Functional characterization of fatty acid synthase gene (FAS) in the reproduction of mirid predator, Cyrtorhinus lividipennis (Hemiptera: Miridae).

Natural enemies serve a crucial role in crop protection through the regulation of pest population dynamics. Cyrtorhinus lividipennis is an important natural enemy of rice planthoppers. Fatty acid synthase (FAS), a multifunctional enzyme crucial for fatty acid biosynthesis, serves as a vital energy source for insect reproduction. However, the function of FAS in the reproductive processes of C. lividipennis remains incompletely understood. In this study, the ClFAS gene was successfully cloned from C. lividipennis. The open reading frame of ClFAS was 7224 bp, encoding a putative protein of 2407 a

The innate immune adaptor protein STING modulates lipid metabolism in an AMPK-dependent manner in Tribolium castaneum.

Stimulator of interferon genes (STING) not only plays an important role in innate immunity, but also acts as a regulator of lipid metabolism in insects. AMP-activated protein kinase (AMPK) serves as a crucial cellular energy sensor that regulates lipid homeostasis. However, the interplay between STING and AMPK remains largely unknown in insects. In this study, STING was cloned from Tribolium castaneum. Spatial-temporal expression analysis revealed that TcSTING was highly expressed in the fat body and in 7-day-old adult females, respectively. Knockdown of TcSTING significantly reduced TAG level

Intersecting experimental evolution and CRISPR screens to identify novel toxin resistance loci.

Understanding toxin resistance in insects is key to appreciating niche adaptations but remains challenging due to its often-polygenic basis. A well-known example is the specialized association of Drosophila sechellia with noni fruit (Morinda citrifolia), which is toxic to other insects, including Drosophila simulans and Drosophila melanogaster. The main noni toxin is octanoic acid (OA), but the mechanisms that determine sensitivity or resistance to OA in different species remain unclear. Here, we experimentally evolved D. simulans with increased OA resistance, identifying multiple loci under s

Immunolocalization of Beet Curly Top Virus (BCTV) and GroEL Chaperon Protein of Endosymbionts in Beet Leafhopper (Circulifer tenellus) Vector Tissue.

Beet curly top virus (BCTV, curtovirus, geminiviridae) causes one of the most economically significant viral diseases in crops in the Western United States and is transmitted only by the beet leafhopper (Circulifer tenellus) in a non-propagative circulative manner. A better understanding of how this virus overcomes insect vector cellular barriers is essential to understanding virus-vector interactions. The distribution of BCTV in its beet leafhopper vector was investigated using immunofluorescence confocal laser scanning microscope analysis (iCLSM) on the whole-mount-dissected organs of leafho

Molecular and functional characterization of heat-shock protein 70 in Aphis gossypii under thermal and xenobiotic stresses.

Aphis gossypii, a globally distributed and economically significant pest of several crops, is known to infest a wide range of host plants. Heat shock proteins (Hsps), acting as molecular chaperones, are essential for the insect's environmental stress responses. The present study investigated the molecular characteristics and expression patterns of AgHsp70, a heat shock protein gene, in Aphis gossypii. Our phylogenetic analysis revealed that AgHsp70 shared high similarity with homologs from other insects, suggesting a conserved function across species. The developmental expression profiles of A

Mining Lygus hesperus (western tarnished plant bug) transcriptomic data for transient receptor potential channels: Expression profiling and functional characterization of a Painless homolog.

The transient receptor potential (TRP) family of cation channels are evolutionarily conserved proteins with critical roles in sensory physiology. Despite extensive studies in model species, knowledge of TRP channel functional diversity and physiological impact remains limited in many non-model insect species. To assess the TRP channel repertoire in a non-model agriculture pest species (Lygus hesperus), publicly available transcriptomic datasets were mined for potential homologs. Among the transcripts identified, 30 are predicted to encompass complete open reading frames that encode proteins re

Farnesyl biliverdins IXα are novel ligands of biliproteins from moths of the Noctuoidea superfamily: A chemosystematic view of the Lepidoptera.

Bilins, derived from biliverdin IXα, are known from animals, plants and microorganisms, where they play vital roles as light-absorbing pigments. Bilins occur also in many insects. Recently, we discovered in insects a novel structural type of bilins with a farnesyl substituent at pyrrole ring A of biliverdin IXα. The first of these unusual bilins with a molecular mass of 852 (C48H60O10N4) was identified in Cerura vinula, subsequently in Spodoptera littoralis; both species are members of the Noctuoidea superfamily of moths. From an evolutionary point of view, it was of interest to examine other

Molecular cloning of bruchid (Zabrotes subfasciatus) alpha-amylase cDNA and interactions of the expressed enzyme with bean amylase inhibitors.

alpha-Amylases are important digestive enzymes in weevils that infest starchy seeds, and plants have evolved proteinaceous alpha-amylase inhibitors (alpha AI) for protection. To gain a better understanding of the interaction between weevil alpha-amylases and alpha AIs, we cloned the alpha-amylase cDNA of Zabrotes subfasciatus larvae. Larvae of this bruchid infest seeds of cultivated varieties of the common bean, Phaseolus vulgaris, although the seeds contain high levels of an alpha AI. The alpha-amylase cDNA, called ZsAmy, encodes a mature protein of 466 amino acids with a signal peptide of 17

Functional characterization of vitellogenin unveils novel roles in RHBP uptake and lifespan regulation in the insect vector Rhodnius prolixus.

In insects, vitellogenesis plays a critical role in providing the energy reserves needed for embryonic development as it ensures the accumulation of yolk in the oocytes. Vitellogenin (Vg), the precursor to vitellin (Vt), is primarily synthesized in the fat body of females and transported to the oocytes via receptor-mediated endocytosis. In Rhodnius prolixus, a key vector of Chagas disease, two Vg genes, Vg1 and Vg2, were characterized. These genes share 65 % amino acid identity and present the conserved Vitellogenin_N, DUF1943, and VWD domains typical of Vg proteins across various species. We

RNAi assay in the 28-spotted ladybeetle Henosepilachna vigintioctopunctata suggests nuclear receptor HR3 as a potential molecular target for pest control.

BACKGROUND: Nuclear hormone receptors play crucial roles in embryonic development, neural regulation, metabolism, and organogenesis in insects. This study aimed to investigate the expression and function of the nuclear hormone receptor 3 (HR3) in the 28-spotted ladybeetle, Henosepilachna vigintioctopunctata. RESULTS: Our findings revealed that HvHR3 was expressed at all developmental stages, with the highest expression levels in the 1st instar larvae, 3rd instar larvae, and pupa. HvHR3 silencing in both the 1st and 3rd instar larvae resulted in high mortality rates. Reduced HvHR3 expression i

Identification of chemosensory genes and antennal sensilla in Nassophasis sp. (Coleoptera: Rhynchophorinae).

Phytophagous insects rely on plant volatiles to select and locate hosts for feeding or reproduction and their olfactory system is essential for detecting plant volatiles. The stem-boring pest, Nassophasis sp. damages Dendrobium and causes economic losses. Currently, there are no effective methods for its control. However, understanding the morphological and molecular basis of its olfactory system may identify new pathways for their management and control. In this study, we observed the stemborer's antennal sensilla using scanning electron microscopy, and transcriptome sequencing was undertaken

Identification of Venom Proteins of the Indigenous Endoparasitoid Chouioia cunea (Hymenoptera: Eulophidae).

Chouioia cunea (Yang) (Hymenoptera: Eulophidae) is an indigenous pupal endoparasitoid that effectively attacks the exotic fall webworm Hyphantria cunea (Drury) (Lepidoptera: Arctiidae) in China. In this novel association, the parasitoid's venom has played an important role in subduing the immune defense of the host although little is known about the composition and functions of the parasitoid's venom. We therefore first identified the parasitoid's major venom proteins using electrospray ionization-mass spectrometry (ESI-MS). Approximately 207 different proteins were identified from C. cunea's

Exoproteome of Aspergillus flavus corneal isolates and saprophytes: identification of proteoforms of an oversecreted alkaline protease.

UNLABELLED: Aspergillus flavus infects the human eye leading to keratitis. Extracellular proteins, the earliest proteins that come in contact with the host and virulence related exoproteins, were identified in the fungus isolated from infected cornea. Virulence of the corneal isolates was tested in the Galleria mellonella larvae model and those isolates showing higher virulence were taken for subsequent exoproteome analysis. High resolution two-dimensional electrophoresis and mass spectrometry were used to generate A. flavus exoproteome reference map as well as to profile most of the exoprotei

Recombinant expression and biochemical characterization of the catalytic domain of acetylcholinesterase-1 from the African malaria mosquito, Anopheles gambiae.

Acetylcholinesterases (AChEs) and their genes from susceptible and resistant insects have been extensively studied to understand the molecular basis of target site insensitivity. Due to the existence of other resistance mechanisms, however, it can be problematic to correlate directly a mutation with the resistant phenotype. An alternative approach involves recombinant expression and characterization of highly purified wild-type and mutant AChEs, which serves as a reliable platform for studying structure-function relationships. We expressed the catalytic domain of Anopheles gambiae AChE1 (r-AgA

S1 domain of the porcine epidemic diarrhea virus spike protein as a vaccine antigen.

BACKGROUND: Porcine epidemic diarrhea virus (PEDV) is a highly contagious virus infecting pigs of all ages with high morbidity and mortality among newborn piglets. Currently, there is no effective vaccine available to protect the pigs from PEDV. The N-terminal subunit of spike protein (S1) is responsible for virus binding to the cellular receptor and contains a number of neutralizing antibody epitopes. Thus, we expressed and produced recombinant S1 protein to protect newborn piglets by immunization of sows. METHODS: Affinity tagged PEDV S1 protein was expressed in a secretory form in yeast, i

A seamless connection from the burst sequence to the start codon is essential for polyhedrin hyperexpression in alphabaculoviruses.

Alphabaculoviruses produce a large number of occlusion bodies (OBs) in host cells during the late stage of infection. OBs are mainly composed of polyhedrin (POLH), and high-level transcription of the polh gene has been exploited to express foreign proteins in insect cells. While making Bombyx mori nucleopolyhedrovirus (BmNPV) polh mutants using a conventional transfer vector-based method, we noticed that a virus with a short sequence insertion just before the polh start codon produces fewer very small OBs. Detailed analysis of several BmNPV mutants revealed that insertions between the burst se

The Nuts and Bolts of SARS-CoV-2 Spike Receptor-Binding Domain Heterologous Expression.

COVID-19 is a highly infectious disease caused by a newly emerged coronavirus (SARS-CoV-2) that has rapidly progressed into a pandemic. This unprecedent emergency has stressed the significance of developing effective therapeutics to fight the current and future outbreaks. The receptor-binding domain (RBD) of the SARS-CoV-2 surface Spike protein is the main target for vaccines and represents a helpful "tool" to produce neutralizing antibodies or diagnostic kits. In this work, we provide a detailed characterization of the native RBD produced in three major model systems: Escherichia coli, insect

Molecular cloning and characterization of major vault protein of Echinococcus multilocularis.

The cDNA clone coding a major vault protein (MVP)-like protein was derived from Echinococcus multilocularis cysts. MVP is a main component of vault particles, which are the largest cytoplasmic ribonucleoprotein particles in eukaryotic cells. We sequenced and characterized E. multilocularis MVP (EmMVP). The nucleotide sequence of the emmvp cDNA clone was 2607 bp in the full length open reading frame and its deduced amino acid sequence had several signature motifs which were specific to MVP families. Immunoblot analysis with mouse anti-EmMVP antiserum revealed that crude antigens of E. multilocu

Phlebotomus papatasi sand fly predicted salivary protein diversity and immune response potential based on in silico prediction in Egypt and Jordan populations.

Phlebotomus papatasi sand flies inject their hosts with a myriad of pharmacologically active salivary proteins to assist with blood feeding and to modulate host defenses. In addition, salivary proteins can influence cutaneous leishmaniasis disease outcome, highlighting the potential of the salivary components to be used as a vaccine. Variability of vaccine targets in natural populations influences antigen choice for vaccine development. Therefore, the objective of this study was to investigate the variability in the predicted protein sequences of nine of the most abundantly expressed salivary
