Immunohistochemical localization of somatostatin in endocrine cells of the rat stomach.

Using an immunohistochemical technique involving unlabeled antibody and the peroxidase-anti-peroxidase complex, we have localized somatostatin (or growth hormone-release inhibiting hormone), a hypothalamic hormone which can also inhibit gastrin secretion, in the rat stomach. Somatostatin was found to be present in a few cells in the mucosa of the pyloric antrum. These cells are characterized by the presence of secretory granules of about 150-250 nm in diameter and are probably endocrine cells.

TFOS DEWS II Sex, Gender, and Hormones Report.

One of the most compelling features of dry eye disease (DED) is that it occurs more frequently in women than men. In fact, the female sex is a significant risk factor for the development of DED. This sex-related difference in DED prevalence is attributed in large part to the effects of sex steroids (e.g. androgens, estrogens), hypothalamic-pituitary hormones, glucocorticoids, insulin, insulin-like growth factor 1 and thyroid hormones, as well as to the sex chromosome complement, sex-specific autosomal factors and epigenetics (e.g. microRNAs). In addition to sex, gender also appears to be a ris

Opioid modulation of magnocellular neurosecretory cell activity.

Magnocellular neurosecretory cells of the hypothalamic supraoptic and paraventricular nuclei secrete the hormones, oxytocin and vasopressin, into the systemic circulation from the posterior pituitary gland. Oxytocin is important for parturition and is essential for lactation. Vasopressin regulates body fluid homeostasis. The secretion of these hormones is altered in response to peripheral stimuli that are conveyed via projections from other parts of the brain. Endogenous opioid peptide systems interact with the magnocellular neurosecretory system at several levels to restrain the basal secreti

Management of Craniopharyngioma - Perspectives beyond Surgery and Endocrinology.

The excess mortality in craniopharyngiomas is attributable to their size, site and the traditional surgical approach; aggressive resection predisposes to hypothalamic complications such as obesity, somnolence, thirst disorders and neurocognitive dysfunction. Recently, treatment has been modified to partial resection and radiotherapy. The role of the endocrinologist has expanded from identification and replacement of hormone deficits to include management of hypothalamic disease. Future treatment of craniopharyngioma with neo-adjuvant chemotherapy to minimise surgical resection may improve the

Hypothalamic function in patients with tumors of the pineal area.

10 patients, aged 4-17 years, were investigated in order to study the influence of tumors of the pineal area on the hypothalamic endocrine function. Immunoreactive growth hormone (GH) failed to increase sufficiently in 9 patients after insulin induced hypoglycemia (IIH) and in seven patients after propranolol-glucagon (PG). Secondary adrenocortical insufficiency was present in four patients. Three of these patients showed biochemically hypothalamic hypothyroidism with elevated basal prolactin levels, hypogonadotropic hypogonadism and neurohumoral diabetes insipidus. The study shows that hypoth

Treatment of hypothalamic obesity in people with hypothalamic injury: new drugs are on the horizon.

Hypothalamic obesity (HO) is a complex and rare disorder affecting multiple regulatory pathways of energy intake and expenditure in the brain as well as the regulation of the autonomic nervous system and peripheral hormonal signaling. It can be related to monogenic obesity syndromes which often affect the central leptin-melanocortin pathways or due to injury of the hypothalamus from pituitary and hypothalamic tumors, such as craniopharyngioma, surgery, trauma, or radiation to the hypothalamus. Traditional treatments of obesity, such as lifestyle intervention and specific diets, are still a the

Induction of ovulation with clomiphene citrate in combination with metoclopramide in patients with amenorrhea of hypothalamic origin.

Literature data have demonstrated that the chronic use of metoclopramide (MCP), a dopamine antagonist, causes increased gonadotropin secretion in patients with hypothalamic amenorrhea but without triggering ovulation. It has also been observed that women with hypothalamic amenorrhea respond poorly to ovulation induction with clomiphene citrate (CC). On this basis, the objective of the present study was to determine the effect of MCP on the response to CC in patients with hypothalamic amenorrhea in order to evaluate the validity of the simultaneous use of these drugs as ovulation inducers in th

Hormonal basis during pregnancy for the onset of maternal behavior in the rat.

This article reviews the current state of our knowledge about the hormonal basis of maternal behavior in the rat. Considered are the ovarian hormones estrogen and progesterone, the pituitary hormones beta-endorphin and prolactin, and the hormone oxytocin, secreted by several hypothalamic nuclei and associated brain regions. The hormones of pregnancy, estrogen and progesterone, prime the female to respond to a terminal rise in estrogen that stimulates a high level of maternal responsiveness even before parturition begins. Studies on the role of prolactin, using hypophysectomy, prolactin release

Emerging concepts in ventilation-induced lung injury.

Previous research has demonstrated significant associations between increased levels of ovarian hormones and increased rates of binge eating (BE) in women. However, whereas all women experience fluctuations in ovarian hormones across the menstrual cycle, not all women binge eat in response to these fluctuations, suggesting that other factors must contribute. Stress is one potential contributing factor. Specifically, it may be that hormone-BE associations are stronger in women who experience high levels of stress, particularly as stress has been shown to be a precipitant to BE episodes in women

Fluperlapine vs haloperidol: a comparison of their neuroendocrinological profiles and the influence on serum lipids.

Twelve male patients suffering from acute paranoid schizophrenia, randomly receiving fluperlapine - a new dibenzodiazepine very similar to clozapine - or haloperidol were included in a double blind study. Plasma levels of triglyceride, cholesterol and HDL-cholesterol, prolactin, thyroxine, triiodothyronine, ACTH and cortisol were determined. There was no evidence, that fluperlapine has any relevant influence on those hypothalamic-pituitary axis mediated hormones with the possible exception of plasma lipids. These findings, together with the clinical experiences, suggest that fluperlapine might

[The mode of action of neurohormones on the central nervous system (author's transl)].

We tried to understand how hypophysiotropic polypeptides could be carried into some areas of the central nervous system. Granulations of neurosecretion in the ependymal cells and the presence of releasing hormones in the cerebrospinal fluid (CSF) suggest that CSF may transport these substances between the hypothalamus and other areas in the brain. Direct connections between the hypothalamus and other parts of the brain through secreting neurons are not necessary, the CSF transmitting to the cerebral hemispheres the hypothalamic polypeptides by the ventricles at the infundibulum.

Molecular delineation of a retina-dependent photoneuroendocrine pathway in zebrafish.

Many vertebrates, including fish, amphibians, and reptiles, dynamically adjust their body color to the perceived brightness of the ambient background. This response takes tens of minutes and involves the aggregation or dispersion of pigment granules within the melanophores of the skin, resulting in the pale or dark appearance of the animal, respectively.1 In teleosts, ambient light detection depends on the retina, which transmits the signal to the hypothalamus, leading to the secretion of peptide hormones via the pituitary gland into the bloodstream. Melanin-concentrating hormone (MCH) is rele

Sleep in neurodegenerative diseases: A focus on melatonin, melanin-concentrating hormone and orexin.

Sleep and circadian rest-activity rhythm alterations are recognised as inherent clinical features of various neurodegenerative diseases. Traditionally viewed as secondary manifestations of neurodegeneration, recent studies have revealed that disruptions in circadian rhythm and sleep-wake cycles can precede clinical symptoms and significantly contribute to the underlying pathophysiological progression. In this review, we summarise recent research on the impact of sleep and circadian rhythm alterations in ageing and major neurodegenerative diseases, including Alzheimer's, Parkinson's, Huntington

Pituitary atrophy in patients with Sheehan's syndrome.

The cases of two women with anterior hypopituitarism after obstetrical catastrophies with blood loss and hypovolemia are reported. Endocrine studies demonstrated unresponsiveness of all the pituitary hormones on provocative testing in both patients. In one woman, clinically apparent diabetes insipidus was also present. Computed tomography of the hypothalamic pituitary area disclosed empty sella in both patients. A combination of appropriate biochemical and radiologic techniques permits the in vivo documentation of pituitary atrophy, and Sheehan's syndrome should be added to the list of conditi

[Arterial pressure in the ophthalmic artery as a diagnostic test in autoimmune thyroiditis].

Arterial pressure in the ophthalmic artery was studied in 73 patients (135 eyes) with autoimmune thyroiditis in different thyroid function. A rise of diastolic and systolic pressure in the ophthalmic artery was noted in all groups of patients. It could be accounted for by a high degree of activation of the hypothalamic region as a result of sharp deficiency of thyroid hormones. Raised arterial pressure in the ophthalmic artery in patients with euthyroidism made it possible to recommend this method for early diagnosis of autoimmune thyroiditis.

Sheehan's syndrome followed by successful pregnancy: A case report.

Sheehan's syndrome is a complication of pregnancy rarely encountered in modern obstetric practice. The management of this condition involves substitution therapy of the hormones produced by the various endocrine glands under hypothalamic and pituitary control. There have been few reports on the occurrence of spontaneous pregnancy in patients with Sheehan's syndrome. This paper describes the investigation and management of a patient with long-standing panhypopituitarism. Hormone replacement therapy, and specifically induction of ovulation with human menopausal gonadatrophins, resulted in a succ

Hormonal regulation of ovarian follicle growth in humans: Model-based exploration of cycle variability and parameter sensitivities.

We present a modelling and simulation framework for the dynamics of ovarian follicles and key hormones along the hypothalamic-pituitary-gonadal axis throughout consecutive human menstrual cycles. All simulation results (hormone concentrations and ovarian follicle sizes) are in biological units and can easily be compared to clinical data. The model takes into account variability in follicles' response to stimulating hormones, which introduces variability between cycles. The growth of ovarian follicles in waves is an emergent property in our model simulations and further supports the hypothesis

Chronic kidney disease as a potential risk factor for pituitary hormone dysregulation: a review of the underlying mechanisms.

Chronic Kidney Disease (CKD) is a condition of abnormal kidney structure or function, pathologically characterized by chronic inflammation, tubular atrophy, vascular rarefaction, glomerulosclerosis, and interstitial fibrosis. CKD potentially represents a risk factor for several disease conditions, such as cardiovascular disease, mineral bone disorder, and electrolyte imbalance. Hormone dysregulation within the pituitary axis is yet another condition that CKD influences. Notably, CKD dysregulates several anterior pituitary hormones, either by directly influencing hypothalamic-pituitary secretio

The dual burden of obesity: decoding metabolism and female reproductive endocrinology.

The global prevalence of obesity continues to rise, posing a threat to health, especially among women, where obesity can lead to reproductive endocrine disorders. Adipose tissue interacts with endocrine hormones, including insulin, leptin, and sex hormones, resulting in functional abnormalities of the female hypothalamic-pituitary-ovarian axis through various central and peripheral mechanisms. At the same time, systemic inflammation, intestinal microbiota, and metabolites are also implicated in these processes, further linking metabolic imbalance to reproductive endocrine dysfunction.
