The Recently Discovered Bokeloh Bat Lyssavirus: Insights Into Its Genetic Heterogeneity and Spatial Distribution in Europe and the Population Genetics of Its Primary Host.

In 2010, a novel lyssavirus named Bokeloh bat lyssavirus (BBLV) was isolated from a Natterer's bat (Myotis nattereri) in Germany. Two further viruses were isolated in the same country and in France in recent years, all from the same bat species and all found in moribund or dead bats. Here we report the description and the full-length genome sequence of five additional BBLV isolates from Germany (n=4) and France (n=1). Interestingly, all of them were isolated from the Natterer's bat, except one from Germany, which was found in a common Pipistrelle bat (Pipistrellus pipistrellus), a widespread a

Genetic diversity, population structure, and benzimidazole resistance profiling of Ascaris lumbricoides in northwestern Thailand.

Ascaris lumbricoides remains one of the most prevalent soil-transmitted helminths worldwide, with ongoing transmission in endemic regions despite long-standing control programs. This study investigated the genetic diversity, population structure, and benzimidazole resistance of A. lumbricoides across three endemic regions of Thailand using mitochondrial (cox1 and nad1) and nuclear (β-tubulin) markers. Haplotype network analysis identified 13 haplotypes within the Thailand dataset, including three newly identified haplotypes, indicating measurable genetic diversity at the local scale. When anal

An Exploratory Analysis of Genetic Structure and Introgression in Salmo trutta Populations from Campania Rivers.

The Salmo trutta complex exhibits high genetic and phenotypic diversity due also to the historical climatic events and colonization processes. In Europe, several evolutionary lineages have been identified, including the Atlantic and Mediterranean. In Italy, native Mediterranean trout populations have been threatened by restocking practices involving hatchery-reared fish of Atlantic origin, thus resulting in widespread hybridization and genetic introgression. This study aimed to genetically characterize trout populations from selected river basins of the Campania region (southern Italy) and to

Shared and Sex-Specific Genetic Risk for Parkinson's Disease Risk Across European Populations.

INTRODUCTION: Parkinson's disease (PD) affects females and males differently, with differences in prevalence, clinical phenotypes, and therapeutic response, suggesting that biological sex may influence the underlying molecular mechanisms of PD. The extent to which genetic factors may contribute to these differences remains largely unknown. OBJECTIVE: To investigate sex-specific autosomal genetic factors associated with PD risk. METHODS: We performed a sex-stratified autosomal GWAS meta-analysis leveraging data from the Global Parkinson's Genetics Program, the International Parkinson's Diseas

Genetic transformation patterns along a longitude gradient in Ukrainian populations of Schizophyllum commune fungus.

The research of fungal biodiversity is important under global climate change and increasing anthropogenic pressure on ecosystems. The most studies focusing on fungal biogeography oriented on species richness. There is a short supply researches on how genetic diversity shifts along a longitudinal gradient within one species. Model fungi with well-characterized genomes and simple cultivation requirements allow detailed research of evolutionary and ecological processes. The aim was to study the genetic profile transformation of the Schizophyllum commune populations by DNA microsatellite loci alon

Genetic diversity of Caracu cattle reveals contrasting trends between general and closed populations.

The conservation of genetic diversity and control of inbreeding rate are fundamental factors in animal breeding programs, especially programs for adapted cattle breeds such as Caracu. However, there are still knowledge gaps regarding the population structure of this breed as a whole and the genetic dynamics in subpopulations, like closed populations reared in experimental farms in Brazil. Within this context, this study aimed to evaluate the population structure of the Caracu cattle breed by investigating inbreeding levels and genetic diversity based on pedigree analyses. Two populations were

Large-Scale Cohort Study on the Genetic and Phenotypic Findings of Palmoplantar Keratodermas in the Chinese Population.

Palmoplantar keratoderma (PPK) is a heterogeneous group of skin diseases, characterised by excessive keratinization and hyperplasia of the palms and soles. However, accurate molecular diagnosis remains challenging due to the low prevalence of hereditary forms. This study analysed the phenotype and genotype distribution in a large cohort of Chinese patients with PPK, aiming to clarify the genetic aetiology and genotype-phenotype correlations. We further validated the pathogenicity of biallelic SERPINA12 variants identified in four patients. A total of 424 patients with PPK were enrolled from 20

Population-genetic and evolutionary context of a CRIP3 variant associated with age-related hearing loss.

Age-related hearing loss is a widespread sensory impairment affecting one third of the elderly population, yet the genetic underpinnings of this condition remain incompletely understood. In this study, we investigate the evolutionary context of a non-synonymous variant (rs2242416) in the CRIP3 gene, which is expressed in auditory hair cells, and previously associated with hearing loss. We first confirm that rs2242416 shows strong and consistent association with hearing loss across multiple genome-wide association studies. Notably, rs2242416 is also associated with reduced risk of heart related

Cell Type-Specific Targeting of Different Smooth Muscle Cell Populations by Intersectional Genetics.

BACKGROUND: The Cre/loxP recombination system is the main tool for cell type-specific lineage tracing and gene targeting. Currently available smooth muscle cell (SMC)-specific Cre mouse lines show off-target activity outside the SMC lineage and are unable to distinguish among arterial SMCs (ASMCs), venous SMCs, and nonvascular SMCs (NVSMCs). These limitations prevent ASMC- and NVSMC-specific gene targeting, which is required to characterize the role of SMCs in different organs and diseases. METHODS: To achieve precise manipulation of ASMCs in vivo, we combined alleles for Cspg4-Dre and rox-St

A reappraisal of APOE genetic effects on Alzheimer's disease risk in the Japanese population: a meta-analysis.

Our meta-analysis demonstrates that the risk effect of APOE-e4*4 relative to APOE-e3*3 for Alzheimer's disease in the Japanese population is approximately 12-15-fold, comparable to that reported in Caucasian populations, rather than greater than 20-fold as previously reported.

Evaluating the Effectiveness of Early Genetic Screening for Fanconi Anemia in High-Risk Pediatric Populations.

INTRODUCTION: Fanconi anemia (FA) is the most prevalent inherited disorder leading to bone marrow failure, resulting from a rare autosomal recessive genetic condition that affects all three types of blood cells. A key characteristic of FA is the body's heightened sensitivity to DNA-damaging agents, particularly those that induce crosslinking, which serves as an important diagnostic marker. Children at higher risk-such as those with unexplained growth delays, congenital defects, or a family history of FA-can significantly benefit from genetic testing. METHODS: This study involved 140 pediatric

Extensive genetic interactions (epistasis) linked to alcohol use disorder in a high-risk population.

Alcohol use disorder (AUD) is known to have a significant genetic component, yet there remains a gap between its heritability and findings from genome-wide association studies. One potential explanation for this could be genetic interactions, or epistasis, which remain largely unexplored in the context of AUD. We investigated the role of epistasis in AUD susceptibility among 742 American Indians. By analyzing 467 K variants in 3,736 genes and regulatory elements linked to AUD, we identified 97 interacting gene pairs significantly associated with AUD severity in an American Indian cohort. Inbreeding Implications on Genetic Diversity and Population Evolution of South American Brown Swiss Cattle.

Inbreeding is a critical issue in both conservation and animal breeding, as it can severely impact survival, reproductive success and genetic diversity (GD) in domestic species. The Brown Swiss cattle were introduced in South America around a century ago; however, the breeding scheme-derived consequences have been neglected by breeders for numerous generations. The present research aimed to elucidate the population evolution, inbreeding and GD in introduced South American Brown Swiss cattle using genealogical databases. The study was based on official registered records from 8686 animals acros

Genome-Wide Association of Genetic Variants with Intestinal Cholesterol Absorption Markers in a European Population.

Background: Interindividual variability in intestinal cholesterol absorption contributes to differences in serum lipid concentrations and cardiovascular risk. Total cholesterol (TC)-standardized campesterol and sitosterol levels are established markers of cholesterol absorption. However, genetic variants in Europeans associated with these markers remain incompletely characterized. Methods: A genome-wide association study (GWAS) was performed in 398 healthy individuals of European ancestry. Samples were genotyped using the Precision Medicine Research Array (PMRA). After quality control, 166,037

The role of apoptotic genetic polymorphisms in prostate cancer susceptibility in a North West Spain population.

Apoptosis and tumor suppression prevent cell proliferation in response to genomic damage. Polymorphisms in genes involved in these pathways can alter their function, facilitating the onset of cancer. Detecting these polymorphisms would assess the individual risk of developing different types of cancer. Therefore, the main goal of this study is to analyze the association between a set of genetic polymorphisms involved in apoptosis and tumor suppression and the risk of developing prostate cancer (PCa) in a population from Galicia. Accordingly, we performed a case-control study with 291 patients

Cross-trait genetic enrichment between GERD and psychiatric disorders in East Asian populations.

Gastroesophageal reflux disease (GERD) exhibits significant epidemiological comorbidity with psychiatric disorders, yet their shared genetic architecture remains poorly characterized in East Asian populations. Leveraging ancestry-specific genome-wide association study (GWAS) summary statistics from East Asian cohorts, we employed linkage disequilibrium score regression and conditional false discovery rate (condFDR) approaches to investigate cross-trait genetic enrichment between GERD and major psychiatric disorders, including major depressive disorder (MDD), schizophrenia (SCZ), and bipolar di

Phylogenetic and Population Genetic Analyses Reveal Patterns of Divergence Among Isolates of Ceratocystis manginecans.

The taxonomic boundaries of Ceratocystis species related to C. manginecans have remained contentious due to limited morphological variation, interfertility in laboratory mating studies, and the application of different species concepts. However, recent studies have highlighted significant differences among species recently reduced to synonymy with C. manginecans, including host associations and biological traits, all with important implications for disease management. We examined the phylogenetic relationships, genetic diversity and population structure of isolates considered broadly as Cerato

Association of IL4 and IL17A genetic polymorphisms with COVID-19 severity in a southern Brazilian population.

COVID-19 has demonstrated marked differences in clinical manifestations and outcomes, with severe cases presenting excessive inflammatory responses known as cytokine storms. Polymorphisms in cytokine genes, such as interleukin-4 (IL4) and interleukin-17A (IL17A), may influence immune responses and contribute to disease severity. This study investigated the association between the IL4 -34C/T and IL17A -197G/A polymorphisms and COVID-19 severity in a population from Southern Brazil. A total of 528 individuals were included, with 157 severe and 371 mild cases. DNA was extracted from buccal swabs

Genetic variation near ROBO1 is associated with craniofacial microsomia and related phenotypes in the Finnish population.

Craniofacial microsomia (CFM) encompasses a phenotypic continuum of congenital anomalies ranging from isolated microtia to more complex manifestations within the oculo-auriculo-vertebral spectrum, including Goldenhar syndrome, reflecting abnormal development of first and second pharyngeal arch-derived structures. While several genetic susceptibility loci have been reported, population-based evidence in individuals of European ancestry remains limited. Using nationwide data from FinnGen in the Finnish founder population, we identified a genome-wide significant association at a conserved interge
