Functional consequences of unilateral olfactory deprivation: time-course and age sensitivity.

Unilateral olfactory deprivation in the rat profoundly modifies olfactory bulb anatomy, chemistry and function. The present report examined the time-course of the functional effects of unilateral deprivation on inhibition in the olfactory bulb using paired-pulse stimulation of the lateral olfactory tract and olfactory nerve. In addition, an attempt was made to correlate these physiological measures with olfactory bulb dopamine and norepinephrine levels and tyrosine hydroxylase immunoreactivity. Deprivation from postnatal day 1 to postnatal day 20 or postnatal day 40 significantly enhanced late…

Anatomical evidence for scent guided foraging in the turkey vulture.

The turkey vulture (Cathartes aura) is a widespread, scavenging species in the Western Hemisphere that locates carrion by smell. Scent guided foraging is associated with an expansion of the olfactory bulbs of the brain in vertebrates, but no such neuroanatomical data exists for vultures. We provide the first measurements of turkey vulture brains, including the size of their olfactory bulbs and numbers of mitral cells, which provide the primary output of the olfactory bulbs. Comparative analyses show that the turkey vulture has olfactory bulbs that are 4× larger and contain twice as many mitral…

Misrouting of mitral cell progenitors in the Pax6/small eye rat telencephalon.

The olfactory bulb is a protruding structure formed at the rostral end of the telencephalon. Pax6-mutant mice and rats lack the olfactory bulb and, instead, develop an olfactory bulb-like structure at the lateral part of the telencephalon. Here, we report that ectopic formation of the olfactory bulb-like structure in these mutants is caused by the abnormal migration of mitral cell progenitors, which first differentiate within the olfactory bulb. Cell-tracing experiments in whole embryos in culture indicate that, in the mutants, the mitral cell progenitors that originate from the rostral part o…

Expression of hepatocyte growth factor/scatter factor, its receptor, c-met, and tissue-type plasminogen activator during development of the murine olfactory system.

The expression of hepatocyte growth factor/scatter factor (HGF/SF) and its receptor, the c-met proto-oncogene product, was examined by in situ hybridization in the developing and adult murine olfactory system and compared with the expression of a known activator of HGF/SF, tissue-type plasminogen activator (tPA). In the developing olfactory canal, expression of both c-met and tPA was observed in the olfactory neuroepithelium, whereas HGF/SF expression appeared to be confined to the mucosa adjacent to the neuroepithelium. During development of the olfactory bulb, HGF/SF and tPA were expressed w…

Expression of galectin-1 in the olfactory nerve pathway of rat.

The olfactory neuroepithelium is characterised by the mosaic distribution of primary olfactory neurons that express different odorant recept … ssed by olfactory ensheathing cells both in olfactory nerve and within the nerve fibre layer of the olfactory bulb of the embryonic and adult rat. In the adult rat, galectin-1 was preferentially expressed by olfactory ensheathing cells in the nerve fibre layer of the ventromedial and lateral surfaces of the olfactory bulb. Galectin-1 was also expressed by subset …

Cerebral Blood Flow Responses to Trigeminal Olfactory Stimulation and Their Nicotinic Cholinergic Regulation in Anesthetized Rats.

Information about the sense of smell is conveyed by the olfactory nerve, or by the trigeminal nerve when irritating odors. We recently reported that olfactory nerve stimulation increases regional cerebral blood flow (BF) in the olfactory bulb, and the activation of nicotinic receptors in the brain potentiates olfactory bulb BF responses in anesthetized rats. This study aimed to elucidate the effects of intranasal trigeminal nerve stimulation on regional cerebral BF compared to the olfactory nerves, as well as the impact of nicotinic receptor activation on these BF responses. Regional BF in the…

Distribution of neuronal intermediate filament proteins in the developing mouse olfactory system.

The distribution of neuronal intermediate filament proteins in the developing mouse olfactory bulb and olfactory epithelium was characterized by immunocytochemical approach. Antibodies against alpha-internexin, neurofilament triplet proteins (NFTPs; NF-L, NF-M, and NF-H) and peripherin were used to determine their expression at different developmental stages. Alpha-internexin and peripherin were first found to be co-localized in the olfactory neuroepithelium during early development. At the perinatal stage, expression patterns of alpha-internexin and peripherin are distinguishable by spatial a…

Subclasses of olfactory receptor cells and their segregated central projections demonstrated by a monoclonal antibody.

A library of monoclonal antibodies (MAbs) was generated against a homogenate of the rabbit olfactory bulb. One of them immunohistochemically distinguished a subgroup of olfactory nerves. Both in the olfactory bulb and the epithelium, this MAb labeled most olfactory receptor axons in the lateral but only a small fraction in the medial portion. These findings demonstrate a molecular heterogeneity among olfactory receptor cells and suggest a functional division between the lateral and the medial portions of the epithelium and the bulb.

Quantitative analysis of the olfactory pathway for drug delivery to the brain.

Following intranasal administration to rats, wheat germ agglutinin-horseradish peroxidase (WGA-HRP) concentrated in the olfactory nerve and glomerular layers of the olfactory bulb resulting in a mean olfactory bulb concentration of 140 nM. A negligible amount of label was detected in the olfactory bulb following intravenous administration of WGA-HRP or intranasal administration of unconjugated HRP. This is the first quantitative assessment of intraneuronal transport of a protein into the brain using the olfactory route.

Olfactory projections in the lepidosirenid lungfishes.

Olfactory nerve and olfactory bulb projections in lepidosirenid lungfishes were experimentally determined with neural tracers. Unilateral injections of DiI into the olfactory nerve labeled the accessory and main olfactory bulbs as well as fibers of the anterior root of the terminal nerve, which terminates extensively in cell groups of the medial hemispheric wall, the dorsal and lateral pallia, and the preoptic nuclei and posterior tubercle. Lepidosirenid lungfishes do not exhibit separate vomeronasal nerves, but previous data indicate that calbindin-positive receptors within basal crypts of th…

Intranasal delivery of insulin via the olfactory nerve pathway.

OBJECTIVES: Intranasal delivery has been shown to target peptide therapeutics to the central nervous system (CNS) of animal models and induce specific neurological responses. In an investigation into the pathways by which intranasal administration delivers insulin to the CNS, this study has focused on the direct delivery of insulin from the olfactory mucosa to the olfactory bulbs via the olfactory nerve pathway. METHODS: Nasal and olfactory tissues of mice were imaged with fluorescent and electron microscopy 30 min following intranasal administration. KEY FINDINGS: Macroscopic analysis confi…

The spatial organization of olfactory nerve projections.

The spatial organization of olfactory nerve projections was examined in the rat. The pathway was traced by orthograde transport of HRP following nasal lavage and by retrograde transport of HRP following injections into the olfactory bulb. The results indicated that there was a broad relationship between the epithelium and the olfactory bulb. Specific regions of the olfactory bulb received input from a large region of the epithelium. However, there was evidence that olfactory nerve terminations were not uniformly dense across their terminal fields. The results suggest that there may be finer so…

Lectin histochemistry of cell-surface glycoconjugates in the primary olfactory projections of the newt.

The binding of 14 lectins were performed on paraffin-embedded sections of the olfactory bulb of Triturus to identify specific glycoconjugates on the cell surface of primary olfactory projections. The histochemical lectin staining patterns indicate that the membrane of olfactory neurons terminating in the main olfactory bulb contained prevalently oligosaccharides with alpha-acetyl-D-galactosamine as terminal residues. In the accessory olfactory bulb, instead, the primary olfactory projections possess a high density of alpha-D-galactose as sugar residues. The selective lectin binding on the surf…

[Clinical and MRI Findings in Patients with Congenital Anosmia].

The clinical characteristics of 16 patients with congenital anosmia were examined retrospectively. MRI (magnetic resonance imaging) was used to assess the morphological changes in the olfactory bulbs and olfactory sulci according to the method of P. Rombaux (2009). Congenital anosmia was divided into two forms: syndromic forms in association with a syndrome, and isolated forms without evidence of other defects. Only three patients (19%) in our series had syndromic forms of congenital anosmia, such as the Kallmann syndrome. Most cases (13 patients, 81%) had isolated congenital anosmia. Psychoph…
