radiation-induced oral mucositis (RIOM) is an intractable inflammatory disease whose pathogenesis needs to be clarified. "Kouchuangling" (KCL), a traditional Chinese medicine formula, is composed of Lonicerae Japonicae Flos, Radix Paeoniae Rubra, and Radix Sanguisorbae. Although all of them are Chinese folk medicines which have long been utilized for ameliorating inflammation, the mechanism of KCL to RIOM remains unclear. PURPOSE: To predict the active ingredients of KCL and identify the mechanism of KCL on RIOM. MATERIALS AND METHODS: We identified the chemical ingredients in KC

Structure-based pharmacophore modeling, virtual screening, and molecular dynamics simulation studies for identification of Plasmodium falciparum 5-aminolevulinate synthase inhibitors.

Plasmodium falciparum (Pf) 5-aminolevulinic acid synthase (5-ALAS) is an essential enzyme with high selectivity during liver stage development, signifying its potential as a prophylactic antimalarial drug target. The aim of this study was to identify important potential lead compounds which can serve as inhibitors of Pf 5-ALAS using pharmacophore modeling, virtual screening, qualitative structural assessment, in silico ADMET (Absorption, Distribution, Metabolism, Excretion and Toxicity) evaluation and molecular dynamics simulation. The best model of the tertiary structure of Pf 5-ALAS was obta

Drug-induced liver injury: a real-world pharmacovigilance study using the FDA Adverse Event Reporting System database (2004-2024).

INTRODUCTION AND OBJECTIVES: Timely identification and discontinuation of culprit-drug is the cornerstone in managing drug-induced liver injury (DILI), and this study aims to thoroughly investigate the potential hepatotoxic drugs by using the FDA Adverse Event Reporting System (FAERS) database. MATERIALS AND METHODS: DILI-related reports in the FAERS database between 2004 and 2024 were used to compile a reported culprit-drug list, and the single drug and drug class with the highest reporting frequency were counted accordingly. The disproportionality analysis was used to assess the DILI risk o

Drug-induced hepatic injury in children: a case/non-case study of suspected adverse drug reactions in VigiBase.

AIM: To identify which drugs are associated with reports of suspected hepatic injury in children and adolescents. METHODS: Using a worldwide pharmacovigilance database, VigiBase, we conducted a case/non-case study on suspected adverse drug reactions (ADRs) occurring in the population <18 years old. Cases were all the records with hepatic ADRs and non-cases were all the other ADR records. Records regarding topically administered drugs were excluded from both groups. The association between drug and suspected hepatic ADRs was calculated using the reporting odds ratio (ROR) as a measure of dispr

Prediction of acute toxicity in fish by using QSAR methods and chemical modes of action.

Three quantitative structure-activity relationship (QSAR) models were evaluated for their power to predict the toxicity of chemicals in two datasets: (1) EPAFHM (US Environmental Protection Agency-Fathead Minnow) and (2) derivatives having a high production volume (HPV), as compiled by the European Chemical Bureau. For all three QSAR models, the quality of the predictions was found to be highly dependent on the mode of action of the chemicals. An analysis of outliers from the three models gives some clues for improving the QSAR models. Two classification methods, Toxtree and a Bayesian approac

Computer simulation of heterogeneous single nucleotide polymorphisms in the catalase gene indicates structural changes in the enzyme active site, NADPH-binding and tetramerization domains: a genetic predisposition for an altered catalase in patients with vitiligo?

Patients with vitiligo have low levels/activities of catalase in their lesional and non-lesional epidermis as well as in their epidermal melanocytes under in vitro conditions while the levels of catalase mRNA are unaltered. This defect leads to a build-up of hydrogen peroxide (H(2)O(2)) in the 10(-3) m range in the epidermis of these patients. In this context, it was realized that 10(-3) m H(2)O(2) deactivates catalase. Along this line, it was also suspected that catalase in patients with vitiligo possesses a special sensitivity to this reactive oxygen species (ROS), and indeed several heteroz

A multiparametric pharmacogenomic strategy for drug repositioning predicts therapeutic efficacy for glioblastoma cell lines.

BACKGROUND: Poor prognosis of glioblastoma patients and the extensive heterogeneity of glioblastoma at both the molecular and cellular level necessitates developing novel individualized treatment modalities via genomics-driven approaches. METHODS: This study leverages numerous pharmacogenomic and tissue databases to examine drug repositioning for glioblastoma. RNA-seq of glioblastoma tumor samples from The Cancer Genome Atlas (TCGA, n = 117) were compared to "normal" frontal lobe samples from Genotype-Tissue Expression Portal (GTEX, n = 120) to find differentially expressed genes (DEGs). Limited representation of drinking-water contaminants in pregnancy-birth cohorts.

Water contamination and noise have been consistently the least assessed environmental/lifestyle exposures in pregnancy-birth cohorts (PBC). Water quality surveillance data collected during the past decade within urban drinking-water distribution systems call for re-evaluation of water and health issues in the developed world. The objectives of this scientific commentary were to (i) highlight the extent of appraisal of water contamination in exposure assessment studies of PBC, worldwide, and (ii) propose recommendations to increase awareness of emerging water-related risks through their improve

Metabolic biomarkers of prenatal alcohol exposure in human embryonic stem cell-derived neural lineages.

BACKGROUND: Fetal alcohol spectrum disorders (FASD) are a leading cause of neurodevelopmental disability. The mechanisms underlying FASD are incompletely understood, and biomarkers to identify those at risk are lacking. Here, we perform metabolomic analysis of embryoid bodies and neural lineages derived from human embryonic stem (hES) cells to identify the neural secretome produced in response to ethanol (EtOH) exposure. METHODS: WA01 and WA09 hES cells were differentiated into embryoid bodies, neural progenitors, or neurons. Cells along this progression were cultured for 4 days with 0, 0.1,

Fermentation-induced molecular remodeling in African indigenous tubers: cassava and cocoyam.

Cassava and cocoyam are major dietary staples in sub-Saharan Africa, commonly processed by natural fermentation before consumption. Although fermentation reduces antinutritional compounds and improves food quality, its molecular effects remain poorly characterized. We used untargeted mass spectrometry-based metabolomics with a computational annotation pipeline to compare fermentation-induced molecular remodeling in the two tubers, which showed distinct responses. In cassava, 718 of 773 significant features (92.9%) were decreased, indicating a predominantly catabolic process. In cocoyam, the re

Durability Degradation Mechanisms and Coupled Deterioration Behaviors of Geopolymer Concrete Under Multi-Hazard Service Environments: A Critical Review.

Geopolymer concrete (GPC), here defined primarily as concrete based on low-calcium alkali-activated aluminosilicate binders, exhibits durability responses that depend strongly on precursor chemistry, gel assemblage, pore structure and exposure conditions; high-calcium or slag-rich systems are therefore discussed separately as related alkali-activated material (AAM) systems. This critical review synthesizes representative peer-reviewed evidence on freeze-thaw cycling, drying and autogenous shrinkage, elevated-temperature/fire exposure, chloride, sulfate and acid attack, carbonation, reinforceme

Definition of Metabolite: Size as a Critical Criterion.

Metabolites have traditionally been defined as organic molecules smaller than 1500 daltons (Da). However, recent advances in analytical technologies and chemoinformatics have uncovered a wider chemical diversity, including biologically significant metabolites exceeding this conventional size cutoff─such as polypeptides, glycosphingolipids, and bacterial lipopolysaccharides. Detecting these larger metabolites challenges standard metabolomics approaches and necessitates optimized mass spectrometry acquisition parameters. In this perspective, the analysis of multiple databases (HMDB blood, GNPS,

Natural radioactivity in mineral phosphate fertilizers and its impacts on human health: an overview.

Humans are constantly exposed to radioactivity present in rocks, soils, and water, mainly from materials in the Earth's crust that contain chemical elements belonging to the radioactive series of uranium and thorium. An important anthropogenic source of these natural radioisotopes to the environment is fertilizers, widely used to increase agricultural productivity. Exposure to ionizing radiation can become a public health problem worldwide, since it is related to the development of different cancers in humans. The present study aimed to survey research on the radioactive content in different t
