Development of a chimeric multi-epitope vaccine targeting surface-exposed proteins of Chlamydia psittaci through immunoinformatics and structural vaccinology.

Chlamydia psittaci is an obligate intracellular zoonotic pathogen that causes psittacosis and severe respiratory illness in humans. Despite its clinical importance, no licensed vaccine is currently available, and antibiotic treatment may not provide long-term protection or prevent recurrent infection. To address this challenge, a computational strategy integrating structural vaccinology and immunoinformatics was used to design a multi-epitope vaccine targeting the major outer membrane porin and the virulence-associated pGP3-D protein of C. psittaci. Cytotoxic T-cell and helper T-cell epitopes

Immunoinformatics approach: Developing a multi-epitope vaccine with novel carriers targeting monkeypox virus.

Since May 2022, the global spread of monkeypox virus (MPXV) has presented a significant threat to public health. Despite this, there are limited preventive measures available. In this study, different computational tools were employed to design a multi-epitope vaccine targeting MPXV. Three key MPXV proteins, M1R, B6R, and F3L, were chosen for epitope selection, guided by bioinformatic analyses to identify immunodominant epitopes for T- and B-cell activation. To enhance immune stimulation and facilitate targeted delivery of the vaccine to specific cells, the selected epitopes were linked to nov

Computational Design and Preliminary Serological Analysis of a Novel Multi-Epitope Vaccine Candidate against Onchocerciasis and Related Filarial Diseases.

: Onchocerciasis is a skin and eye disease that exerts a heavy socio-economic burden, particularly in sub-Saharan Africa, a region which harbours greater than 96% of either infected or at-risk populations. The elimination plan for the disease is currently challenged by many factors including amongst others; the potential emergence of resistance to the main chemotherapeutic agent, ivermectin (IVM). Novel tools, including preventative and therapeutic vaccines, could provide additional impetus to the disease elimination tool portfolio. Several observations in both humans and animals have provided

In silico design and experimental validation of a multi-epitope vaccine candidate against Helicobacter hepaticus associated chronic liver inflammation.

BACKGROUND: Helicobacter hepaticus is an emerging enterohepatic pathogen associated with chronic gastrointestinal and hepatic inflammatory disorders. Despite its clinical relevance, effective vaccination strategies remain unavailable due to antigenic variability, immune evasion, and limited induction of durable cellular immunity. METHODS: An integrated reverse vaccinology and experimental validation approach was employed to design a multi-epitope vaccine against H. hepaticus. Two conserved outer membrane proteins, BamA and an organic solvent tolerance protein, were selected based on antigenic

In silico design and testing of a multi-epitope camel mastitis vaccine candidate for development.

INTRODUCTION: Camel mastitis caused by a diverse array of pathogenic bacteria including Staphylococcus aureus and Streptococcus agalactiae is responsible for enormous socioeconomic losses in the arid and semi-arid regions of Africa where camels are an invaluable source of livelihood and nutrition. Although S. aureus and S. agalactiae are environmental contaminants, the latter is an infectious zoonotic agent. The development of a safe, efficacious vaccine for camel mastitis is a compelling priority particularly against the backdrop of climate change and antimicrobial resistance. METHODS: We ha

Integrated in-silico design and in vivo validation of multi-epitope vaccines for norovirus.

BACKGROUND: Norovirus (NoVs) is a foodborne pathogen that causes acute gastroenteritis. The diversity of its principal antigenic protein poses a significant challenge to vaccine development and the prevention of large-scale outbreaks globally. Currently, no licensed vaccines against norovirus have been approved. METHODS: We developed a novel pipeline that integrates multiple bioinformatics tools to design broad-spectrum vaccines against NoVs. Specifically, broad-spectrum T-cell epitope vaccines were designed based on consensus sequences and optimized epitope screening, while broad-spectrum B-

In silico design of a multi-epitope vaccine against Cryptosporidium parvum using structural and immunoinformatics approaches.

BACKGROUND: Cryptosporidium parvum is a waterborne protozoan parasite responsible for diarrheal illness in humans and animals. The lack of effective vaccines and the emergence of antimicrobial resistance underscore the urgent need for novel prophylactic strategies. METHODS: A structure-based immunoinformatics approach was used to design a multi-epitope subunit vaccine (MESV) targeting immunogenic regions of C. parvum. Three proteins, Cp15, Cp23, and CpP2, were selected based on antigenicity, non-allergenicity, non-homology with host proteins, and absence of transmembrane domains. B-cell, CD4+

Variants of two major T cell epitopes within the hepatitis B surface antigen are not recognized by specific T helper cells of vaccinated individuals.

Several naturally occurring variants of immunogenic T cell epitopes were identified within the hepatitis B surface antigen (HBsAg). The effect of these variants on the cellular immune response was studied in individuals vaccinated against HBV. Class-II restricted T-cell responses of 30 vaccinees were analyzed after stimulation of peripheral blood mononuclear cells (PBMCs) with 4 synthetic peptides representing the 4 T-cell epitopes of HBsAg known as of yet. The 2 epitopes P1 (aa 16-33) and P4 (aa 213-226) could be identified as the dominant ones in our vaccinees by proliferation assays and enz

Identification of human leucocyte antigen (HLA)-A*0201-restricted cytotoxic T lymphocyte epitopes derived from HLA-DOβ as a novel target for multiple myeloma.

Despite the recent development of effective therapeutic agents against multiple myeloma (MM), new therapeutic approaches, including immunotherapies, remain to be developed. Here we identified novel human leucocyte antigen (HLA)-A*0201 (HLA-A2)-restricted cytotoxic T lymphocyte (CTL) epitopes from a B cell specific molecule HLA-DOβ (DOB) as a potential target for MM. By DNA microarray analysis, the HLA-DOB expression in MM cells was significantly higher than that in normal plasma cells. Twenty-five peptides were predicted to bind to HLA-A2 from the amino acid sequence of HLA-DOB. When screened

Receptors for toxic shock syndrome toxin-1 and staphylococcal enterotoxin A on human blood monocytes.

Staphylococcal toxic shock syndrome toxin-1 (TSST-1) as well as staphylococcal enterotoxin A (SEA) and B (SEB) have recently been shown to bind directly to the class II major histocompatibility antigen, HLA-DR. Whereas others have characterized TSST-1 and SEA binding to HLA-DR on transfected L cells or B lymphoma cell lines, we sought evidence for direct binding of TSST-1 and SEA to HLA-DR on purified human monocytes. A single class of high-affinity receptors was found for both TSST-1 (dissociation constant (Kd) 40 nM, 3.4 x 10(4) receptors per cell) and SEA (Kd 12 nM, 3.2 x 10(4) receptors pe
