Field tests of hexabutyldistannoxane (TBTO) in slow-release formulations against Biomphalaria spp.

Hexabutyldistannoxane (TBTO) in an asphalt base was found to retain molluscicidal activity for more than a year in the field. It was not deactivated by immersion in mud or by drying and exposure to the sun. Complete elimination of planorbid snails was achieved and maintained when repopulation pressure was sporadic, but control of a continuously entering population was not practicable. Fixing the product at the site is important, and a formulation in fragments of rubber that floated failed after 1-2 months. TBTO apparently acts cumulatively in snails, but is only initially toxic to aquatic inse

The linker governs the organelle targeting of TPP⁺-modified zinc phthalocyanine photosensitizers.

Mitochondria are key targets in anticancer drug design owing to their central roles in cancer cell apoptosis and energy metabolism. In this study, we design and synthesize two triphenylphosphonium (TPP⁺)-modified zinc phthalocyanine (ZnPc) photosensitizers: ZnPc-O3-P(ph)3 linked by a polyethylene glycol (PEG) chain and ZnPc-C5-P(ph)3 linked by an alkyl chain. Subcellular localization studies show that ZnPc-O3-P(ph)3 precisely targets mitochondria, whereas ZnPc-C5-P(ph)3 unexpectedly accumulates in lysosomes. This finding overturns the conventional assumption that TPP⁺ confers universal mitocho

Developmental Toxicology of Metal Mixtures in Drosophila: Unique Properties of Potency and Interactions of Mercury Isoforms.

Mercury ranks third on the U.S. Agency of Toxic Substances and Disease Registry priority list of hazardous substances, behind only arsenic and lead. We have undertaken uncovering the mechanisms underlying the developmental toxicity of methylmercury (MeHg), inorganic mercury (HgCl2), lead acetate (Pb), and sodium arsenite (As). To probe these differences, we used the Drosophila model, taking advantage of three developmental transitions-pupariation, metamorphosis, and eclosion-to differentiate potentially unique windows of toxicity. We elaborated dose response profiles for each individual metal

2-(2'-Pyridyl)-4,6-diphenylphosphinine versus 2-(2'-pyridyl)-4,6-diphenylpyridine: synthesis, characterization, and reactivity of cationic Rh(III) and Ir(III) complexes based on aromatic phosphorus heterocycles.

The bidentate P,N hybrid ligand 1 allows access for the first time to novel cationic phosphinine-based Rh(III) and Ir(III) complexes, broadening significantly the scope of low-coordinate aromatic phosphorus heterocycles for potential applications. The coordination chemistry of 1 towards Rh(III) and Ir(III) was investigated and compared with the analogous 2,2'-bipyridine derivative, 2-(2'-pyridyl)-4,6-diphenylpyridine (2), which showed significant differences. The molecular structures of [RhCl(Cp*)(1)]Cl and [IrCl(Cp*)(1)]Cl (Cp*=pentamethylcyclopentadienyl) were determined by means of X-ray di

Bi- and poly-metallic cyanide-bridged complexes of the redox-active cyanomanganese nitrosyl unit [Mn(CN)(PR3)(NO)(eta-C5H4Me)].

Cationic nitrile complexes and neutral halide and cyanide complexes, with the general formula [MnL1L2(NO)(eta-C5H4Me)]z, undergo one-electron oxidation at a Pt electrode in CH2Cl2. Linear plots of oxidation potential, Eo', vs. nu(NO) or the Lever parameters, EL, for L1 and L2, allow Eo' to be estimated for unknown analogues. In the presence of TlPF6, [MnIL'(NO)(eta-C5H4Me)] reacts with [Mn(CN)L(NO)(eta-C5H4Me)] to give [(eta5-C5H4Me)(ON)LMn(mu-CN)MnL'(NO)(eta5-C5H4Me)][PF6] which undergoes two reversible one-electron oxidations; DeltaE, the difference between the potentials for the two process

The relation of the neurotoxicity of organic tin and lead compounds to neurotubule disaggregation.

The effect of organic lead and tin compounds upon the integrity of cerebral neurotubules has been studies in vitro and in vivo, using 3H-colchicine binding as an index of tubulin aggregation. Triethyl tin, trimethyl lead and triethyl lead chlorides at concentrations above 5 X 10(-5)M, all prevented the polymerization of tubulin. Inorganic lead had a similar effect. Trimethyl tin had a much lesser capacity to prevent such polymerization. Rats received a single dose of each organometal, by subcutaneous injection and the capacity of hippocampal soluble protein to bind 3H colchicine was assessed.

Inactivation of yeast hexokinase B by triethyltin bromide.

Triethyltin bromide was found to demonstrate temperature-dependent inactivation of yeast hexokinase B. At temperatures of 20 degrees C or lower, little or no inactivation of the enzyme was detected after 2 h of reaction with 50-300 microM concentrations of the reagent. However, incubation at 25 degrees C or higher resulted in an increased rate and extent of loss of the enzyme activity with increasing incubation temperatures. The Arrhenius plot for the inactivation process showed a sharp break at approximately 30 degrees C, with a heat of activation (delta H*) above this temperature of 55.2 kca

A novel screening method for transition metal-based anticancer compounds using zebrafish embryo-larval assay and inductively coupled plasma-mass spectrometry analysis.

As novel metallodrugs continue to emerge, they are evaluated using models, including zebrafish, that offer unique sublethal endpoints. Testing metal-based anticancer compounds with high-throughput zebrafish toxicological assays requires analytical methods with the sensitivity to detect these sublethal tissue doses in very small sample masses (e.g., egg mass 100 μg). A robust bioanalytical model, zebrafish embryos coupled with inductively coupled plasma-mass spectrometry (ICPMS) for measurement of delivered dose, creates a very effective means for screening metal-based chemotherapeutic agents.

Elucidating the Supramolecular Interaction of Positively Supercharged Fluorescent Protein with Anionic Phthalocyanines.

Developing bioinspired materials to convert sunlight into electricity efficiently is paramount for sustainable energy production. Fluorescent proteins are promising candidates as photoactive materials due to their high fluorescence quantum yield and absorption extinction coefficients in aqueous media. However, developing artificial bioinspired photosynthetic systems requires a detailed understanding of molecular interactions and energy transfer mechanisms in the required operating conditions. Here, the supramolecular self-assembly and photophysical properties of fluorescent proteins complexed

Comparison of the biological effects of gadodiamide (Omniscan) and gadoteridol (ProHance) by means of multi-organ and plasma metabolomics.

Gadolinium-based contrast agents (GBCAs) are massively employed in radiology to increase the diagnostic power of MRI. However, investigations aiming at detecting possible metabolic perturbations or adverse health effects due to gadolinium deposition are still lacking. In this work, aqueous organs extract and plasma samples were analyzed by GC-MS and 1H-NMR, respectively, to investigate the effects of multiple administrations of one linear (Omniscan) and one macrocyclic (ProHance) GBCA, on the main metabolic pathways in healthy mice. Multivariate analysis revealed that plasma metabolome was not

A phase I/II study of the safety and efficacy of [177Lu]Lu-satoreotide tetraxetan in advanced somatostatin receptor-positive neuroendocrine tumours.

PURPOSE: We present the results of an open-label, phase I/II study evaluating the safety and efficacy of the novel somatostatin receptor (SSTR) antagonist [177Lu]Lu-satoreotide tetraxetan in 40 patients with previously treated, progressive neuroendocrine tumours (NETs), in which dosimetry was used to guide maximum administered activity. METHODS: This study was conducted in two parts. Part A consisted of 15 patients who completed three cycles of [177Lu]Lu-satoreotide tetraxetan at a fixed administered activity and peptide amount per cycle (4.5 GBq/300 µg). Part B, which included 25 patients wh

Light-Driven Tetra- and Octa-β-substituted Cationic Zinc(II) Phthalocyanines for Eradicating Fusarium oxysporum Conidia.

Photodynamic inactivation (PDI) is an emerging therapeutic approach that can effectively inactivate diverse microbial forms, including vegetative forms and spores, while preserving host tissues and avoiding the development of resistance to the photosensitization procedure. This study evaluates the antifungal and sporicidal photodynamic activity of two water-soluble amphiphilic tetra- and octa-β-substituted zinc(II) phthalocyanine (ZnPc) dyes with dimethylaminopyridinium groups at the periphery (ZnPcs 1, 2) and their quaternized derivatives (ZnPcs 1a, 2a). Safety and Gadolinium Distribution of the New High-Relaxivity Gadolinium Chelate Gadopiclenol in a Rat Model of Severe Renal Failure.

OBJECTIVE: The aim of this study was to investigate the toxicological profile of gadopiclenol, a new high-relaxivity macrocyclic gadolinium-based contrast agent (GBCA), in renally impaired rats, in comparison with 2 other macrocyclic GBCAs, gadoterate meglumine and gadobutrol, and 1 linear and nonionic GBCA, gadodiamide. METHODS: Renal failure was induced by adding 0.75% wt/wt adenine to the diet for 3 weeks. During the second week of adenine-enriched diet, the animals (n = 8/group × 5 groups) received 5 consecutive intravenous injections of GBCA at 2.5 mmol/kg per injection, resulting in a c
