Postnatal deletion of Wnt7a inhibits uterine gland morphogenesis and compromises adult fertility in mice.

The success of postnatal uterine morphogenesis dictates, in part, the embryotrophic potential and functional capacity of the adult uterus. The definitive role of Wnt7a in postnatal uterine development and adult function requires a conditional knockout, because global deletion disrupts müllerian duct patterning, specification, and cell fate in the fetus. The Wnt7a-null uterus appears to be posteriorized because of developmental defects in the embryo, as evidenced by the stratified luminal epithelium that is normally found in the vagina and the presence of short and uncoiled oviducts. To underst…

Identification of a Tbx1/Tbx2/Tbx3 genetic pathway governing pharyngeal and arterial pole morphogenesis.

The 22q11.2 deletion syndrome (22q11.2DS) is the most common microdeletion disorder and is characterized by abnormal development of the pharyngeal apparatus and heart. Cardiovascular malformations (CVMs) affecting the outflow tract (OFT) are frequently observed in 22q11.2DS and are among the most commonly occurring heart defects. The gene encoding T-box transcription factor 1 (Tbx1) has been identified as a major candidate for 22q11.2DS. However, CVMs are generally considered to have a multigenic basis and single-gene mutations underlying these malformations are rare. The T-box family members

C. elegans CAND-1 regulates cullin neddylation, cell proliferation and morphogenesis in specific tissues.

Cullin-RING ubiquitin ligases (CRLs) are critical regulators of multiple developmental and cellular processes in eukaryotes. CAND1 is a biochemical inhibitor of CRLs, yet has been shown to promote CRL activity in plant and mammalian cells. Here we analyze CAND1 function in the context of a developing metazoan organism. Caenorhabditis elegans CAND-1 is capable of binding to all of the cullins, and we show that it physically interacts with CUL-2 and CUL-4 in vivo. The covalent attachment of the ubiquitin-like protein Nedd8 is required for cullin activity in animals and plants. In cand-1 mutants,

Discordant influence of Blastomyces dermatitidis yeast-phase-specific gene BYS1 on morphogenesis and virulence.

Blastomyces dermatitidis is a thermally induced dimorphic fungus capable of causing lung and systemic infections in immunocompetent animal hosts. With the publication of genomic sequences from three different strains of B. dermatitidis and the development of RNA interference as a gene-silencing tool, it has become possible to easily ascertain the virulence and morphological effects of knocking down the expression of candidate genes of interest. BYS1 (Blastomyces yeast-phase-specific 1), first identified by Burg and Smith, is expressed at high levels in yeast cells and is undetectable in mold.

MT2-MMP-dependent release of collagen IV NC1 domains regulates submandibular gland branching morphogenesis.

Proteolysis is essential during branching morphogenesis, but the roles of MT-MMPs and their proteolytic products are not clearly understood. Here, we discover that decreasing MT-MMP activity during submandibular gland branching morphogenesis decreases proliferation and increases collagen IV and MT-MMP expression. Specifically, reducing epithelial MT2-MMP profoundly decreases proliferation and morphogenesis, increases Col4a2 and intracellular accumulation of collagen IV, and decreases the proteolytic release of collagen IV NC1 domains. Importantly, we demonstrate the presence of collagen IV NC1…

Salivary gland branching morphogenesis: a quantitative systems analysis of the Eda/Edar/NFkappaB paradigm.

BACKGROUND: Ectodysplasin-A appears to be a critical component of branching morphogenesis. Mutations in mouse Eda or human EDA are associated with absent or hypoplastic sweat glands, sebaceous glands, lacrimal glands, salivary glands (SMGs), mammary glands and/or nipples, and mucous glands of the bronchial, esophageal and colonic mucosa. In this study, we utilized EdaTa (Tabby) mutant mice to investigate how a marked reduction in functional Eda propagates with time through a defined genetic subcircuit and to test the proposition that canonical NFkappaB signaling is sufficient to account for th…

Protein phosphatase PP1 gamma 2 in sperm morphogenesis and epididymal initiation of sperm motility.

The serine/threonine phosphatase (PP1) isoform PP1 gamma 2, predominantly expressed in the testis, is a key enzyme in spermatozoa. High PP1 gamma 2 catalytic activity holds motility in check in immature spermatozoa. Inhibition of PP1 gamma 2 causes motility initiation in immature spermatozoa and motility stimulation and changes in flagellar beat parameters in mature spermatozoa. The PP1 gamma 2 isoform is present in all mammalian spermatozoa studied: mouse, rat, hamster, bovine, non-human primate and man. We have now identified at least four of its regulatory proteins that regulate distinct po

Angiotensin II type 1 receptor-EGF receptor cross-talk regulates ureteric bud branching morphogenesis.

Angiotensinogen-, angiotensin-converting enzyme-, and angiotensin II (Ang II) type 1 receptor (AT(1)R)-deficient mice exhibit a dilated rena … inase inhibitor. These data demonstrate that Ang II, acting via the AT(1)R, stimulates UB branching morphogenesis. This process depends on tyrosine phosphorylation of the EGFR. Cooperation of AT(1)R and EGFR signaling therefore is important in the development of the renal collecting system.

FGF10/FGFR2b signaling plays essential roles during in vivo embryonic submandibular salivary gland morphogenesis.

BACKGROUND: Analyses of Fgf10 and Fgfr2b mutant mice, as well as human studies, suggest that FGF10/FGFR2b signaling may play an essential, nonredundant role during embryonic SMG development. To address this question, we have analyzed the SMG phenotype in Fgf10 and Fgfr2b heterozygous and null mutant mice. In addition, although previous studies suggest that the FGF10/FGFR2b and FGF8/FGFR2c signaling pathways are functionally interrelated, little is known about the functional relationship between these two pathways during SMG development. We have designed in vivo and in vitro experiments to addr

Morphogenesis of adaptation and compensatory reactions in mouse adrenals during restitution after thermal exposure.

Single hyperthermia session causes an appreciable decrease in adrenocorticocyte count in the adrenal cortex in mice on day 3 of postthermal restitution. Morphological changes in the adrenals under conditions of developing stress reaction manifested in increased structural and functional heterogeneity of adrenocorticocytes in all cortical layers, exhaustion of lipid incorporations in the zona fasciculata cells, pronounced shrinkage of zona reticularis, developing against the background of hemodynamic disorders. Recovery of the total count of adrenocorticocyte population was associated with the

Compartment-selective sensitivity of cardiovascular morphogenesis to combinations of retinoic acid receptor gene mutations.

Several aspects of normal cardiovascular development require signaling by the vitamin A metabolite retinoic acid. We have previously established germ-line mutations in mice in the genes that encode the RAR alpha 1, RAR beta, and RXR alpha retinoic acid receptors as a means of studying the function of these receptors in vivo. Although mutation of RXR alpha results in fetal ventricular defects, the RAR alpha 1 and RAR beta mutations are apparently nonphenotypic in the heart and elsewhere. In this study, we have established and analyzed combinations of these receptor gene mutations. Malformations

Morphogenesis of yellow fever virus in Aedes aegypti cultured cells. II. An ultrastructural study.

The growth and intracytoplasmic development of two yellow fever virus strains (wild and French neurotropic) were studied in Aedes aegypti cells (clone C 17). Despite a longer period of latency for the vaccine virus, infected cells appeared similar. The cisternae of the rough endoplasmic reticulum (RER) were swollen and formed vesicles which contained the virus. This RER appeared to be the predominant locus of viral synthesis and maturation. Cytopathic effect appeared when the cells were filled with vesicles, and it was characterized by cell degeneration and lysis.

A predictive model for coupling cell division orientation to tissue mechanics during epithelial morphogenesis.

Stratified epithelial tissues such as the skin epidermis maintain barrier integrity during development and homeostasis through the coordinated action of cell proliferation, differentiation, delamination, and tissue-scale mechanical forces. During development, the orientation of cell division within the basal layer plays a pivotal role in tissue stratification; however, the mechanical principles linking the orientation of the division plane to these processes across developmental stages remain poorly understood. Here, we expand a recently developed three-dimensional vertex model for stratified

Counterion-Controlled Nanoscale Morphogenesis in Interfacial Polymerization via Decoupled Interfacial Transport and Reaction Kinetics.

The morphology of thin-film composite membranes is encoded during interfacial polymerization (IP) by competing monomer transport and reaction kinetics, yet molecular control remains limited due to their intrinsic coupling. Herein, we show that counterions decouple solvation-controlled transport from transition-state energetics, enabling deterministic control over nonequilibrium thin-film morphogenesis. Using a model polyester IP system, we demonstrate that varying phenoxide counterions (Li+, Na+, K+, TBA+) reveals anomalous energetic trends, with transition-state energies (ΔG‡ ≈ 6.9-21.7 kcal …

Hac1-independent functions of Ire1 drive morphogenesis in Candida albicans through distinct transcriptional programs.

The pathogenic yeast Candida albicans relies on morphogenesis - the transition from spherical yeast to filamentous hyphal forms - for infection. Although morphogenesis requires Ire1, a transmembrane protein that canonically initiates the unfolded protein response (UPR) through HAC1 mRNA splicing, the specific mechanisms linking Ire1 to filamentation remain unclear. Using transcriptome analysis, we found that the Ire1-dependent transcriptional response driving morphogenesis was fundamentally distinct from the canonical UPR response to proteotoxic stress, with minimal overlap between programs. M…
