Breast and mammary epithelial cells experience different local environments during tissue development and tumorigenesis. Microenvironmental heterogeneity gives rise to distinct cell regulatory states whose identity and importance are just beginning to be appreciated. Cellular states diversify when clonal three-dimensional (3D) spheroids are cultured in basement membrane, and one such state is associated with stress tolerance and poor response to anticancer therapeutics. Here, we found that this state was jointly coordinated by the NRF2 and p53 pathways, which were costabilized by spontaneous o

Cysteine (Cys) is crucial for redox balance and metabolism but real-time in situ detection of Cys in plants limited by tissue penetration, sensitivity, and response time. Herein, a novel fluorescent probe, DCI-Cys, is developed for the detection of Cys, which exhibits characteristics of near-infrared emission (650 nm), high sensitivity (30.32 nM), and rapid response (80 s). With the aid of DCI-Cys, real-time imaging of Cys in various plant tissues is innovatively achieved, and fluctuations in endogenous Cys levels in Nicotiana tabacum L. seedlings during exposure to chromium (Cr) and drought s

Dephosphorylation of translation initiation factor 2 (eIF2α) terminates signalling in the mammalian integrated stress response (ISR) and has emerged as a promising target for modifying the course of protein misfolding diseases. The [(o-chlorobenzylidene)amino]guanidines (Guanabenz and Sephin1) have been proposed to exert protective effects against misfolding by interfering with eIF2α-P dephosphorylation through selective disruption of a PP1-PPP1R15A holophosphatase complex. Surprisingly, they proved inert in vitro affecting neither stability of the PP1-PPP1R15A complex nor substrate-specific d…

Cytokines are key factors in the cross talk between the immune system and other systems including hepatic, nervous, cardiac and cutaneous systems, leading to an adaptive and integrated response of the organism to stress. They are also involved in the regulation of many processes, including hematopoiesis, the immune response and inflammation. IL-10 is one of the most important anti-inflammatory cytokines. Five cytokines structurally related to IL-10 have been described and presently form this family of cytokines: IL-19, IL-20, IL-22, IL-24 and IL-26. In contrast to IL-10, these cytokines displa

BACKGROUND: Proline plays a critical regulatory role in microbial stress responses; however, its specific mechanism of action under ethanol stress in Wickerhamomyces anomalus during logarithmic growth remains poorly understood.
RESULTS: This study investigated the protective effects of proline against ethanol-induced injury and elucidated the mechanism by which it enhances resistance to 9% (v/v) ethanol stress. The results indicated that proline supplementation significantly increased the viability of ethanol-stressed yeast and stimulated cellular proline transport and catabolism. Furthermore

Finally, we present preliminary evidence of integrated stress response activity that may serve as an alternative hypothesis to what drives entorhinal degeneration after silencing. Our data demonstrate that c-Jun is activated in response to neuronal silencing in the entorhinal cortex but is decoupled from subsequent neu …

Stress signaling is important for determining the fates of neurons following axonal insults. Previously, we showed that the stress-responsive kinase PERK contributes to injury-induced neurodegeneration (Larhammar et al., 2017). Here, we show that PERK acts primarily through activating transcription factor-4 (ATF4) to stimulate not only pro-apoptotic but also pro-regenerative responses following optic nerve damage. Using conditional knockout mice, we find an extensive PERK/ATF4-dependent transcriptional response that includes canonical ATF4 target genes and modest contributions by C/EBP Homolog

Waterlogging stress significantly impairs early development and yield in Brassica napus, yet the underlying epigenetic regulatory mechanisms remain poorly understood. We applied a multi-omics strategy integrating RNA-seq, WGBS, and ChIP-seq to dissect multi-layered regulatory dynamics during waterlogging stress and recovery. We found that waterlogging stress globally repressed gene expression and enhanced expression divergence between homoeologous genes of the two subgenomes. Transposable element (TE) activation, together with dynamic epigenetic changes, is associated with altered expression o

Proton (H+) fluxes in plant roots play critical roles in maintaining root growth and facilitating plant responses to multiple soil stresses, including fluctuations in nutrient supply, salt infiltration, and water stress. Soil mining for nutrients and water, rates of nutrient uptake, and the modulation of cell expansion all depend on the regulation of root H+ fluxes, particularly at the root apex, mediated primarily by the activity of plasma membrane (PM) H+-ATPases. Here, we summarize recent findings on the regulatory mechanisms of H+ fluxes at the root apex under three abiotic stress conditio

Metal-based nanoparticles are increasingly encountered in human exposure scenarios, yet the relationship between residue-level exposure and cellular hazard remains unclear. In this study, iron (Fe), copper (Cu), and silver (Ag) nanoparticles were applied to chilli pepper plants to estimate plant-associated metal nanoparticle residues in the edible fruits. Subsequently, exposure-relevant concentration ranges were defined and examined in normal human WI-38 fibroblasts using integrated cytological and transcriptomic approaches. Metal accumulation was higher in vegetative tissues than fruits, with

Tardive dyskinesia (TD) is a persistent hyperkinetic movement disorder associated with prolonged dopamine D2 receptor blockade, particularly during chronic haloperidol (HP) exposure. Emerging evidence suggests that TD-like pathology is sustained by an interconnected redox-mitochondrial-inflammatory network within striatal circuits; however, the regulatory architecture of this network remains incompletely defined. Hyperoside (HS), a flavonol glycoside with cytoprotective properties, has been implicated in cellular stress-response modulation, yet its role in antipsychotic-induced motor dysfuncti

During infection, Mycobacterium tuberculosis (Mtb) encounters multiple environmental stressors, including nitric oxide (NO) and iron limitation, and an ability to mount an integrated response is essential for the bacterium's adaptation and continued survival. Iron-containing prosthetic groups in key enzymes are critical for Mtb sensing and detoxification of NO, and there is significant overlap between NO- and low iron-responsive genes. However, how Mtb adapts to these two stressors concurrently is largely unknown. Here, we find that exposure to NO globally augments expression of low iron-respo

Benzophenone (BP) is a widely used organic UV filter in personal care products. Despite its frequent detection in soil ecosystems, its toxicity to soil biota has not been extensively explored. This study investigates the effects of BP on earthworms (Eisenia fetida) exposed in artificial soil. Soil degradation kinetics followed a single first-order model at low (1.69 mg/kg) and medium (3.39 mg/kg) concentrations, and a biphasic double first-order in parallel model at high (16.9 mg/kg) concentration. The calculated DT50 (time to 50 % degradation) and DT90 (time to 90 % degradation) ranged from 0

Climate is becoming more variable, and the frequency and magnitude of drought and pathogen epidemics are changing. This creates a need to understand how limited water availability influences plant responses to biotic challenges. In Brassica napus L., drought decreases the disease progression and fungal colonization of the vascular pathogen Verticillium longisporum, yet the underlying regulatory mechanisms remain unclear. Here, we used an integrative approach combining transcriptome profiling, co-expression network analysis, and DNA methylation analysis to investigate host responses in hypocoty

Etomidate has been linked to addictive behaviors and neurotoxic and hepatotoxic effects. We established a mouse exposure model across graded etomidate doses and assessed reward using conditioned place preference (CPP). Proteomic and metabolomic profiling of cerebral tissue and right hepatic lobe tissue was performed using data-independent acquisition (DIA) liquid chromatography-tandem mass spectrometry and quadrupole time-of-flight mass spectrometry, respectively; integrated multi-omics candidates were verified by parallel reaction monitoring (PRM). Both low- and high-dose etomidate significan

Nickel (Ni) contamination has become an increasingly important environmental issue, and phytoremediation is considered a sustainable strategy for its remediation. Leersia hexandra Swartz shows strong tolerance to multiple heavy metals, yet the mechanisms underlying its root response to Ni stress remain poorly understood. In this study, L. hexandra was exposed to Ni concentration gradient treatments, followed by inhibitor assays, synchrotron-based microspatial imaging, and integrated root transcriptomic and metabolomic analyses of roots to investigate its root-level responses to Ni stress. Cancer stem cells (CSCs) contribute to therapeutic resistance, metastatic progression, and tumor recurrence, yet the metabolic pathways that sustain their survival remain incompletely understood. Here, we identify 3-mercaptopyruvate sulfurtransferase (3-MST), a hydrogen sulfide-producing enzyme encoded by MPST, as a metabolic dependency of colorectal CSCs. 3-MST expression was increased in human colorectal tumors and cancer cell lines and strongly correlated with proliferative capacity. HCT116-derived CSCs exhibited elevated 3-MST expression, increased hydrogen sulfide and reactive sulfur spec
