Ionic mechanisms of GABA-induced long-term potentiation in the rat superior colliculus.

GABA-induced excitation and long-term potentiation (LTPG) have been demonstrated recently in the superficial layers of the superior colliculus (SC). In other regions of the nervous system, GABA elicits excitatory responses via ionotropic GABA receptors under certain conditions. This excitation is proposed to be due to either a high neuronal chloride concentration favouring a depolarising chloride efflux, or to a bicarbonate efflux coupled to a chloride influx. The aim of this study was to characterise the mechanisms underlying excitation and prolonged increase in synaptic transmission induced

Mercury suppression of a potassium current in human B lymphocytes.

Mercury is a recognized environmental toxin. Several organ systems are targeted by this substance and impairment of immune function is known to result from exposure to mercury. Using the patch clamp technique in the whole cell configuration on resting human B lymphocytes we have identified an outward potassium current and studied the effects of mercury on this current. We present data that demonstrate: (i) the absence of inward currents; (ii) a time and voltage dependent outward current with a threshold of -40 mV and reversal potential near EK+; (iii) blocking of this current by TEA (tetraethy

Hepatoportal bumetanide-sensitive K(+)-sensor mechanism controls urinary K(+) excretion.

To determine whether a K(+)-sensor mechanism exists in the hepatoportal region, periarterial hepatic afferent nerve activity responses to intraportal injection of KCl were examined in anesthetized rats. Hepatic afferent nerve activity increased in response to intraportal injection in a K(+) concentration-dependent manner, and the increase was attenuated by inhibition of the Na(+)-K(+)-2Cl(-) cotransporter by bumetanide in a dose-dependent manner. These results suggest that a bumetanide-sensitive K(+)-sensor mechanism exists in the hepatoportal region. Stimulation of this mechanism by intraport

Vasopressin regulation of sodium transport in the distal nephron and collecting duct.

Arginine vasopressin (AVP) is released from the posterior pituitary gland during states of hyperosmolality or hypovolemia. AVP is a peptide hormone, with antidiuretic and antinatriuretic properties. It allows the kidneys to increase body water retention predominantly by increasing the cell surface expression of aquaporin water channels in the collecting duct alongside increasing the osmotic driving forces for water reabsorption. The antinatriuretic effects of AVP are mediated by the regulation of sodium transport throughout the distal nephron, from the thick ascending limb through to the colle

Loop diuretics are K+-sparing in the presence of a low-Na+, high-K+ diet.

Under most conditions, loop diuretics are K+-wasting, requiring potassium supplementation. In this issue, Wang and colleagues demonstrate that in mice fed a low-Na+, high-K+ diet, loop diuretics, in contrast, are K+-sparing. This observation suggests that possible elevations in plasma K+ should be monitored when using a loop diuretic with a low-Na+, high-K+ diet, particularly when in combination with a potassium supplement.

Effect of acute metabolic acidemia on renal electrolyte transport in man.

The effect of acute NH4C1-induced metabolic acidemia on renal electrolyte excretion was examined in nine healthy subjects during steady state water diuresis. Following oral NH4C1, venous pH and bicarbonate concentration declined significantly (p less than 0.01) while inulin and PAH clearances remained unchanged. Mean sodium excretion (UNaV) increased from 142 +/- 16 mueq/min (mean +/- SEM) to 310 +/- 49 mueq/min (p less than 0.01) at 8 hr without change in plasma aldosterone or renin levels. Urine flow remained unchanged while CH2O/(CH2O + CCl) declined significantly, suggesting that acute met

Serum electrolytes and capillary blood gases in the management of hypertrophic pyloric stenosis.

The purpose of this study was to assess whether the concentration of serum chloride and other variables, namely serum sodium, potassium and bicarbonate, can be used to predict metabolic acid-base status in infants with hypertrophic pyloric stenosis (HPS) and to assess whether such a prediction is influenced by the state of hydration. One hundred and sixty-three infants with HPS who had at least one set of serum electrolyte and capillary blood gas estimations performed after admission were studied retrospectively. A further 25 infants who had their electrolyte and blood gases measured after at

Potassium restores vasorelaxation of resistance arterioles in non-hypertensive DOCA/salt fed mice.

BACKGROUND: Potassium-enriched diets exert renal and cardiovascular protective effects, but the underlying mechanisms are largely unknown. METHODS: Using the dorsal skinfold chamber model for intravital microscopy, we examined endothelium-dependent vasorelaxation of precapillary resistance arterioles in response to acetylcholine or the NO donor SNAP in awake mice. Experiments were performed in uni-nephrectomized one renin gene (Ren-1c) C57BL/6 mice (control group) and in mice having received a continuous administration of deoxycorticosterone acetate and a dietary supplementation of 1% sodium

A girl having congenital chloride diarrhea treated with spironolactone for seven years.

We report on a girl having congenital chloride diarrhea (CCD) who has been followed for 7 years and 6 months sequentially. Dilated intestina … entified. She was diagnosed as having CCD. Because it was difficult to administer a large volume of potassium chloride (KCl), and sodium chloride (NaCl), we decided to administer spironolactone. After administration of spironolactone, we could generate correct serum electrolytes using less amounts of KCl. At 7 years and 6 months of age, her body size was within norm …

[The pre- and postoperative course of pH values and net acid-base excretion in the urine of cows with abomasal displacement].

The pre- and postoperative course of acid-base parameters and electrolyte concentrations in urine and blood was studied in 67 cows affected with left- or right-sided abomasal displacement. Paradoxic aciduria was detected in 21 of 37 cattle with preoperative alkalosis. The validity of serum electrolyte determinations for the detection of electrolyte disturbances is limited. The evaluated parameters returned to normal in most of the animals within the following intervals post-op.: 1 day: blood gas parameters 2 days: hematocrit; 3 days: general condition, serum chloride and serum sodium concentra

Congenital chloride diarrhoea in Kuwaiti children.

Congenital chloride diarrhoea was diagnosed in 16 Kuwaiti children over a 7 year period (1980-1986) with an estimated incidence of 7.6 per 100,000 live births. The mean age at diagnosis was 3.2 months (range 1 week to 5 months). There were 9 boys and 7 girls with a mean age of 3 years 10 months (range 10 months to 7 years). All children had a shortened gestational period, abdominal distension and chronic diarrhoea. The serum electrolytes in all patients prior to treatment showed hyponatraemia, hypokalaemia, hypochloraemia and metabolic alkalosis. The diagnosis was confirmed by a stool chloride

Elution and decomposition of cyanide in soil contaminated with various cyanocompounds.

Standard soil samples contaminated with various standard cyanocompounds were prepared. Column elution experiments and analyses were conducted. Compounds with an easy capacity for dissociation to ions, such as KCN and potassium hexacyanoferrate(III), were found to be eluted by forming free cyanide even in fresh water. Hexacyanoferrate(II) salts, such as potassium hexacyanoferrate(II) and iron(III) hexacyanoferrate(II), were found not to be dissociated in water, but were dissociated and diffused under alkaline conditions (pH >13). Hexacyanoferrate(II) ion was found to be more easily dissociated

New selective inhibitors of calcium-activated chloride channels - T16A(inh) -A01, CaCC(inh) -A01 and MONNA - what do they inhibit?

BACKGROUND AND PURPOSE: T16A(inh)-A01, CaCC(inh)-A01 and MONNA are identified as selective inhibitors of the TMEM16A calcium-activated chloride channel (CaCC). The aim of this study was to examine the chloride-specificity of these compounds on isolated resistance arteries in the presence and absence (±) of extracellular chloride. EXPERIMENTAL APPROACH: Isolated resistance arteries were maintained in a myograph and tension recorded, in some instances combined with microelectrode impalement for membrane potential measurements or intracellular calcium monitoring using fura-2. Voltage-dependent c

Continuous muscle fibre activity: a case treated with acetazolamide.

A case is reported of the continuous muscle fibre activity syndrome, which includes a group of disorders characterised by sustained motor unit activity due to hyperactivity of peripheral nerve motor axons. In this patient the muscle stiffness and myokymic movements were successfully treated with acetazolamide, which acts as a membrane stabiliser either by blockade of chloride and bicarbonate membrane transport or by producing kaliuresis and raising the transmembrane potential by decreasing extracellular potassium.

[Gordon's syndrome--a combination of hyperkalemia and arterial hypertension with preserved kidney function].

A case of Gordon's syndrome in a 38 years old man is presented--hyperpotassemia and arterial hypertension with preserved renal function, normal supra-adrenal function, no data for hemolysis, normal coagulation state, mild compensated metabolic acidosis. Renal tubular defect, for excretion of potassium and chlorides, sodium reabsorption, tubular resistance to mineralocorticoids are discussed. The combination of hyperpotassemia and arterial hypertension might be due to phenotype manifestations of a single gene.
