The MYST family of lysine acetyltransferase (KAT) are required for many critical nuclear functions, including transcription and DNA damage repair in somatic cells; however, the functions of individual MYST members in mammalian oocyte meiosis remain largely unclear. Here, we show that KAT7, as a key MYST family member, might support mitochondrial function during porcine oocyte maturation. We found that KAT7 localized to the cytoplasm during meiotic progression. Moreover, impaired oocyte maturation was observed following its inhibition, which was associated with compromised mitochondrial ATP pro

DEK is a chromatin-associated, DNA-binding protein with unique properties that place it in its own protein class. First discovered in 1992 as a fusion protein in acute myeloid leukemia, DEK gained further attention in the mid-2000s as a growing number of studies identified its connections to chromatin architecture and subsequent impact on pathologies such as cancer and autoimmune diseases. Current evidence indicates that DEK can alter the topology of nucleic acids in a variety of biological processes, including DNA replication, DNA repair, chromatin organization, epigenetic modification, trans

Background/Objectives: Prostate cancer (PCa) remains a prevalent malignancy among men, often complicated by recurrence and unfavorable clinical outcomes. Consequently, precise risk stratification and timely clinical intervention are paramount. Initially, we delineated distinct expression profiles of histone modification regulators via unsupervised clustering, identifying PCa subtypes with divergent survival probabilities and biological phenotypes. Subsequently, we sought to develop a prognostic gene signature, derived from the transcriptomic variations among these regulator-defined subtypes, t

Formation of neuronal progenitor cells, neural crest cells and surface ectoderm cells is a complex process and differential gene expression allows for generation of these three cell types during neurulation process. Histone modifiers such as writers and erasers may play an important role in achieving the differential gene expression during neurulation event, similar to how DNMTs and TET enzymes function in germ cell development. We had previously reported that Polycomb group (PcG) protein RING1B which catalysed repressive H2AK119ub1 mark and prevented precocious activation of neuronal genes in

Microglia-mediated neuroinflammation is a key driver of neurodegenerative disease progression, yet the metabolic mechanisms underlying microglial dysfunction remain poorly understood. Recent studies highlight glycolytic reprogramming in activated microglia, which generates lactate that, in turn, promotes histone lactylation, an epigenetic modification that significantly alters gene expression. This glycolysis-histone lactylation axis has been implicated in Alzheimer's disease, Parkinson's disease, and other neurodegenerative disorders, where its dysregulation exacerbates chronic neuroinflammat

Hepatocellular carcinoma (HCC) has a high mortality rate, primarily driven by metastasis. The role of the histone deacetylase inhibitor (HDACi) entinostat in this process remains controversial, limiting its clinical application. This study aims to define entinostat's function and mechanism in HCC metastasis. We employed in vitro models, transcriptomic sequencing, chromatin immunoprecipitation-qPCR, and an orthotopic mouse model to assess the effects of entinostat on epithelial-mesenchymal transition (EMT), invasion, and tumor growth. Our findings demonstrate that entinostat potently prevented

BACKGROUND: Liver cancer remains a major global challenge, with limited benefit from immune checkpoint inhibitors due to strong immune evasion. Emerging evidence indicates that chromatin modifiers shape tumor immunogenicity, but their roles in liver cancer immunity are not fully understood.
METHODS: We investigated the role of the histone demethylase KDM2A in liver cancer using genetic silencing in vitro and in vivo. We evaluated mitochondrial function, redox balance, genomic stability, and performed transcriptomic and epigenomic analyses to assess immune pathway changes and H3K36me2 distribu

The cohesin complex structures the interphase genome of human cells by extruding loops and organizing topologically associating domains (TADs), yet how chromatin state regulates cohesin-chromatin interactions remains unclear. Here, we show that histone hyperacetylation induced by trichostatin A (TSA) selectively disrupts short-range intra-TAD interactions while largely preserving CTCF-anchored loops. These distinct responses define two functional cohesin populations: a TSA-sensitive pool associated with dynamic loop extrusion, and a TSA-resistant pool at CTCF sites maintained by topological en

Cancer remains a leading cause of global mortality, with tumor heterogeneity, recurrence, metastasis, and drug resistance posing significant challenges to conventional therapies, which are often limited by toxicity and efficacy. Epigenetic regulation, particularly histone modifications such as methylation, acetylation, phosphorylation, and lactylation, plays a pivotal role in orchestrating gene expression and tumor progression, representing promising yet complex therapeutic targets. Bioactive compounds derived from natural plants, characterized by multi-target activity and low toxicity, have e

Histone post-translational modifications play a major role in gametogenesis and early embryogenesis, being closely associated with in vitro fertilization (IVF). Proper regulation of histone modifications enables chromatin compaction, transcriptional activity, parental imprint establishment and genomic reprogramming which are essential events for zygotic genome activation and lineage specification post-fertilization. Aberrant histone regulation can disrupt epigenetic integrity, leading to impaired gamete quality, defective embryonic development, imprinting errors and potential long-term offspri

Importins transport numerous proteins into cell nuclei. They also chaperone highly positively charged cargoes against ionic aggregation during transit. An extreme example is histone H1, which requires two importins, Importin 7 (Imp7) and Importin β (Impβ), for safe import. Here, we present a combined cryoelectron microscopy (cryo-EM)/molecular dynamics (MD) structure of the Imp7·Impβ·H1 complex, correcting a previous model (PDB 6N88) with misassigned importin chains. Our structure reveals that Imp7 contacts Impβ via three interfaces and that this heterodimerization creates a large negatively c

Conjugative plasmids drive bacterial evolution and antibiotic resistance spread, yet their gene expression must be silenced to protect the host. A histone-like protein H-NS represses many mobile and sedentary xenogenes but fails to silence the conjugal transfer vir operon of R6K, a prototype IncX plasmid. Instead, R6K encodes its own H-NS homolog, Sfx, to repress the vir operon. Here, we show that, unlike other plasmid silencers that target promoters, Sfx cooperates with Rho factor to arrest transcription elongation. ChIP-seq reveals that Sfx and H-NS share similar DNA motifs and a preference

Spike-in normalization enables quantitative analysis of chromatin immunoprecipitation sequencing (ChIP-seq) signal. Here we introduce a robust dual spike-in normalization approach for ChIP-seq (ChIP-wrangler), optimize parameters and verify its accuracy in quantifying changes in ChIP-seq signal and detecting technical artifacts. We use ChIP-wrangler to revisit recent claims that active histone marks depend on transcription. We show that acute depletion of RNA polymerase II (RNAPII) has a modest impact on H3K27ac levels, with only 6% of peaks significantly changing after RNAPII depletion, indic

The glutathione (GSH) metabolism pathway plays a pivotal role in maintaining redox homeostasis, yet its coordination with mitochondrial function and reactive oxygen species (ROS) dynamics during this process remains poorly defined. Here, we report that a developmentally programmed decline in endogenous GSH occurs from the zygote to the 2-cell stage, while ROS levels and mitochondrial activity remain low. Perturbation of this physiological GSH decrease-either by depletion or excessive supplementation-led to developmental arrest at the 2-cell stage, accompanied by aberrant histone acetylation, p

Sepsis-induced immunosuppression is associated with both autophagy impairment and glycolytic dysfunction. However, it is unclear how metabolic dysfunction drives epigenetic reprogramming, thereby reducing autophagy in sepsis. Here, we demonstrate that glycolytic dysfunction and consequent lactate deficiency epigenetically silence autophagy by reducing histone H3 lysine 18 lactylation (H3K18la). Using LPS-tolerant macrophages and CLP-induced immunosuppressive murine models, we observed that reduced lactate levels and global lactylation correlated with impaired autophagic flux and diminished bac

Optimization of tree architecture is essential for advancing sustainable forestry, as appropriate canopy structure can contribute to growth and development. However, the molecular mechanisms that control tree architecture have not been fully characterized. While the putative NAD-dependent histone deacetylase HST1 was shown to regulate branching architecture in multiple crop species, whether epigenetic mechanism also influence tree architecture is unknown. Here, we investigated the genetic regulation of branching angle in white birch (Betula platyphylla Suk.), a pioneer tree of the temperate an

Poly (ADP-ribose) polymerase inhibitors (PARPi) have revolutionized the treatment of homologous recombination (HR)-deficient tumors. Nevertheless, their clinical utility is often compromised by the emergence of resistance. Although genetic restoration of HR is a well-established resistance mechanism, emerging evidence suggests that epigenetic and chromatin responses are also central determinants of cell survival under PARPi-induced stress. Indeed, PARP inhibition drives the acute depletion of H3-H4 histones, thereby creating a cellular reliance on efficient histone recycling to ensure genome i…

BACKGROUND: Neurodevelopmental disorders, including autism spectrum disorder, involve widespread transcriptional dysregulation. Copy number variations at 16p11.2 are among the strongest genetic risk factors for autism spectrum disorder, yet the molecular mechanisms by which these copy number variations contribute to neurodevelopmental pathology remain unclear.
RESULTS: We identify significant genetic associations between autism spectrum disorder susceptibility and the HIST1 histone gene cluster through genome-wide analysis. Transcriptomic profiling across post-mortem brain tissue, patient-der

Flower size is a key ornamental trait in roses (Rosa spp.), determined by coordinated regulation of cell proliferation and expansion. Although jasmonic acid (JA) influences plant growth, its role in flower size control remains unclear. Here, we showed that methyl jasmonate (MeJA) treatment reduces flower size in Rosa chinensis, accompanied by elevated expression of the JA-responsive transcription factor RcMYC2. Silencing RcMYC2 resulted in larger flowers due to enhanced petal cell expansion and upregulation of cell expansion-related genes. Mechanistically, RcMYC2 directly bound to the promoter
