A previous laboratory study using Haemaphysalis longicornis Neumann (Acari: Ixodidae) ticks of North American origin showed that larvae could acquire the Lyme disease spirochete, Borrelia burgdorferi sensu stricto (s.s.) (Spirochaetales: Spirochaetaceae) while feeding to completion on infected mice. However, the infection was lost during the molt to the nymphal stage. Nonetheless, questing H. longicornis nymphs and adults collected by drag sampling in the northeastern United States have been reported infected with B. burgdorferi s.s. DNA; occasionally these ticks appeared to be partially engor

Aspergillosis is a major cause of morbidity and mortality in penguins, with triazole antifungal drugs being commonly used for prophylaxis and treatment. This report describes 15 cases of fatal hemolysis associated with liquid itraconazole and voriconazole formulations administered to African penguins (Spheniscus demersus) from four institutions. All penguins underwent stressful events (e.g. relocation, induced molt) and were administered commercial liquid itraconazole formulations or compounded voriconazole liquid suspension. Observed clinical signs in affected penguins prior to death included

ETHNOPHARMACOLOGICAL RELEVANCE: Periostracum Cicadae (PC), the molted exoskeleton of the cicada Cryptotympana pustulata Fabricius, is frequently employed in Chinese herbal medicine. Based on traditional therapies and pharmacological studies, PC appears to have immunomodulatory activity. However, the specific impact of PC on immunomodulation, particularly its effect on dendritic cells (DCs), remains unknown. DCs act professionally as antigen-presenting cells that trigger adaptive immune responses, making them critical for immunomodulation.
MATERIALS AND METHODS: The DCs derived from mouse bone

Lyme disease is the most common vector-borne disease in the United States impacting the Northeast and Midwest at the highest rates. Recently, it has become established in southeastern and south-central regions of Canada. In these regions, Lyme disease is caused by Borrelia burgdorferi, which is transmitted to humans by an infected Ixodes scapularis tick. Understanding the parasite-host interaction is critical as the white-footed mouse is one of the most competent reservoir for B. burgdorferi. The cycle of infection is driven by tick larvae feeding on infected mice that molt into infected nymph

To explore the molecular mechanisms of action underlying the antileukemia activities of darinaparsin, an organic arsenical approved for the treatment of peripheral T-cell lymphoma in Japan, cytotoxicity of darinaparsin was evaluated in leukemia cell lines NB4, U-937, MOLT-4 and HL-60. Darinaparsin was a more potent cytotoxic than sodium arsenite, and induced apoptosis/necrosis in NB4 and HL-60 cells. In NB4 cells exhibiting the highest susceptibility to darinaparsin, apoptosis induction was accompanied by the activation of caspase-8/-9/-3, a substantial decrease in Bid expression, and was supp

BACKGROUND: The behaviors and ontogeny of Aedes aegypti are closely related to the spread of diseases caused by dengue (DENV), chikungunya (CHIKV), Zika (ZIKV), and yellow fever (YFV) viruses. During the life cycle, Ae. aegypti undergoes drastic morphological, metabolic, and functional changes triggered by gene regulation and other molecular mechanisms. Some essential regulatory factors that regulate insect ontogeny have been revealed in other species, but their roles are still poorly investigated in the mosquito.
RESULTS: Our study identified 6 gene modules and their intramodular hub genes t

The morphology of juveniles of Mithraculus forceps A. Milne-Edwards, 1875, raised in the laboratory, is described. The larvae were obtained from ovigerous females collected in the wild, maintained in the rearing system for culture of crabs (400 L). A total of 229 larvae (zoea I), from 7 females were distributed in lots of 20 larvae kept in glass flasks (500 ml), with seawater circulation and temperature and salinity constants, which were checked daily for molts or deaths. After the larvae reached the juvenile stage, exuviae and dead specimens were deposited in a scientific collection. We analy

Leukemia is a group of cancer caused by the abnormal proliferation and differentiation of hematopoietic stem cells. Efforts geared toward effective therapeutic strategies with minimal side effects are underway. Peptides derived from natural resources have recently gained special attention as alternative chemotherapeutic agents due to their minimal adverse effects. In the present study, the aim was to isolate peptides from garlic (Allium sativum) and investigate their anticancer activity against leukemic cell lines. The protein extract of A. sativum was pepsin-digested to obtain protein hydroly

Marine sponges have been recognized as a rich source of potential anti-proliferative metabolites. Currently, there are two sponge-derived anti-cancer agents (a macrolide and a nucleoside) isolated from the Porifera phylum, suggesting the great potential of this sponge as a rich source for anti-neoplastic agents. To search for more bioactive metabolites from this phylum, we examined the EtOAc extract of Theonella sp. sponge. We isolated seven compounds (1-7), including four 4-methylene sterols (1-4), two nucleosides (5 and 6), and one macrolide (7). Among them, theonellasterol L (1) was identif

The blacklegged tick (Ixodes scapularis Say) is the primary vector of Borrelia burgdorferi sensu stricto (Spirochaetales: Spirochaetaceae), the Lyme disease agent in North America. The basic reproduction number (R0) for B. burgdorferi in I. scapularis in the Northeast is highly sensitive to the probability that engorged larvae survive the winter, molt into nymphs, and find a host. These processes are dependent on local environmental variables, including climate, host population size and movement, and tick behavior. A simple model is presented for estimating host-finding success from the ratio

The reproductive system of Mothocya renardi (Bleeker, 1857), a protandrous hermaphroditic cymothoid that infects the belonid fish Strongylura leiura Bleeker, 1850, is characterized using light and electron microscopy. Three protandrous hermaphroditic adult phases are identified: male, transitional and female. Each phase includes a paired reproductive system, one on either side of the gut. Each consists of three lobed testes, followed by an ovary, then a vas deferens that opens into a penis on the same side. During the male phase, all testis lobes are filled with germ cells at various stages of

This study reports the third collection of Triatoma nitida Usinger in Mexico, with a brief description of the collection area and an investigation of parameters related to its vectorial capacity. Whether a triatomine (Hemiptera: Reduviidae) species is a primary or secondary vector is determined by factors that include vectorial capacity, anthropophilic habits, geographic distribution, and capacity to invade and colonize human dwellings. However, when the primary vectors are removed, secondary vectors, such as T. nitida, can become important transmitters of Trypanosoma cruzi Chagas to humans. T

Fluid clearance from the respiratory system during developmental transitions is critically important for achieving optimal gas exchange in animals. During insect development from embryo to adult, airway clearance occurs episodically each time the molt is completed by performance of the ecdysis sequence, coordinated by a peptide-signaling cascade initiated by ecdysis-triggering hormone (ETH). We find that the neuropeptide Kinin (also known as Drosokinin or Leukokinin) is required for normal respiratory fluid clearance or "tracheal air-filling" in Drosophila larvae. Disruption of Kinin signaling

We tested whether emphysema progression accompanies enhanced tissue loss in other body compartments in 1817 patients from the ECLIPSE chronic obstructive pulmonary disease (COPD) cohort.Clinical and selected systemic biomarker measurements were compared in subjects grouped by quantitative tomography scan emphysema quartiles using the percentage of low attenuation area (LAA%). Lowest and highest quartile patients had amino-acid metabolomic profiles. We related LAA% to 3 years decline in lung function (forced expiratory volume in 1 s (FEV1)), body mass index (BMI), fat-free mass index (FFMI) and

The pine moth Dendrolimus pini effectively resists many insecticides, but it can be controlled by the use of bioinsecticides such as entomopathogenic fungi. In the use of microbial agents for the biocontrol of D. pini, it is important to identify the cuticular lipids of this pest if we are to understand the factors responsible for the preferential adhesion or selective repulsion of entomopathogenic fungi that are potentially useful in biocontrol. In this work the qualitative and quantitative analyses of free fatty acids in two exuviae extracts (petroleum ether and dichloromethane) and two deve

LIN-42, the Caenorhabditis elegans homolog of the Period (Per) family of circadian rhythm proteins, functions as a member of the heterochronic pathway, regulating temporal cell identities. We demonstrate that lin-42 acts broadly, timing developmental events in the gonad, vulva, and sex myoblasts, in addition to its well-established role in timing terminal differentiation of the hypodermis. In the vulva, sex myoblasts, and hypodermis, lin-42 activity prevents stage-specific cell division patterns from occurring too early. This general function of timing stage-appropriate cell division patterns

The deficiency of methylthioadenosine phosphorylase (MTAP) in T-cell acute lymphoblastic leukemia (T-ALL) and other cancers, while constitutively expressed in normal cells, allows for selective therapy using L-alanosine, an inhibitor of de novo AMP synthesis. We demonstrate that MTAP- T-ALL cells obtained at relapse are as sensitive to L-alanosine toxicity as diagnosis samples. The therapeutic index of L-alanosine can be increased by the use of a MTAP substrate, which protects MTAP+ normal cells. Since MTAP substrates MTA and 5'deoxyadenosine are prone to toxicities associated with adenosine,

OBJECTIVE: Natural killer (NK) cell-mediated cytotoxicity contributes to the innate immune response against numerous malignancies, including leukemias. Acute lymphoblastic leukemias (ALL) often display a high degree of resistance, the mechanisms of which have not been elucidated.
METHODS: We used the well-characterized NK cell line NK-92 as a model to investigate whether mechanisms commonly implicated in tumor escape from NK cell killing are relevant for ALL.
RESULTS: We demonstrate selective resistance of B-precursor ALL to NK-92 cytotoxicity even in the absence of inhibitory killer cell im

We examined the functional role of the phosphatidylinositol 3'-kinase pathway in the growth and survival of cell lines of T-cell origin. Pharmacological inhibition of PI3'-kinase using LY294002 resulted in apoptosis of acute lymphoblastic T-cell leukemia (T-ALL) cell lines including CEM, Jurkat, and MOLT-4. On the other hand, the cutaneous T-cell lymphoma cell line HUT-78 was found to be refractory to LY294002- inducible apoptosis. Sensitivity or resistance to pharmacological inhibitors of PI3'-kinase correlated with tumor suppressor PTEN gene expression, as sensitive T-ALL cells do not expres

Infection with Anaplasma phagocytophilum in white-footed mice results in partial protection against reinfection with the same agent. However, humans and domestic animals may be sequentially exposed to different isolates of the agent circulating in the same or adjacent foci. We investigated whether immune response to a tick-borne infection with A. phagocytophilum provides protection against homologous and heterologous challenges. BALB/c mice were infected with one of the two sympatric isolates of A. phagocytophilum via tick bite and challenged 16 weeks later by Ixodes scapularis nymphs infected
