New Therapies for Advanced Thyroid Cancer.

Thyroid cancer is the most common endocrine cancer. The discovery of new biomarkers for thyroid cancer has significantly improved the understanding of the molecular pathogenesis of thyroid cancer, thus allowing more personalized treatments for patients with thyroid cancer. Most of the recently discovered targeted therapies inhibit the known oncogenic mechanisms in thyroid cancer initiation and progression such as MAPK pathway, PI3K/Akt-mTOR pathways, or VEGF. Despite the significant advances in molecular testing and the discoveries of new and promising therapeutics, effective treatments for ad

LncRNAs in cardiac hypertrophy: From basic science to clinical application.

Cardiac hypertrophy is a typical pathological phenotype of cardiomyopathy and a result from pathological remodelling of cardiomyocytes in humans. At present, emerging evidence demonstrated the roles of long non-coding RNAs (lncRNAs) in regulating the pathophysiological process of cardiac hypertrophy. Herein, we would like to review the recent researches on this issue and try to analysis the potential therapeutic targets on lncRNA sites. Studies have revealed both genetic mutations related hypertrophic cardiomyopathy and the compensative cardiac hypertrophy due to pressure overload, inflammatio

Dystroglycanopathy: From Elucidation of Molecular and Pathological Mechanisms to Development of Treatment Methods.

Dystroglycanopathy is a collective term referring to muscular dystrophies with abnormal glycosylation of dystroglycan. At least 18 causative genes of dystroglycanopathy have been identified, and its clinical symptoms are diverse, ranging from severe congenital to adult-onset limb-girdle types. Moreover, some cases are associated with symptoms involving the central nervous system. In the 2010s, the structure of sugar chains involved in the onset of dystroglycanopathy and the functions of its causative gene products began to be identified as if they were filling the missing pieces of a jigsaw pu

Current therapy and the evolving molecular landscape of paediatric ependymoma.

Ependymomas are the third commonest paediatric central nervous system (CNS) tumour, accounting for 6-12% of brain tumours in children. The management of these tumours has seen considerable changes over the last two decades, leading to a significant improvement in outcomes. However, despite advances in neurosurgical, neuroimaging and postoperative adjuvant therapy, management of these tumours remain challenging, and recurrence occurs in over 50% of cases, particularly when complete resection is not achieved prior to conformal radiotherapy. To-date no chemotherapeutic regimen has proven to be of

Neuron-tumor interplay in colorectal cancer: from mechanisms of onset and progression to targeted therapies.

Colorectal cancer (CRC) is a major cause of cancer-related mortality worldwide, traditionally attributed to genetic mutations and epigenetic alterations. However, with the emergence of cancer neuroscience, a paradigm shift has occurred, revealing the central role of the nervous system in the initiation, progression, and metastasis of CRC. The gastrointestinal tract, being the only organ in the body with an independent peripheral nervous system - the enteric nervous system (ENS) - provides a unique anatomical and functional basis for neuro-tumor interactions. This review systematically explores

Notch signaling in the tumor microenvironment: recent advances and targeted therapeutics.

The Notch signaling pathway, an evolutionarily conserved mechanism governing cell-cell communication, plays a pivotal and multifaceted role in shaping the tumor microenvironment (TME). This review provides a comprehensive overview of the core components and signaling mechanisms of the Notch pathway, encompassing both canonical and non-canonical signaling cascades. It further examines the dual functionality of Notch in tumorigenesis, functioning as either an oncogene or a tumor suppressor. A central focus of this review is the detailed investigation of the molecular mechanisms through which Not

Precision oncology in gastric cancer: navigating molecular subtypes, therapeutic targets, and future horizons.

Gastric cancer (GC) remains a leading cause of cancer-related mortality globally and is characterized by significant inter- and intra-tumoral heterogeneity, which poses major challenges to effective treatment. Although traditional "one-size-fits-all" chemotherapy regimens have improved outcomes, the prognosis for advanced disease remains poor, necessitating a paradigm shift towards personalized medicine. This review provides a comprehensive synthesis of the current landscape of precision oncology in GC. We systematically analyze the clinical implications of major molecular classification syste

Mitochondrial dynamics and cancer: mechanisms and targeted therapy.

Mitochondria, as key organelles in eukaryotic cells, regulate cellular energy homeostasis and functional integrity through dynamic processes, including fission, fusion, autophagy, and transport, underpinned by precise molecular mechanisms. In healthy cells, the equilibrium of mitochondrial dynamics ensures network quality control and metabolic coordination. In contrast, cancer cells exhibit multi-dimensional dysregulation characterized by hyperactive fission, impaired fusion, and compensatory upregulation of respiratory chain assembly factors, creating a state of "morphology-function decouplin

SnapShot: The hallmarks of cancer treatment.

Cancer therapy evolved from cytotoxic chemotherapy to targeted and immune treatments guided by molecular profiling. Early cytotoxic drugs, rooted in mustard-gas observations, remain effective but toxic. Targeted agents exploit oncogenic vulnerabilities yet commonly select resistant clones. Checkpoint inhibitors and engineered modalities (ADCs, BiTEs, CAR-T) yield responses but are limited by toxicity and adaptive antigen loss. Future treatment combinations will require targeting the evolution of resistance. To view this SnapShot, open or download the PDF.

Role of HMGB1 in tumors and its targeted therapy.

High-mobility group box 1 (HMGB1), a protein with context-dependent pro- and anti-tumor functions, is expressed in tumor cells, tumor-associated macrophages, tumor-associated neutrophils, cancer-associated fibroblasts, and T cells within the tumor microenvironment. The identity of HMGB1 has undergone a transformation from a nuclear architectural protein to a secreted damage-associated molecular pattern with pivotal roles in inflammation and cancer. The heterogeneity of HMGB1 stems from the diversity of post-translational modifications, proteolytic cleavage and lysis, partial independence of do

Cancer Drug Development in Never-Smoker Lung Cancer: Targeted and Immune-Based Therapeutic Strategies.

Lung cancer in individuals who have never smoked (LCINS) represents a clinically and biologically distinct subset of non-small cell lung cancer, driven predominantly by oncogenic alterations rather than tobacco-related mutagenesis. This review aims to summarize current and emerging targeted and immune-based therapeutic strategies in LCINS individuals. These patients present a molecular profile that differs substantially from tobacco-associated disease and has direct consequences for treatment selection. Evidence published over the past five years has clarified how these molecular features shap

Targeting claudins in gastric cancer: A novel GLOWing strategy in the SPOTLIGHT.

Gastric cancer is one of the most aggressive and lethal malignancies, with limited reliable biomarkers to inform prognosis and therapeutic decision-making. Claudins, which are tight junction proteins that establish epithelial polarity, have recently emerged as clinically actionable targets. CLDN18.2, typically sequestered within the tight junctions of gastric epithelial cells, becomes accessible during malignant transformation and is currently regarded as a critical standard marker in patients with metastatic gastric or gastroesophageal junction adenocarcinoma. Global research trends and hotspots in targeted therapy for IgA nephropathy: a bibliometric and visualization analysis (1999-2025).

Targeted therapy has emerged as a promising precision medicine strategy for immunoglobulin A nephropathy (IgAN) through the modulation of specific pathogenic pathways. Although research in this area has accelerated, the literature remains scattered, and no bibliometric study has mapped its global knowledge structure or evolving hotspots. We conducted a bibliometric and visualization analysis of 678 publications indexed in the Web of Science Core Collection (1999-2025). CiteSpace 6.4R1, VOSviewer 1.6.20, and the R‑based bibliometric package were used to assess publication and citation trends; i

Toward targeted therapeutics for lobular breast cancer.

Despite growing recognition of invasive lobular carcinoma (ILC) as a biologically and clinically distinct subtype of breast cancer, ILC remains understudied. Most contemporary therapeutic trials continue to enroll patients predominantly with invasive ductal carcinoma/invasive carcinoma of no special type and rarely stratify by histology. As a result, ILC's unique disease biology, characteristic loss of E-cadherin function, diffuse growth pattern, and distinct metastatic tropism remain underrepresented in evidence that guides systemic therapy recommendations. In this Review, we examine key mole

Evolving Therapeutic Strategies in Neuroendocrine Neoplasms: A New Era of Personalized Therapies.

Neuroendocrine neoplasms (NENs) represent a heterogeneous group of malignancies ranging from relatively indolent well-differentiated neuroendocrine tumors (NETs) to more aggressive poorly differentiated neuroendocrine carcinomas (NECs). The advent of somatostatin receptor (SSTR)-targeting therapies, particularly radioligand therapy (RLT), has transformed the therapeutic landscape of NETs. Ongoing efforts are aimed at maximizing the therapeutic benefit of RLT and include retreatment and combination strategies as well as novel α-emitter-based radiopharmaceuticals. The increasing use of molecular

Treatments for human epidermal growth factor receptor 2 (HER2)-altered cancers evaluated using a tumor‑agnostic approach: A narrative review.

Human epidermal growth factor receptor 2 (HER2) is a critical driver of tumorigenesis across a wide range of cancers, positioning it as a key target for tumor-agnostic therapies. Advances in HER2 therapies have shifted the treatment paradigm from histology-driven toward biomarker-driven, pan-tumor approaches. This review discusses the outcomes of approved and emerging HER2 therapies, highlighting the variability in efficacy across tumor types due to a combination of factors, such as HER2 heterogeneity, co-alterations, and tumor-specific resistance mechanisms. Although tumor-agnostic basket tri
