Understudied PFAS precursors dominate household dust: insights from a pilot study in Rochester, NY.

BACKGROUND: Indoor exposure to per- and polyfluoroalkyl substances (PFAS) may contribute substantially to total PFAS exposure, yet few studies characterize indoor concentrations of precursor PFAS. OBJECTIVE AND METHODS: We conducted a pilot study targeting 46 ionic and neutral PFAS in vacuumed dust from 20 low-income owner-occupied homes in Rochester, New York. RESULTS: Forty-three PFAS were detected; polyfluoroalkyl phosphate esters (PAPs), fluorotelomer alcohols (FTOHs), and perfluorocarboxylic acids (PFCAs) were detected in every sample. The highest median concentrations were 6:2 diPAP (7

Prospector enables defined enriched consortium selection for PFAS degradation via defluorination and Chain-Shortening.

Per- and polyfluoroalkyl substances (PFAS) are recalcitrant environmental contaminants whose biodegradation remains challenging due to the exceptional stability of carbon-fluorine bonds. Here, we employed Prospector® nanowell array technology to isolate enriched bacterial consortia from cotton detritusphere capable of degrading perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) as selective growth-promoting substrates. Screening of 96,000 nanowells across four inoculum-loading regimes identified high-density loading (20-33 genera per community) as the sole condition that yielde

PFAS and One Health: integrating human, animal, and environmental perspectives.

Per- and polyfluoroalkyl substances (PFAS)-ubiquitous, persistent, and bioaccumulative-pose a cross-cutting threat to human, animal, and environmental health. Their resistance to degradation and global dissemination demand an integrated One Health framework to address their complex, interdependent risks across ecosystems, wildlife, and public health. This review synthesizes current knowledge on PFAS, highlighting their anthropogenic origins, environmental persistence, and global dispersion. Common exposure pathways, such as dietary intake, contaminated drinking water, and maternal transfer, le

Tracking the impact of PFAS regulations: Long-Term analysis of roe deer livers from 1998 until 2022.

PER: and polyfluoroalkyl substances (PFAS) are persistent environmental pollutants with widespread occurrence and bioaccumulation potential. Regulatory measures have been introduced to limit the use of certain PFAS, yet their presence remains detectable in wildlife. This study examines long-term trends in PFAS concentrations in one-year-old roe deer livers (n = 28) collected from the Bavarian Forest National Park between 1998 and 2022. Targeted analysis revealed that ΣPFAStarget significantly declined over the years (64-8 ng gꟷ1), driven by the decrease in perfluorooctane sulfonic acid (PFOS)

Emulsified Vegetable Oil Droplet Interfaces Sustain PFOA Defluorination in Groundwater through Carbon-Centered Radicals.

Perfluorooctanoic acid (PFOA) is highly resistant to degradation due to the extremely high bond energy of its C-F bonds. Current remediation approaches lead to incomplete defluorination and generally rely on external energy input, which limits their applicability for groundwater remediation. Here, we develop a novel emulsified vegetable oil (EVO)-Fe(II)/O2 system that achieves near-complete defluorination of PFOA (>99%) under dark conditions without external oxidants. Mechanistic investigations reveal that EVO droplets create droplet-associated interfaces that confine PFOA and reactive species

Separation of imidazolium-based ionic liquid cations with various substituents on a pentafluorophenylpropyl stationary phase using UHPLC-MS.

Ionic liquid cations are inherently fixed-charge and, when paired with smaller or polar substituents, not well-suited for reverse phase columns; they are also bulky organic species which can make methods traditionally used for monoatomic cation analysis less than ideal. For aromatic IL cations, such as those built upon the popular imidazolium scaffold, the pentafluorophenylpropyl (PFPP) column allows π-π stacking to improve retention. The PFPP stationary phase also has the potential to better retain polar substituents via additional dipole-dipole and/or H-bonding interactions. Previous related

Co-Occurrence, Bioaccumulation, and Dietary Risk Assessment of Per- and Polyfluoroalkyl Substances and Heavy Metals in Rice.

The co-occurrence and combined dietary health risks of per- and polyfluoroalkyl substances (PFASs) and heavy metals (HMs) in rice remain poorly understood. This study analyzed 29 PFAS compounds and 4 HMs in rice tissues collected from paddy fields. HFPO-DA was dominant in brown rice (4.84-15.99 ng/g), while PFOA primarily accumulated in leaves and straw. Cd and Cu showed elevated grain concentrations. PFASs exhibited higher bioaccumulation and translocation factors than HMs, especially in aboveground tissues, whereas Cd and As accumulated most in roots. Correlation analyses suggest dual root a

Method development and validation for neutral PFAS in food packaging using fast low pressure gas chromatography - mass spectrometry.

A new method for the targeted analysis of 37 volatile and semi-volatile PFAS was developed using fast low pressure gas chromatography-triple quadrupole mass spectrometry (GC-MS/MS). The targeted analytes included fluorotelomer alcohols, fluorotelomer acrylates and methacrylates, perfluoroalkane sulfonamides, and perfluoroalkane sulfonamido alcohols. Two GC phases for volatile PFAS were evaluated under low pressure vacuum outlet conditions, enabling a total GC run time of 8 min. Inter- and intra-day performance demonstrated high accuracy and reproducibility. The method was further validated for

Impact of understudied PFAS on Lipid Metabolism in Hepatocyte 2D and 3D in vitro model.

Perfluoroalkyl substances (PFAS) are industrial chemicals with several applications, including heat and water resistance. These chemicals are ubiquitous in our environment, and human exposure to them has been linked to liver injury and disruption of lipid metabolism. This study investigated the effects of PFAS (fluorotelomer sulfonic acids 6:2 FTSA and 8:2 FTSA, and perfluorooctane sulfonamide (PFOSA)) on hepatic lipid metabolism using in vitro human cell models. Initially, we assessed the effects of 10 μM PFOSA, 100 μM 6:2 FTSA, and 100 μM 8:2 FTSA in HepG2 monolayer (2D) cultures, focusing o

Drinking waters contamination by per- and poly-fluoroalkyl substances of the anthropized area of Fos-Berre, France.

Per- and poly-fluoroalkyl substances (PFAS) contamination and geographical distribution were studied in drinking water from the industrial harbor of Fos-Berre, a highly industrialized area in southern France. An analytical method for 38 PFAS was developed, using large volume injection ultra-high performance liquid chromatography-tandem mass spectrometry. To minimize the risk of contamination during preparation, samples were directly injected onto a SCIEX Triple Quad 7500 system after dilution in acidified methanol. Method limits of quantification ranged from 0.081 to 2.2 ng L-1, except for 8:2

Brønsted Acid-Catalyzed Direct Dehydroxylative Allylation of Benzylic Alcohols with Allylsilanes in 1,1,1,3,3,3-Hexafluoro-2-propanol.

A direct dehydroxylative allylation reaction of benzylic alcohols with allylsilanes catalyzed by Brønsted acids in 1,1,1,3,3,3-hexafluoro-2-propanol was developed. A wide variety of secondary and tertiary benzylic alcohols are applicable to the reaction with various allyltrimethylsilanes, to provide the corresponding coupling products in high yields. Using this catalytic transformation, a concise and short-step synthesis of (±)-crucudiol was achieved.

The price of protection: an integrated toxicological and modelling assessment of flubendiamide exposure in Anabas testudineus.

The widespread use of pesticides such as flubendiamide raises increasing concerns regarding their ecological impact on non-target aquatic organisms. This study evaluated the acute and chronic toxicity of flubendiamide in the freshwater fish Anabas testudineus by integrating hematological, biochemical, oxidative stress, and genotoxic biomarkers with advanced modelling approaches. Fish were exposed to sublethal concentrations (2.66 and 5.32 mg/L) for 45 days, and responses were assessed at multiple time points. Acute toxicity assays revealed a time-dependent decrease in LC50 values, confirming c

Enhancement of Piezoelectric Performance in PVDF via ZnO Doping and Its Application in Wearable Real-Time Monitoring of Human Radial Pulse.

Flexible piezoelectric materials demonstrate broad application potential in wearable health monitoring, human-machine interaction, and biosensing. However, the piezoelectric response of pure PVDF-TrFE is limited and insufficient to meet the requirements for highly sensitive sensing. In this study, ZnO/PVDF-TrFE composite films with varying ZnO doping contents (3-11 wt%) were fabricated and systematically characterized in terms of their structural, thermal, and electrical properties. The results indicate that ZnO significantly promotes the formation of the polar β-phase in PVDF-TrFE, with the m

The pharmacokinetics of perfluorobutanoic acid in female rats.

Perfluorobutanoic acid (PFBA) is of increasing concern due to its widespread occurrence, high environmental mobility, and potential for human exposure. This study investigated the toxicokinetics of PFBA in female Sprague Dawley rats and assessed implications for human exposure. Under repeated oral exposure (0.2 mg/kg/day for 16 days), concentrations of PFBA in blood increased rapidly and stabilized within the range of 230-300 ng/mL. Owing to its large tissue mass, muscle served as the major reservoir for the overall body burden of PFBA. Among the various organs, kidney, stomach, liver and gut

Environmental exposure to perfluorooctanoic acid promotes early gastric carcinogenesis via PPAR-γ-mediated ferroptosis.

Perfluorooctanoic Acid (PFOA) has gained our attention for it is resistant to natural degradation and can migrate over long distances. We have already found that PFASs were related to gastric cancer in previous work. However, the systematic evidence on the gastric toxicity of PFOA and its mechanisms remains scarce. In this study, network toxicology pinpointed the peroxisome proliferator-activated receptor (PPAR) pathway as the pivotal driver between PFOA exposure and gastric cancer progression. In the cell experiments, PPAR inhibitor (10 μM) and ferroptosis inhibitor (2 μM) protect gastric cel

Systemic cardiac migration of perfluorocarbon liquid following unsuccessful endoresection of choroidal melanoma: a case report.

Choroidal melanoma (CM) is the most common intraocular malignant tumor in adults, with a high rate of metastasis and mortality. This study reports a case of systemic cardiac migration of perfluorocarbon liquid (PFCL) following an unsuccessful endoresection of CM. A 61-year-old woman underwent gamma knife radiosurgery and pars plana vitrectomy endoresection for CM in her left eye. The procedure was complicated by intraocular bleeding, leading to incomplete tumor removal, with the eye filled with PFCL and blood before closure. The patient refused further surgery and later developed secondary gla

PFOS Disrupts Oocyte Maturation and Early Embryonic Development via Ovarian FOXK1 O-GlcNAcylation in Mice.

Perfluorooctane sulfonate (PFOS) is of great concern due to its accumulation in living organisms and reproductive toxicity. Although prior studies indicate that PFOS exposure causes female reproductive disorders, the underlying mechanism remains obscure. This study investigates the molecular mechanisms underlying PFOS-induced female reproductive toxicity at human-relevant exposure levels. These results demonstrate that PFOS exposure (0.2 and 20 µM) significantly reduces polar body extrusion (PBE) and delays germinal vesicle breakdown (GVBD) in oocytes. Additionally, PFOS exposure (1 mg kg-1 da

Assessment of modeled serum per- and polyfluoroalkyl substances concentrations from exposure estimates for pregnant women in the general population in comparison to previously measured serum concentrations.

When drinking water is uncontaminated, exposure to PFAS is thought to occur primarily via ingestion of food and indoor dust. To understand the background exposure during prenatal periods, this study examined whether published estimates of PFAS exposure rates from dietary and dust ingestion provide reasonable predictions of PFAS serum concentrations among pregnant women in the general population. This study estimated serum concentrations of four PFAS during pregnancy based on published PFAS intake rates for food and indoor dust reported in the peer-reviewed literature, a pharmacokinetic model u

Dynamic interplay of protein-humic complexes dictates perfluoroalkyl acids phase partitioning during sequential sludge treatment: multiscale mechanistic insights from composition-structure-energy interdependencies.

Sewage sludge acts as a substantial reservoir and potential secondary source of perfluoroalkyl acids (PFAAs) in the environment. The phase partitioning and interaction mechanisms of PFAAs during sludge treatment and disposal are not fully understood. Herein, a holistic approach that incorporated the analysis of sludge composition, structural evolution, and surface energy dynamics was employed to systematically elucidate PFAA occurrence, migration, and fate throughout the sludge treatment process. Various treatment processes minimally affected the overall PFAA concentration. PFAAs partitioning
