Differential Regulations of Antioxidant Metabolism and Cold-Responsive Genes in Three Bermudagrass Genotypes under Chilling and Freezing Stress.

As a typical warm-season grass, bermudagrass growth and turf quality begin to decrease when the environmental temperature drops below 20 °C. The current study investigated the differential responses of three bermudagrass genotypes to chilling stress (8/4 °C) for 15 days and then freezing stress (2/-2 °C) for 2 days. The three genotypes exhibited significant variation in chilling and freezing tolerance, and Chuannong-3, common bermudagrass 001, and Tifdwarf were ranked as cold-tolerant, -intermediate, and -sensitive genotypes based on evaluations of chlorophyll content, the photochemical effici

MaMYB4 is a negative regulator and a substrate of RING-type E3 ligases MaBRG2/3 in controlling banana fruit ripening.

Fruit ripening is a complex developmental process, which is modulated by both transcriptional and post-translational events. Control of fruit ripening is important in maintaining moderate quality traits and minimizing postharvest deterioration. In this study, we discovered that the transcription factor MaMYB4 acts as a negative regulator of fruit ripening in banana. The protein levels of MaMYB4 decreased gradually with banana fruit ripening, paralleling ethylene production, and decline in firmness. DNA affinity purification sequencing combined with RNA-sequencing analyses showed that MaMYB4 pr

A Constitutively Active Cytokinin Receptor Variant Increases Cambial Activity and Stem Growth in Poplar.

The cambial meristem is responsible for bark and wood formation in woody plants. The activity of the cambial meristem is controlled by various factors; one of them is the plant hormone cytokinin. Here, we have explored different approaches to genetically engineering cambial activity in poplar plants by the ectopic expression of a cytokinin biosynthesis gene with enhanced activity (named ROCK4) or of a gene encoding a constitutively active cytokinin receptor variant (ROCK3). Both genes are derived from Arabidopsis thaliana and were expressed in poplar trees under the control of their own promot

TaMOR is essential for root initiation and improvement of root system architecture in wheat.

Optimal root system architecture is beneficial for water-fertilizer use efficiency, stress tolerance and yield improvement of crops. However, because of the complexity of root traits and difficulty in phenotyping deep roots, the study on mechanisms of root development is rarely reported in wheat (Triticum aestivum L.). In this study, we identified that the LBD (LATERAL ORGAN BOUNDARIES DOMAIN) gene TaMOR (MORE ROOT in wheat) determines wheat crown root initiation. The mor mutants exhibited less or even no crown root, dwarfism, less grain number and lodging caused by few roots. The observation

Mutation of the imprinted gene OsEMF2a induces autonomous endosperm development and delayed cellularization in rice.

In angiosperms, endosperm development comprises a series of developmental transitions controlled by genetic and epigenetic mechanisms that are initiated after double fertilization. Polycomb repressive complex 2 (PRC2) is a key component of these mechanisms that mediate histone H3 lysine 27 trimethylation (H3K27me3); the action of PRC2 is well described in Arabidopsis thaliana but remains uncertain in cereals. In this study, we demonstrate that mutation of the rice (Oryza sativa) gene EMBRYONIC FLOWER2a (OsEMF2a), encoding a zinc-finger containing component of PRC2, causes an autonomous endospe

Hydrogen sulphide signalling in plant response to abiotic stress.

Throughout their whole life cycle, higher plants are often exposed to diverse environmental stresses, such as drought, salinity, heavy metals and extreme temperatures. In response to such stress, plant cells initiate signalling transduction, resulting in downstream responses, such as specific gene transcription and protein expression. Accumulating evidence has revealed that hydrogen sulphide (H2 S) serves as a signalling molecule in plant acclimation to stressful conditions. More important, H2 S interacts with other signalling molecules and phytohormones, contributing to transcriptional regula

A single nucleotide mutation in GID1c disrupts its interaction with DELLA1 and causes a GA-insensitive dwarf phenotype in peach.

Plant stature is one important factor that affects the productivity of peach orchards. However, little is known about the molecular mechanism(s) underlying the dwarf phenotype of peach tree. Here, we report a dwarfing mechanism in the peach cv. FenHuaShouXingTao (FHSXT). The dwarf phenotype of 'FHSXT' was caused by shorter cell length compared to the standard cv. QiuMiHong (QMH). 'FHSXT' contained higher endogenous GA levels than did 'QMH' and did not response to exogenous GA treatment (internode elongation). These results indicated that 'FHSXT' is a GA-insensitive dwarf mutant. A dwarf phenot

3'O-Methyltransferase, Ps3'OMT, from opium poppy: involvement in papaverine biosynthesis.

Using, in silico, in vitro and in planta functional assays, we demonstrate that Ps3'OMT, an 3'-O methyl transferase is linked to papaverine biosynthesis in opium poppy. Papaverine, one of the benzylisoquinoline alkaloids (BIA) synthesized in the medicinally important plant, Papaver somniferum, is known for the potent pharmacological properties. Papaverine biosynthesis has remained debatable as two different pathways, NH (involving N-desmethylated intermediates) and the NCH3 (involving N-methylated intermediates), have been proposed. In addition, there are several intermediate steps in both the

Exploring natural variation for rice sheath blight resistance in Brachypodium distachyon.

Sheath blight caused by the soil borne fungus Rhizoctonia solani AG1-IA is one of the major diseases of rice in the world. Genetic resistance in rice against this disease has not been very successful. Brachypodium distachyon is considered as a model species for several cereal crops and it has been studied in the past to identify novel sources of disease resistance against cereal crop diseases. Therefore, the current study was designed to explore nonhost disease resistance in Brachypodium accessions against sheath blight pathogen of rice, Rhizoctonia solani. A total of 19 Brachypodium distachyo

Transcript profiling of a bitter variety of narrow-leafed lupin to discover alkaloid biosynthetic genes.

Lupins (Lupinus spp.) are nitrogen-fixing legumes that accumulate toxic alkaloids in their protein-rich beans. These anti-nutritional compounds belong to the family of quinolizidine alkaloids (QAs), which are of interest to the pharmaceutical and chemical industries. To unleash the potential of lupins as protein crops and as sources of QAs, a thorough understanding of the QA pathway is needed. However, only the first enzyme in the pathway, lysine decarboxylase (LDC), is known. Here, we report the transcriptome of a high-QA variety of narrow-leafed lupin (L. angustifolius), obtained using eight

Bacterial Outer Membrane Vesicles Induce Plant Immune Responses.

Gram-negative bacteria continuously pinch off portions of their outer membrane, releasing membrane vesicles. These outer membrane vesicles (OMVs) are involved in multiple processes including cell-to-cell communication, biofilm formation, stress tolerance, horizontal gene transfer, and virulence. OMVs are also known modulators of the mammalian immune response. Despite the well-documented role of OMVs in mammalian-bacterial communication, their interaction with plants is not well studied. To examine whether OMVs of plant pathogens modulate the plant immune response, we purified OMVs from four di

Role of Fe-responsive genes in bioreduction and transport of ionic gold to roots of Arabidopsis thaliana during synthesis of gold nanoparticles.

Several studies have shown potassium chloroaurate (KAuCl4)-mediated synthesis of gold nanoparticles (AuNPs) by using extracts of different parts of diverse plant species. However, the mechanism underlying the formation of AuNPs in planta has far from being elucidated. Here, we report the molecular evidence towards the role of genes involved in iron (Fe) homeostasis during in planta synthesis of AuNPs in roots of Arabidopsis thaliana. Firstly, we examined the dosage-dependent effects of KAuCl4 treatment on primary root length (PRL), and meristematic activity of roots in transgenic CycB1;1::uidA

Arabidopsis thaliana cdd1 mutant uncouples the constitutive activation of salicylic acid signalling from growth defects.

Arabidopsis genotypes with a hyperactive salicylic acid-mediated signalling pathway exhibit enhanced disease resistance, which is often coupled with growth and developmental defects, such as dwarfing and spontaneous necrotic lesions on the leaves, resulting in reduced biomass yield. In this article, we report a novel recessive mutant of Arabidopsis, cdd1 (constitutive defence without defect in growth and development1), that exhibits enhanced disease resistance associated with constitutive salicylic acid signalling, but without any observable pleiotropic phenotype. Both NPR1 (NON-EXPRESSOR OF P

Melatonin-mediated metabolic reprogramming and stress adaptation in plants.

Melatonin acts as a multifunctional regulator of plant stress adaptation by reprogramming primary and secondary metabolism and coordinating ROS, nitric oxide, and phytohormone signalling. Integrating metabolomics with transcriptomics and proteomics provides new insights into these mechanisms and highlights melatonin as a promising strategy for enhancing crop resilience and productivity under diverse environmental stresses. Plants, being sessile, are continuously exposed to diverse environmental challenges, which disrupt cellular homeostasis and reduce crop productivity. To survive under such c

Blue or far-red light supplementation induced pre-hardening in the leaves of the Rht12 wheat dwarfing line: hormonal changes and freezing tolerance.

Reduced height (Rht) genes have revolutionised wheat cultivation, but they can compromise freezing tolerance, and only a few alleles are in use. Thus, evaluating the role of other Rht alleles in stress responses is crucial. Far-red supplementation of white light (W+FR) can induce pre-hardening in cereals at 15°C. However, the relevant effect of blue light enrichment (W+B) is poorly described. This study investigates the influence of W+FR or W+B exposure in young winter wheat leaves of a tall (wild-type, rht12) and a dwarf, gibberellin-deficient (near-isogenic line, Rht12) genotype in cv. The Characteristics and Expression Analysis of the Tomato SlRBOH Gene Family under Exogenous Phytohormone Treatments and Abiotic Stresses.

Respiratory burst oxidase homologs (RBOHs), also known as NADPH oxidases, contribute significantly to the production of ROS in plants, alongside other major sources such as photosynthesis and electron transport in chloroplasts. It has been shown that plant RBOHs play an active role in plant adversity response and electron transport. However, the phylogenetic analysis and characterization of the SlRBOH gene family in tomatoes have not been systematically studied. This study identified 11 SlRBOH genes in the tomato genome using a genome-wide search approach. The physicochemical properties, chrom

Whole-Mount In Situ Hybridization Using Microwave Treatment.

In situ mRNA hybridization, which relies on the complementarity between antisense riboprobes and specific RNA populations, was originally used on thin sections with colorimetric dyes to visualize mRNA distribution. The use of fluorescent probes and confocal imaging enabled whole-mount in situ hybridization (WISH) and 3D reconstruction of gene expression domains. Here, we present a versatile protocol based on microwave treatment that results in high and homogeneous hybridization yields. When used with a cell wall marker and tissue clearing, it enables simultaneous visualization of gene expressi

CSLF6 Mediates Cadmium Accumulation in Rice by Modulating Cellulose Synthesis.

Cadmium (Cd) pollution severely limits rice growth and yield. The plant cell wall acts as the first barrier against Cd, but how cell wall biosynthesis affects Cd accumulation is unclear. This study found that CSLF6, a cellulose synthase-like F, is involved in cellulose synthesis and Cd accumulation. Cd treatment strongly induced CSLF6 expression in roots. Under Cd stress, cell wall thickness and cellulose content were increased by CSLF6 overexpression and decreased when CSLF6 was mutated. CSLF6 overexpression enhanced Cd tolerance, with higher Cd accumulation in roots and shoots. In contrast,
