Direct anticryptococcal activity of lymphocytes from Cryptococcus neoformans-immunized mice.

Assessment of the direct anticryptococcal activity of murine lymphocytes from both Cryptococcus neoformans-immunized and control mice was the focus of this investigation. We demonstrate that at a 2:1 effector cell-to-cryptococcal target cell ratio, effector cell populations comprised of alpha beta T-cell receptor-positive T lymphocytes (98 to 99% CD3+) from C. neoformans-immunized mice inhibited the growth of cryptococcal cells better than similar populations of lymphocytes from nonimmunized control mice. Almost immediately after mixing of cryptococci with the effector cells, C. neoformans-lym

Sustained growth-promoting effects of vosoritide in children with achondroplasia from an ongoing phase 3 extension study.

BACKGROUND: Vosoritide is a C-type natriuretic peptide analog that addresses an underlying pathway causing reduced bone growth in achondroplasia. Understanding the vosoritide treatment effect requires evaluation over an extended duration and comparison with outcomes in untreated children. METHODS: After completing ≥6 months of a baseline observational growth study and 52 weeks in a double-blind, placebo-controlled study (ClinicalTrials.gov: NCT03197766), participants were eligible to continue treatment in an open-label extension (ClinicalTrials.gov: NCT03424018) wherein all received 15 μg/kg

Clinical research lags behind biomedical, population-based health, and health services research at multiple levels.

PURPOSE: Concerns regarding a decline in clinical research have been raised internationally. In this study, research initiatives and competitiveness of investigators seeking funding for clinical research were compared with those for three other health research themes in Canada, namely, biomedical, population-based, and health services research. METHODS: A retrospective, multi-level descriptive study was conducted using administrative data from the Canadian Institutes for Health Research (CIHR) research grants program. Annual growth rates in numbers of proposals submitted since year 2000 (leve

Customized versus population birth weight charts for identification of newborns at risk of long-term adverse cardio-metabolic and respiratory outcomes: a population-based prospective cohort study.

BACKGROUND: Customized birth weight charts take into account physiological maternal characteristics that are known to influence fetal growth to differentiate between physiological and pathological abnormal size at birth. It is unknown whether customized birth weight charts better identify newborns at risk of long-term adverse outcomes than population birth weight charts. We aimed to examine whether birth weight classification according to customized charts is superior to population charts at identification of newborns at risk of adverse cardio-metabolic and respiratory health outcomes. METHOD

Steroid inhibition of limbal blood and lymphatic vascular cell growth.

Steroids are widely used in the prevention of corneal neovascularization in a wide range of natural and experimental situations. However, no information is available on their effect on the growth of the individual limbal blood vascular cells or of lymphatic cells involved in corneal neovascularization. In addition, tritiated thymidine labelling index is commonly used as an indicator of cell population but doubt exists as to whether it truly represents cell growth. Remote thermal cautery of the rat cornea was used to elicit corneal neovascularization. New cell growth was measured by tritiated t

Navigating rural challenges and promoting posttraumatic growth via expressive writing: a qualitative study among rural breast cancer survivors.

PURPOSE: Breast cancer is the second most prevalent cancer among women in the USA. Rural breast cancer survivors (RBCS) face unique challenges because geographical isolation limits access to facilities, support groups, and mental health services. Expressive writing (EW), a therapeutic intervention navigates individuals through traumatic experiences, has demonstrated benefits for RBCS. However, first-person accounts of how RBCS experience EW remain limited. This study addresses this gap. METHODS: Virtual semi-structured in-depth interviews were conducted with participants (N = 14) who previous

Percentile Charts for Body Mass Index of Indian Down Syndrome Children.

Growth charts are used to detect growth impairment, overweight, and obesity among Down syndrome (DS) children belonging to different population groups. Due to nonavailability of similar information, age, and gender specific body mass index (BMI) charts for DS children of Indian origin, based on serial data, have been developed. A total of 752 boys and 373 girls diagnosed as cases of DS at <1 month to 10 years of age enrolled from the "genetic clinic" were followed up in the "growth clinic/growth laboratory" of the institute, following a mixed-longitudinal growth research design. BMI was calcul

Higher prevalence of stunting and poor growth outcomes in HIV-exposed uninfected than HIV-unexposed infants in Kenya.

BACKGROUND: With the growing population of HIV-exposed uninfected (HEU) children globally, it is important to determine population-level growth differences between HEU and HIV-unexposed uninfected (HUU) children. METHODS: We analyzed data from a population-level survey enrolling mother-infant pairs attending 6-week and 9-month immunizations in 140 clinics across Kenya. Weight-for-age (WAZ), length-for-age (LAZ), head circumference-for-age (HCAZ) z-scores and underweight (WAZ < -2), stunting (LAZ < -2), and microcephaly (HCAZ < -2), were compared between HEU and HUU. Key gut bacterial taxa and their correlative relationships with host genes during Musca domestica aggregation.

Aggregation enhances survival and reproduction by regulating body coloration, physiology, and immunity. The housefly (Musca domestica) is a notable sanitary pest that exhibits group-breeding behavior during its larval stage, which promotes its growth and development. However, the mechanisms underlying how high larval density regulates growth remain unclear. Previous studies have shown that gut bacteria are important regulators of larval growth and development. To further investigate this, the present study aimed to assess the impact of group rearing on both the gut microbiota and host gene exp

Air Quality and Health Impact of Future Fossil Fuel Use for Electricity Generation and Transport in Africa.

Africa has ambitious plans to address energy deficits and sustain economic growth with fossil fueled power plants. The continent is also experiencing faster population growth than anywhere else in the world that will lead to proliferation of vehicles. Here, we estimate air pollutant emissions in Africa from future (2030) electricity generation and transport. We find that annual emissions of two precursors of fine particles (PM2.5) hazardous to health, sulfur dioxide (SO2) and nitrogen oxides (NOx), approximately double by 2030 relative to 2012, increasing from 2.5 to 5.5 Tg SO2 and 1.5 to 2.8 …

A new method for the detection of Lesch-Nyhan heterozygotes by peripheral blood T cell culture using T cell growth factor.

Thioguanine-resistant T lymphoblast populations were selectively amplified using T cell growth factor in the cultures of peripheral blood T cells from four Lesch-Nyhan heterozygotes. Although Lesch-Nyhan T lymphoblasts were all thioguanine-resistant, none of the cultures from 13 control subjects yielded the growth of such defective cell populations. These data provide direct evidence for the existence of a small percentage (5%-40%) of hypoxanthine-guanine phosphoribosyltransferase (HGPRT) deficient T cells in the heterozygotes, but not in normal individuals. Conversely, culture of the T lympho

Exploring posttraumatic outcomes as a function of childhood sexual abuse.

There is sparse systematic examination of the potential for growth as well as distress that may occur for some adult survivors of childhood sexual abuse. The presented study explored posttraumatic growth and its relationship with negative posttrauma outcomes within the specific population of survivors of childhood sexual abuse (N = 40). Results showed that 95% of the participants experienced clinically significant post-traumatic stress disorder symptomatology related to their childhood sexual abuse. In conjunction with these high levels of negative symptoms, the population evidenced posttrauma

Rat heart-derived endothelial and smooth muscle cell cultures: isolation, cloning and characterization.

This report describes the initiation, cloning and establishment of long-term serial cultures of rat heart-derived vascular endothelial (EC) and smooth muscle cells (SMC). Populations of these cells derived from both the macro-and microcirculation were obtained utilizing isolated heart perfusion technique. Elimination of potential mesothelial cell contamination was achieved by ethanol fixation of the pericardial surface prior to perfusion. Initial outgrowths from perfusate yielded both endothelial (rapid adhering) and smooth muscle (slow adhering) appearing cell populations. Subsequent pooling

Locomotory and physiological responses induced by clove and cinnamon essential oils in the maize weevil Sitophilus zeamais.

Plant essential oils have been suggested as a suitable alternative for controlling stored pests worldwide. However, very little is known about the physiological or behavioral responses induced by these compounds in insect populations that are resistant to traditional insecticides. Thus, this investigation evaluated the toxicity (including the impacts on population growth) as well as the locomotory and respiratory responses induced by clove, Syzygium aromaticum L., and cinnamon, Cinnamomum zeylanicum L., essential oils in Brazilian populations of the maize weevil Sitophilus zeamais. We used pop…

Recent evolution of extreme cestode growth suppression by a vertebrate host.

Parasites can be a major cause of natural selection on hosts, which consequently evolve a variety of strategies to avoid, eliminate, or tolerate infection. When ecologically similar host populations present disparate infection loads, this natural variation can reveal immunological strategies underlying adaptation to infection and population divergence. For instance, the tapeworm Schistocephalus solidus persistently infects 0-80% of threespine stickleback (Gasterosteus aculeatus) in lakes on Vancouver Island. To test whether these heterogeneous infection rates result from evolved differences in

[Short stature in chronic kidney disease: physiopathology and treatment with growth hormone].

Growth failure is frequent and a clinically important issue in children with chronic kidney disease (CKD). Many factors contribute to impaired growth in these children, including abnormalities in the growth hormone (GH)--insulin-like growth factor 1 (IGF-1) axis, malnutrition, acidosis, renal bone disease and glucocorticoid associated treatment. The management of growth failure in children with CKD is complicated by the presence of other-disease related complications requiring medical intervention. Despite evidence of GH efficacy and safety in this population, this therapy is still underutiliz

Chromosome replication as a measure of bacterial growth rate during Escherichia coli infection in the mouse peritonitis model.

The efficacy of most antibiotics is dependent on active bacterial growth, yet little is known about the growth dynamics during infection. Therefore, means to measure in-host bacterial growth rate is of importance. Here, we use chromosome replication as readout for in situ bacterial growth rate during infection; obtained from a single biological specimen. We have applied two independent methods: quantitative PCR (qPCR) and fluorescence microscopy, to quantify the level of chromosome replication present during Escherichia coli propagation in the mouse peritonitis model. We find that the methods
