Exosome nanotechnology: an emerging paradigm shift in drug delivery: exploitation of exosome nanovesicles for systemic in vivo delivery of RNAi heralds new horizons for drug delivery across biological barriers.

The demonstration that dendritic cell (DC)-derived exosomes can be exploited for targeted RNAi delivery to the brain after systemic injection provides the first proof-of-concept for the potential of these naturally occurring vesicles as vehicles of drug delivery. As well as being amenable to existing in vivo targeting strategies already in use for viruses and liposomes, this novel approach offers the added advantages of in vivo safety and low immunogenicity. Fulfilment of the potential of exosome delivery methods warrants a better understanding of their biology, as well as the development of n

Engineering Oxidative Stress for Precision Therapy: Reactive Oxygen Species-Responsive Hydrogel-Nanocarrier Systems as Translational Redox Drug-Delivery Platforms.

Reactive oxygen species (ROS) are regulated chemical signals that become pathological when production exceeds antioxidant buffering. This duality creates a translational drug-delivery opportunity: therapeutic systems can be engineered to remain stable under basal redox conditions while activating in oxidatively stressed tissues. This Review critically evaluates ROS-responsive hydrogel-nanocarrier systems as multiscale platforms for localized redox therapy. The discussion is organized around quantitative and clinically relevant design logic rather than broad cataloging. We first define hydrogel

Comparative analysis between peristaltic and syringe nebulization systems: Advancing high-pressure drug delivery for pressurized intraperitoneal aerosol chemotherapy (PIPAC).

BACKGROUND: The high-pressure syringe pump (HP-SP) is the most widely applied device for PIPAC. However, HP-SP systems exhibited several limitations: relatively more drug assembly procedures and significant start-up delays. Herein, the performance of a high-pressure peristaltic pump (HP-PP) was compared to the existing HP-SP for PIPAC. METHODS: Mechanical and ex vivo experiments were conducted to compare the performance of the HP-PP and HP-SP for PIPAC. The evaluated parameters included (1) particle size distribution, operational pressure, and its influence on particle size distribution in th

Toward oral nanomaterial-based drug delivery systems for hepatocellular carcinoma therapy: evidence mapping, route-specific validation, and translational challenges.

Hepatocellular carcinoma (HCC) remains a major cause of cancer-related mortality worldwide, and current systemic therapies are limited by advanced-stage diagnosis, dose-limiting toxicity, drug resistance, and incomplete response rates. Oral nano-drug delivery systems (nano-DDS) are being explored as patient-friendly platforms to improve gastrointestinal protection, intestinal absorption, and hepatic exposure of anticancer agents. However, the evidence base remains uneven: only a minority of HCC nano-DDS studies have been validated through oral administration, whereas many mechanistically impor…

Oral Lipid Nanoparticles for Improving the Efficiency of Drug Delivery Systems in Ulcerative Colitis: Recent Advances and Future Prospects.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by persistent, recurrent, and relapsing inflammation of the mucosal layer. Its pathogenesis is complex and not yet fully understood, with current treatments mainly focused on alleviating symptoms through pharmacological methods. Direct drug administration for UC often leads to poor intestinal bioavailability, suboptimal targeting, and an increased risk of resistance. Therefore, there is an urgent need for effective drug delivery systems. Lipid nanoparticles (LNPs) are promising candidates for UC drug delivery due to …
