Do Autistic and Depressed Rats Express the Same Type of Maternal Care?

Autism is a neurodevelopmental disorder that is more frequently diagnosed in men. Nevertheless, through current diagnostic tools, women have also been found to be affected by this disorder, but in different ways. Few studies have been conducted regarding unique periods of life, such as motherhood. Yet, extant literature has already described the existence of a comorbidity between autism and postpartum depression. Thus, this study aimed to compare the maternal care sphere between two animal models of these diseases. Lactating rats were subdivided into three groups (n = 8 animals/group): 1) cont

Development of a constant pressure perfused ex vivo model of the equine larynx.

Distal axonopathy is seen in a broad range of species including equine patients. In horses, this degenerative disorder of the recurrent laryngeal nerve is described as recurrent laryngeal neuropathy (RLN). The dysfunctional innervation of the cricoarytenoideus dorsalis muscle (CAD) leads to a loss of performance in affected horses. In general, ex vivo models of the larynx are rare and for equine patients, just one short report is available. To allow for testing new therapy approaches in an isolated organ model, we examined equine larynges in a constant pressure perfused setup. In order to chec

Impaired CO2 sensitivity of astrocytes in a mouse model of Rett syndrome.

Rett syndrome, a prototypical neurological disorder caused by loss of function of the transcriptional regulator methyl-CpG-binding protein 2 (MeCP2) gene, is associated with a severely disordered breathing pattern and reduced ventilatory CO2 sensitivity. In a mouse model of Rett syndrome (MeCP2 knockout), re-introduction of the MeCP2 gene selectively in astrocytes rescues normal respiratory phenotype. In the present study we determined whether the metabolic and/or signalling functions of astrocytes are affected by testing the hypotheses that in conditions of MeCP2 deficiency, medullary astrocy

Systemic Modulation of Group II mGluRs Increases Extracellular Lactate in the Rat Prefrontal Cortex.

BACKGROUND: Lactate is increasingly recognized as a dynamic indicator of neuronal activity and brain energy metabolism and is closely linked to glutamatergic neurotransmission via the astrocyte-neuron lactate shuttle. Group II metabotropic glutamate receptors (mGluR2/3) regulate glutamate release and have been implicated in psychiatric disorders; however, their role in cortical lactate metabolism remains unclear. METHODS: In this study, we examined the effects of pharmacological modulation of mGluR2/3 on extracellular lactate concentration (eLAC) in the medial prefrontal cortex of naïve rats

Radiosynthesis and Preclinical Evaluation of Enantiopure (R)-[18F]Fluorolactate for PET Imaging of Metabolic Processes.

L-Lactate is a key substrate in energy metabolism and a potential biomarker for cancer and metabolic disorders. We developed an enantiomerically pure positron emission tomography (PET) tracer, (R)-[18F]fluorolactate, to mimic natural L-lactate. (R)-[18F]Fluorolactate was synthesized via nucleophilic ring-opening of benzyl (S)-oxirane-2-carboxylate, optimized to yield >91% radiochemical purity, > 95% enantiomeric excess, pH 6, and >5.56 GBq/μmol molar activity. PET/computed tomography (CT) imaging, biodistribution, autoradiography, and stability analyses were conducted in fasted and nonfasted r

Ultramicronized palmitoylethanolamide restores astrocyte-neuron metabolic coupling and Klotho/FGF21 signaling in a triple-transgenic mouse model of Alzheimer's disease.

Alzheimer's disease (AD), a multifactorial neurodegenerative disorder, is characterized by metabolic deficiency, neuroinflammation, and synaptic impairment. Astrocyte-neuron metabolic coupling regulates cerebral energy homeostasis through key metabolites such as lactate, glutamate, and taurine. We investigated the therapeutic potential of ultramicronized-palmitoylethanolamide (um-PEA) in restoring the homeostasis of these metabolites in the triple transgenic (3 ×Tg-AD) mouse model of AD. Using in vivo magnetic resonance imaging and spectroscopy (MRI/MRS) combined with Western blot, we evaluate

Juvenile social isolation leads to schizophrenia-like behaviors via excess lactate production by astrocytes.

Repeated early environmental deprivation is regarded as a typical paradigm to mimic the behavioral abnormalities and brain dysfunction that occur in psychiatric disorders. Previously, we reported that social isolation could disrupt prepulse inhibition (PPI) in Sprague-Dawley (SD) rats, producing the typical characteristics of a schizophrenia animal model. Based on further analysis of previous proteomic and transcriptomic data, a disrupted balance of glucose metabolism was found in the prefrontal cortex (PFC) of isolated rats. Subsequently, in the first experiment of this study, we investigated

Lactate Administration Reduces Brain Injury and Ameliorates Behavioral Outcomes Following Neonatal Hypoxia-Ischemia.

Neonatal hypoxic-ischemic encephalopathy is a major cause of mortality and disability in newborns and the only standard approach for treating this condition is therapeutic hypothermia, which shows some limitations. Thus, putative neuroprotective agents have been tested in animal models. The present study evaluated the administration of lactate, a potential energy substrate of the central nervous system (CNS) in an animal model of hypoxia-ischemia (HI), that mimics in neonatal rats the brain damage observed in human newborns. Seven-day-old (P7) male and female Wistar rats underwent permanent co

Clinical response of quetiapine in rapid cycling manic bipolar patients and lactate level changes in proton magnetic resonance spectroscopy.

The aim of the current study was to evaluate the relationship between quetiapine's effect on the improvement of mood symptoms in bipolar patients and brain metabolite level changes as measured by proton magnetic resonance spectroscopy ((1)H-MRS). Rapid cycling bipolar patients in the manic state were recruited and treated with quetiapine for 12 weeks. Clinical assessment was performed using the Young Mania Rating Scale (YMRS), the 17-item Hamilton Depression Rating Scale (HDRS) and the Clinical Global Impression-Severity scale (CGI-S) at baseline and weekly intervals during the 12-week period.

Dysphoric Milk Ejection Reflex: Prevalence, persistence, and implications.

BACKGROUND: Dysphoric Milk Ejection Reflex (D-MER), characterized by an abrupt wave of negative emotions during milk-letdown, affects a relevant proportion of lactating parents and may significantly impact feeding choices, but remains poorly understood. METHODS: We conducted a cross-sectional study using an online survey of lactating parents of children 18 months and younger who were nursed for any period of time. RESULTS: The prevalence of D-MER in our cohort was 14.2 % (n = 209/1469). Among the 209 women who reported having experienced D-MER, the most frequently selected descriptions were:

Reasons for giving up breastfeeding and support during problems with lactation in the north-western part of Poland.

INTRODUCTION: Breastfeeding is the optimal way of feeding for neonates and infants. Its duration is of vital importance for the child's development and health. The World Health Organization recommends exclusive breastfeeding until the end of the sixth month of life. Many mothers give up breastfeeding earlier than recommended. Professional and non-professional support plays a great role in motivation to start and continue breastfeeding. The aim of this study was to determine how long infants from Szczecin, the capital city of the North-Western region of Poland, are breastfed, what are the reaso

[Localization of pathologically secreting lactiferous ducts].

During the investigation of ductographic findings in 200 patients with secretory disease of the breasts it was discovered that pathologically secreting lactiferous ducts were statistically predominantly localized in the retroareolar region and in the lower quadrates of the breasts. This prevalence does not correspond with the regions of the most voluminous portions of the mammary gland and with the most frequent localization of mammography manifestations of its fibroepithelial dysplasia.

Galactorrhea: a rare complication following reduction mammaplasty.

A case of galactorrhea immediately following a reduction mammaplasty is presented. Prolactin levels were elevated postoperatively and could be reduced to normal by a prolactin inhibitor. No other disease or disorder could be found. Based on a review of the literature, it seems that various factors have to contribute to galactorrhea, including stress-related prolactin secretion, rebound phenomenon after discontinuation of birth control pills, and hypersensitivity to prolactin receptors. Their coincidental occurrence, resulting in abundant milk production after surgery, is discussed.

[Psychopharmacology of panic attacks].

A new type of anxiety state has recently been identified: the panic attack. This condition is qualitatively distinct from other anxiety disturbances and its treatment is different. Sodium lactate seems to play an essential role in the triggering of panic attacks through the intermediary of catabolic, physicochemical mechanisms or by alteration of adrenergic neurotransmission. Anxiety may be autonomised into two sub-groups, based on the effects of psychotropic drugs: phobic and anxiety states. The presence or absence of panic attacks is essential as it has diagnostic and therapeutic implication
