Mutations in exons 5-8 of the p53 gene, independent of their type and location, are associated with increased apoptosis and mitosis in invasive breast carcinoma.

In breast cancer, mutations located in the zinc-binding functional domains of the p53 gene have been reported to predict a worse prognosis and a worse response to treatment with doxorubicin, compared with mutations in other parts within exons 5-8 of the gene. Similarly, mutations in residues of p53 that directly contact DNA have been associated with a poor prognosis. To investigate whether these specific p53 mutations are associated with differences in the rate of apoptosis and/or mitosis, or expression of the anti-apoptotic Bcl-2 protein, these parameters were evaluated in 89 invasive breast …

Reduced induction of P53 protein by gamma-irradiation in ataxia telangiectasia cells without constitutional mutations in exons 5, 6, 7, and 8 of the p53 gene.

Ataxia telangiectasia (AT) is an autosomal recessive disease of childhood with several phenotypic characteristics. One of the hallmarks of t … d. We used the PCR amplification and DNA sequencing methods to detect mutations in the hypermutable exons (5-8) of germline p53 in fibroblast cells from 3 AT homozygotes. No mutation was found in any of these exons. In order to determine the role of the p53 protein in G1 arrest, its levels were measured before and after gamma-irradiation by flow cytomet …

Clustering of FBN2 mutations in patients with congenital contractural arachnodactyly indicates an important role of the domains encoded by exons 24 through 34 during human development.

Congenital contractural arachnodactyly (CCA) is an autosomal dominant condition phenotypically related to Marfan syndrome (MFS). CCA is caused by mutations in FBN2, whereas MFS results from mutations in FBN1. FBN2 mRNA extracted from 12 unrelated CCA patient cell strains was screened for mutations, and FBN2 mutations were identified in six of these samples. All of the identified FBN2 mutations cluster in a limited region of the gene, a region where mutations in FBN1 produce the severe, congenital form of MFS (so-called neonatal MFS). Furthermore, three of the identified mutations occur in the

SIRT1 Exon 2-Encoded IDR Gates CREB-Dependent Transcriptional Timing During Fasting.

Metabolic adaptation to fasting requires precise temporal control of gene expression. Although transcription factors and upstream co-regulators that mediate the fasting response are well studied, the role of intrinsically disordered regions in coordinating transcriptional timing remains unclear. Here, we identify the exon 2-encoded intrinsically disordered region of SIRT1 as a regulator of fasting-responsive transcription in the liver. Using a physiological SIRT1ΔE2 mouse model, we show that loss of this region alters the sequence and magnitude of starvation-induced transcriptional responses,

Furmonertinib in lung adenocarcinoma with EGFR exon 20 insertion mutation: A case report of positive outcome.

RATIONALE: epidermal growth factor receptor (EGFR) mutations play a pivotal role in non-small cell lung cancer (NSCLC), with the EGFR exon 20 insertion (EGFR20ins) mutation being the third most prevalent alteration in the EGFR gene. Patients harboring this mutation typically exhibit a greater tumor burden and poorer overall prognosis. This mutation is strongly associated with resistance to EGFR-TKIs, presenting significant challenges for clinical management. Furmonertinib, an innovative third-generation EGFR-TKI, has been validated in numerous trials for its effectiveness in targeting the EGFR

Improving angiogenesis ameliorates the efficacy of ASO-based exon skipping for the treatment of Duchenne muscular dystrophy.

Duchenne muscular dystrophy (DMD) is a severe X-linked disease caused by pathogenic variants in the DMD gene, resulting in the absence of functional dystrophin. Antisense oligonucleotide (ASO)-based therapies aim to restore the open reading frame and produce a truncated but functional dystrophin protein. Although several ASOs are approved in the United States and Japan via accelerated approval procedures, dystrophin restoration in patient biopsies remains low, underlining the need to improve ASO potency. One major limitation is poor ASO biodistribution to skeletal muscle, influenced by both AS

Evaluation of pediatric FMF patients with biallelic pathogenic exon 10 variants: focus on chest pain.

UNLABELLED: Chest pain occurs in approximately one-quarter of patients with FMF, the most common monogenic autoinflammatory disease characterized by recurrent attacks of fever and serositis. This study evaluated a large, genetically homogeneous pediatric FMF cohort carrying biallelic pathogenic exon 10 MEFV variants to describe their demographic, clinical, and genetic characteristics, and to compare patients with and without chest pain to better define this manifestation within the FMF spectrum. This cross-sectional single-center study included 918 pediatric FMF patients with biallelic pathoge

Tumor patterns and cancer risk in carriers of TP53 exonic germline variants that alter mRNA splicing.

Abnormal RNA splicing is an underrecognized driver of pathogenicity in germline TP53 - the cause of Li-Fraumeni syndrome (LFS). We re-evaluated exonic single-nucleotide variants (SNVs) that yield missense or synonymous changes for spliceogenic effects by integrating SpliceAI prediction, in-vitro minigene assays, and analysis of tumor RNA-seq from TCGA, and assessed genotype-phenotype correlations using clinical data from multiple databases and national registries. We identified 58 spliceogenic exonic SNVs (SE-SNVs) across the TP53 gene (40 missense, 18 synonymous). Experimental validation conf

Rapidly progressive steroid-resistant focal segmental glomerulosclerosis associated with an INF2 exon 6 variant.

Variants in the inverted formin-2 (INF2) gene are a known cause of hereditary focal segmental glomerulosclerosis (FSGS) and Charcot-Marie-Tooth disease. We report a case of rapidly progressive FSGS associated with a rare INF2 variant. A 12-year-old boy developed proteinuria and was diagnosed with FSGS at age 14 following a renal biopsy. Steroid therapy and subsequent immunosuppressive treatments, including plasma exchange, were ineffective. At age 15, a heterozygous missense variant in exon 6 of the INF2 gene (c.763G>A, p.Asp255Asn) was identified. Despite conservative management, the patient

Cholesterol Conjugation Strategies to Enhance In Vivo Antisense Oligonucleotide-Mediated Exon Skipping in the Mouse Retina.

PURPOSE: Antisense oligonucleotide (AON)-mediated exon skipping is a potential therapeutic approach to certain inherited retinal diseases, including Usher's Syndrome Type 2 A. Heteroduplex AONs (HAONs) have been reported to enhance RNA-modulating activity in vivo; this study evaluated whether the HAON strategy improves Ush2a exon skipping activity in the mouse retina. METHODS: AONs of different chemical makeup targeting Ush2a exon 12 were formulated as HAONs with and without lipid conjugation. Activity was evaluated in 2D and 3D cell models and in the mouse eye following intravitreal administ

Evaluating exon skipping in the central nervous system in Duchenne muscular dystrophy using spatial transcriptomics.

Duchenne muscular dystrophy (DMD) causes progressive muscle degeneration due to dystrophin deficiency. Dystrophin is also expressed in the brain during development and postnatally, yet a characterization of dystrophin isoform expression across brain cells and regions is lacking, limiting our understanding of the cognitive impairment affecting one-third of the patients and hampering the development of dystrophin-restoring drugs in the central nervous system (CNS). Here, we applied spatial transcriptomics to map Dmd isoforms across mouse brain regions and cell types. Mdx52 mice received exon 51-

Microglial TDP-43 mediates myelin refinement and represses Tyrobp cryptic exon inclusion in mice.

TDP-43 proteinopathy is a hallmark of neurodegenerative disorders such as amyotrophic lateral sclerosis and frontotemporal dementia where mislocalization of TDP-43 has been observed in neurons and glial cells. However, the role of TDP-43 in microglia and the consequences of its loss of function remain unexplored. Combining magnetic resonance imaging, and confocal, and electron microscopy, we uncovered structural changes and myelin abnormalities in the early postnatal brain of mice lacking microglial TDP-43. Spatial transcriptomics further revealed an enriched interferon-responsive signature as

HNRNPF-driven retention of exon 14 in HIF1A pre-mRNA promotes breast cancer metastasis.

Hypoxia-inducible factor 1-alpha (HIF1A) is a core regulator of cellular adaptation to hypoxic environments and is extensively involved in various cancer processes. Although it is known that HIF1A produces two isoforms, HIF1A-L and HIF1A-S, through the alternative splicing of exon 14, the specific functional differences between these isoforms in cancer development and progression remain unclear, and the molecular mechanisms regulating this exon skipping event have yet to be elucidated. Clinical IHC and FISH analyses demonstrate that HIF1A-L expression is elevated in high-grade breast cancer ti

SLC31A1 exon 1 methylation reduces intracellular copper ion and promotes diabetic cardiac fibrosis.

BACKGROUND: Metal homeostasis is linked to cellular organelles modification. Dynamic lipid droplet (LD)-mitochondria interactions contribute to diabetes-related complications. However, little is known about whether and how copper regulates LD-mitochondria contacts in diabetic cardiac fibrosis (CF). METHODS: Leptin receptor-deficient (db/db) mice were used to induce diabetic CF. To investigate target gene function in vivo, AAV9 vectors carrying the cardiac fibroblast-specific POSTN promoter-driven small hairpin RNAs were administered. SLC31A1 overexpression was achieved via AAV9 plasmid specifi

Generation of a novel mouse model of nemaline myopathy due to recurrent NEB exon 55 deletion.

Biallelic pathogenic variants in the nebulin (NEB) gene lead to the congenital muscle disease nemaline myopathy. In-frame deletion of exon 55 (ΔExon55) is the most common disease-causing variant in NEB. Previously, a mouse model of NebΔExon55 was developed; however, it presented an uncharacteristically severe phenotype with a near complete reduction in Neb transcript expression that is not observed in NEB exon 55 patients. We identified by RNA sequencing that the cause of this unexpectedly severe presentation in mice is the generation of a pseudoexon containing two premature termination codons
