Deoxynivalenol exposure-related male reproductive toxicity in mammals: Molecular mechanisms, detoxification and future directions.

An increasing body of evidence indicates that exposure to widespread, environmental and food contaminants such as mycotoxins may cause endocrine disorders and infertility. Deoxynivalenol (DON), which is a toxic secondary metabolite produced by Fusarium fungi, can lead to multiple harmful effects in humans and animals, such as hepatotoxicity, nephrotoxicity, immunotoxicity, gastrointestinal toxicity, neurotoxicity, genetic toxicity and carcinogenicity. Recently, there has been growing concern about DON-induced male infertility. Exposure to DON and its metabolites can damage the structure and fu…

Oat Leaf Diseases: Secondary Metabolites of Pathogenic Fungi and Infected Plants and Their Toxicity.

Oats (Avena sativa), a globally vital cereal and forage crop, incur heavy losses from foliar fungal pathogens, whose virulence is largely mediated by secondary metabolites. These compounds are toxic to the host plant, normal cells, and livestock. This review summarizes the current knowledge on 31 pathogenic fungi and their 310 secondary metabolites, which are classified into 9 chemical classes: lactones, benzene derivatives, terpenoids, nitrogen-containing compounds, peptides, flavonoids, pyranones, steroids, and organic acids. Of these 310 secondary metabolites, 68 compounds display phytotoxi

Toxicity induced by orellanine from the mushroom Cortinarius orellanus in primary renal tubular proximal epithelial cells (RPTEC): Novel mechanisms of action.

The toxicity of Orellanine (OR), a significant factor in mushroom poisoning, has severe effects on the kidneys, particularly the proximal tubules. This study investigated the acute toxicity of OR from the Cortinarius orellanus mushroom in human Primary Renal Tubular Proximal Epithelial Cells (RPTEC). Additionally, the half maximal inhibitory concentration (IC50) of OR in MCF-7 cells was established. RPTEC were subjected to a 6.25 μg/ml dose of orellanine for 24 h, while Control cells were exposed to 0.05% DMSO (vehicle). The RT2 Profiler™ PCR Array Human Nephrotoxicity was utilized to identify

Development and application of a quantitative fluorescence-based immunochromatographic assay for fumonisin b1 in maize.

A fluorescence-based immunochromatographic assay (ICA) for fumonisin B1 (FB1) that employs conjugates of fluorescent microspheres and monoclonal antibodies (FM-mAbs) as detection reporters is described. The ICA is based on the competitive reaction between FB1-bovine serum albumin (BSA; test line) and the target FB1 for binding to the FM-mAb conjugates. A limit of detection (LOD) for FB1 of 0.12 ng/mL was obtained, with an analytical working range of 0.25-2.0 ng/mL (corresponding to 250-2000 μg/kg in maize flour samples, according to the extraction procedure). The recoveries of the ICA to detec

Microbial diversity, functional activities, and safety risks in fermented tea: a comprehensive review.

Microbial fermented teas are gaining global popularity due to their unique sensory profiles and health benefits. The quality and safety of these products are governed by complex microbial ecosystems that orchestrate the biotransformation of tea leaf components. This review addresses a critical paradox in the field: the same microbial activities that generate desirable bioactive metabolites, such as theabrownins and organic acids, also create ecological niches for mycotoxigenic fungi, posing significant health risks from contaminants like ochratoxin A, citrinin, and aflatoxins. While extensive

Marine toxin C17-SAMT causes major structural damage to vital organs in mice following subchronic toxicity trials.

C17-sphinganine analog mycotoxin (C17-SAMT) has been characterized as the contaminant responsible for the atypical toxicity reported in mussels from the Bizerte lagoon (northern Tunisia) over the past decade. C17-SAMT exhibited common symptoms of toxicity in mice, including flaccid paralysis and severe respiratory distress, followed by rapid death. To determine the potential health risks of this neurotoxin, we assessed its subchronic toxicity according to the recommendations of OCDE n° 407. The body weight and the structural changes of vital organs were recorded. Biochemical and hematological

Natural toxins and their therapeutic potential.

Plants have been extensively investigated for exploring their therapeutic potentials, but there are comparatively scanty reports on drugs derived from animal kingdom, except for hormones. During last decade, the toxins that are used for defense by the animals, have been isolated and found useful tools for physiological and pharmacological studies, besides giving valuable leads to drug development. Toxins with interesting results have been isolated from the venoms of snakes, scorpions, spiders, snails, lizards, frogs and fish. The present review describe about some toxins as drugs and their bio

Killer toxin of Kluyveromyces phaffii DBVPG 6076 as a biopreservative agent to control apiculate wine Yeasts.

The use of Kluyveromyces phaffii DBVPG 6076 killer toxin against apiculate wine yeasts has been investigated. The killer toxin of K. phaffii DBVPG 6076 showed extensive anti-Hanseniaspora activity against strains isolated from grape samples. The proteinaceous killer toxin was found to be active in the pH range of 3 to 5 and at temperatures lower than 40 degrees C. These biochemical properties would allow the use of K. phaffii killer toxin in wine making. Fungicidal or fungistatic effects depend on the toxin concentration. Toxin concentrations present in the supernatant during optimal condition

The green colour effect (GCE) of the killer strain Cryptococcus laurentii CBS 139 on Staib agar.

Attention is drawn to the observation that the type strain Cryptococcus laurentii CBS 139, producing killer toxins (mycocins) directed at Cr. neoformans var. gattii, causes a green colour effect (GCE) on Staib agar (Guizotia abyssinica creatinine agar) in combination with an intense assimilation of creatinine. Five (9.6%) out of 52 strains of Cr. laurentii of various origin, showed a GCE and intense creatinine assimilation. Further research must show if all Cr. laurentii strains, characterized by a GCE similar to that of the strain CBS 139, are also capable of producing killer toxins. For furt

Granulocytopaenia and thrombocytopaenia in dairy cattle--a suspected mycotoxicosis.

During a 6-week period, 22 Dairy Shorthorn cows and heifers died with granulocytopaenia and thrombocytopaenia. Clinical signs observed in the affected animals included increased salivation, pyrexia, depression, rumenal stasis, bilateral epistaxis, melaena, increased bleeding after removal of retained foetal membranes and rapid weight loss. Despite intensive antibiotic and vitamin K therapy and blood transfusions, all affected animals died. The aetiological agent, thought to be a fungal toxin, could not be isolated from post mortem specimens or pasture samples.

Highly efficient electrochemiluminescent properties of porphyrin-based metal-organic framework Zn-TCPP and its immunoassay application to the detection of ochratoxin A.

BACKGROUND: Ochratoxin A (OTA) is a group of mycotoxins that are widely distributed in food and feed and are closely associated with human health, so it is particularly important to detect OTA in cereal-based foods. Porphyrins and their derivatives have been widely investigated for their excellent electrochemical luminescence properties. Tetrakis 4-carboxyphenyl) porphyrin (TCPP) has limited applications because of its tendency to aggregate in water. RESULTS: To enhance the luminescence efficiency of TCPP, the porphyrin can be immobilized as an organic ligand in a metal-organic framework. Thi…

Photosensitisation diseases of animals: Classification and a weight of evidence approach to primary causes.

Clare's (1952) classification system for photosensitisation diseases (types I, II, III and Uncertain) has endured many years of use despite … s; however, the weight of evidence for all but a few is sparse. With respect to plants (and certain mycotoxins and insects) implicated in primary photosensitisation outbreaks, McKenzie's "toxicity confidence rankings" (Australia's Poisonous Plants, Fungi and Cyanobacteria, 2012) has been adapted to "phototoxic agent confidence rankings". Thus, plants, mycoto …

Effects of curcumin on performance, antioxidation, intestinal barrier and mitochondrial function in ducks fed corn contaminated with ochratoxin A.

Curcumin has been attributed with antioxidant, anti-inflammatory, antibacterial activities, and has shown highly protective effects against enteropathogenic bacteria and mycotoxins. Ochratoxin A (OTA) is one of the major intestinal pathogenic mycotoxins. The possible effect of curcumin on the alleviation of enterotoxicity induced by OTA is unknown. The effects of dietary curcumin supplementation on OTA-induced oxidative stress, intestinal barrier and mitochondrial dysfunctions were examined in young ducks. A total of 540 mixed-sex 1-day-old White Pekin ducklings with initial BW (43.4±0.1 g) we…

PRESENCE OF CITRININ IN GRAINS AND ITS POSSIBLE HEALTH EFFECTS.

BACKGROUND: Citrinin is a mycotoxin produced by several species of the genera Aspergillus, Penicillium and Monascus and it occurs mainly in stored grain. Citrinin is generally formed after harvest and occurs mainly in stored grains, it also occurs in other plant products. Often, the co-occurrence with other mycotoxins is observed, especially ochratoxin A, which is usually associated with endemic nephropathy. At the European Union level, systematic monitoring of Citrinin in grains began with the aim of determining its highest permissible amount in food. Thus, far the systematic monitoring of th…
