Mechanosensitive ion channels in intestinal homeostasis and disease.

Mechanical deformation is integral to intestinal physiology, linking luminal transit to epithelial adaptation and motor control. These physical cues engage multiple mechanotransduction pathways that coordinate cellular adaptation and help preserve intestinal homeostasis. Inflammation, obstruction and matrix remodelling alter both the physical input and the state of the responding cell, allowing normally transient signals to persist or acquire pathological consequences. Ion channels act at several levels of this process. PIEZO1 and PIEZO2 provide a direct route for force-gated cation entry; mec

Ion Mix Can Invert Centrifugal Confinement.

Centrifugal plasma traps, in which plasma is confined partly by centrifugal forces, represent a possible path to fusion energy production. In centrifugal plasma traps, electric fields naturally arise in the direction parallel to the magnetic field in order to ensure quasineutrality. These electric fields are sensitive to the particular mix of ion species present. This Letter uses analytic and numerical calculations to consider how these mix effects could be used to improve device performance. Changing the composition of the plasma can mix or demix different ion species, improve confinement of …

A three-dimensional scanning trapped-ion probe.

The performance of a wide range of physical systems suffers from undesired fields from nearby surfaces, placing a premium on developing methods for understanding these imperfections. Trapped ions excel at sensing surface electric fields, but spatial scanning was limited to linear translations in previous work using radio-frequency traps, restricting the conclusions that could be drawn. Here, we demonstrate a single ion sensor confined using only static fields, consequently allowing three-dimensional position scanning and the possibility to isolate the trap from external noise. We measure stati…

Voltage gated ion channels and breast cancer.

Breast cancer is the leading form of cancer in women worldwide. Despite extensive research efforts to develop detection methods and treatment, mortality remains high. Biomarkers and therapeutic targets are needed. Tumoral cell express ion channels which may serve both functions. Expression of specific ion channels by neoplastic cells within tissues that normally do not express those channels, or expression of embryonic splice variants may be useful markers for early detection. Ion channels associated with metastatic potential or cell migration may also be useful therapeutic targets.

Spectrum and functions of ion channels and transporters in osteoclasts.

Osteoclasts are essential for bone resorption and interact with osteoblasts during bone remodeling. Ion channels and transporters located in the ruffled border or intracellular vesicles coordinate the transport of various ions and substrates, which is fundamental to the primary functions of osteoclasts. Numerous channels and transporters are implicated in bone metabolic disorders and genetic diseases. Among these, the voltage-gated chloride channel 7 (ClC-7) and vacuolar proton ATPases (V-ATPase) represent the most well-characterized examples in osteoclasts. Using the classification system of …

Vitamin D Activation and Mineral Ion Regulation.

Nutrients frequently act in coordination with the absorption and metabolism of one nutrient often interdependent on the presence of others. For example, magnesium is essential for the activation of vitamin D, which, in turn regulates calcium and phosphate homeostasis, both of which are critical for cellular and organ function. A comprehensive understanding of disorders arising from disruptions in vitamin D and mineral ion metabolism is essential for explaining the pathophysiological mechanisms associated with their imbalance. The chapters of this book are selected to provide in-depth insights …

Ionic Sliding Ferroelectricity in Layered Ion Conductors.

Recent research on sliding ferroelectricity in various two-dimensional bilayers and multilayers has shown its promising application potential and emerging new physics distinct from conventional ferroelectricity. However, it stems from van der Waals interactions of asymmetrically stacked layers, so the polarizations are weak. Here, we propose that in most layered ion conductors, the asymmetric stacking of layers will give rise to a unique type of ionic sliding ferroelectricity. The major weakness is overcome since the vertical polarizations are generally enhanced by an order of magnitude, which…

Hydride Ion Conductors with Polyanionic Complex Anions.

Hydride ion (H-)-conducting solid-state materials have lately received great attention for advanced electrochemical energy storage/conversion systems, including hydride ion-based batteries, electrolysis, and fuel cells. However, the highly reactive nature of hydride ions has posed challenges in diversifying anion systems, which are crucial for designing their effective transport. This study reports perovskite-type hydride ion conductors, Sr1-yNayLiH3-x-y(BH4)x (0 ≤ x ≤ 1 and 0 ≤ y ≤ 0.1), employing polyanionic borohydride (BH4-) as the anions in the structure. Structural characterization indic…

Copper ion incorporation in α-synuclein amyloids.

Copper ion dys-homeostasis is linked to neurodegenerative diseases involving amyloid formation. Even if many amyloidogenic proteins can bind copper ions as monomers, little is known about copper interactions with the resulting amyloid fibers. Here, we investigate copper interactions with α-synuclein, the amyloid-forming protein in Parkinson's disease. Copper (Cu(II)) binds tightly to monomeric α-synuclein in vitro involving the N-terminal amine and the side chain of His50. Using purified protein and biophysical methods in vitro, we reveal that copper ions are readily incorporated into the form…

Oxidative stress and ion channels in neurodegenerative diseases.

Numerous neurodegenerative diseases result from altered ion channel function and mutations. The intracellular redox status can significantly alter the gating characteristics of ion channels. Abundant neurodegenerative diseases associated with oxidative stress have been documented, including Parkinson's, Alzheimer's, spinocerebellar ataxia, amyotrophic lateral sclerosis, and Huntington's disease. Reactive oxygen and nitrogen species compounds trigger posttranslational alterations that target specific sites within the subunits responsible for channel assembly. These alterations include the adjus…
