c-Cbl tyrosine phosphorylation and subcellular localization in human primary leukemic cells.

Several studies indicate that a number of signal-transducing molecules involved in the proliferation, differentiation, and functional activation of normal hemopoietic cells may be constitutively activated in primary leukemic cells and play a role in the outcome or in the progression of these neoplastic disorders. In this study we show that the product of the proto-oncogene c-Cbl, whose function is still unknown, is constitutively tyrosine phosphorylated not only in cells from chronic myelogenous leukemias (CMLs) in the blast phase, but also in cells from acute myeloblastic leukemias (AMLs), Ph

Collagen induces tyrosine phosphorylation of Wiskott-Aldrich syndrome protein in human platelets.

Wiskott-Aldrich syndrome (WAS) and X-linked thrombocytopenia (XLT) are caused by mutations of the WAS protein (WASP) gene. All hematopoietic stem cell-derived lineages, including platelets, express WASP. Platelets from WAS patients are smaller than their normal counterparts and defects in platelet aggregation and actin polymerization have been reported. To determine if WASP is important for normal platelet function, we examined its role in signal transduction. We found that collagen but not thrombopoietin or thrombin induces a rapid and robust increase in tyrosine phosphorylation of platelet-a…

Phosphorylation of specific sets of tau isoforms reflects different neurofibrillary degeneration processes.

Tau proteins are the basic components of filaments that accumulate within neurons during neurofibrillary degeneration, a degenerating process with disease-specific phenotypes. This specificity is likely to be sustained by both phosphorylation state and isoform content of tau aggregates that form neuronal inclusions. In the present study, characterization of tau isoforms involved in neurofibrillary degeneration in Alzheimer's disease, Pick's disease, corticobasal degeneration and progressive supranuclear palsy was performed. Both analyses by immunoblotting using specific tau antibodies and cell…

Aberrant tyrosine phosphorylation in head and neck tumours and potentially premalignant tissues.

Protein tyrosine kinases (PTK) and protein tyrosine phosphatases (PTPase) activity in tumor tissue, obtained from 107 patients with squamous cell carcinoma of the upper aerodigestive tract, was determined. In 79 patients samples were also taken from a histologically proven non-tumourous part of the mucosa. In this extensive study we confirmed our previous findings of increased enzyme activities (cytosol PTK, membranes PTK, cytosol PTPase) in non-tumour tissues compared with control tissues. These biochemical changes suggesting a premalignant mucosa could not be correlated to a higher probabili

Control of oxidative phosphorylation in rat muscle mitochondria: implications for mitochondrial myopathies.

The control of oxidative phosphorylation has been studied in normal skeletal muscle isolated from the hind legs of rats. The control coefficients of different steps of oxidative phosphorylation on the flux of O2 consumption were determined by the inhibitor method and calculation was done according to the model of Gellerich et al. (FEBS Lett. 274 (1990) 167-170) using a non-linear regression fitting procedure. The respiration was recorded with pyruvate (+malate) and palmitoyl-L-carnitine (+malate) as respiratory substrates, which are the main substrates oxidized in the muscle. It appears that t…

Phosphorylation of elongation factor 2 in normal and malignant rat glial cells.

Certain calmodulin (CaM)-dependent protein kinases phosphorylate substrates have been implicated in regulating cellular proliferation. In this study, CaM-dependent phosphorylation has been examined in normal and tumor tissue from rat brain to determine whether differences exist. Using in vitro phosphorylation reactions, we compared endogenous substrates for Ca2+/CaM-dependent protein kinases in rat brain white matter (RBWM), a tissue rich in normal glia, to those of C6 rat glioma cells. A major phosphoprotein having a M(r) of 100,000 was observed in proliferating C6 cells that was not present …

Subnuclear localization and phosphorylation of Epstein-Barr virus latent infection nuclear proteins.

Functions of the six Epstein-Barr virus latent infection nuclear proteins (EBNA-1, -2, -3A, -3B, -3C, or -LP) in maintaining latent infection or cell growth transformation are only partially understood. Using antibodies specific for each EBNA in immunofluorescence microscopy, EBNA-2, -3A, and -3C localized to subnuclear granules which fill much of the nucleus, excluding nucleoli. EBNA-LP localized to a small number of discrete subnuclear particles, also excluding nucleoli. Only EBNA-1 associated with metaphase chromosomes. Concordantly, in biochemical nuclear fractionation studies, EBNA-1 was

Influence of cerebral ischemia and post-ischemic reperfusion on mitochondrial oxidative phosphorylation.

Unilateral ischemia in the right cerebral hemisphere of the rat was induced by ligation of the right common carotid artery coupled with controlled hemorrhage to produce hypotension (25 +/- 8 mm/Hg). Where indicated after 30 min of ischemia, the withdrawn blood was reinfused to restore arterial pressure to normal. Mitochondria isolated from the ipsilateral hemisphere after 30 min of ischemia showed significantly lower respiratory rates than the organelles isolated from the contralateral side. Oxidation of NAD(+)-linked substrates was more sensitive to inhibition in ischemia (30%) than was of fe

Defective phosphorylation of a calmodulin-binding protein in cystic-fibrosis submandibular glands.

Calmodulin-binding proteins in fractions purified from human submandibular glands by calmodulin-Sepharose were phosphorylated with [gamma-32P]ATP, in the absence of exogenous protein kinase. The major proteins phosphorylated had molecular masses of 45, 51 and 61 kDa. Phosphorylation was increased by activators of protein kinase C and inhibited by H-7. Phosphorylation of the 61 kDa band was markedly decreased in cystic-fibrosis submandibular glands.

Relationship between phosphorylation and synthesis of goldfish optic nerve proteins during regeneration.

After intraocular injection of radiolabeled phosphate and 3H-proline, the labeling of goldfish optic nerve proteins was monitored over a 7 w … decrease in 32P incorporation, even though 3H-proline incorporation was increased. For example, the phosphorylation of ON2, a neuronal intermediate filament protein, showed a long-lasting decline, which was already evident at 1 week and had not yet returned to normal by 7 weeks. Other proteins in this group (33, 37, and 46) showed a faster recovery.(ABSTRACT TRUN …

Protein phosphorylation of human lysosomal arylsulfatase B from normal and cancer tissues.

Previous studies from this laboratory have demonstrated that arylsulfatase B (ASB) is phosphorylated by a protein kinase, which is the first finding of phosphorylation in lysosomal hydrolases. The present study was undertaken to characterize the sites of phosphorylation in ASB from transplanted human lung cancer and from normal human tissues, and to identify type of tumor protein kinase responsible for the phosphorylation of ASB. When ASB purified from liver and placenta was phosphorylated in vitro by a cAMP-dependent protein kinase, it gave a single tryptic phosphopeptide (X) and phosphothreo…

Phosphorylation of alpha-tubulin carboxyl-terminal tyrosine prevents its incorporation into microtubules.

Insulin receptor kinase phosphorylated tubulin in an insulin-dependent fashion. Two different populations of phosphotubulin were found. In tubulin dimers containing tyrosine at the carboxyl-terminal of their alpha subunit, phosphate was incorporated in that residue, and the phosphorylated protein did not assemble into polymers. In tubulin dimers lacking this tyrosine residue, phosphate was incorporated into different tyrosine residues located in other parts of the molecule, and the phosphoprotein retained its capacity to polymerize.

Ascaris suum: regulation of myosin light chain phosphorylation from adult skeletal muscle.

The regulatory myosin light chain in Ascaris suum muscle was isolated in a phospho and dephosphoform. In freshly dissected tissue or in control muscle segments perfused with saline, approximately 48% of the total myosin light chain pool was phosphorylated. Perfusion with acetylcholine caused a dose dependent increase in muscle contraction but no change in myosin light chain phosphorylation. In contrast, perfusion with gamma-aminobutyric acid resulted in a dose dependent relaxation of A. suum muscle and a decrease in myosin light chain phosphorylation so that only 18% of the total myosin light …

Phosphorylation of phospholamban in intact myocardium. Role of Ca2+-calmodulin-dependent mechanisms.

Phospholamban, a putative regulator of cardiac sarcoplasmic reticulum Ca2+ transport, has been shown to be phosphorylated in vitro by cAMP-dependent protein kinase and an intrinsic Ca2+-calmodulin-dependent protein kinase activity. This study was conducted to determine if Ca2+-calmodulin-dependent phosphorylation of phospholamban occurs in response to physiologic increases in intracellular Ca2+ in intact myocardium. Isolated guinea pig and rat ventricles were perfused with 32Pi after which membrane vesicles were isolated from individual hearts by differential centrifugation. Administration of …

Ethanol tolerance and enhanced calcium/calmodulin-dependent phosphorylation of synaptic membrane proteins.

The chronic effects of ethanol on synaptic membrane proteins was studied in ethanol-tolerant rats. Synaptic plasma membranes and postsynaptic densities were prepared from homogenates of forebrain and incubated in vitro with [gamma-32P]adenosine triphosphate in the presence and absence of calcium and calmodulin. In ethanol-tolerant animals, enhanced phosphorylation of synaptic plasma membrane but not postsynaptic density proteins was demonstrated in the presence of calcium and calmodulin. These results suggest that the development of tolerance to ethanol may involve alteration in neuronal sensi…

Pi transport, phosphorylation, and dephosphorylation in renal membranes from HYP/Y mice.

To ascertain whether cAMP-dependent phosphorylation could be demonstrated in brush border membrane vesicles (BBMV) isolated from kidneys of mice with X-linked hypophosphatemic rickets (HYP/Y) and normal littermates (+/Y) and, if so, to determine whether the absence of dephosphorylation might underlie differences in Na+-dependent 32Pi transport in BBMV, we measured 1) 32Pi transport, 2) cAMP-dependent phosphorylation, and 3) dephosphorylation in BBMV from +/Y and HYP/Y mice. Na+ gradient-dependent 32Pi transport was decreased in BBMV from HYP/Y mice as reflected in a decreased apparent Vmax. cA…

Urea cycle regulation: I. Coupling of ornithine metabolism to mitochondrial oxidative phosphorylation.

Ornithine metabolism is coupled to oxidative phosphorylation in isolated rat liver mitochondria. The pathway involving ornithine: alpha-ketoglutarate transaminase (OKT), glutamic semialdehyde dehydrogenase (GSDH), and glutamate dehydrogenase (GDH) with cycling of alpha-ketoglutarate-glutamate at the OKT reaction appears to be involved. Ornithine may be utilized by this pathway to sustain ATP levels during mitochondrial energy-deficiency states with resultant decreased urea-cycle flux and increased ammonia production. This pathophysiologic mechanism suggests that hyperammonemia is a consequence…

Altered erythrocyte membrane protein phosphorylation in an unusual case of hereditary spherocytosis.

Membrane protein phosphorylation was examined in several members from a family with an unusual form of hereditary sperocytosis. After incubation of membrane ghosts with (gamma-32 P) ATP, the phosphorylation of spectrin component II was diminished both in the absence of cAMP. In the presence of this nucleotide, the phosphorylation of components IV5 and IV8 was also decreased. Along with a previously reported alteration of a membrane neutral phosphatase in this family, these abnormalities remove the present condition from the usual form of hereditary spherocytosis.
