Forebrain-specific glutamate receptor B deletion impairs spatial memory but not hippocampal field long-term potentiation.

We demonstrate the fundamental importance of glutamate receptor B (GluR-B) containing AMPA receptors in hippocampal function by analyzing mice with conditional GluR-B deficiency in postnatal forebrain principal neurons (GluR-B(deltaFb)). These mice are as adults sufficiently robust to permit comparative cellular, physiological, and behavioral studies. GluR-B loss induced moderate long-term changes in the hippocampus of GluR-B(deltaFb) mice. Parvalbumin-expressing interneurons in the dentate gyrus and the pyramidal cells in CA3 were decreased in number, and neurogenesis in the subgranular zone

Labeled lines in the retinotectal system: markers for retinorecipient sublaminae and the retinal ganglion cell subsets that innervate them.

Axons of retinal ganglion cells (RGCs) carry visual information to the brain. In most vertebrates, the major synaptic target of RGCs is the optic tectum. In the chick, RGC axons form synapses in just 4 of 16 histologically recognizable laminae (the retinorecipient laminae [RRLs]), and arbors of individual RGCs are confined to a single RRL. To analyze the development and function of these parallel pathways, markers are required that selectively label them. Here, we have identified molecular markers for individual RRLs and for RGCs that project to them. Some of the markers may mediate or modulat

Neurochemical markers for schizophrenia, bipolar disorder, and major depression in postmortem brains.

BACKGROUND: Previous studies of postmortem neurochemical markers in severe psychiatric disorders have been carried out on different brain collections, making it difficult to compare results. METHODS: One hundred RNA, protein, and other neurochemical markers were assessed in a single set of 60 postmortem brains (15 each with schizophrenia, bipolar disorder, major depression without psychosis, and unaffected control subjects) in relation to seven neurochemical systems. Quantitative measures of continuous variables for prefrontal, hippocampus, anterior cingulate, superior temporal cortex, or a c

Rotenone induces non-specific central nervous system and systemic toxicity.

We investigated the dopaminergic (DA) neuronal degeneration in animals subjected to systemic treatment of rotenone via subcutaneous delivery. Behavioral observations revealed a hypokinetic period in rats sacrificed at 3 and 5 days, and dystonic episodes in animals sacrificed at 8 days. Less than 20% of the total number of animals given rotenone depicted brain lesions after 8 days of treatment, as demonstrated by a significant loss of DA fibers in the striatum, but not of DA nigral neurons. Tyrosine hydroxylase-negative striatal territories were characterized by post-synaptic toxicity as demons

TRPM6 forms the Mg2+ influx channel involved in intestinal and renal Mg2+ absorption.

Mg2+ is an essential ion involved in a multitude of physiological and biochemical processes and a major constituent of bone tissue. Mg2+ homeostasis in mammals depends on the equilibrium between intestinal Mg2+ absorption and renal Mg2+ excretion, but little is known about the molecular nature of the proteins involved in the transepithelial transport of Mg2+ in these organs. Recently, it was shown that patients with mutations in TRPM6, a member of the transient receptor potential family of cation channels, suffer from hypomagnesemia with secondary hypocalcemia (HSH) as a result of impaired ren

Clinical findings in sporadic Creutzfeldt-Jakob disease correlate with thalamic pathology.

The pathogenesis underlying the typical findings in Creutzfeldt-Jakob disease (CJD) such as periodic EEG changes or myoclonus is not fully understood. The thalamus possesses a high density of inhibitory neurones and serves as a crucial pacemaker of rhythmic EEG activity. As inhibitory neurones expressing parvalbumin (PV) are reduced in the cerebral cortex and hippocampus in sporadic CJD (sCJD), we studied the distribution and number of PV-immunoreactive neurones in sCJD thalami in order to determine whether damage to them could account for certain clinical findings. Immuno histochemical analys

Effect of fetal hydrocephalus on the distribution patterns of calcium-binding proteins in the human occipital cortex.

Neuronal pathology in the cerebral cortex (CC) of fetal hydrocephalus brains is quite subtle when applying routine Nissl staining. This study is aimed at investigating alterations of interneurons which can be immunolabelled by antibodies against the calcium-binding proteins calretinin (CR), calbindin (CB) and parvalbumin (PV). The subplate (SP) subjacent to the cortex anlage is included as this transient zone plays a pivotal role in the establishment of cortical connections. Nine occipital lobes from cases of fetal hydrocephalus and 9 controls were categorized according to age: 21-25 weeks of

Morphological alterations in neocortical and cerebellar GABAergic neurons in a canine model of juvenile Batten disease.

The pathogenesis of brain dysfunction in a canine model of juvenile Batten disease was studied with techniques designed to determine sequential changes in mitochondrial morphology and cytochrome oxidase (CO) activity, and in neurons and synapses using gamma-aminobutyric acid (GABA) as a neurotransmitter. Histochemical and immunocytochemical methods were employed. Mitochondrial alterations were found in a select population of nonpyramidal neurons in neocortex and claustrum, and in cerebellar basket cells. Proportions of affected neurons at any one time remained constant over the disease course,

p75NTR modulation prevents cellular, cortical activity and cognitive dysfunctions caused by perinatal hypoxia.

Children who have experienced moderate perinatal hypoxia are at risk of developing long-lasting subtle cognitive and behavioural deficits, including learning disabilities and emotional problems. Understanding the underlying mechanisms is an essential step for designing targeted therapy. Fast-spiking, parvalbumin (PV)-positive GABAergic interneurons modulate the generation of gamma oscillations, which, in turn, regulate many cognitive functions, including goal-directed attentional processing and cognitive flexibility. Owing to their fast firing rate, PV cell function requires high levels of ene

"Ependymal-in" Gradient of Thalamic Damage in Progressive Multiple Sclerosis.

Leptomeningeal and perivenular infiltrates are important contributors to cortical grey matter damage and disease progression in multiple sclerosis (MS). Whereas perivenular inflammation induces vasculocentric lesions, leptomeningeal involvement follows a subpial "surface-in" gradient. To determine whether similar gradient of damage occurs in deep grey matter nuclei, we examined the dorsomedial thalamic nuclei and cerebrospinal fluid (CSF) samples from 41 postmortem secondary progressive MS cases compared with 5 non-neurological controls and 12 controls with other neurological diseases. CSF/epe

EF-hand protein Ca2+ buffers regulate Ca2+ influx and exocytosis in sensory hair cells.

EF-hand Ca(2+)-binding proteins are thought to shape the spatiotemporal properties of cellular Ca(2+) signaling and are prominently expressed in sensory hair cells in the ear. Here, we combined genetic disruption of parvalbumin-α, calbindin-D28k, and calretinin in mice with patch-clamp recording, in vivo physiology, and mathematical modeling to study their role in Ca(2+) signaling, exocytosis, and sound encoding at the synapses of inner hair cells (IHCs). IHCs lacking all three proteins showed excessive exocytosis during prolonged depolarizations, despite enhanced Ca(2+)-dependent inactivation

Redox Homeostasis and Oxidative Stress in Schizophrenia: Glutathione-NMDA-Neuroimmune Convergence and a Hypothesis-Generating Iron-Lipid Redox Extension.

Schizophrenia is a heterogeneous neurodevelopmental disorder in which genetic liability, developmental exposures, illness stage, treatment, and metabolic or inflammatory comorbidity may shape redox biology. This narrative review evaluates the human and mechanistic evidence for impaired redox adaptation as a convergence mechanism linking glutathione (GSH) regulation, N-methyl-D-aspartate receptor hypofunction, parvalbumin-interneuron and perineuronal-net vulnerability, mitochondrial-glial dysfunction, and neuroimmune signalling. The findings do not support a uniform oxidative abnormality across

Hippocampal mechanisms and neural circuitry of negative symptoms in schizophrenia.

Negative symptoms represent a debilitating core of schizophrenia symptomatology that fail to respond to treatment with D2 antagonist antipsychotic drugs and may even be exacerbated by D2 antagonism. The neurobiological mechanisms underlying these symptoms are proposed to be related to limbic hippocampal hyperactivity, which is associated with the loss of parvalbumin-expressing GABAergic interneurons. This hippocampal hyperexcitability generates aberrant excitatory and dysrhythmic output to key limbic regions. Dysregulation of limbic hippocampal projections to the ventral striatum could impair

ECM remodeling in the mPFC exacerbates cocaine-induced hyperactivity and impairs threat vigilance.

The extracellular matrix (ECM) plays a crucial role in addiction-related neuroplasticity. While chondroitinase ABC (chABC)-mediated ECM degradation has been shown to impair drug-conditioned place preference, its effects on cocaine-induced locomotor activity remain unknown. Using 3D motion capture combined with unsupervised behavioral clustering in a chronic cocaine exposure mouse model, we demonstrate that cocaine treatment significantly increases locomotor activity while impairing environmental threat perception. Mechanistically, we observed elevated Wisteria floribunda agglutinin (WFA) inten

Neurodevelopmental Alterations in Inhibitory Interneurons of Cortex, Hippocampus, and Striatum of Genetic Absence Epilepsy Rats.

AIMS: Absence seizures, characterized by spike-and-wave discharges (SWDs), are mediated by reciprocal thalamocortical interactions; however, the contribution of developing inhibitory networks to SWDs remains unclear. We investigated the developmental trajectory of inhibitory interneurons in Genetic Absence Epilepsy Rats from Strasbourg (GAERS) by analyzing their distribution across postnatal development in the somatosensory (S1) and motor (M1) cortices, the hippocampus, and striatum. METHODS: The neurodevelopmental trajectory of parvalbumin-positive (PV+) and somatostatin-positive (SST+) inte

Dysfunction of GABAergic interneurons underlies altered neural network oscillations associated with epileptiform activity in PPT1-deficient mice.

The neuronal ceroid lipofuscinosis family of lysosomal storage diseases, also called CLN1 disease, is characterized by the deficiency of palmitoyl-protein thioesterase 1 (PPT1). In this study, we investigated the impact of PPT1 deficiency on hippocampal GABAergic interneurons (INs) and associated neural network oscillations in a PPT1-KI (CLN1 c.451 C > T (p.R151X)) mouse model. Using a combination of in vivo electrophysiology, immunostaining, and fiber photometry, we observed that PPT1 deficiency led to the activation of caspase 3 in parvalbumin-positive (PV+) INs, an increased activity of pyr

Optogenetic Inhibition of Striatal Parvalbuminergic Interneurons Unmasks Impaired GABA and Adenosine Signaling in DYT1 Knock-In Mice.

Neurochemical imbalances in the striatum are thought to contribute to the pathophysiology of DYT1 dystonia (TOR1A), a severe movement disorder. Parvalbumin-positive GABAergic fast-spiking interneurons (PV+ FSI) exert a powerful inhibition within the striatal microcircuitry. To elucidate the impact of PV+ FSI on striatal neurotransmitter dynamics in a DYT1 knock-in (KI) mouse model, we combined optogenetic inhibition of PV+ FSI with in vivo microdialysis (optodialysis) and LC-MS/MS analysis. Dialysates were collected across baseline (light off), stimulation (light on, 595 nm), and post-stimulat

A nose-to-brain circuit underlies anxiety regulation by nasal afferent frequency in mice.

Slow nasal breathing alleviates negative moods, but the precise nose-to-limbic pathways and their causal contributions remain unclear. Here, we identify a pathway in mice from olfactory sensory neurons (OSNs) in the nasal cavity to mitral cells in the olfactory bulb (OB), then to parvalbumin-positive (PV+) long-projecting interneurons in the perirhinal cortex (PRC), and subsequently to glutamatergic neurons in the posterior basolateral amygdala (pBLA), through which nasal afferent activity bidirectionally regulates anxiety in a frequency-dependent manner. Low-frequency nasal airflow or optogen

Minimally-invasive manipulation of spared and hypoactive interneurons reduces CA1 synchronization and nonspatial behavior alterations in epilepsy models.

Temporal lobe epilepsy (TLE) is associated with seizures and severe cognitive impairments, including memory deficits. The dysfunction of a large population of hippocampal interneurons, composed of various subtypes, is proposed as a key mechanism and a therapeutic target. However, the nature and extent of alterations in hippocampal inhibitory neurons remain unclear, as does their impact on TLE pathology. To address this issue, we combined immunolabeling, calcium imaging, electrophysiology, chemogenetic tools, and behavioral assays in mouse pilocarpine TLE models to investigate the survival and
