S100A9 as a Candidate Molecular Bridge in Hepato-Ocular Crosstalk.

Increased S100 calcium-binding protein A9 (S100A9)-related signals have been reported in selected hepatic and ocular inflammatory settings. This structured narrative review evaluates S100A9-related species as candidate participants in hepato-ocular crosstalk. Across human, ocular fluid, animal, and cellular studies, the available findings provide context-specific support for disease-associated hepatic expression and ocular responsiveness, with stronger evidence for selected local S100A9-Toll-like receptor 4 (TLR4)-associated effects than for S100A9-specific receptor for advanced glycation end

Water-accommodated fractions of crude oil and its mixture with chemical dispersant impairs oxidase stress and energy metabolism disorders in Oryzias melastigma embryos.

In this study, marine medaka (Oryzias melastigma) embryos were exposed to different concentrations of water-accommodated fractions (WAFs) and chemically enhanced water-accommodated fractions (CEWAFs) of Oman crude oil for 14 d by semi-static exposure methods. The effects on growth and development and energy metabolism process were evaluated. Results showed that embryo survival and hatchability were decreased in a dose-dependent manner with an increase in the concentration of petroleum hydrocarbon compounds, whereas the malformation exhibited a dose-dependent increase. Compared to the control,

Metabolic Flexibility and Its Impact on Health Outcomes.

A metabolically flexible state exists when there is a rapid switch between glucose and fatty acids during the transition between the fed and fasting state. This flexibility in fuel choice serves to prevent hyperglycemia following a meal and simultaneously ensures an adequate amount of blood glucose is available for delivery to the brain and exclusively glycolytic tissues during fasting. The modern era is characterized by chronic overnutrition in which a mixture of fuels is delivered to the mitochondria in an unabated manner thereby uncoupling the feast and famine situation. The continuous infl

Ideal lipid profile and genes for an extended life span.

PURPOSE OF REVIEW: The world population is aging and a rapid increase is being seen in the very elderly (aged >80 years). Cholesterol levels in general rise with age and high cholesterol has been associated with extreme longevity. The relationship between lipids and cardiovascular events in the extreme elderly is unclear. RECENT FINDINGS: A number of genetic factors associated with lipid metabolism have also been described as having potential antiaging roles, including the genes encoding lipoprotein-associated factors - apolipoprotein E and cholesterol ester transfer protein; adipose tissue m

The Effect of Rivastigmine on Enhanced Glucose Metabolism and Enhanced Synaptic Plasticity in the Hippocampus of Aged Rats: A Sex-Dependent Study.

The aim of this study was to investigate the enhancing effects of rivastigmine administration on hippocampal glucose metabolism, insulin signaling pathways, and synaptic plasticity, independently of cognitive function, in an aged rat model. Thirteen-month-old male and female Wistar rats were randomly divided into four groups: male control, male rivastigmine, female control, and female rivastigmine. Rivastigmine (2 mg/kg) or an equivalent volume of physiological saline was administered intraperitoneally for 21 days. At the end of the experiment, the animals were sacrificed, and hippocampal tiss

Transcriptional and biochemical changes in mouse liver following exposure to a metal/drug cocktail. Attenuating effect of a selenium-enriched diet.

Real-life pollution usually involves simultaneous co-exposure to different chemicals. Metals and drugs are frequently and abundantly released into the environment, where they interact and bioaccumulate. Few studies analyze potential interactions between metals and pharmaceuticals in these mixtures, although their joint effects cannot be inferred from their individual properties. We have previously demonstrated that the mixture (PC) of the metals Cd and Hg, the metalloid As and the pharmaceuticals diclofenac (DCF) and flumequine (FLQ) impairs hepatic proteostasis. To gain a deeper vision of how

Utility of human induced pluripotent stem cell-derived small intestinal epithelial cells for pharmacokinetic, toxicological, and immunological studies.

The small intestine, which plays a crucial role in the absorption and metabolism of drugs and foods, serves as a target organ for drug-induced toxicity and immune interactions with functional foods and intestinal bacteria. Current alternative models of the human small intestine, such as Caco-2 cells and experimental animals, have limitations due to variations in the expression levels of metabolic enzymes, transporters, and receptors. This study presents investigations into the utility of human induced pluripotent stem cell-derived small intestinal epithelial cells (hiSIECs) for pharmacokinetic

Rhesus monkeys as a translational model for late-onset Alzheimer's disease.

Age is a major risk factor for late-onset Alzheimer's disease (AD) but seldom features in laboratory models of the disease. Furthermore, heterogeneity in size and density of AD plaques observed in individuals are not recapitulated in transgenic mouse models, presenting an incomplete picture. We show that the amyloid plaque microenvironment is not equivalent between rodent and primate species, and that differences in the impact of AD pathology on local metabolism and inflammation might explain established differences in neurodegeneration and functional decline. Using brain tissue from transgeni

Hypercholesterolemia and Ecto-enzymes of Purinergic System: Effects of Paullinia cupana.

Hypercholesterolemia is a metabolic disorder characterized by high levels of low-density lipoprotein and blood cholesterol, causing inflammatory lesion. Purinergic signaling modulates the inflammatory and immune responses through adenine nucleotides and nucleoside. Guaraná has hypocholesterolemic and antiinflammatory properties. Considering that there are few studies demonstrating the effects of guaraná powder on the metabolism of adenine nucleotides, we investigated its effects on the activity of ecto-nucleoside triphosphate diphosphohydrolase (E-NTPDase) and ecto-adenosine deaminase activity

Weight regulation, leptin and growth hormone.

Leptin, the product of the adipose tissue-specific ob gene, is a newly recognized hormone involved in the regulation of metabolism and body composition. Leptin appears to provide information to the central nervous system on the amount of energy stored in the adipose tissue. Serum leptin levels are highly correlated with body fat mass in adults, children and newborns. Obese individuals have significantly higher circulating leptin than normal, lean subjects. In addition, females have higher serum leptin than males with equivalent fat mass. Although leptin correlates with fat mass, circulating co

Metabolism and disposition of difluoromethane (HFC32) in the mouse.

1. Difluoromethane (HFC32) is under development as a replacement for chlorofluorocarbons (CFCs) in some refrigeration applications. 2. The metabolism and disposition of [14C]-difluoromethane ([14C]-HFC32) was determined in male Swiss mice as a consequence of a single 6 h inhalation exposure to atmospheres of 10 000 p.p.m. 3. Of the inhaled dose, about 1-2% was recovered in expired air, urine, faeces and carcass suggesting that systemic absorption of this hydrofluorocarbon from the alveolar air space of the lung into blood is poor. Upon cessation of exposure the majority of the systemically abs

Ethanol metabolism: The good, the bad, and the ugly.

Throughout the world, ethanol is both an important commercial commodity and a source of major medical and social problems. Ethanol readily passes through biological membranes and distributes throughout the body. It is oxidized, first to acetaldehyde and then to acetate, and finally by the citric acid cycle in virtually all tissues. The oxidation of ethanol is irreversible and unregulated, making the rate dependent only on local concentration and enzyme activity. This unregulated input of reducing equivalents increases reduction of both cytoplasmic and intramitochondrial NAD and, through the la

Effect of surface polishing on roughness, biofilm formation, and biocompatibility of LCD-printed denture base polymer.

This study investigated the effect of surface polishing on roughness, mechanical performance, and biological properties of a liquid crystal display (LCD)‑printed denture base polymer. Disc‑shaped samples of three-dimensional (3D) printing polymer were fabricated and allocated to polished (P) or not polished (NP) groups for comparative analysis. Polishing was performed manually in a standardized manner using sequential silicon carbide sandpapers, while the NP samples were sanded to reproduce the texture generated by the printing lines. Surface roughness was measured by contact and non‑contact p

Mechanistic link of obesity to depression and cognitive decline in Alzheimer's disease.

The global rise in obesity and metabolic dysfunction has heightened concern about their links to depression, cognitive impairment, and Alzheimer's disease (AD)-related pathology. High-fat diet (HFD) exposure, excess adiposity, and metabolic dysfunction may each contribute to these outcomes, but they represent biologically distinct conditions that should not be treated as interchangeable. Epidemiological evidence suggests links between these conditions, yet the molecular pathways connecting them remain poorly characterized. This systematic review synthesizes current evidence on how HFD exposure

Contribution of liver alcohol dehydrogenase to metabolism of alcohols in rats.

The kinetics of oxidation of various alcohols by purified rat liver alcohol dehydrogenase (ADH) were compared with the kinetics of elimination of the alcohols in rats in order to investigate the roles of ADH and other factors that contribute to the rates of metabolism of alcohols. Primary alcohols (ethanol, 1-propanol, 1-butanol, 2-methyl-1-propanol, 3-methyl-1-butanol) and diols (1,3-propanediol, 1,3-butanediol, 1,4-butanediol, 1,5-pentanediol) were eliminated in rats with zero-order kinetics at doses of 5-20 mmol/kg. Ethanol was eliminated most rapidly, at 7.9 mmol/kgh. Secondary alcohols (2

Exogenous 20-Hydroxyecdysone (20E) rescues chlorantraniliprole-induced developmental impairment in Bombyx mori based on Ca2+ homeostasis.

Chlorantraniliprole (CAP), a widely used diamide insecticide, can exert effects on insect development and metabolic homeostasis through low-concentration environmental residues. However, it remains unclear whether exogenous 20-hydroxyecdysone (20E) can rescue CAP-induced developmental impairment. In this study, we systematically investigated CAP-induced developmental delay and its rescue by exogenous 20E in fifth-instar Bombyx mori larvae. The results showed that CAP exposure (0.01 mg/L) significantly prolonged the duration of the fifth instar and reduced whole-cocoon weight. Supplementation w

Direct comparison of UDP-glucuronosyltransferase and cytochrome P450 activities in human liver microsomes, plated and suspended primary human hepatocytes from five liver donors.

UDP-glucuronosyltransferases (UGTs) and cytochrome P450s (CYPs) are the major enzymes involved in hepatic metabolism of drugs. Hepatic drug metabolism is commonly investigated using human liver microsomes (HLM) or primary human hepatocytes (PHH). We describe the development of a sensitive assay to phenotype activities of six major hepatic UGT isoforms (UGT1A1, UGT1A3, UGT1A4, UGT1A6, UGT1A9 and UGT2B7) in intact PHH by analysis of glucuronidation of selective probe substrates. The non-selective, general substrate 7-hydroxycoumarin was included for comparison. For each liver donor preparation (

Pharmacokinetics of Tecarfarin and Warfarin in Patients with Severe Chronic Kidney Disease.

Chronic kidney disease (CKD) complicates warfarin anticoagulation partially through its effect on CYP2C9 activity. Tecarfarin, a novel vitamin K antagonist, is not metabolized by CYP2C9. To evaluate the effect of CKD on their metabolism, we measured PK parameters of warfarin and tecarfarin in subjects with and without CKD. CKD subjects with estimated glomerular filtration rate < 30 mL/min not on dialysis (n = 13) were matched to healthy volunteers (HVs) (n = 10). Each subject was randomized to either warfarin 10 mg or tecarfarin 30 mg and was later crossed over to the other drug. PK parameters

Nutrient digestibility, nitrogen balance, and energy partitioning in laying hens subjected to different molt induction programs.

Induced molting is a widely used strategy to extend the productive lifespan of laying hens; however, limited information is available regarding the effects of different molting programs on nutrient utilization and energy metabolism. The objective of this study was to compare nutrient digestibility, nitrogen balance, and energy partitioning among nutritionally distinct molt induction programs (MIP), which differed in ingredient composition and metabolizable energy availability, during molt induction and early post-molt recovery. A total of 75 Rhode Island Red laying hens were assigned to 1 of 5
