Does the host matter? Variable influence of host traits on parasitism rates.

Parasitism of mammals is ubiquitous, but the processes driving parasite aggregation on hosts are poorly understood, as each system seems to show unique correlations between parasitism and host traits such as sex, age, size and body mass. Genetic diversity is also posited to influence susceptibility to parasitism, and provides a quantifiable measure of an intrinsic unchanging host property, but this link has not been well established. A lack of consistency in host traits predicting parasite heterogeneity may derive from the contribution of environmental factors to parasite aggregation. To evalu

Unraveling the Role of Proteinopathies in Parasitic Infections.

Proteinopathies, characterized by the misfolding, aggregation, and deposition of proteins, are hallmarks of various neurodegenerative and sy … is. By causing oxidative stress, persistent inflammation, and disturbance of cellular proteostasis, host-parasite interactions also contribute to proteinopathy. For instance, Plasmodium falciparum disrupts normal protein homeostasis and encourages the accumulation of misfolded proteins by influencing host redox systems involved in protein folding. In addition to interfering …

Bacterial cell-to-cell signaling promotes the evolution of resistance to parasitic bacteriophages.

The evolution of host-parasite interactions could be affected by intraspecies variation between different host and parasite genotypes. Here we studied how bacterial host cell-to-cell signaling affects the interaction with parasites using two bacteria-specific viruses (bacteriophages) and the host bacterium Pseudomonas aeruginosa that communicates by secreting and responding to quorum sensing (QS) signal molecules. We found that a QS-signaling proficient strain was able to evolve higher levels of resistance to phages during a short-term selection experiment. This was unlikely driven by demograp…

Clustering of host N-glycans licenses Toxoplasma rhoptry discharge.

Apicomplexan parasites must discharge the contents of specialized organelles called rhoptries into host cells to initiate the process of invasion. This process requires the prior recognition and binding of the host cell by proteins released from another set of parasite organelles, the micronemes. However, the host-parasite interactions required for rhoptry discharge are largely unknown. Here we performed a host-cell directed genome-wide screen for host factors required for rhoptry discharge from Toxoplasma gondii, the causative agent of toxoplasmosis. The screen identified host N-glycosylation…

Experimental infection of wild Mastomys natalensis with the parasitic nematode Trichuris mastomysi.

Trichuris muris infections in laboratory mice underpin current understanding of host-parasite interactions, yet the extent to which these insights apply to natural systems remains largely untested. In this study, we experimentally infected wild-caught Mastomys natalensis with Trichuris mastomysi under controlled conditions. Mice were assigned to either a single high-dose infection or repeated low-dose exposure. Larval infection success and intensity were assessed 21 days post-infection. Infection success was high in both exposure groups, and infection dose strongly influenced parasite burden, …

Seasonal rainfall drives temporal niche partitioning in the helminth community of scorpion mud turtle (Kinosternon scorpioides) from Marajó Island.

The alternation between wet and dry seasons in tropical regions can dramatically affect parasite infection dynamics by influencing larval survival, intermediate-host density, definitive-host foraging behaviour, and host immune function. Freshwater chelonians are excellent models for studying parasite-host ecology. Their longevity, site fidelity, and dietary breadth make them valuable sentinel species for aquatic ecosystem health. Here, we identified the gastrointestinal helminth fauna of scorpion mud turtle (Kinosternon scorpioides) from Marajó Island and evaluated seasonal effects on parasite

Immunological clues to sex differences in parasitic diseases.

The effect of sex on the prevalence and severity of parasitic diseases is an emerging area of research. Several factors underlie sex-based differences, including sociocultural influences that affect exposure to parasites, and physiological disparities linked to biological sex. Hence, human studies must be interpreted cautiously; however, studies conducted under controlled laboratory conditions are important to validate findings in humans. Such research can more effectively elucidate the role of sex-determining physiological factors (particularly their impact on immune responses), as well as th

Phylogenetic and host-parasite relationship analyses of Henneguya caquetaia sp. nov (Myxosporea: Myxobolidae) infecting an Amazonian cichlid fish.

The present study describes a new species of Henneguya infecting the ornamental fish Caquetaia spectabilis from the Brazilian Amazon. Fish specimens were collected where the Tapajós and Amazon rivers merge, municipality of Santarém in the State of Pará, Brazil. Infections were intense, with several plasmodia spread on the opercula, fins and eye. Phylogenetic characterization and host-parasite relationship studies of the new Henneguya species used a combination of small subunit ribosomal DNA (ssrDNA) and morphological (photonic and transmission electron microscopy) analyses. Plasmodia were whit

Trematode parasites exceed aquatic insect biomass in Oregon stream food webs.

Although parasites are increasingly recognized for their ecosystem roles, it is often assumed that free-living organisms dominate animal biomass in most ecosystems and therefore provide the primary pathways for energy transfer. To examine the contributions of parasites to ecosystem energetics in freshwater streams, we quantified the standing biomass of trematodes and free-living organisms at nine sites in three streams in western Oregon, USA. We then compared the rates of biomass flow from snails Juga plicifera into trematode parasites relative to aquatic vertebrate predators (sculpin, cutthro

Allee effect in a manipulative parasite within poikilothermic host under temperature change.

Temperature and intraspecific competition are important factors influencing the growth of all organisms, including parasites. The temperature increase is suggested to stimulate the development of parasites within poikilothermic hosts. However, at high parasite densities, this effect could be diminished, due to stronger intraspecific competition. Our study, for the first time, addressed the joint effects of warming and parasite abundances on parasite growth in poikilothermic hosts. The growth of the common fish parasite larvae (trematode Diplostomum pseudospathaceum) within the rainbow trout at

Apicomplexan co-infections impair with phagocytic activity in avian macrophages.

Mixed infections of Toxoplasma gondii and Eimeria tenella are likely to occur frequently due to the high prevalence of both pathogens in free-ranging chickens. In this study, we investigated the co-occurrence of the two parasites in the same immune-competent host cell towards altered patterns of parasite-host interactions. Chicken blood monocyte-derived macrophages were co-infected with T. gondii RH tachyzoites and E. tenella Houghton sporozoites in vitro for 24 h. Through monitoring the uptake of pH-sensitive pHrodo™ Zymosan BioParticles ("Zymosan") by macrophages, we created a three-dimensio

Associations between Gut Microbiota and Common Luminal Intestinal Parasites.

The development and integration of DNA-based methods in research and clinical microbiology laboratories have enabled standardised and comprehensive detection and differentiation of the microbes colonising our guts. For instance, the single-celled parasites Blastocystis and Dientamoeba appear to be much more common than previously thought, especially so in healthy individuals. While increasing evidence appears to suggest limited pathogenicity of these parasites, next-generation-sequencing-based studies have helped us to appreciate links between parasite colonisation and certain host phenotypica

Parasites, pathogens and commensals in the "low-impact" non-native amphipod host Gammarus roeselii.

BACKGROUND: Whilst vastly understudied, pathogens of non-native species (NNS) are increasingly recognised as important threats to native wildlife. This study builds upon recent recommendations for improved screening for pathogens in NNS by focusing on populations of Gammarus roeselii in Chojna, north-western Poland. At this location, and in other parts of continental Europe, G. roeselii is considered a well-established and relatively 'low-impact' invader, with little understanding about its underlying pathogen profile and even less on potential spill-over of these pathogens to native species.

Drug resistance in vectorborne parasites: multiple actors and scenarios for an evolutionary arms race.

Drug-resistant pathogens emerge faster than new drugs come out of drug discovery pipelines. Current and future drug options should therefore be better protected, requiring a clear understanding of the factors that contribute to the natural history of drug resistance. Although many of these factors are relatively well understood for most bacteria, this proves to be more complex for vectorborne parasites. In this review, we discuss considering three key models (Plasmodium, Leishmania and Schistosoma) how drug resistance can emerge, spread and persist. We demonstrate a multiplicity of scenarios,

Modelling contact spread of infection in host-parasitoid systems: vertical transmission of pathogens can cause chaos.

All animals and plants are, to some extent, susceptible to disease caused by varying combinations of parasites, viruses and bacteria. In this paper, we develop a mathematical model of contact spread infection to investigate the effect of introducing a parasitoid-vectored infection into a one-host-two-parasitoid competition model. We use a system of ordinary differential equations to investigate the separate influences of horizontal and vertical pathogen transmission on a model system appropriate for a variety of competitive situations. Computational simulations and steady-state analysis show t

Role of host feeding niches and host refuges in habitat-related behaviour of Hyssopus pallidus (Hymenoptera: Eulophidae), a larval parasitoid of the codling moth.

Parasitoid fitness depends largely on the capability to locate a host in an ecosystem. A parasitoid of a polyphagous host might not be able to find or to access the host in all its feeding niches. This study evaluated the niche selection of Hyssopus pallidus (Askew), a larval parasitoid of Cydia pomonella (Linnaeus), at the plant level with the goal of assessing its potential for biological control on different fruit crops throughout the plant cycle. Parasitoid behaviour during host location and reproduction rate were investigated on host caterpillars actively feeding on apple, pear, apricot o

Lead poisoning and parasitism in a flock of mute swans (Cygnus olor) in Scotland.

Increased mortality in a flock of non-breeding mute swans (Cygnus olor) on a Scottish loch was investigated. Postmortem examinations were carried out on eight adult and six immature swans. The commonest cause of death, found in eight birds, was lead poisoning associated with the ingestion of large lead fishing weights. Heavy parasitic burdens were found in five immature birds, involving combinations of the gizzard worm Amidostomum species, the thornyheaded worms Polymorphus minutus and Profilicollis anatis, and the tracheal trematode Orchipedum tracheicola. Other parasites of lesser significan

Noninvasively measured immune responses reflect current parasite infections in a wild carnivore and are linked to longevity.

Host immune defenses are important components of host-parasite interactions that affect the outcome of infection and may have fitness consequences for hosts when increased allocation of resources to immune responses undermines other essential life processes. Research on host-parasite interactions in large free-ranging wild mammals is currently hampered by a lack of verified noninvasive assays. We successfully adapted existing assays to measure innate and adaptive immune responses produced by the gastrointestinal mucosa in spotted hyena (Crocuta crocuta) feces, including enzyme-linked immunosor…

Advances in imaging the innate and adaptive immune response to Toxoplasma gondii.

Toxoplasma gondii is an intracellular protozoan parasite that infects a wide variety of warm-blooded hosts and can have devastating effects in the developing fetus as well as the immunocompromised host. An appreciation of how this organism interacts with the host immune system is crucial to understanding the pathogenesis of this disease. The last decade has been marked by the application of various imaging techniques, such as bioluminescent imaging as well as confocal and multiphoton microscopy to study toxoplasmosis. The ability to manipulate parasites to express fluorescent/bioluminescent ma
