BACKGROUND: Children whose mothers consumed alcohol during pregnancy exhibit widespread brain abnormalities and a complex array of behavioral disturbances. Here, we used a mouse model of fetal alcohol exposure to investigate relationships between brain abnormalities and specific behavioral alterations during adulthood.
RESULTS: Mice drank a 10% ethanol solution throughout pregnancy. When fetal alcohol-exposed offspring reached adulthood, we used high resolution MRI to conduct a brain-wide screen for structural changes and found that the largest reduction in volume occurred in the olfactory bu

Alcohol-exposed pregnancies are especially of concern for American Indians. The Indian Health Service reported that 47% to 56% of pregnant patients admitted to drinking alcohol during their pregnancy. In addition, rates of Fetal Alcohol Syndrome are estimated to be as high as 3.9 to 9.0 per 1,000 live births among American Indians in the Northern Plains, making prevention of alcohol-exposed pregnancies an important public health effort for this population. The goal of this article is to add to the literature on universal prevention of Fetal Alcohol Spectrum disorders by describing the developm

PURPOSE: In the human body, the orbital muscle is a limited smooth-muscle tissue extending between hard tissues. To provide better understanding of its function, the authors re-examined its development in fetuses.
METHODS: Using 20 human fetuses (12-25 weeks of gestation), semiserial horizontal or sagittal paraffin sections were prepared at intervals of 20 to 100 μm. In addition to routine histology, the authors performed silver staining as well as immunohistochemistry for alpha smooth-muscle actin (SMA), vimentin, S100 protein, and tyrosine hydroxylase.
RESULTS: Up to 12 weeks, the orbital

BACKGROUND: The 20th week gestational age (GA) is at mid-gestation and corresponds to the age at which the termination of pregnancy in several countries and the first Magnetic Resonance Imaging (MRI) can be performed, and at which the premature babies may survive. However, at present, very little is known about the exact anatomical character at this GA.
OBJECTIVE: To delineate the developing fetal brain of 20 weeks GA and obtain the three dimensional visualization model.
MATERIALS AND METHODS: 20 fetal specimens were scanned by 3.0 T and 7.0 T post-mortem MRI, and the three dimensional visua

Fetal alcohol syndrome is a serious developmental disorder and exposure of the fetal heart to alcohol results in disturbances in the biochemistry of all cellular substructures. Mitochondrial effects include diminished respiratory function and physical alteration of the membrane secondary to interaction of ethanol with the hydrophobic region of the bilayer. Cardiolipin is a major mitochondrial membrane phospholipid in the heart and plays an important role in the function of mitochondrial enzymes involved in cellular respiration. We examined the activity of cardiac monolysocardiolipin acyltransf

Prenatal standards of bi-iliac width were not found in the literature based on autopsy investigations, nor was the caudo-cranial position of … The caudo-cranial position of the iliac bones was evaluated. The present study shows that in normal fetal development there is a continuous linear enlargement of the pelvic region in the transverse and vertical planes. The upper iliac contour stays at the level of the first sacral vertebral body, whereas the lower iliac line moves caudally. Significant differences …

During three and a half years we observed 83 single pregnancies all delivered by caesarean section. All of them had a mild (systolic blood pressure > 140, diastolic > 90 and proteinuria > 0.5 g/dl) or a severe preeclampsia (systolic blood pressure > 160, diastolic > 100 and proteinuria > 3.0 g/dl). We found significantly twice as many abnormal uteroplacental blood flow velocities in the severe preeclampsia group than in the mild one. These results draw us to the conclusion that possible pathological changing of the vessels is due to preeclampsia which does not need to correlate with a placenta

The transverse growth of long bones during intrauterine development was studied in rat fetuses subjected to experimental oligohydramnios in order to determine whether the skeletal changes, if any, in extrinsic fetal akinesia were similar to those observed in curarized rat fetuses with the fetal akinesia deformation sequence. Oligohydramnios was induced by daily extraction of amniotic fluid from day 17 of gestation until term. Experimental fetuses were compared with a sham-operated control group. The total area and perimeter, the absolute and relative amount of periosteum and bone trabeculae, t

The effect of thyroid hormone on maturation of fetal rabbit lung was studied with maternal treatment using 3,5-dimethyl-3'-isopropyl-L-thyronine (DIMIT), a synthetic analogue of triiodothyronine. To investigate the in vivo kinetics and distribution of DIMIT, we prepared [3H]DIMIT and injected both pregnant rats (18-21 d gestation) and rabbits (25 d gestation). In the rat, maximal concentrations of radioactivity in maternal plasma, fetal plasma, and amniotic fluid occurred within 10 min, 1-2 h, and 4-6 h, respectively, after intramuscular injection. After 7 h the concentration of radioactivity

Evidence from epidemiologic, clinical, and experimental studies has shown that a suboptimal intrauterine environment during early pregnancy can alter fetal growth and gestation length and is associated with an increased prevalence of adult hypertension and cardiovascular disease. It has been postulated that maternal nutrient restriction may act to reprogram the development of the pituitary-adrenal axis, resulting in excess glucocorticoid exposure and adverse health outcomes in later life. It is unknown, however, whether maternal nutrient restriction during the periconceptional period alters th

BACKGROUND: Fetal hypoxia is a leading cause of neonatal morbidity and mortality. Cardiotocography (CTG) is widely used to predict fetal hypoxia during labor, but its interpretation remains suboptimal. Artificial intelligence (AI) models have been developed for CTG interpretation, but their clinical utility is limited by two major challenges: demonstrating superiority over human experts and ensuring explainability in real-world settings.
METHODS: A large dataset containing CTG traces from three tertiary hospitals between January 2014 and May 2022 was built for model development. Deep learning

INTRODUCTION: The FTO gene is strongly linked to obesity, but its mechanisms in fetal growth and placental function under maternal obesity remain unclear. This study investigated FTO-related biological processes in the placental transcriptome and fetal development using a diet-induced obesity model.
METHODS: Swiss mice were fed a 45% high-fat diet for 4 weeks and classified as obesity-prone (HFP) or obesity-resistant (HFR). Transcriptomic analysis identified differentially expressed genes (DEGs) in placentas. Maternal and fetal parameters were correlated with placental FTO expression, and in …

This study examined the adverse impacts of furan, a pervasive environmental toxin, on fetal development in Wistar rats. Twenty-eight pregnant rats (seven animals per group) were orally administered different doses of furan (18 and 28 mg/kg), distilled water, or corn oil as vehicles from gestation days 6-12, and their progeny at gestation day 20 were assessed for developmental anomalies, oxidative stress, apoptosis, and placental impairment. The findings indicated substantial decreases in fetal weight, length, and survival rates after furan exposure. Histopathological analyses revealed damage t…

Understanding the timing of fetal brain vulnerability to inflammatory changes in pregnancy complications is crucial for predicting neurodevelopmental risks. Beyond the placenta, the developing brain's vascular system is believed to form a secondary defense, the blood-brain barrier (BBB), which restricts harmful substances that could disrupt neurodevelopment. However, the precise timing and mechanisms underlying BBB development are poorly understood. In this study, we examined the spatiotemporal expression of key BBB components and fetal brain vascularization in mice from gestational days (GD)

Maternal proline (Pro) supplementation enhances fetal survival and placental development in mice. However, the effect of Pro on fetal and placental development in gilts remains to be investigated, particularly in the context of obesity-induced impaired pregnancy. Here, we investigated the effect of dietary Pro on fetal and placental development in obese gilts. Exp.1: On day 60 of gestation, 48 gilts with similar delivery times were selected and followed up until delivery to determine the relationship between maternal obesity, litter performance, and Pro abundance in term placentae. The results

Human cerebellum encompasses numerous neurons, exhibiting a distinct developmental paradigm from cerebrum. Here we conducted scRNA-seq, scATAC-seq and spatial transcriptomic analyses of fetal samples from gestational week (GW) 13 to 18 to explore the emergence of cellular diversity and developmental programs in the developing human cerebellum. We identified transitory granule cell progenitors that are conserved across species. Special patterns in both granule cells and Purkinje cells were dissected multidimensionally. Species-specific gene expression patterns of cerebellar lobes were character

Prenatal alcohol exposure (PAE) and prenatal stress (PS) are highly prevalent conditions known to affect fetal programming of the hypothalamic-pituitary-adrenal (HPA) axis. The objectives of this study were to assess the effect of light PAE, PS, and PAE-PS interaction on fetal HPA axis activity assessed via placental and umbilical cord blood biomarkers. Participants of the ENRICH-2 cohort were recruited during the second trimester and classified into the PAE and unexposed control groups. PS was assessed by the Perceived Stress Scale. Placental tissue was collected promptly after delivery; gene

Successful embryo implantation relies on a receptive endometrium and a maternofetal dialogue. Abnormal receptivity is a common cause of implantation failure in assisted reproductive techniques. This study aimed to develop a novel transcriptomic-based diagnostic assay, Adhesio, for assessing endometrial receptivity and guiding personalized embryo transfer. Adhesio was developed based on an initial dataset of 74 endometrial biopsies. Two types of biopsy samples were involved: 45 endometrial biopsies collected during the optimal theoretical window of implantation (WOI) and 29 endometrial biopsies
