A study of the copper-binding proteins in liver and kidney tissue of neonatal normal and mottled mutant mice.

The Cu2+-binding proteins from liver and kidney tissue of 7--8-day-old brindled (Mobr) mice and their normal littermates were compared. (1) … Cl2-treated mutant and normal animals revealed clos similarities. The proteins showed high glycine, glutamic acid, serine, alanine and lysine contents and a rather variable cysteine content. Differences were apparent in the normal liver subfractions, which showed a higher cysteine content and lower glutamic acid content than did either the mutant liver or norma …

Neuromuscular synaptic transmission in Limulus polyphemus--II. Release of amino acid putative transmitters from the neuromuscular preparation.

High-performance liquid chromatography with fluorescence detection was used to assay the release of putative amino acid transmitters from the Limulus neuromuscular preparation. Motor axon stimulation increased the concentrations of aspartate, glutamate and eight other amino acids in fluid bathing the neuromuscular preparation. Pentobarbital, which attenuates the excitatory postsynaptic potential of Limulus muscle, was used to block both synaptic activation of muscle fibers and any amino acid release that may have resulted from this activation. Stimulus-induced release of glutamate and five oth

Evidence for transporter-mediated uptake of environmental L-glutamate in a freshwater sponge, Ephydatia muelleri.

The freshwater sponge, Ephydatia muelleri, lacks a nervous or endocrine system and yet it exhibits a coordinated whole-body action known as a "sneeze" that can be triggered by exposure to L-glutamate. It is not known how L-glutamate is obtained by E. muelleri in sufficient quantities (i.e., 70 µM) to mediate this response endogenously. The present study tested the hypothesis that L-glutamate can be directly acquired from the environment across the body surface of E. muelleri. We demonstrate carrier mediated uptake of two distinct saturable systems with maximal transport rates (Jmax) of 64.27 ±

Elevated glutamate levels in the vitreous body of humans and monkeys with glaucoma.

OBJECTIVE: To explore the possibility that the excitatory amino acid glutamate might be associated with the disease process of glaucoma, which is characterized by the death of retinal ganglion cell neurons and subsequent visual dysfunction. METHODS: Amino acid analyses were performed on vitreous specimens that were obtained from patients who were undergoing cataract extraction. Samples were collected prospectively from those patients who sustained inadvertent rupture of the posterior capsule between 1988 and 1993. An additional set of specimens, obtained from both eyes of monkeys, was analyze

Glutamate uptake activity in retina Müller cells: Circadian modulation.

Signal transmission in the visual pathway is carried by glutamate, the major excitatory neurotransmitter in the vertebrate brain. Ionotropic and metabotropic receptors mediate the actions of this amino acid. A tight regulation of extra-synaptic glutamate concentrations is needed for the selective activation of its receptors as a function of the day/night cycle. Moreover, proper extra synaptic glutamate levels regulation is also important for the avoidance of an excitotoxic insult. A family of plasma membrane glutamate transporters expressed in glia cells carries out the clearance of this amino

Decreased glutamate transport by the brain and spinal cord in amyotrophic lateral sclerosis.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a chronic degenerative neurologic disorder characterized by the death of motor neurons in the cerebral cortex and spinal cord. Recent studies have suggested that the metabolism of glutamate, a potentially neurotoxic amino acid, is abnormal in patients with ALS. We hypothesized that the high-affinity glutamate transporter is the site of the defect. METHODS: We measured high-affinity, sodium-dependent glutamate transport in synaptosomes from neural tissue obtained from 13 patients with ALS, 17 patients with no neurologic disease, and 27 patient

Elevated extracellular glutamate levels increased the formation of hydroxyl radical in the striatum of anesthetized rat.

Results from various in vitro experiments have indicated that excitotoxicity and oxidative stress are two interrelated major mechanisms in causing neuronal damage in brain disorders such as cerebral ischemia/reperfusion. Thus, we have conducted experiments to investigate whether in the striatum of anesthetized rats the elevated brain extracellular concentrations of glutamate could increase the formation of hydroxyl radical. Elevation of glutamate levels and trapping of hydroxyl radical were accomplished by perfusing Ringer solutions containing both glutamate and salicylic acid through microdia

The effect of thiopental on glutamate metabolism in mouse cerebellar astrocytes in vitro.

The effect of thiopental on [U-(13)C]glutamate metabolism was studied in cerebellar astrocytes from mice using (13)C magnetic resonance spectroscopy. The cells were incubated with 0.5 mM [U-(13)C]glutamate for 2 h in the presence of 1 mM thiopental and 1 mM thiopental plus 0.2 mM gammaamino butyric acid (GABA). Labeled glutamate, glutamine, aspartate and glutathione were observed in cell extracts, and glutamine, aspartate and lactate in the media. Not only uniformly labeled glutamate was present in the medium, but also glutamate derived from the tricarboxylic acid (TCA) cycle. In the presence

Contribution of glutamatergic systems in locus coeruleus to nucleus paragigantocellularis stimulation-evoked behavior.

The role of extracellular glutamate, within the locus coeruleus, in mediation of the behavioral signs elicited by electrical stimulation of the nucleus paragigantocellularis (PGi) was investigated in conscious, opioid-naive rats. Each rat was prepared with a chronically implanted unilateral electrode within the PGi and a microdialysis guide cannula directed at the ipsilateral locus coeruleus. Opioid withdrawal-like behaviors (rearing, teeth-chattering, wet-dog shakes, etc.) and increases in extracellular glutamate concentrations within the locus coeruleus were evoked, in a frequency-dependent

Visible-Light Photoredox Synthesis of Chiral α-Selenoamino Acids.

N-Acetoxyphthalimide derivatives of two genetically coded proteinogenic amino acids, l-aspartic acid and glutamic acid, were used as visible light photoredox chiral sources and radical precursors, diorganyl diselenides were used as the radical acceptors, and the diverse chiral α-selenoamino acid derivatives were prepared in good yields at room temperature. Furthermore, decarboxylative coupling of N-protected dipeptide active ester with diphenyl diselenide provided the corresponding selenodipeptide. The simple protocol, mild reaction conditions, high efficiency, and chiral keeping of this metho…

Plasma amino acid levels in children with autism and their families.

Plasma amino acid levels were measured in autistic and Asperger syndrome patients, their siblings, and parents. The results were compared with values from age-matched controls. Patients with autism or Asperger syndrome and their siblings and parents all had raised glutamic acid, phenylalanine, asparagine, tyrosine, alanine, and lysine (p < .05) than controls, with reduced plasma glutamine. Other amino acids were at normal levels. These results show that children with autistic spectrum disorders come from a family background of dysregulated amino acid metabolism and provide further evidence for…

Taurine and other free amino acids in the retina, vitreous, lens, iris-ciliary body, and cornea of the rat eye.

Levels of free amino acids were determined quantitatively in whole ocular tissues of the rat eye with aid of a sensitive amino acid analyzer. The tissues studied were the retina, vitreous, lens, iris-ciliary body, and cornea. The retina and lens contained a more concentrated free amino acid pool than other tissues. The neuroactive amino acids taurine. GABA, glutamic acid, aspartic acid, and glycine were clearly enriched in the retina. Taurine was the most abundant amino acid in all five tissues studied, and its high concentration in non-neural tissues, especially the lens, suggests that it mus…

The role of glutamate in swim initiation in the medicinal leech.

Antagonists were used to investigate the role of the excitatory amino acid, L-glutamate, in the swim motor program of Hirudo medicinalis. In previous experiments, focal application of L-glutamate or its non-NMDA agonists onto either the segmental swim-gating interneuron (cell 204) or the serotonergic Retzius cell resulted in prolonged excitation of the two cells and often in fictive swimming. Since brief stimulation of the subesophageal trigger interneuron (cell Tr1) evoked a similar response, we investigated the role of glutamate at these synapses. Kynurenic acid and two non-NMDA antagonists,

D- and L-Amino Acid Blood Concentrations Are Affected in Children With Duchenne Muscular Dystrophy.

Duchenne muscular dystrophy (DMD) is an X-linked disease caused by the absence of functional dystrophin in the muscle cells. Recent untargeted metabolomics studies identified amino acid metabolism alterations as biochemical pathways potentially involved in DMD pathogenesis. Here, in a well-characterised cohort of DMD children and paediatric controls, we investigated by high-performance liquid chromatography (HPLC) the serum profile of a selected pool of amino acids in D- and L-configuration, including L-glutamate, L-glutamine, glycine, L-aspartate, D-aspartate, L-asparagine, L-serine, and D-se

Effects of pharmacological manipulation on neurotransmitter and other amino acid levels in the CSF of the Senegalese baboon Papio papio.

The concentrations of GABA, glutamate, aspartate, glycine, taurine, glutamine, asparagine and alanine were determined in the CSF of 10 Senegalese baboons (Papio papio) following initial ketamine anaesthesia and subsequent administration (4 h later) of different compounds known to alter either inhibitory or excitatory neurotransmission. Ketamine itself was apparently without effect as the administration of a second dose of ketamine did not significantly alter the levels of any of the amino acids studied, although GABA levels tended to decrease. The presence of haemolysed material in occasional

Intracellular acidification of the leech giant glial cell evoked by glutamate and aspartate.

Glutamate is an excitatory receptor agonist in both neurones and glial cells, and, in addition, glutamate is also a substrate for glutamate transporter in glial cells. We have measured intracellular and extracellular pH changes induced by bath application of glutamate, its receptor agonist kainate, and its transporter agonist aspartate, in the giant neuropile glial cell in the central nervous system of the leech Hirudo medicinalis, using double-barrelled pH-sensitive microelectrodes. The giant glial cells responded to glutamate and aspartate (100-500 microM), and kainate (5-20 microM) with a m

A selective reduction of excitatory amino acids in cerebrospinal fluid of patients with Alzheimer type dementia compared with vascular dementia of the Binswanger type.

We determined the concentrations of the putative transmitter amino acids in the cerebrospinal fluid of patients with Alzheimer type dementia (ATD) and vascular dementia of the Binswanger type (VDBT). In ATD, aspartate and glutamate concentrations were significantly and selectively reduced, while in VDBT, concentrations of aspartate, glutamate, gamma-aminobutyric acid (GABA) and many other amino acids were decreased non-selectively. In both ATD and VDBT, we found a tendency for all amino acids to increase with progression of the disease, and this reached statistical significance for some amino

Adenosinergic approaches to stroke therapeutics.

Basic neuroscience research findings during the past five years have established a clear relationship between the excitatory amino acid (EAA) neurotransmitters (glutamic and aspartic acid) and various pathological states. A major mechanism of neural tissue degeneration following cerebral ischemia, and perhaps other neurodegenerative diseases, seems to involve overactivity of the EAA system in brain. This process is called delayed excitotoxicity and it has become a focal point for the design of new drugs that inhibit its course (EAA receptor blockers). Very recently it has been shown that it is

Characterization of saliva proteins from 'stainers' and 'non-stainers' adsorbed to hydroxyapatite.

Stainers and non-stainers were selected on the basis of their individual tendency to develop extrinsic tooth discolorations from a chlorhexidine mouth rinse. Saliva proteins adsorbed to hydroxyapatite in vitro and in vivo pellicle from the participants were analyzed by gel filtration and ion-exchange chromatography. Two distinct anionic components were isolated. The elution patterns from stainers and non-stainers were identical. Amino acid analyses of the main peaks demonstrated a prevalence of serine, glycine, and glutamic acid.
